Inhibition of Quorum Sensing Regulated Virulence Factors and Biofilm Formation by Eucalyptus globulus against Multidrug-Resistant Pseudomonas aeruginosa

  • Objectives: The quorum-sensing-inhibitory and anti-biofilm activities of the methanol extract of E. globulus leaves were determined against clinically isolated multidrug-resistant Pseudomonas aeruginosa. Methods: The preliminary anti-quorum-sensing (AQS) activity of eucalyptus was investigated against a biosensor strain Chromobacterium violaceum ATCC 12472 (CV12472) by using the agar well diffusion method. The effect of sub-minimum inhibitory concentrations (sub-MICs) of the methanol extract of eucalyptus on different quorum-sensing-regulated virulence factors, such as swarming motility, pyocyanin pigment, exopolysaccharide (EPS), and biofilm formation, against clinical isolates (CIs 2, 3, and 4) and reference PA01 of Pseudomonas aeruginosa were determined using the swarm diameter (mm)-measurement method, chloroform extraction method, phenol (5%)-sulphuric acid (concentrated) method, and the microtiter plate assay respectively, and the inhibition (%) in formation were calculated. Results: The preliminary AQS activity (violacein pigment inhibition) of eucalyptus was confirmed against Chromobacterium violaceum ATCC 12472 (CV12472). The eucalyptus extract also showed concentration-dependent inhibition (%) of swarming motility, pyocyanin pigment, EPS, and biofilm formation in different CIs and PA01 of P. aeruginosa. Conclusion: Our results revealed the effectiveness of the E. globulus extract for the regulation of quorum-sensing-dependent virulence factors and biofilm formation at a reduced dose (sub-MICs) and suggest that E. globulus may be a therapeutic agent for curing and controlling bacterial infection and thereby reducing the possibility of resistance development in pathogenic strains.

  • Coix lacryma-jobi (Hanjeli) is known to posses anti-microbial properties. Therefore, phytochemical compounds of C. lacryma-jobi have been studied to produce novel antimicrobial agents as treatments against antibiotic-resistant bacteria.The objective of this study was to determine the phytochemical composition and antibacterial activity of the C. lacryma-jobi oil against Escherichia coli, Staphylococcus aureus, and Bacillus subtilis. The phytochemical composition of the oil was determined via gas chromatography mass spectrophotometry (GC-MS). Moreover, agar disk and agar well diffusion were employed to screen the antibacterial activity of the oil. An agar well diffusion test was implemented to determinate MIC's (minimum inhibitory concentrations). Dodecanoic acid, tetradecanoic acid, 2,3-dihydroxypropylester, 1,3-dioctanoin, N-methoxy-N-methyl-3,4-dihydro-2H-thiopyran6-carboxamide, propanamide, 5-Amino-1-(quinolin-8-yl)-1,2,3-triazole-4-carboxamide, and pyridine were identified in the C. lacryma-jobi oil. The MIC value of the oil was 0.031 g/L and the MBC of the oil was 0.125 g/L effective in all test bacteria. Dodecanoic acid displayed inhibitory activity against gram-positive and gram-negative bacteria. Therefore, our research demonstrated C. lacryma-jobi (Hanjeli) oil exhibited antibacterial activity against E. coli, S. aureus, and B. subtilis. These research suggest that C. lacryma-jobi root oil could be used for medicinal purposes; however clinical and in vivo tests must be performed to evaluate its potential as an antibacterial agent.

The Effects of Temperature, pH, and Bactericides on the Growth of Erwinia pyrifoliae and Erwinia amylovora

  • In this comparative study, the effects of temperature, pH, and bactericides on the growth of Erwinia pyrifoliae and Erwinia amylovora were investigated. The maximum temperature for the growth of both Erwinia species was estimated to be $36{\circ}C$. The maximum specific growth rates of E. pyrifoliae and E. amylovora were observed at $27{\circ}C$ and $28{\circ}C$, respectively, and no significant growth differences were shown at their optimum temperatures. However, at lower temperatures ranging from 12-$21{\circ}C$, E. pyrifoliae showed higher growth rates with doubling times shorter than those of E. amylovora. Distinct growth rates at these temperatures revealed that E. pyrifoliae is more cold-tolerant than E. amylovora. The optimum pH for the growth of both pathogens was 7.5 and growth was not seen at pH ${\le}$ 5.0 and ${\ge}$ 10.0. These results showed that the effect of pH on the growth of E. pyrifoliae and E. amylovora was similar. Minimum inhibitory concentrations (MICs) of copper sulfate, oxolinic acid, streptomycin, and tetracycline, which inhibited growth of E. pyrifoliae and E. amylovora, were determined. The strains of both pathogens were able to grow at 0.08-0.32 mM copper sulfate, but not at higher concentrations. However, none of the tested strains grew in the presence of oxolinic acid (0.001 mM), streptomycin (0.1 mM), and tetracycline (0.01 mM) concentrations. These results suggested that all strains of both Erwinia species were sensitive to tested bactericides and indicated no occurrence of resistant strains of E. pyrifoliae in Korea.

치아우식증유발세균에 대한 단삼 메탄올추출물과 항생제와의 병용효과 (Synergistic Effect of Methanol Extract of Salvia Miltiorrhiza and Antibiotics against Dental Caries Pathogens)

  • 단삼(Salvia miltiorrhiza Bunge)은 우리나라에서 전통적으로 부종, 관절염, 간염 등과 같은 염증질환의 치료에 사용해왔다. 본 연구에서는 단삼 메탄올 추출물을 이용하여 구강미생물에 대한 항균활성을 확인하였다. 그 결과 메탄올추출물은 실험 되어진 모든 구강미생물에서 강한 항균활성을 나타내었다(MICs, 8 to $64\;{\mu}g/mL$; MBCs, 16 to $64\;{\mu}g/mL$). 추가적으로 ampicillin이나 gentamicin과의 병용투여서 최소억제농도(MIC)와 최소살균농도(MBC)가 감소하는 병용효과를 나타내었다. 더불어 메탄올추출물 단독 사용시 보다 항생제와 병용투여시 빠른 사멸효과를 보였다. 결론적으로 단삼 메탄올추출물이 치아우식을 유발하는 많은 세균들에 대한 항균효과가 뛰어남을 확인 할 수 있었으며, 안정성이 높은 천연 치아우식예방제로서의 개발가능성을 확인하였다.

Augmentin(Amoxycillin/clavulanic acid)의 시험관내 항균효과 (Antibacterial Activity of Amoxycillin/Clavulanic Acid(Augmentin) in Vitro)

  • Strains of bacteria resistant to beta-lactam antibiotics have been increasing in number and are becoming troublesome in clinical medicine. The in vitro antibacterial activity of augmentin, a combination drug consisting of two parts amoxycillin to one part clavulanic acid, a potent beta-lactamase inhibitor, and their minimum inhibitory concentrations were determined by an agar dilution technique against ampicillin-resistant clinical isolates in Korea. Of the 226 strains tested, 140 strains(62%) were resistant to ampicillin. Among the 140 ampicillin-resistant strains, all Salmonella spp. Proteus spp. the majority of S. aureus and Shigella spp. were sensitive to augmentin. Ps. aeruginosa remained 100% resistant and there has been a considerable decline in resistant strains in E. coli and K. pneumoniae although a significant percentage of strains showed intermediate sensitivity. The minimum inhibitory concentrations of augmentin were ranged in $8{\mu}g/ml$ to $32{\mu}g/ml$ in most bacteria and all S. aureus were inhibited by $8{\mu}g/ml$. In our microbiological studies we have shown that augmentin is active against ampicillin-resistant strains of Staphylococci and Gram-negative bacteria. In this hospital there would appear to be a significant number of strains of E. coli and K. pneumoniae showing intermediate resistance to augmentin. Most of these strains should be susceptible to augmentin given by mouth or by the intravenous route depending on the concentrations of both amoxycillin and clavulanic acid obtainable in the various tissues.

Antibacterial Effect of Allium ampeloprasum and Allium porrum Extracts on Staphylococcus aureus and Pseudomonas aeruginosa

  • Objectives: The most prominent microorganisms that cause hospital infections and acquire antibiotic resistance are Staphylococcus aureus and Pseudomonas aeruginosa. The present study aimed to compare the phenolic and flavonoid compounds of various Allium ampeloprasum and Allium porrum extracts and evaluate the antibacterial effects of these extracts against these two microorganisms. Methods: The total phenolic and flavonoid contents of the acetone, methanol, aqueous, and hexane leeks extracts from A. ampeloprasum and A. porrum were measured. The antibacterial activity of these extracts against S. aureus and P. aeruginosa was tested using the disk diffusion method for 24, 48, and 72 hours. Further, the minimum inhibitory concentrations and the minimum bactericidal concentrations of these extracts for these two bacteria were evaluated and compared with those of common antibiotics. Results: The aqueous extracts showed the highest phenolic and flavonoid contents and at concentrations of 35 and 40 mg per disk, showed the most antibacterial activity against S. aureus and P. aeruginosa; P. aeruginosa showed more sensitivity to the aqueous extracts than S. aureus. Conclusion: Aqueous A. ampeloprasum and A. porrum extracts may prevent the growth of hospital pathogens, especially P. aeruginosa; our findings will aid the discovery of new antimicrobial substances against antibiotic-resistant bacteria.

Delamanid, Bedaquiline, and Linezolid Minimum Inhibitory Concentration Distributions and Resistance-related Gene Mutations in Multidrug-resistant and Extensively Drug-resistant Tuberculosis in Korea

  • Background: Delamanid, bedaquiline, and linezolid have recently been approved for the treatment of multidrug- and extensively drug-resistant (MDR and XDR, respectively) tuberculosis (TB). To use these drugs effectively, drug susceptibility tests, including rapid molecular techniques, are required for accurate diagnosis and treatment. Furthermore, mutation analyses are needed to assess the potential for resistance. We evaluated the minimum inhibitory concentrations (MICs) of these three anti-TB drugs for Korean MDR and XDR clinical strains and mutations in genes related to resistance to these drugs. Methods: MICs were determined for delamanid, bedaquiline, and linezolid using a microdilution method. The PCR products of drug resistance-related genes from 420 clinical Mycobacterium tuberculosis strains were sequenced and aligned to those of M. tuberculosis H37Rv. Results: The overall MICs for delamanid, bedaquiline, and linezolid ranged from ${\leq}0.025$ to >1.6 mg/L, ${\leq}0.0312$ to >4 mg/L, and ${\leq}0.125$ to 1 mg/L, respectively. Numerous mutations were found in drug-susceptible and -resistant strains. We did not detect specific mutations associated with resistance to bedaquiline and linezolid. However, the Gly81Ser and Gly81Asp mutations were associated with resistance to delamanid. Conclusions: We determined the MICs of three anti-TB drugs for Korean MDR and XDR strains and identified various mutations in resistance-related genes. Further studies are needed to determine the genetic mechanisms underlying resistance to these drugs.

맥문동 에탄올 추출물(Liriope platyphylla ethanol extract)의 Streptococcus mutnas와 Porphyromonas gingivalis에 대한 항균력과 성장억제 효과 (Antibacterial and Growth Inhibitory Effects of Liriope Platyphylla Ethanol Extract on Streptococcus Mutnas and Porphyromonas Gingivalis)

  • Purpose : This study aimed to analyze the antibacterial activity of Liriope platyphylla ethanol extract (LPEE) against Streptococcus mutans and Porphyromonas gingivalis and to validate its potential for the prevention and treatment of dental caries, gingivitis, and periodontal disease. Methods : To verify the antibacterial effect of L. pulsatilla ethanolic extract (LPEE) against S. mutans and P. gingivalis, the disk diffusion method was used to determine the inhibition zones at concentrations of 50, 100, 200, and 300 mg/㎖. To determine the minimum inhibition concentration (MIC), the final dose of LPEE was .2, .4, .8, 1.6, 2.5, and 5.0 mg/㎖, and the minimum bactericidal concentration (MBC) was determined based on the MIC results. To confirm the growth inhibitory effect of LPEE on both pathogens, the absorbance was measured at 600 nm after each incubation for 0, 3, 6, 12, and 24 hr at concentrations of .8, 1.6, 2.5, and 5.0 mg/㎖. Results : The cytotoxicity of LPEE was evaluated and the cell viability was more than 70 % at 400 mg/㎖. Therefore, concentrations of 50, 100, 200, and 300 mg/㎖ were used in this study. The antimicrobial effect against S. mutans was seen at 100 mg/㎖ and grew in a concentration-dependent manner, while P. gingivalis was effective at 50 mg/㎖ with the dose dependency. The MIC was .8 mg/㎖ for both strains, and the MBC was 1.6 mg/㎖ with the same results. The growth inhibitory effect of LPEE on S. mutans and P. gingivalis was observed, even at low concentrations. Conclusion : The antibacterial effect of LPEE was evaluated through the analysis of MIC, MBC, and growth inhibition effect on S. mutans and P. gingivalis, which suggests LPEE might have the possibility of utilization as a preventive and therapeutic composition for oral diseases.

부유 상태와 바이오필름 상태에서 Mutans streptococci의 Erythrosine에 대한 감수성 평가 (Susceptibility of Mutans streptococci in the Planktonic and Biofilm State to Erythrosine)

  • 이 연구의 목적은 부유 및 바이오필름 상태의 Mutans streptococci에서 치태와 치석 착색제로 치과 진료실에서 사용하는 식용색소인 에리스로신에 대한 감수성을 조사하는 것이다. 세균은 호기성 조건에 배양하였고, 에리스로신은 $0.02-10000{\mu}g/L$의 농도로 희석하였다. 부유 상태 S. mutans의 최소 억제 농도(MIC)와 최소 살균 농도(MBC)를 측정하였다. 세균을 배양하여 바이오필름을 형성한 후, 최소 바이오필름 억제 농도(MBIC) 및 최소 바이오필름 박멸 농도(MBEC)를 측정하였다. MIC와 MBC는 $19.5{\mu}g/L$로 동일하게 측정되었으며 따라서 에리스로신은 부유 상태의 S. mutans에 대해 살균 효과를 가지는 것으로 나타났다. MBIC, MBEC 값은 각각 $313{\mu}g/L$, $2500{\mu}g/L$로 측정되었다. 측정된 MIC, MBC, MBIC는 실제로 치과에서 사용되는 농도에 비해 낮은 값이며 따라서 인체에 무해한 농도에서 S. mutans를 억제할 수 있을 것으로 보인다.

Isoflavonoid의 피부염증균에 대한 항균 및 항산화효과 (Antimicrobial and Antioxidative Effects of Isoflavonoids on Skin Microbial Inflammation Pathogens)

  • Puerarin and daidzin have been isolated from Puerariae thunbergiana Benth. Structures were determined by spectroscopic methods. Compounds showed weak antimicrobial activity against S. mutans, S. epidermidis, S. aureus, and C. albicans (MIC, $800{\mu}g/ml$). However, these compounds were not antioxidative. Puerariae thunbergiana Benth. extracts against microorganisms were evaluated in terms of the minimum inhibitory concentrations (MIC). In general, C. albicans was stronger antimicrobial activity than the other microorganisms. The antioxidative activity of was observed in the etyl acetate extract ($IC_{50},\;119.87{\pm}0.16\;{\mu}g/ml$). The DPPH radical scavenging effect ($IC_{50},\;1,673.3{\pm}0.54\;{\mu}M$) of the puerarin was comparable with that of synthetic antioxidant, BHA $(IC_{50},\;88.39{\pm}1.1){\times}10^{3}{\mu}M$. These results suggest that puerarin and daidzin have a potential antimicrobial activity.
