수용성 프로폴리스의 항균성 (Antimicrobial Activity of Water Soluble Propolis)

  • In this study, the minimum inhibition concentration(MIC), growth inhibition activity, and colony forming inhibitory activity of water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae and Salmonella enteritidis were tested. The MICs of the water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 312.5 ppm, below 156.3 ppm, 625 ppm, 10,000 ppm, above 10,000 ppm, 10,000 ppm, above 10,000 ppm, above 10,000 ppm, 10,000 ppm, and above 10,000 ppm, respectively. The growth inhibition concentrations against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, and Klebsiella pneumoniae were 156.3 ppm, below 156.3 ppm, 625 ppm, 5,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, and 5,000 ppm, respectively. However, 10,000 ppm did not inhibit the growth of Salmonella enteritidis. Finally, the colony forming inhibitory activities against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 98.0%, 99.8%, 69.8%, 98.1%, 62.0%, 63.1%, 79.5%, 61.9%, 79.6%, and 0.0%, respectively.

Antibacterial Effects of Extracts of Thuja Orientalis cv Aurea Nana Cones against Food-spoilage and Food-borne Pathogens

  • BACKGROUND: Nowadays, Chemical antiseptics have become great problems for health and environmental, so that developing of new substitutes for chemical antiseptics is more and more important. Natural product is a kind of environment-friendly additive that could be used as antiseptic in food industry. Thuja orientalis cv Aurea Nana is a gymnospermous plant of the family Cupressaceae, native to northwestern China and widely naturalised elsewhere in Korea and Japan. This study was aimed to investigate the antibacterial potential of various organic extracts from T. orientalis cones against some food-borne and food-spoilage bacteria. METHODS AND RESULTS: Hexane extract (HE), chloroform extract (CE), ethyl acetate extract (EAE) and methanol extract (ME) were obtained from female cones of T. orientalis. The antibacterial activities of various extracts were tested by standard agar diffusion and minimum inhibitory concentrations (MICs) against five gram-positive and six gram-negative bacteria. Cell viability and morphology change of L. monocytogenes ATCC 10943 treated with hexane extract were also observed. The various extracts displayed remarkable antibacterial effects against all the gram-positive bacteria but did not show any effect against the gram-negative bacteria. Hexane extract has the highest inhibitory effect on cell viability of L. monocytogenes ATCC 10943. SEM observation also demonstrated the damaging effect of the hexane extract on the morphology of L. monocytogenes ATCC 10943 at the minimum inhibitory concentration. CONCLUSION(s): The tested gram-positive bacteria were significantly inhibited by organic extracts of T. orientalis cone. Hexane extract was the most potent against Listeria monocytogenes ATCC 10943, as evidenced by the lowest MIC level and the complete inhibition of cell viability within shortest exposure time, along with SEM observation.

황금작약탕(黃芩芍藥湯)의 MRSA에 대한 항균활성에 관한 연구 (Antibacterial Effect of Huanggeumjakyak-tang against Methicillin-resistant Staphylococcus aureus)

  • Objectives The aim of this study was to investigate antimicrobial activity of Huanggeumjakyak- tang water extract against MRSA. Methods The antibacterial activities of Huanggeumjakyak-tang were evaluated against 3 strains of Methicillin-resistant Staphylococcus aureus (MRSA) and 1 standard Methicillinsusceptible S. aureus (MSSA) strain by using the disc diffusion method, minimal inhibitory concentrations (MICs) assay, colorimetric assay using MTT test, checkerboard dilution test and time-kill assay was performed under dark. Results The MIC (minimum inhibitory concentration) of Huanggeumjakyak-tang water extract against S. aureus strains ranged from 1,000 to $2,000{\mu}g/ml$. So we confirmed that it has a strong antibacterial effect. Also the combinations of Huanggeumjakyak-tang water extract and conventional antibiotics exhibited improved inhibition of MRSA with synergy effect. Conclusions The results obtained in this study suggest that Huanggeumjakyak-tang water extract showed antibacterial effect against MRSA, and it also showed reducing effect on the side-effect problems that are the major weak points of traditional antibiotics.

매실 추출물을 함유한 음료가 식중독 유발균의 성장에 미치는 영향 (Effect of Prunus mume Extract Containing Beverages on the Proliferation of Food-borne Pathogens)

  • Prunus mume has been used for the folk medicine by many old civilizations to treat food-borne diseases or enteric disorders. The purpose of this study was to investigate the antimicrobial activity of beverages containing Prunus mume extracts against Staphylococcus aureus, Shigella dysenteriae, Shigella flexneri, Shigella sonnei, Escherichia coli and Pseudomonas aeruginosa. Seven different types of Prunus mume extracts containing beverages have been prepared for the test in which minimum inhibitory concentration for each microorganism has been determined by serial dilution method using either TSA or TSB medium. Pseudomonas aeruginosa showed least resistance and 0.3g/$m\ell$ concentrations of 5% Prunus mume extracts containing beverage had antimicrobial property against the organism. Beverages containing more than 15% of Prunus mume extracts showed antimicrobial activity against all tested microorganisms at the MIC value of less than 0.25g/$m\ell$. The effect of Prunus mume on the growth of food-borne pathogens has been also studied using a spectrophotometer. In the growth assay, each of the Prunus mume extracts containing beverage was added to the medium at the concentration of 25% (v/v). Beverage containing 20% Prunus mume extracts showed inhibitory effect on the growth of all tested microorganisms.

Antifungal and synergistic effects of an ethyl acetate extract of the edible brown seaweed Eisenia bicyclis against Candida species

  • With the continuing demand for new solutions in the development of effective and safe candidiasis therapies, we investigated the efficacy of an antifungal agent from the marine brown alga Eisenia bicyclis. The methanolic extract of E. bicyclis evinced potential antifungal activity against Candida species. The ethyl acetate (EtOAc)-soluble extract from E. bicyclis demonstrated the strongest antifungal activity against Candida species among five solvent-soluble extracts. Indeed, the EtOAc-soluble extract showed minimum inhibitory concentrations (MICs) ranging from 4 to 8 mg/mL. Furthermore, the EtOAc-soluble extract considerably reversed high-level fluconazole resistance of Candida species. The MIC values of fluconazole against Candida species decreased substantially (from 64 to $4{\mu}g/mL$) in combination with the MIC of the EtOAc-soluble extract (4 mg/mL). The fractional inhibitory concentration indices of fluconazole ranged from 0.531 to 0.625 in combination with 4, 2, or 1 mg/mL of the EtOAc-soluble extract against Candida isolates, indicating that these combinations exert a marked synergistic effect against Candida isolates. These findings imply that compounds derived from E. bicyclis can be a potential source of natural antifungal agents against Candida species.

Antimicrobial Activity of Berberine against Oral Bacteria Related to Endodontic Infections

  • It has been established that berberine has strong antimicrobial effects. Little is known however regarding the antimicrobial activity of berberine against endodontic pathogenic bacteria or its cytotoxicity in human oral tissue cells. The antibacterial properties of berberine were tested against 5 strains of Enterococcus faecalis and type strains of Aggregatibacter actinomycetemcomitans, Prevotella nigrescens, Prevotella intermedia, and Tannerella forsythia, which are involved in endodontic infections. Antimicrobial activity was evaluated through minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) measurements. The viability of normal human gingival fibroblast (NHGF) cells after exposure to berberine was measured using a methyl thiazolyl tetrazolium (MTT) assay. The data showed that berberine has antimicrobial effects against A. actinomycetemcomitans with an MIC and MBC of $12.5{\mu}g/ml$ and $25{\mu}g/ml$, respectively. In the cytotoxicity studies, cell viability was maintained at 66.1% following exposure to $31.3{\mu}g/ml$ berberine. Overall, these findings suggest that berberine has antimicrobial activity against the tested bacteria. Nevertheless, lower concentrations in combination with other reagents will need to be tested before these in vitro results can be translated to clinical use.

Anticaries Effect of Ethanol Extract of Terminalia chebula

  • Background: Dental caries is mainly composed of various cellular components and is deposited around the tooth surface and gums, causing a number of periodontal diseases. Streptococcus mutans is commonly found in the human oral cavity and is a significant contributor to tooth decay. The use of antibacterial ingredients in oral hygiene products has demonstrated usefulness in the management of dental caries. This study investigated the anticaries effect of the ethanol extract of Terminalia chebula (EETC) against S. mutans and their cytotoxicity to gingival epithelial cells. Methods: The EETC was prepared from T. chebula fruit using ethanol extraction. Disk diffusion, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and colony forming unit (CFU) were analyzed to investigate the antimicrobial activity of the EETC. Glucan formation was measured using the filtrate of the bacterial culture medium and sucrose. Gene expression was analyzed using reverse transcription-polymerase chain reaction (RT-PCR). Cytotoxicity was analyzed via the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay. Results: The antibacterial activity of the EETC was explored using disc diffusion and CFU measurements. The MIC and MBC of the EETC were 10 and 20 ㎍/ml, respectively. EETC treatment decreased insoluble glucan formation by S. mutans enzymes and also resulted in reduced glycosyltransferase B (gtf B), gtf C, gtf D, and fructosyltransferase (ftf), expressions on RT-PCR. In addition, at effective antibacterial concentrations, EETC treatment was not cytotoxic to gingival epithelial cells. Conclusion: These results demonstrate that the EETC is an effective anticaries ingredient with low cytotoxicity to gingival epithelial cells. The EETC may be useful in antibacterial oral hygiene products for the management of dental caries.

Dichlofluanid 저항성 및 감수성 잿빛곰팡이병균(Botryits cinerea)의 약제 반응과 생태 적응력과 관련된 특성 (Chemical Responses and Fitness-Related Characteristics of Dichlofluanid-Resistant and -Sensitive Botrytis cinerea Isolates)

  • Nine hundred and ninety-two isolates of Botrytis cinerea were obtained from infected strawberries, tomatoes and cucumbers in Taejon, Gongju, Puyo, Nonsan and Kimhae in Korea. Six hundred forty-two (64.7%) isolates were benomyl resistant (BR), 245 (24.7%) were procymidone resistant (PR), and 105 (10.6%) were dichlofluanid resistant (DR). In the minimum inhibitory concentration (MIC) test, DR isolates showed mycelial growth on the PDA incorporated with 100 or 500 $\mu\textrm{g}$/ml of dichlofluanid, while dichlofluanid sensitive (DS) isolates did not grow on the PDA incorporated even with 10 $\mu\textrm{g}$/ml of dichlofluanid. Chemical concentrations for inhibition of spore germination were much lower than those for inhibition of mycelial growth. IC50 values, the effective concentrations for 50% inhibition of spore germination, for DR were 0.11~0.29 $\mu\textrm{g}$/ml, whereas they were 0.04~0.09 $\mu\textrm{g}$/ml for DS isolates. In comparison of fitness-related characteristics such as virulence, sclerotial formation, and sporulation, DR isolates were inferior to DS isolates. However, mycelial growth was little different between DR isolates and DS isolates.

밤 부위별 추출물의 항균활성 (Antimicrobial Activity of the Solvent Extracts from of Chestnut)

  • 새로운 식품개발 및 천연 보존료 개발의 일환으로 물, 에탄올, 에칠아세테이트로 추출한 항균활성 물질을 몇 종의 병원균과 식중독균 식품과 관련이 있는 세균, 젖산균 및 효모 등 10개 균주에 대하여 항균활성 검색을 하였으며, 항균력이 강한 밤잎과 수피 에탄올 추출물을 용매 계통분획하여 각 분획별 항균활성을 조사하였다. 또한 에탄올 추출물의 최소저해농도, 추출물에 함유된 항균성물질의 열안정성 pH 등을 조사하였다. 밤 잎과 수피의 항균활성은 에탄올 추출물과 물 추출물이 10균주 중 세균에 대하여 항균활성이 강하였으나, 젖산균 및 효모에 대해서는 항균활성이 나타나지 않았다. 밤잎과 수피를 핵산, 디에칠에테르, 에칠아세테이트 및 물로 용매계통분획하여 얻은 각 분획물의 항균활성은 에칠아세테이트와 물층에서 강한 항균활성을 보였고 균주별로 일반세균은 항균활성이 나타났으며, 효모와 젖산균에서는 항균활성이 나타나지 않았다. 최소저해농도는 밤나무 잎 0.5 mg/mL 이었고, 수피 추출물에서는 1 mg/mL이었다. 밤 잎과 수피의 에탄올 추출물에 함유되어 있는 항균활성 물질은 $121^{\circ}C$에서 15분간 가열한 후에도 그 활성이 유지된 것으로 보아 열에 안정하였으며, pH의 변화에도 항균활성의 변화가 거의 없는 것으로 나타났다.

블렌딩 에센션오일의 항균효과 증진 (Improved Antibacterial Effect of Blending Essential Oils)

  • 식물로부터 유래되는 휘발성 유기 화합물인 에센셜 오일은 인체에 약리효과를 나타낸다고 알려져 있다. 본 연구에서는 화농성 질환과 병독성 질환 비뇨기계통 감염에 나올 수 있는 병원성 세균 Staphylococcus aureus (그람양성알균), Escherichia coli (그람음성막대균), Pseudomonas aeruginosa (그람음성막대균, 극성단모균)을 대상으로 천연 허브 오일 중에 비교적 많이 알려져 있는 라벤더(lavender), 티트리(tea tree), 로즈마리(rosemary), 멜리사(melisa) 4종류의 단일 오일과 비교해, 티트리와 로즈마리, 멜리사를 블렌딩하였을 때 항균력이 상승되는 효과가 있는지 연구하였다. 그 결과 디스크 확산법에서 블렌딩 오일(티트리, 로즈마리, 멜리사)의 경우 단독의 허브오일보다 S. aureus에서18 mm, E. coli 에서 22 mm 와 P. aeruginosa는13 mm의 우수한 항균능력을 확인할 수 있었다. 뿐만 아니라 블렌딩 오일의 경우 최소억제농도(Minimum Inhibitory Concentrations, MIC) 분석에서 S. aureus, E. coli 에서는 0.3125%에서, P. aeruginosa는 1.25%에서 성장억제효과가 확인되었다. 따라서 본 실험에서는 블렌딩 오일이 Oxacillin과 로즈마리, 티트리보다 항균효과가 우수한 것으로 증명되어 단독의 허브오일보다 블렌딩 오일이 우수한 항균력을 나타나는 것을 확인할 수 있었다. 그러므로 허브의 블렌딩을 활용하면 다제 내성균에 대한 항균시너지효과를 발휘할 수 있음을 입증했을 뿐만 아니라 향후 천연보존제와 건강기능성 식품 및 화장품에 활용 가능성을 기대해본다.
