The buffalo (Bubalus bubalis) is an important contributor to milk, meat, power, fuel and leather production in many developing countries. Buffaloes can be categorized into Asian and Mediterranean buffaloes. Asian buffalo includes two subspecies known as Riverine and Swamp types. Riverine (water buffalo) and Swamp buffaloes possess different genetics (50 vs. 48 chromosomes, respectively), morphology (body frame, body weight, horn shape and skin color) and behavior (wallowing in mud or water) and thus, are reared and used for different purposes. Low per head milk yield, poor reproductive performance (seasonal breeding behavior, anestrous, and longer calving interval) and low growth rate in buffaloes have been attributed to insufficient supply of nutrients. In many parts of Asia, where the buffalo is an integral part of the food chain and rural economy, irregular and inadequate availability of quality feedstuffs and their utilization are hampering the performance of this unique animal. Balanced nutrition and better management can enhance buffalo productivity. Many efforts have been made in the last few decades to improve nutrient supply and utilization in buffaloes. Recent research on locally available feed resources such as crop residues, and industrial by-products, dietary addition of micronutrients, use of performance modifiers and use of ruminally protected fat and protein sources have shown significant potential to improve growth, milk yield and reproductive performance of buffaloes. However, a number of issues, including establishment of nutrient requirements for dairy and beef, development of buffalo calf feeding systems, nutritional management of metabolic and reproductive anomalies, and understanding and exploitation of the buffalo gut ecosystem, need to be addressed. Extensive coordinated research and extension efforts are required for improved buffalo nutrition in developing countries.
We investigated the effect of reproductive herd health program (RHHP) on the reproductive performance in high yielding dairy herds. Data collected from 205 lactations at Boeun County, Chungbuk from April 2001 to Dec. 2003 included postpartum reproductive and metabolic diseases, body condition score (BCS), reproductive performance and milking records. First we compared the reproductive performance of cows without RHHP (pre-RHHP group) and cows with RHHP (RHHP group). Intervals from calving to first service (99.6 vs. 81.8 days) and to conception (160.3 vs. 131.8 days) were shorter in the RHHP group than in the pre-RHHP group. Secondly, we determined the effect of milk yield (305 days) on the occurrence of postpartum diseases, BCS changes, and reproductive performance in cows under RHHP. The occurrence of metritis and metabolic diseases were greater in the over-10,000 kg group than in the under-10,000 kg group. The over-10,000 kg group lost more body condition than the under-10,000 kg group from calving to month 3 postpartum. Nevertheless, reproductive performance did not differ between the two groups. These results indicate that reproductive performance can be improved by the application of RHHP in high yielding dairy herds and that reproductive performance in the cows under RHHP are not affected by high milk yielding.
Productivity of bacterial cellulose by Acetobacter xylinum GS11 was investigated in the several culture conditions. In various carbon sources, others with the exception of glucose were not found to be effective for cellulose production, and 2% was better in yield than other concentration of glucose. Yeast extract and soytone among several organic nitrogens were effective, but inorganic nitrogen sources tested were not efficient for cellulose production by A. xylinum GS11. The effects of various inorganic salts, amino acids and vitamins were also investigated: $MgSO_4$, phenylalanine and $\alpha$-tocopherol gave the cellulose yield of 1.5, 1.4 and 1.4 fold, respectively, compared with basal medium. In our experiment, cellulose production by A. xylinum GS11 added with 10% coconut milk and 0.5% lignosulfonate in basal medium, was the most efficient among the several material sources employed here, and these were 2.2 and 2.1 fold, respectively.
Intensive genetic selection has resulted in modern dairy cow with very high milk yields but reduced fertility, due mainly to an increase in reproductive disorders. The study was conducted to estimate the proportion of reproductive disorders among dairy cattle. The factors analyzed were; milk yield, cow parity, periparturient disorders, and reproductive status. The result of this study showed the incidence of reproductive disorders in high yielding dairy cows was prevalent. Repeat breeding was the major postpartum reproductive problem comprising (42.3%) of the samples. Higher reproductive disorders were also associated with lower parity. These disorders subsequently reduced reproductive performance by prolonged intervals from calving to first artificial insemination and from calving to pregnancy. Abnormalities in the reproductive system such as repeat breeder, silent heat, anestrous and ovarian cysts, as well as lower fertility have been recorded during the summer months (June to August). In conclusion, the study revealed that incidence of repeat breeder is mostly prevalent in dairy cows. Moreover, the incidence of reproductive disorders was higher in summer season and hence appropriate preventive and or therapeutic measures as per the type of abnormalities need to be undertaken.

This study aimed to extend the retention of flavor in coffee-containing milk beverage by microencapsulation. The core material was caramel flavor, and the primary and secondary coating materials were medium-chain triglyceride and maltodextrin, respectively. Polyglycerol polyricinoleate was used as the primary emulsifier, and the secondary emulsifier was polyoxyethylene sorbitan monolaurate. Response surface methodology was employed to determine optimum microencapsulation conditions, and headspace solid-phase microextraction was used to detect the caramel flavor during storage. The microencapsulation yield of the caramel flavor increased as the ratio of primary to secondary coating material increased. The optimum ratio of core to primary coating material for the water-in-oil (W/O) phase was 1:9, and that of the W/O phase to the secondary coating material was also 1:9. Microencapsulation yield was observed to be approximately 93.43%. In case of in vitro release behavior, the release rate of the capsules in the simulated gastric environment was feeble; however, the release rate in the simulated intestinal environment rapidly increased within 30 min, and nearly 70% of the core material was released within 120 min. The caramel flavor-supplemented beverage sample exhibited an exponential degradation in its flavor components. However, microcapsules containing flavor samples showed sustained flavor release compared to caramel flavor-filled samples under higher storage temperatures. In conclusion, the addition of coffee flavor microcapsules to coffee-containing milk beverages effectively extended the retention of the coffee flavor during the storage period.

Objective: The aim of this research was to explore the effects of dietary substitution of alfalfa hay by amaranth hay on production performance, rumen fermentation, nutrient digestibility, serum biochemical parameters and antioxidant ability in dairy cows. Methods: A total of 45 healthy Holstein cows with same parity and similar milk yield and body weight were randomly divided into 3 groups: control diet without amaranth hay (CON) or 50% and 100% alfalfa hay replaced by an equal amount of amaranth hay (dry matter basis, AH1 and AH2, respectively). All the cows were fed regularly 3 times a day at 06:30, 14:30, and 22:30 and had free access to water. The experiment lasted for 60 d. Results: The dry matter intake of CON and AH1 groups was higher (p<0.05) than that of AH2 group. Compared with AH1 group, the milk yield of AH2 group was reduced (p<0.05). Moreover, dietary substitution of alfalfa hay by amaranth hay increased (p<0.05) milk fat, ammonia nitrogen and acetate concentrations. However, the crude protein digestibility of AH2 group was lower (p<0.05) than that of CON group, while an opposite tendency of serum urea nitrogen was found between two groups. The neutral detergent fiber digestibility of AH1 group was increased (p<0.05) when compared to AH2 group. Amaranth hay treatment increased (p<0.05) the serum concentration of glutathione peroxidase in dairy cows. Compared with CON group, the malonaldehyde activity of AH1 group was decreased (p<0.05). Conclusion: Dietary replacing alfalfa hay with amaranth hay (50% ratio) in dairy cows did not affect production performance but improved their antioxidant ability.
A feeding experiment with 40 lactating Holstein cows and 4 dietary treatments was conducted to investigate supplementation with different levels of alcohol fermented feed to the TMR on lactating performance, blood metabolites, milk fatty acid profile and cholesterol concentration of blood and milk. Forty Holstein lactating cows ($106{\pm}24$ d post-partum; mean${\pm}$SD) were distributed into four groups and randomly assigned to one of four treatments with each containing 10 cows per treatment. The treatment supplemented with TMR (DM basis) as the control (CON), and CON mixed with alcohol-fermented feeds (AFF) at a level of 5%, 10% and 15% of the TMR as T1, T2 and T3, respectively. Dry matter intake and milk yield were not affected by supplementation of AFF. An increased 4% FCM in the milk occurred in cows fed T3 diet compared with CON, while T1 and T2 diets decreased 4% FCM in a dose dependent manner. Supplementation of AFF increased the concentration of albumin, total protein (TP), ammonia, and high density lipoprotein-cholesterol in serum compared with CON. In contrast, supplementation with AFF clearly decreased concentration of blood urea nitrogen (BUN) and total cholesterol (TC) compare with CON. AFF supplementation increased the proportion of C18:1n9 and C18:2n6 compared to CON. A decrease in the concentration of saturated fatty acid (SFA) for T1, T2 and T3 resulted in an increased unsaturated fatty acid (USFA) to SFA ratio compared to CON. Concentration of cholesterol in milk fat was reduced in proportion to the supplemental level of AFF. Feeding a diet supplemented with a moderate level AFF to lactating cows could be a way to alter the feed efficiency and fatty acid profile of milk by increasing potentially human consumer healthy fatty acid without detrimental effects on feed intake and milk production. A substantially decreased cholesterol proportion in milk induced by supplementation AFF suggests that alcohol fermented feed may improve milk cholesterol levels without any negative effects in lactating cows.
Objective: Dairy cattle nutrient requirement systems acknowledge amino acid (AAs) requirements in aggregate as metabolizable protein (MP) and assume fixed efficiencies of MP used for milk protein. Regulation of mammary protein synthesis may be associated with AA input and milk protein output. The aim of this study was to evaluate the effect of nanoemulsified methionine and cysteine on the in-vitro expression of milk protein (casein) in bovine mammary epithelial cells (MAC-T cells). Methods: Methionine and cysteine were nonionized using Lipoid S 75 by high-speed homogenizer. The nanoemulsified AA particle size and polydispersity index were determined by dynamic light scattering correlation spectroscopy using a high-performance particle sizer instrument. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was performed to determine the cytotoxicity effect of AAs with and without nanoionization at various concentrations (100 to $500{\mu}g/mL$) in mammary epithelial cells. MAC-T cells were subjected to 100% of free AA and nanoemulsified AA concentration in Dulbecco's modified Eagle medium/nutrient mixture F-12 (DMEM/F12) for the analysis of milk protein (casein) expression by the quantitative reverse transcription polymerase chain reaction method. Results: The AA-treated cells showed that cell viability tended to decrease (80%) in proportion to the concentration before nanogenesis, but cell viability increased as much as 90% after nanogenesis. The analysis of the expression of genetic markers related to milk protein indicated that; ${\alpha}_{s2}$-casein increased 2-fold, ${\kappa}$-casein increased 5-fold, and the amount of unchanged ${\beta}$-casein expression was nearly doubled in the nanoemulsified methionine-treated group when compared with the free-nanoemulsified methionine-supplemented group. On the contrary, the non-emulsified cysteine-administered group showed higher expression of genetic markers related to milk protein ${\alpha}_{s2}$-casein, ${\kappa}$-casein, and ${\beta}$-casein, but all the genetic markers related to milk protein decreased significantly after nanoemulsification. Conclusion: Detailed knowledge of factors, such nanogenesis of methionine, associated with increasing cysteine and decreasing production of genetic markers related to milk protein (casein) will help guide future recommendations to producers for maximizing milk yield with a high level of milk protein casein.
This study was conducted to investigate the effects of Aspergillus niger-derived multi-enzyme complex supplementation to feedrestricted lactating sows on performances, milk yield, blood profiles, and manure excretion as compared with ad libitum-fed sows without supplementation of enzyme. Fifty multiparous lactating Berkshire sows were allotted to 5 treatments of 10 sows per treatment during a 28-d lactation period and litter per sow was standardized to 9 suckling piglets. Treatments were ad libitum-fed sows without enzyme and feed-restricted sows supplemented with four increasing levels (0, 0.02, 0.04 and 0.08%) of multi-enzyme complex derived from Aspergillus niger. Blood samples from all sows were collected to determine serum metabolite concentrations before the morning feeding on d 27 of lactation. Litter body weight and a piglet weight at weaning, and litter weight gain significantly (P<0.05) increased with increasing levels of multi-enzyme complex, but there was no significant difference between ad libitum-fed sows without enzyme and feed-restricted sows supplemented with multi-enzyme complex. Body condition score and backfat depth at weaning significantly (P<0.05) increased as multi-enzyme complex level increased. Lactational backfat depth tended (P>0.05) to less decrease with increasing levels of enzyme complex. Serum inorganic phosphorus and non-esterified fatty acid concentrations significantly (P<0.05) increased with increasing levels of enzyme complex. Daily milk yield was not significantly different across treatments, but milk fat yield significantly (P<0.05) increased as multi-enzyme complex level increased. Manure output was significantly (P<0.01) higher for ad libitum-fed sows than for feed-restricted sows, but there was no significant difference among feed-restricted sows supplemented with increasing levels of multi-enzyme complex. Fecal phosphorus amount significantly (P<0.05) decreased with increasing levels of multi-enzyme complex. Feed costs of sows per litter weight gain were reduced by 1.25% to 9.67% with increasing levels of multi-enzyme complex as compared with ad libitum-fed sows without enzyme. The results indicated that multi-enzyme supplementation to feed-restricted lactating sows not only increased litter performances, but also was comparable to ad libitum-fed sows, resulting in reduced feed costs. Moreover, the reduction of fecal phosphorus amount with increasing levels of enzyme complex would contribute to the reduction of environmental pollution.
This study was aimed to solve the problems of current national genetic evaluation systems in Korea and its development to pass the verification processes as required by International Bull Evaluation Service (Interbull). This will enable Korea to participate in international genetic evaluation program. A total of 1,416,589 test-day milk records with calving dates used in this study were collected by National Agricultural Cooperative Federation from 2001 to 2009. Parity was limited up to fifth calving and milk production records were adjusted to cumulative 305 day lactation. The pedigree consisted of 2,279,741 animals where 2,467 bulls had 535,409 parents. A newly developed multiple trait model was used in calculation of breeding values for milk yield, milk fat, and protein yield. Data were edited with SAS (version 9.2) and R programs, and genetic parameters were estimated using VCE 6.0. Results showed a continuous increase in genetic potentials, in general, and no remarkable differences were found between performances by parity. Except fat yield, potentials in milk yield and protein yield were well calculated. We found an increased number of daughters per each top ranked 1,000 bulls in recent years of calf births compared to the cases of previous evaluations. Of the bulls ranked top 100 by our new models (multiple-trait models) we found that increased numbers of bulls were included. Of twenty eight bulls born in 2006, twenty bulls born in 2007 and eight bulls born in 2008 that were listed by new models, only 23, 12, and 2 bulls born in respective years were represented on top 100 by old single-trait models. Re-ranking of the daughters or sires by multiple-trait models suggest that this new multiple trait approach should be used for dairy cattle genetic evaluation and seed-stock selection in the future to increase the accuracy of multiple trait selection. Breeding values for these traits should also be calculated by new method for international genetic evaluation.
