Optimal Milieu for Culturing Porcine Sertoli Cell

  • The purpose of the present study was to establish culture conditions for the in vitro study of the neonatal piglet Sertoli cell. Isolation for the culture of Sertoli cell was established using collagenase and pancreatin digestion of testicular tissues. The effects of various culture media, fetal bovine serum(FBS), follicular stimulating hormone(FSH), epidermal growth factor(EGF) and insulin-transferrin-sodium selenite(ITS) on growth of neonatal piglet Sertoli cells were investigated. The mitogenic effects of Dulbecco's modified Eagle's medium+Ham's F-12 medium was higher than other media used in this experiment. The addition of 1% FBS in cultures was necessary for attachment of Sertoli cell clusters. However, except FBS and EGF, FSH and ITS did not stimulate Sertoli cell proliferation. When Sertoli cells isolated from neonatal piglets were cultured in Dulbecco's modified Eagle's medium+Ham's F-12 medium supplemented with 1% FBS, FSH EGF and ITS, the yield and plating efficiency of Sertoli cells were largely increased. Confluency of Sertoli cells was reached as early as 4 days of culture. The method described here reduces or eliminates many of the drawbacks of the conventional procedures used to isolate and culture of Sertoli cells, thus providing a useful tool in studies of growth kinetics and regulation of cell proliferation in vitro.

  • Purpose: For the last few decades, human civilization has observed an enormous change due to digitalization. However, question remains whether and how management accounting has been transformed to meet the demand of the digitalization. This paper provides a literary illustration of such transformation in the context of a digitalized world. Research design, data and methodology: This is a conceptual study based on extant literature. Therefore, the secondary sources of materials were used. Materials include literatures published in this domain, archival documents, and other publicly available data. Results: Drawing from literary evidence, the paper first highlights the evolving character of management accounting. Then it illustrates the changing landscape in the milieu of global and institutional settings with a focus in accounting profession. It further demonstrates the digitalization impact in management accounting. Here it also identifies the changing role of management accountants. Conclusions: The paper is critical for the management accounting professionals in identifying the attributes and nature of management accounting in the contemporary world. It also provides a sketch of the potential skillset that management accounting professionals should embrace to tackle the challenges of the new environment.

Haewon-sangsaeng as a Religio-Ethical Metaphor

  • This paper deals with figurative meanings of Haewon-sangsaeng. It is an investigation which is both semantic and diachronic. In the first part, important implications of sangsaeng (or xiangsheng in Chinese) in the context of correlative cosmology are extensively explored. Among others, saeng (in Chinese sheng) as a powerful metaphor and its related Chinese compounds are broadly discussed. In the second part, the evolution of ideas of yuan (or won in Korean) in Chinese history is explicated. Above all, in the traditional Chinese cultural milieu, wrongful treatments which make victims feel themselves aggrieved are socio-politically orientated. The Scripture on Great Peace (Taiping Jing) is used as reference point to elucidate the essential points of yuan and its knots. However, the advent of Buddhism in East Asia adds a new dimension to the understanding of yuan (won). Accumulated yuan as karmic bond thus gives a new identity of yuan as predetermined animosity. Widely recognized idioms like "adverse relatives and karmic debtors" and indigenous Chinese Buddhist rituals like Repentance Ritual of the Emperor Liang bear witness to this transformation of the meaning of yuan in East Asia. The fruitful yet correlated meanings of yuan also make the endeavor of untying yuan deeply significant and important to human society. Haewon-sangsaeng, as a religio-ethical ideal, brings out an amicable and harmonious relationship among myriad beings in the cosmos.

Clinical Features of Otomycosis Co-occurring with Chronic Otitis Media and the Causative Fungi

  • Background: Otomycosis is a fungal infection that comprises 7~10% of outer ear infections. Although the occurrence is higher in humid climates, relatively few studies have investigated otomycosis occurrences in humid environments. While recurrent chronic otitis media discharge in the ear creates a milieu in which otomycosis is likely to occur, investigations of otomycosis co-occurring with chronic otitis media have been rare. Objective: To examine the characteristics of patients with otomycosis co-occurring with chronic otitis media and identify causative fungi. Methods: The study included 60 patients with chronic otitis media who presented typical otomycosis findings in the outer ear canal and the presence of fungi. Patients were treated in the department of otolaryngology, Daegu Catholic University Medical Center, between July 2011 and June 2018. Results: The mean patient age was 57.77 years, and our study included 20 men and 40 women (p=0.010). The lesion was on the right in 39 patients and on the left in 21 (p=0.020). Ear discharge was the most common chief complaint at diagnosis. Of the 54 patients over age 19, 10 had diabetes (18.5%). Aspergillus was causative in 29 patients and Candida in 31. Aspergillus niger was identified in 15 patients, Aspergillus sp. in 14, Candida parapsilosis in 12, Candida sp. in six, and Candida albicans in five. Conclusion: Otomycosis and chronic otitis media co-occurrences increase with age. The Aspergillus and Candida genera were similar in proportion. A. niger was the most common Aspergillus species, while C. parapsilosis was the most common Candida.

Segmented Filamentous Bacteria Induce Divergent Populations of Antigen-Specific CD4 T Cells in the Small Intestine

  • CD4 T cells differentiate into $ROR{\gamma}t/IL$-17A-expressing cells in the small intestine following colonization by segmented filamentous bacteria (SFB). However, it remains unclear whether SFB-specific CD4 T cells can differentiate directly from naïve precursors, and whether their effector differentiation is solely directed towards the Th17 lineage. In this study, we used adoptive T cell transfer experiments and showed that naïve CD4 T cells can migrate to the small intestinal lamina propria (sLP) and differentiate into effector T cells that synthesize IL-17A in response to SFB colonization. Using single cell RT-PCR analysis, we showed that the progenies of SFB responding T cells are not uniform but composed of transcriptionally divergent populations including Th1, Th17 and follicular helper T cells. We further confirmed this finding using in vitro culture of SFB specific intestinal CD4 T cells in the presence of cognate antigens, which also generated heterogeneous population with similar features. Collectively, these findings indicate that a single species of intestinal bacteria can generate a divergent population of antigen-specific effector CD4 T cells, rather than it provides a cytokine milieu for the development of a particular effector T cell subset.

Development of teacher training program for overseas Korean language teachers of preservice career local milieu: focusing on 2017 Kazakhstan project by National Institute of Korean Language (한국어 예비·경력·현지 교원을 위한 국외 파견 실습 프로그램 개발 연구 -2017 국립국어원 카자흐스탄 파견 실습 프로그램 개발을 중심으로-)

  • The purpose of this paper is to develop a training program for overseas Korean language teachers focus on preservice teacher. This thesis based on the 2017 Korean Language (prospective) Teacher Overseas Dispatch and Practical Training Assistance Project (Almaty, Kazakhstan region). The present task established prospective teachers, career teachers, and local teachers as the targets of each assignment. We focused on developing a program that could match each of these characteristics. For prospective teachers, the program was designed and conducted with the goal of "improving real expertise through practical training," whereas for career teachers the program was developed with the goal of "improving leadership" and "retraining teachers" by focusing on their abilities as middle managers to build and maintain foreign and domestic networks. For local teachers, the goal was to provide "retraining as Korean language teacher certification". The limitations of those unable to attend domestic meetings were alleviated through training, workshops, and meetings, and a program was developed for real education practical training. For prospective teachers and career teachers in particular, the program was designed to center on a system of collaboration in which classes based on international Project Based Learning(iPBL) were conducted, and groups prepared practical training and practice modules.

Insomnia in Patients with Hematopoietic Stem Cell Transplantation(HSCT) (조혈모세포 이식 환자의 불면증)

  • Insomnia in patients with hematopoietic stem cell transplantation(HSCT) has been underdiagnosed and undertreated. This study reviewed the frequency, characteristics, physical and psychological effects, and treatments of insomnia in HSCT patients to highlight clinical importance in this specialized population. Furthermore, the authors intended to suggest a model that would conceptualize insomnia in the context of HSCT. In the pre-transplant period, about half of patients with HSCT suffered from sleep disturbance. A substantial number of patients experienced distressing insomnia during the HSCT procedure and recovered to the level of the pre-transplant period. However, sleep disruption could be a chronic symptom in HSCT survivors and could negatively impact quality of control, cancer-related fatigue(CRF), immune function, and psychological distress. The 3P's model(Predisposing, Precipitating, Perpetuating) explains insomnia in cancer population and could be also relevant to HSCT patients with specific consideration of CRF, graft-versus-host diseases, specific properties of hematological disease, and protective isolated milieu. Effective treatment of insomnia in HSCT includes non-pharmacological(e.g., cognitive behavioral therapy, environmental modification) and pharmacological interventions. The decision of pharmacological treatment should be based on the issue of safety due to high risk of potential drug-drug interactions. Screening, treatment, and further research of insomnia in HSCT patients using validated subjective and/or objective measures are warranted.

Anti-inflammatory Effects of Neuregulin-1 via the Downregulation of IL-6, IL-8, and MCP-1 Secretion

  • The trophic factor Neuregulin-1 (NRG-1) plays a critical role in the development of the peripheral nervous system and the repair of nerve injuries. The regulation of neutrophil apoptosis by cytokine secretion from structural cells is an important process in inflammatory diseases, including asthma. This study aimed to investigate the relationship between NRG-1 and the alteration of neutrophil apoptosis by the regulation of cytokine release in the human lung epithelial BEAS-2B cells. Tumor necrosis factor-alpha (TNF-α) and interferon-gamma (IFN-γ) induce the increase in the release of interleukin (IL)-6, IL-8, and monocyte chemoattractant protein-1 (MCP-1). NRG-1 alone had no effect on the secretion of IL-6, IL-8, and MCP-1. However, co-treatment of TNF-α and IFN-γ with NRG-1 inhibited the secretion of IL-6, IL-8, and MCP-1 that had been increased by TNF-α and IFN-γ. Treatment with NRG-1 did not have a direct effect on neutrophil apoptosis. Co-treatment of TNF-α and IFN-γ with NRG-1 was not effective on suppression of neutrophil apoptosis due to TNF-α and IFN-γ. The supernatant of BEAS-2B cells after co-treatment of TNF-α and IFN-γ with NRG-1 suppressed the inhibition of neutrophil apoptosis that had been caused due to the supernatant treated with TNF-α and IFN-γ. Taken together, NRG-1 has an anti-inflammatory effect in an inflammatory milieu by the regulation of cytokine secretion and neutrophil apoptosis.

Chemical Weathering Trend of Granitic Rock by evaluated with CIA in Southern Korea (화학적 풍화지수(CIA)로 본 한반도 중남부 화강암류의 화학적 풍화 경향성)

  • Grus weathering mantles are widely distributed in Southern Korean Peninsula and are considered to be results of chemical weathering related to palaeoclimate milieu. This paper attempts to address this issue by CIA(chemical index of alteration). The climatic approach to the formation of grus mantles offers limited explanation of field occurrences, as these materials are widespread across climatic zones, from the humid tropics to cool temperate areas, although rates of grusification are likely to be influenced by climatic parameters. CIA values for granitoid weathering mantles in S. Korea are 50, which is the same of unweathered granitic rocks. Grus mantles in Korean peninsula show very low level in chemical alteration by CIA.

Discussion of the Origin and the Properties of Granitic Red Regolith Profile in Goseong, Southern Korea (경남 고성의 화강암 적색 풍화층의 특색과 성인에 관한 논의)

  • Granitic regolith profile in Goseong, southern Korea, reveals various types of corestones from unweathered round fresh rocks to decomposed rock cores with unweathered spheroidal rock-layers on outer surface, deep weathering profile with 10m in depth, and red saprolite usually cited as related to high temperature climatic environment. In respect of this optical features such as the depth and the color of the profile, Goseong granitic profile may be regarded as products of paleoclimate with more high temperature and humidity than the present. But it is revealed that halloysite is a predominant clay mineral in upper part as well as lower part by XRD analysis. Halloysite is dominant mineral in hydrothermal alteration milieu. The leaching rate of chemical components is up to 97%, very high compared to common granite profiles with 60~70% in Korean peninsula. So, it is assumed that granitic red regolith in Goseong is a product of hydrothermal alteration, not meteoric weathering.
