Development and Characterization of 14 Microsatellite Markers for the Antarctic Midge Parochlus steinenii (Diptera, Chironomidae) in Maritime Antarctic

  • A winged midge species, Parochlus steinenii is one of the most abundant species in Antarctica, which is distributed over a wide area from the South American continent to the South Shetland Islands in Antarctica. It was dispersed into islands in the South Shetland Islands from the South American continent, and it adapted to a variety of environments and settled. This species, therefore, is a good model organism to explain the evolutionary process of Antarctic terrestrial fauna. Nevertheless, there are few genetic studies on this species, which are necessary for understanding the genetic diversity, population structure, etc. Here, we developed and characterized 14 polymorphic microsatellite markers. The number of alleles per locus ranged from 2 to 5. The observed and expected heterozygosities were in the range of 0.024 to 0.561 and 0.024 to 0.535, respectively. Identifying genetic differences between populations, they are suitable markers for researches investigating genetic diversity and population structure of P. steinenii, which provide us with clues to dispersion, evolution and ecology of this species.

Annual Fluctuations in Population of the Pine Needle Gall Midge, Thecodiplosis japonensis, and its Parasitoids in Kyeongbuk Province, Korea (경북지역에서의 솔잎혹파리(Thecodiplosis japonensis)와 솔잎혹파리기생봉의 개체군 변동)

  • This study was conducted to investigate the effect of parasititoids on the density-fluctuation of the pine needle gall midge, Thecodiplosis japonensis. The density of T. japonensis and its parasitoids was observed by emergence trap in pine forest in Namsung-dong, Keumho-eup, Youngcheon-shi, Kyeongsangbuk-do, Korea, from 1986 to 2005. After 1987, density fluctuations of adults pine needle gall midge and its parasitoids showed similar trends and density of parasitoids was dependent on density of host population. Sex ratio of the pine needle gall midge, T. japonensis, was female-biased and had some variations in according to years. These trends were similar to those of parasitoid populations and sex ratio of Inostemma seoulis was more female-biased than that of I. matsutama. T. japonensis adults emerged in the field from May 16 to July 27 and the peak of emergence was between late May and early June. The adults of Inostemma matsutama emerged from May 16 to June 11 (peak between late May and early June), whereas those of Inostemma seoulis emerged from June 9 to July 27 (peak: late June).

  • A new species of Platygasteridae, Inostemma hockpari (n. sp.) is described from Korea. This new species is an important biological control agent of the pine gall midge together with Inostemma seoulis, Platygaster matsutama and Inostemma matsutama (Collected in Jinhae by the author) in Korea.

  • Vegetatiion structure and population dynamcis of pine needle gall midge were investigated from 1980 to 1982 at the southern part of Palgong mountain near Daegu. The results of investigation at the areas of A (pure stand), B(30% mixed forest) adn C(60% mixed forest) were as follow; Vegeation of 3 areas were not classified by the species diversity, but by the unequal distribution of Alnus hirsuta, Robinia pseudo-acacia, Lespedeza cyrtobotrya, Quercus variabilis and vitality of Pinus densiflora at the area C was low, due to shadow by broad-leaf trees.   • Huge numbers of non-biting midges emerged from brackish water which were made at the harbor construction field in Jinhae City, Kyongsangnam-do, Korea in late summer in 2005, and caused a serious nuisance to villagers. The midges were collected and identified as Chironomus salinarius (Kieffer, 1921). Although this species was recorded in Korea for the first time in 1998, the morphological descriptions were so brief and simple. A full redescription is made with detailed illustrations for ecological and control workers of this nuisance midge.

  • The ecotoxicological effects of 4-tert-octylphenol were observed on non-biting midge, Chironomus plumosus collected from Anyang stream in Seoul. The survival rate and adult emergence rate on C. plumosus exposed to octylphenol were not significantly affected. However, 4-tert-octylphenol induced developmental delay and disrupted sex ratio in high concentration of octylphenol. The mouth deformity such as tooth deletion or fusion in mentum, and tooth deletion in mandible were observed exposure to 4-tert-octylphenol. The deformity type of the mentum showed deletion (LT, 6.7%), and fusion (LT, 6.3%). Moreover, tooth deletion of mandible was observed in 4-tert-octylphenol treated groups (6.7%, 3 ppm).

  • The aphid-eating gall-midge, Aphidoletes aphidimyza (Rondani), as predator of aphids was wildly used as one of the biological control agents for control of several kinds of aphids. Their some kinds of biological characteristics were examined. The developmental periods of A. aphidimyza from egg to adult with the green peach aphid as prey were 40 and 12 days at 15$^{\circ}C$ and 30$^{\circ}C$, respectively. Supplying the cotton aphid as prey, their developmental periods were shorter than with the green peach aphid. In case of the preservation of pupa in the cold condition, emergence rates were over 90% for 1 and 2 weeks at 5$^{\circ}C$ and l0$^{\circ}C$, respectively. Adult females of A. aphidimyza began mating and laying eggs at 2-3 days after emergence, and they laid about 200 eggs for lo days of average life span. Most of adults were emerged from pupa at 6 to 8 pm during a day, and they mainly acted in the early night.

  • Effect of monoterpene composition in pine needles on the susceptibility to pine gall midge (Thecodiplosis japonensis Uchida et Inouye) has been pursued. The pines studied include 5 susceptible and 2 resistant species. Also included were severely damaged or unaffected P. densiflora in the pine gall midge affected region. From the needles of the pine trees, 9 monoterpenes were identified by GLC. No correlationship, was found to hold in the monoterpene composition between pine gall midge susceptible and nonsusceptible pine species. Concentrations of limonene, however, gradually increased following oviposition in the unaffected P. densiflora while the concentrations of the monoterpene remained constant in the damaged P. densiflora. Effect of high limonene concentration in the needles of P. densiflora is discussed as a possible factor inducing resistance toward the pest in the unaffected Pinus densiflora.
