A Study on a Morphological Identification of Acanthopanacis Cortex (오가피(五加皮)의 형태(形態)에 관한 연구(硏究))

  • Objectives : In order to distinguish morphological characteristics of trunk bark and root bark of Ulmus davidiana var. japonica (Rehder) Nakai and the trunk bark and root bark of Hemiptelea davidii Planchon were sampled and compared in terms of their external and internal features with flour states according to their medical use, through microscopic examination. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and the flour of it was mounted by the liquid made by the same ratio of each of glycerin, acetic acid, and water, and then observed and photographed by olympus-BHT. Results : 1. Internal Features 1) A large parenchymatous cell was observed in the phloem of the slice of both trunk bark and root bark of Ulmi Cortex. However, both of the trunk bark and root bark of Hemipteleae Cortex did not have parenchymatous cell in the phloem; instead, stone cells including much square crystal of calcium oxalate were distributed around fiber bundle, and the parenchymatous cell included much druse crystal of calcium oxalate. 2) In both the Ulmi Cortex and Hemipteleae Cortex, rhytidome was observed in trunk bark, but not in root bark, but in the parenchymatous cell of the root bark of the Ulmi Cortex contained starch grain. 2. Flour States 1) In the flour of root bark of the Ulmi Cortex, a large parenchymatous cell was observed. However, in the flour of trunk bark and root bark of Hemipteleae Cortex, no parenchymatous eel was found; instead, stone cell including square crystal of calcium oxalate and druse crystal of calcium oxalate were observed. 2) There was no remarkable difference between the trunk bark and root bark of Hemipteleae Cortex. However, starch grain was contained in the parenchymatous cell of the root bark of Ulmi Cortex but not in the trunk bark of it. Conclusions : There were some morphological differences in external, internal, and flour parts of Ulmi Cortex and Hemipteleae Cortex. In particular, there was a morphological difference in flour states between the trunk bark and root bark of Ulmi Cortex, it is possible to use microscope to distinguish their flour states.

Comparison of the Efficiency between a Remodeled Bubble Generating Pumps for an Aquarium Fish and the Existed Commercial Air Sampler for the Sampling of Ambient Air Asbestos (공기 중 석면농도 분석시 관상어용 기포발생기를 개조한 장치와 기존의 상업용 시료 채취기와의 성능 비교)

  • Objectives: The purpose of this study is to estimate the applicability of regional sample collection of environmental samples. The concentration of asbestos fibers were analyzed with two devices. One was an existing commercial air sampling pump that has been proved to be accurate and exact, and the other is a remodeled pump for sample collection which was made from an electric bubble generator originally designed for aquarium fish. Samples were collected with the two devices under the same environmental conditions and collection equipment. A comparative analysis of the concentration of ambient asbestos fiber was then performed. Methods: Based on previous research, six farmhouses with asbestos fiber slate roofs known to have high concentrations of asbestos fiber were selected. Using the existing commercial air sampling pump and the remodeled electric bubble generator, four to seven samples were collected each day one meter downwind from the edge of the slate roof at high volume (about 4 L/min) and low volume (about 1.4 L/min). The analyzer responsible for sample quality control of asbestos fibers counted the number of asbestos fibers with a phase microscope. Results: The rates of flow change of the existed sampler and the remodeled pump at high volume were 0.82% and 0.17%, respectively. The rates of flow change at low volume were 3.83% and 1.09%, but there was not significant difference. The rates of flow change are within the error range (${\pm}5%$) of OSHA analyzing methods. For the high volume sampler, the average asbestos fiber concentration in the air collected by the existed sampler is 6.270 fibers/L and for the remodeled one 5.527 fibers/L, not a significant difference. For the low volume sampler, the average asbestos fiber concentration in the air collected by the existed sampler is 7.755 fibers/L and for the remodeled one 7.706 fibers/L, not a significant difference. The total area of the slate roof of the targeted farmhouse has an effect on the concentration of asbestos fibers in the air from the existing pump and the remodeled one (p<0.01). Conclusions: The sampling function between the existing commercial pump and the remodeled one shows little difference. Therefore, the remodeled pump is considered a pump with a good availability for collecting ambient air asbestos samples.

Morphological Studies of the Central Neural Pathways to the Pancreas, Sanyinjiao(Sp6) and Yinlingquan(Sp9) using Pseudorabies Virus (Pseudorabies virus를 이용한 췌장과 삼음교(三陰交)(Sp6), 음릉천(陰陵泉)(Sp9)에서 투사되는 중추 신경로에 관한 형태학적 연구)

  • Objectives : The purpose of this morphological studies was to investigate the relations between Sanyinjiao(Sp6), Yinlingquan(Sp9) and pancreas of rats using peudorabies virus(PRV). Methods : We observed labeled neurons following the injection of PRV, Bartha strain, into the Sanyinjiao(Sp6), Yinlingquan(Sp9) and pancreas of rats. After survival times of 4 days following the injection of PRV, the rats were perfused, and their spinal ganglia, spinal cord and brain stem were frozen sectioned($35{\mu}m$). These sections were strained by PRV immunohistchemical staining methods and observed with light microscope. Results : The results were as follows. 1. In the spinal ganglia, the overlap areas of PRV labeled neurons projecting to Sanyinjiao(Sp6), Yinlingquan(Sp9) and pancreas were observed in T10-13 dorsal root ganglia. 2. In the spinal cord, the overlap areas of PRV labeled neurons projecting to Sanyinjiao(Sp6), Yinlingquan(Sp9) and pancreas were lamina I, IV, V, VII, IX, X, intermediolateral nucleus(IML), intermediomedial nucleus(IMM) in thoracic segments. In lumbar segments, the overlap areas of PRV labeled neuron were lamina I, IV, V, VI, IX, X and IMM. In sacral segments, the overlap areas of PRV labeled neuron were lamina I, IV, V, VI, VII, IX, X. 3. In the brain, the overlap areas of PRV labeled neurons projecting to Sanyinjiao(Sp6), Yinlingquan(Sp9) and pancreas were area postrema, nucleus tractus solitarius, caudoventrolateral reticular nu., medullary reticular nu., lateral paragigantocellular nu., C3 adrenalin cells, gigantocellular nu., raphe pallidus nu., raphe obscurus nu., ambiguus nu., raphe magnus nu., pontine reticular formation, A5 cell group, subcoeruleus nu., locus coeruleus, Barringnton's nu., $K{\ddot{o}}lliker$-Fuse nu., dorsal raphe nu., Edinger-Westphal nu., central gray matter, perifornical nu., dorsomedial hypothalamic nu., arcuate nu., lateral hypothalamic nu., paraventricular hypothalamic nu., hindlimb area. Conclusions : In conclusion, these results suggest that the interrelationship of meridian(spleen meridian), acupoints(Sp6 and Sp9) and viscera(pancreas) may be related the central autonomic centers.

The Effects of Sanpunggosamhwan on the Allergic Contact Dermatitis (산풍고삼환(散風苦蔘丸)이 알레르기성 접촉피부염에 미치는 영향)

  • Objectives: To study the effectiveness of Sanpunggosamhwan(SGH) against Allergic Contact Dermatitis(ACD), the contact hypersensitivity assay, change of cutaneous shape, anti-allergic effect, anti-inflammatory effect, and the effect on skin barrier were observed. Methods: The 200g rats were divided into three groups of 15 rats. The first group is the Normal group which was applied Acetone olive oil only. The second group is the ACD group which has intentionally activated Allergic Contact Dermatitis by DNCB. The third group is the SGH group which was given medication of Sanpunggosamhwan extract after the induction of Allergic Contact Dermatitis by DNCB. Each group of rats was observed after 24, 48 and 72 hours. Results: I. With the result of contact hypersensitivity assay, at 24hours SGH group showed appreciably less ear swelling than ACD group. 2. Regarding general change of skin, SGH group showed less hyperplasia of epidermis, less damage of epidermis than ACD group. 3. Regarding the number of WBC, ACD group showed significantly less than normal and SGH group at 72 hours. 4. Regarding the number of RBC in blood, ACD and SGH group showed significantly more RBC than normal group at 24, 48, 72 hours. 5. Regarding the ratio of neutrophil in WBC, ACD and SGH group showed significantly high percentage than normal group at 24, 48 hours. 6. Regarding the ratio of lymphocyte in WBC, ACD and SGH group showed significantly high percentage than normal group at 48 hours. 7. Regarding the erythema, SGH group showed significantly more erythema than normal and ACD group at 48 hours. 8. Regarding the melanin, SGH group showed significantly less melanin than normal group at 24 hours.9. Regarding the skin hydration, SGH group showed significantly high value than and ACD group at 72 hours. 10. Regarding the skin pH, ACD group showed significantly high value than normal and SGH group af 24 hours. 11. Regarding the number of Total IgE, ACD and SGH group showed more Total IgE than normal. g개up at 24 hours. 12. At Electro microscope-morphologic changes of skin, the damage of epithelium was decreased and regeneration power of skin was increased in the SGH group. Conclusions: The Sanpunggosamhwan extract administration was effective on the mitigation of skin damage in rats with allergic contact dermatitis.

Embryonic Zebrafish Model - A Well-Established Method for Rapidly Assessing the Toxicity of Homeopathic Drugs - Toxicity Evaluation of Homeopathic Drugs Using Zebrafish Embryo Model -

  • Objectives: Advancements in nanotechnology have led to nanoparticle (NP) use in various fields of medicine. Although the potential of NPs is promising, the lack of documented evidence on the toxicological effects of NPs is concerning. A few studies have documented that homeopathy uses NPs. Unfortunately, very few sound scientific studies have explored the toxic effects of homeopathic drugs. Citing this lack of high-quality scientific evidence, regulatory agencies have been reluctant to endorse homeopathic treatment as an alternative or adjunct treatment. This study aimed to enhance our insight into the impact of commercially-available homeopathic drugs, to study the presence of NPs in those drugs and any deleterious effects they might have, and to determine the distribution pattern of NPs in zebrafish embryos (Danio rerio). Methods: Homeopathic dilutions were studied using high-resolution transmission electron microscopy with selected area electron diffraction (SAED). For the toxicity assessment on Zebrafish, embryos were exposed to a test solution from 4 - 6 hours post-fertilization, and embryos/larvae were assessed up to 5 days post-fertilization (dpf ) for viability and morphology. Toxicity was recorded in terms of mortality, hatching delay, phenotypic defects and metal accumulation. Around 5 dpf was found to be the optimum developmental stage for evaluation. Results: The present study aimed to conclusively prove the presence of NPs in all high dilutions of homeopathic drugs. Embryonic zebrafish were exposed to three homeopathic drugs with two potencies (30CH, 200CH) during early embryogenesis. The resulting morphological and cellular responses were observed. Exposure to these potencies produced no visibly significant malformations, pericardial edema, and mortality and no necrotic and apoptotic cellular death. Conclusion: Our findings clearly demonstrate that no toxic effects were observed for these three homeopathic drugs at the potencies and exposure times used in this study. The embryonic zebrafish model is recommended as a well-established method for rapidly assessing the toxicity of homeopathic drugs.

An Investigation on the Airborne Asbestos Concentrations using PCM and TEM in the Public Buildings in Seoul (PCM과 TEM을 이용한 서울지역 일부 공공 건축물의 실내공기 중 석면농도 조사)

  • Objectives: This investigation is purposed to evaluate the airborne asbestos concentrations in the public buildings having asbestos containing materials(ACMs) in Seoul. Methods: The Seoul Metropolitan Government carried out an asbestos survey to the city-owned public buildings to identify the level of risk exposure, classified into low, moderate and high risk. To evaluate the airborne concentration of asbestos, 11 sampling sites in ten buildings based on the survey were selected. The air samples from the eleven sites were analyzed by Phase Contrast Microscopy(PCM) and Transmission Electron Microscopy (TEM), and compared the analytical results from the both. Results: 1. The airborne fiber concentrations by PCM were less than the detection limit($7f/mm^2$) in 9(82%) out of 11 sampling sites. The highest concentration was 0.0043 f/cc, but it was below the guideline value for indoor air quality(0.01 f/cc), proposed by the Ministry of Environment, Korea. 2. In two sampling sites, having moderate risk level, the chrysotile was identified and showed it's concentrations of 0.0102 s/cc and 0.0058 s/cc, less than $5{\mu}m$ lengths. 3. The ACMs identified in the two sampling sites were a packing material(65% of chrysotile) in mechanical area and a thermal system insulation(5% of chrysotile) in a boiler room. Having more possibility of asbestos emission in the mechanical area, it would be required to set up and carry out the asbestos management plan. Conclusions: Based on the result of this study, the airborne asbestos concentrations in the public buildings with ACMs were generally lower than the guideline value for indoor air quality. There are widespread concerns about the possible health risk resulting from the presence of airborne asbestos fibers in the public buildings. Most of the previous studies about airborne asbestos analysis in Korea were performed based on PCM method that asbestos and non-asbestos fibers are counted together. In the public and commercial buildings, having ACMs, it is suggested that the asbestos be analyzed by TEM method to identify asbestos due to concerns about asbestos exposure to workers and unspecified people.

Overexpression of Astrocyte Elevated Gene-1 (AEG-1) in Cervical Cancer and its Correlation with Angiogenesis

  • Objectives: To explore the expression of astrocyte elevated gene-1 (AEG-1) in cervical cancer and analyze its correlation with microvascular density (MVD), nuclear factor kappaB (NF-kB p65) and vascular endothelial growth factor (VEGF). Materials and Methods: Immunohistochemical MaxVision method was adopted to detect the expression level of AEG-1, NF-kB p65 and VEGF in 45 samples of invading cervical cancer and 12 samples of cervicitis from The First Affiliated Hospital of Wenzhou Medical University. Tumor microvascular endothelial marker CD34 combined with Weidner was used to determine the MVD in cervical cancer tissue. The positive expression and staining conditions of AEG-1, NF-kB p65 and VEGF in cervical cancer tissues were observed under a light microscope. Correlations between expression of AEG-1 protein and those of NF-Kb p65 and VEGF, as well as MVD, were analyzed using Pearson correlation. Results: The expression levels of AEG-1 were $0.186{\pm}0.043$ in cervical cancer and $0.051{\pm}0.002$ in chronic cervicitis (p<0.01). Moreover, expression of AEG-1 was related to vascular invasion and lymphatic metastasis of cervical cancer (p<0.01), but not with age of the patients, differentiation degree, tumour size, pathological type and parametrial infiltration (p>0.05). Pearson correlation analysis showed that the expression of AEG-1 was linked with NF-kB p65 (r=0.501, p=0.000), VEGF (r=0.718, p=0.000) as well as MVD in cervical cancer tissue (r=0.815, p=0.000). Conclusions: AEG-1 is highly expressed in cervical cancer and promotes angiogenesis, which might be related to the fact that AEG-1 activating the signal pathway of NF-kB could up-regulate the level of VEGF expression.

The Effect of MK801 on SSEP and Patholoy in Chronic Spinal Cord Injured Rat (척수 손상 백서에서 MK801 투여가 체성감각 유발전위 및 척수 조직에 미치는 효과)

  • Objectives : This study was undertaken to investigate the effect of noncompetitive NMDA receptor blocker, MK801 on motor recovery, SSEP and pathology in spinal cord injured rat. Methods : The effects of MK801 on neuronal function protection, SSEP, and pathology were measured on spinal cord injury rats which were divided into 6 groups according to dose, time of drug delivery and magnitude of injury. Spinal cord injury was made with the magnitude of 25gm-cm and 50gm-cm on 42 rats. BBB locomotor function test was performed to evaluate the motor power recovery in hindlimb for 2 weeks after injury. After motor function test was completed, SSEP was measured. Amplitude and latency of the P1, N1 peak was measured and compared between groups. Finally rats were sacrificed, and pathologic findings including measurement of area of necrotic cord were studied and compared between groups. Results : Motor recovery at 2 weeks was better in MK801 group comparing to saline control group. SSEP at 2 weeks showed no difference in N1, P1 latencies, but significantly greater amplitude in MK801 group, compared to saline control group. On light microscope, there was no specific histologic differences between experimental groups. The cystic necrotic area in coronal plane was measured and compared in each group. The necrotic area was significantly smaller in MK801 1mg/kg group(delivered after injury) than vehicle group. The necrotic area in MK801 5mg/kg group and MK801 1mg/kg group(delivered before injury) was smaller than vehicle group even though it was not statistically significant. Conclusion : From the above result, it is speculated that NMDA blocker, MK801 can improve impaired neuronal function in spinal cord injury.

The effect of initial packaging on the deformation of ear acupuncture needle tips (1차 포장이 이침의 침첨 불량에 미치는 영향)

  • Objectives : The present study is to verify whether the deformation of ear acupuncture needle tips that are currently on the market is caused during the process of initial packaging. Methods : Ear acupuncture needles distributed from three different companies were collected to measure the size of the groove on the transparent plate which is used in for the intial packaging. Then the tolerance range for off-centered tips, which represent the range of the groove size that is safe from damaging the needle tip, was determined based on the vertical length measured from tip to handle. 200 ear acupuncture needles were dipped in alcohol and scanned to confirm the position of the needles. Stereo-microscope was used to observe the needle tips that were located outside the tolerance range. Results : The increased number of ear acupuncture needle tips placed outside the tolerance range were seen from in the order of C company, A company and B company. The increased deformation of the needle tips caused by initial packaging was observed in the order of A company, C company and B company. Conclusion : Only the ear acupuncture needles from B company showed the strong correlation between needle tip deformation and initial packaging. It has been observed that the deformation of the ear acupuncture needles is mainly due to manufacturing process, not the packaging process.

Electric Corrosion of STS304 Acupuncture Needles Used for Electroacupuncture (STS304 스테인리스 강선 호침에서 전침 자극 후 침체에 나타나는 전기적 부식 연구)

  • Objectives : The electroacupuncture(EA) was often used in Oriental medicine clinic and basic researches. However, there have been neither any national standard of acupuncture needles in Korea nor many studies about the property of acupuncture needles for applying electrical stimulations. Therefore we investigated the electrical corrosion property of acupuncture needles used for EA stimulation in clinic. Methods : We observed the occurrence of electrical corrosion like changes in the surface of acupuncture needles after EA stimulation with various parameters(electrical stimulation intensity, treatment duration, diameter of needle) under a light microscope. We performed EA stimulation on Baekhoe($GV_{20}$) or Joksamli($ST_{36}$) of rats with intensity of 0.025, 0.05, 0.075, 0.1, 0.25 or 0.5mA at a 10Hz frequency. Results : The electrical corrosion-like changes were found on the surface of needles when the EA stimulation was given over 0.05mA of intensity. When electrical stimulation was treated for 10min, the electrical corrosion-like changes were low, but the electrical corrosion-like changes gradually increased by the treatment duration(20, 30min). We also found that acupuncture needles of 0.2mm diameter were produced more extensive electrical corrosion-like changes than acupuncture needles of 0.3 or 0.4mm diameter by electrical stimulation. Conclusions: These results suggested that EA stimulation could produce electrical corrosion-like changes of acupuncture needles through intensity of current, duration of electric stimulation and/or diameter of needle.
