Analysis of Microbiological Contamination in the Chosun Dynasty Textiles Exhumed from Hwasung Kupori Burial

  • The purpose of this research was first to analyse and compare the types of microbes inherent among the unwashed and washed Kupori textiles, and second to investigate whether there is a difference between unwashed and washed Kupori textiles on the susceptibility of contamination by microorganism when exposed to the same microbial environment. Microbial identification procedure and the Shake Flask Test for investigating the effect of exposure to microbial environment were carried out separately. The result of microbial identification procedure indicated that a variety of bacteria and fungi were inherent in both unwashed and washed textiles and that the population of contaminated microorganism became more diverse after washing. The result of Shake Flask Test indicated that given the same exposure condition, the unwashed textiles tend to be more susceptible to bacterial contamination than the washed textiles. The results of the present study supported the current conservation procedures adopted in Korean museums which include washing and humigation procedures before long-term storage or display of exhumed textiles.

Extraction and Degradation of Alginate from Brown Seaweed Stem Using Microorganism (미생물을 이용한 미역줄기에서 알긴산염 추출 및 저분자화)

  • We studied a extraction and degradation of alginate from seaweed-stems using microorganism DS-02. DS-02 has a maximum growth rate at $30^{\circ}C$ and the enzyme has a maximum activity of alginate extraction at $35^{\circ}C.$ The yield of alginate extraction using DS-02 is about $16.0{\%}$ for 3.0 hour and molecular weight of the alginate decreased to about 1/8 of initial value after 24 hour extraction. Alginate extraction method by DS-02, compared with general alkali-extraction method, has an advantage of decreasing the molecular weight of alginate during extraction.

  • This paper was carried out to investigate change in the initial microorganism of Pepper which were treated with ozonated water for microbial decontamination. When ozonated water was 30, 60, 90minutes treatment, there was decrease in the overall total microbial of the black & white pepper. Total lactic acid bacteria died completely after contacted with ozonated water for 60minutes but the inhibition rate on the total aerobic bacteria of black & white pepper were 62% and 96%, respectively. Total aerobic bacteria of four packing material was increased after 1 months of storage at 37$^{\circ}C$ when it had treated with 90minutes ozone reaction Then, there was no significant changes in the piperin contents.

  • Titanium(Ti) alloys has been mostly concerned with biocompatibility, corrosion resistance, and biofunctionality. However, very little is known, about the biological effects of titanium on microorganism and in particular on the oral flora. So, the effect of titanium on the in vitro growth of oral microorganism forming dental caries was studied under either aerobic or anaerobic condition. In this study, the mostly bacterial species commonly found in dental plaque or gingival sulcus grew well in an aqueous medium containing $100{\mu}g/ml$ of titanium standard solution.

  • Microorganisms living in clothing cause damage to fabric as well as unhygienic conditions with unpleasant odor fur wearers. Removal or growth of microorganisms are affected by the conditions during washing and storage. The purpose of this research was to study the consumer's knowledge and habits in laundering with respect to microorganisms in clothing. For survey method, questionnaires were administered to 580 housewives, age of 20∼60s living in Seoul. Employing 479 respondents, the data were analyzed by using descriptive statistics. The results are as follows: The level of knowledge about microorganisms of clothing was high in general, but wasn't expert level. Many people had experienced damages of textiles, clothing and unpleasant odor due to microorganisms. Fabric softeners and bleaches were rarely used for disinfection but usually used for antistatic, whitening or removal of stains. There was no relationship between laundering habits, the knowledge of microorganism, and experience of clothing damage by microorganism.

  • This paper presents the study of the effect of microorganism Bacillus pasteurii on the properties such as Atterbergs' limit and unconfined compressive strength of cohesive soils. The results of this study reveal that the liquid limit and plasticity index for all clay soils decreased and the unconfined compressive strength increased. Decrease in plasticity index is very high for Kuttanad clay followed by bentonite and laterite. The unconfined compressive strength increased for all the soils. The increase was high for Kuttanad soil and low for laterite soil. After 24 h of treatment the improvement in the soil properties is comparatively less. Besides the specific bacteria selected Bacillus pasteurii, other microorganisms may also be taking part in calcite precipitation thereby causing soil cementation. But the naturally present microorganisms alone cannot work on the calcite precipitation.

A Survey of Macrofungal Diversity in Da-Lat, Southern Vietnam

  • Da-Lat is a hilly area located in southern Vietnam. Macrofungal diversity of Da-Lat was investigated from 2018 to 2019. A total of 468 macrofungal specimens was collected and identified using the modern species concept and taxonomic and phylogenetic analyses. Among them, internal transcribed spacer(ITS) region of 401 specimens were successfully sequenced and compared with those of related species retrieved from GenBank. In total, 180 specimens were identified at the species level. The sequenced specimens were classified into 2 phyla, 13 orders, 38 families, 93 genera, and 124 species. The remaining 221 specimens (175 species) did not match the species level. This study is the first well-documented taxonomic list of macrofungi collected from southern Vietnam.

Biodegradation Rate of Recycling Soap Prepared from Non-Cooking Oils (폐식용유로 제조된 재생비누의 생분해 속도)

  • A recycling soap was prepared from non-cooking oils. The effects of physlcal and chemical properties of the recycling soap on biodegradation are expected to be different due to the thermal histories of the non-cooking oils. Therefore, the biodegradation rate of the recycling soap was studied by using Klebssella Pneumoniae(K. pneumoniae), and the growth rate of K. pnewoniae in soap solution was observed. The biodegradation rate of the recycling soap appeared to be slower as the thermal histories of the non-cooking oils became larger. This might be resulted from hydrolysis, in which the ester bonds in the oils are broken to produce hydroxyl group. It was also observed that the growth rate of the microorganism decreased with the increase in the thermal histories of the oils. As a result, it is desired that recycling soap should be produced from the non-cooking oils with the prober ranges of thermal histories to reduce water contamination. The non-cooking oils with larger thermal histories are considered to be recycling through the cracking process before used. Opto-electrokinetic Technique for Microfluidic Manipulation of Microorganism (광-전기역학 기술을 이용한 미생물의 미세유체역학적 제어)

  • This paper introduces microfluidic manipulation of microorganism by opto-electrokinetic technique, named rapid electrokinetic patterning (REP). REP is a hybrid method that utilizes the simultaneous application of a uniform electric field and a focused laser to manipulate various kinds and types of colloidal particles. Using the technique in preliminary experiments, we have successfully aggregated, translated, and trapped not only spherical polystyrene, latex, and magnetic particles but also ellipsoidal glass particles. Extending the manipulation target to cells, we attempted to manipulate saccharomyces cerevisiae (S. cerevisiae), the most commonly used microorganism for food fermentation and biomass production. As a result, S. cerevisiae were assembled and dynamically trapped by REP at arbitrary location on an electrode surface. It firmly establishes the usefulness of REP technique for development of a high-performance on-chip bioassay system.

A report of 10 unrecorded bacterial species of Korea, isolated from agricultural soil in 2022

  • In 2022, research for native prokaryotic species in Korea reported 10 unrecorded bacterial strains affiliated to phyla Actinomycetota, Bacillota, and Pseudomonadota. The strains formed monophyletic clades with the most closely related species (with ≥98.7% sequence similarity) in the 16S rRNA gene sequencing. Among them, four species of the phylum Actinomycetota, two species of the phylum Bacillota, and four species of the phylum Pseudomonadota have not been reported in Korea, suggesting unrecorded species in Korea. Information on strains such as Gram staining reaction, colony and cell morphology, biochemical characteristics, and isolation sources were provided in the species description.
