Analysis of Pathogenic Microorganism's Contamination and Heavy Metals on Kimchi Cabbage by Cultivation Methods in Korea (재배농법에 따른 국내산 배추의 위해미생물 및 중금속 오염평가)

  • Kimchi cabbage is one of the four major vegetable crops in Korea. The total annual production of kimchi cabbage, the main material of kimchi, was 20,559 tons in 2015. Kimchi cabbage is one of the majer crops produced by farmers which accounts for about 80% of the total leaf vegetable production in Korea. As the consumption of environmental-friendly agricultural products increases, food safety is one of the major public health concerns. We analyzed the biological hazards of kimchi cabbage produced by two types of cultivation methos such as organic farming and conventional farming using various culture media and microscopy. A total of 432 samples were analysed for presence of sanitary indicator microorganisms (aerobic plate count, coliform count, yeast & mold) and food-borne pathogens (Staphylococcus aureus, environmental Listeria, Bacillus cereus). The population of sanitary indicating microorganisms and food borne pathogens was under 5 Log CFU/g in all tested samples. The results of total microorganism numbers of leaf surface showed a positive correlation to those of soil samples. Additionally, we examined chemical factors such as pesticide residues and heavy metals in soil samples. All tested samples did not shown contamination levels higher than the standard limit.

An Automatic Mobile Cell Counting System for the Analysis of Biological Image (생물학적 영상 분석을 위한 자동 모바일 셀 계수 시스템)

  • This paper presents an automatic method to detect and count the cells from microorganism images based on mobile environments. Cell counting is an important process in the field of biological and pathological image analysis. In the past, cell counting is done manually, which is known as tedious and time consuming process. Moreover, the manual cell counting can lead inconsistent and imprecise results. Therefore, it is necessary to make an automatic method to detect and count cells from biological images to obtain accurate and consistent results. The proposed multi-step cell counting method automatically segments the cells from the image of cultivated microorganism and labels the cells by utilizing topological analysis of the segmented cells. To improve the accuracy of the cell counting, we adopt watershed algorithm in separating agglomerated cells from each other and morphological operation in enhancing the individual cell object from the image. The system is developed by considering the availability in mobile environments. Therefore, the cell images can be obtained by a mobile phone and the processed statistical data of microorganism can be delivered by mobile devices in ubiquitous smart space. From the experiments, by comparing the results between manual and the proposed automatic cell counting we can prove the efficiency of the developed system.

Isolation and characterization of acid-resistanct and halophilic bacteria using cultivation technique in Jeju island (배양기법을 활용한 제주도내 내산 및 호염성 미생물의 분리 및 특성 분석)

  • In this study, we isolated about 70 bacterial strains from terrestrial and marine environments in Jeju island, and finally, total 21 strains were obtained based on the 16S ribosomal RNA gene sequence analysis. These isolated strains were classified into 16 genera of 5 classes and were identified as an unrecorded species in the Republic of Korea. As a result of the substrate utilization and capability for polymer degradation, the physiological phenotypes for acid resistance and halophilic bacteria were observed to be distinct from each other, except for some acid resistance strains. This study might provide basic information on utilization for indigenous microorganisms.

PCE, TCE로 오염된 지하수내 미생물 특성 및 분포

  • Chlorinated aliphatic hydrocarbons (CAHs) especially perchlorethylene (PCE) and trichlooethylene (TCE) are common groundwater contaminants in Korea. PCE and TCE were often reductively dechiorinated in an aquifer. Several isolates dechlorinate PCE to TCE or cis-1,2 dichloroethylene (c-DCE) were obtained from contaminated and pristine sites in USA and Europe. However in Korea, no information on indigenous microorganism being involved in reductive dechlorination of PCE and TCE is available and different dechlorinating microorganisms might be reside in Korea, since geochemical, and hydrogeological conditions are different, compared to those in the other sites. So we evaluate that: 1) if reductive dechlorinating microorganisms are present in PCE-contaminated site in Korea, 2) if so, what kinds of microorganisms are present; 3) to what extent PCE is reductively dechlorinated. As a results in some PCE-contaminated aquifers in Korea other dechlorinating microorganisms but Dehalococcoides ethenogenes may be responsible for PCE dechlorination. More detailed molecular works are required to evaluate that different dechlorinating microorganisms would reside in Korea.

3,5-Bis(aminopyrimidinyl)indole Derivatives: Synthesis and Evaluation of Pim Kinase Inhibitory Activities

  • Pim kinases are promising targets in the treatment of hematopoietic and solid cancers. Meridianin C was chosen as a starting point to discover novel pim kinase inhibitors. Using known pim kinase's structural information, aminopyrimidine was introduced to provide the hydrogen-bonding interactions with the conserved lysine residue in the ATP binding pocket of all three Pim kinases. Synthesized 3,5-bis(aminopyrimidinyl)indole derivatives showed pan-pim inhibitory activity. Aminoalkyl substituent was attached on the aminopyrimidine to further enhance the potency and physicochemical properties of compound. The research reveals a significative way of designing compounds with high potency and kinase selectivity for pan-pim kinases.

  • Melatonin, an indolic tryptophan-derived compound, is secreted rhythmically from the pineal gland, mainly under the regulation of the circadian clock located in the suprachiasmatic nuclei (SCN) of the hypothalamus. Melatonin is widely present in nature, with biological activities in unicellular organisms, plants, and animals. A major function of melatonin is to transmit information to organisms about certain physiological functions in response to daily and seasonal variations in their environment. In this paper, we review a variety of melatonin's functional properties, its occurrence in plants, and its synthesis by yeasts. Fermented milk supplemented with melatonin-rich plants and yeasts can be used for the effective treatment of sleep disorders.

Identification of Actinomycins by High Performance Liquid Chromatography and Fast Atom Bombardment Mass Spectrometry

  • An acinomycin complex isolated from culture broth of a soil microorganism, SNUS 9305-011 has been examined by High performance liquid chromatography (HPLC). From the analysis of the fractions obtained by column chromatography of the ethyl acetate extract, three actinomycin components are confimed . The HPLC analysis is carried out with a CN-bonded nucleosil column. Comparison of the retention times of the components with those of actinomycin D, C complex, $X_{o{\beta}$, and V and suggests that they are different actinomycins. FBA mass spectra fo the coponents also shows different molecular ions from those of standards and other reported actionbmycins. The present work has demonstrated that actinomycin components can be separated by a CN-bonded HPLC column, and that ocmparison of their HPLC chormatograms with authentic smaples and information on their molecular ions can be successfully employed for indentification of actionmycins.

A Review on Identification Methods for TCE Contamination Sources using Stable Isotope Compositions (안정동위원소 조성을 이용한 TCE 오염원 규명방법 소개)

  • This study was performed to summarize application of ${\delta}^{13}C$, ${\delta}^{37}Cl$ and ${\delta}D$ of trichloroethylene (TCE) to studies on environmental forensic field regarding identification of TCE sources and evaluation of contribution of TCE to groundwater using data collected from literatures. ${\delta}^{13}C$, ${\delta}^{37}Cl$ and ${\delta}D$ of TCE give some information regarding sources of TCE because they show specific value according to manufacturing method. Also, TCE do not show a significant isotopic fractionation owing to adsorption and dilution. The isotopic fractionation mainly occurs by biodegradation. In addition, isotopic fractionation factor for TCE is different according to a kind of microorganism participated in biodegradation. However, the isotopic data of TCE have to be applied with chemical compositions of TCE and other hydrogeologic factors because isotopic fractionation of TCE is influenced by various factors.

Deciphering Functions of Uncultured Microorganisms (난배양성 미생물의 기능 분석 방법)

  • Microbes within complex communities show quite different physiology from pure cultured microbes. However, historically the study of microbes has focused on single species in pure culture and most of microbes are unculturable in our labs, so understanding of complex communities lags behind understanding of pure cultured cells. Methodologies including stable isotope probing (SIP), a combination of fluorescence in situ hybridization (FISH) and microautoradiography (MAR), isotope micrarray, and metagenomics have given insights into the uncultivated majority to link phylogenetic and functional information. Here, we review some of the most recent literatures, with an emphasis on methodological improvements to the sensitivity and utilities of these methods to link phylogeny and function in complex microbial communities.

A Study on the Purchase Action of Processed Foods and the Recognition for Food Additives of Urban Housewives (도시주부의 가공식품 구매행동과 식품첨가물에 관한 인식 연구 -서울.경기지역 거주 주부를 대상으로-)

  • This study was carried out to give some help for housewives recognizing the desirable purchase of processed foods and food additives. This results were abtained as follows; In case of purchase action for processed foods of housewives confirmation of manufacturing date was the highest point(4.62). When housewives bought processed foods they considered the taste of them as first, and frequencies of processed foods intake showed the ratio of 46.6%, also it was shown to be used once a week. The order for use of processed foods were milk, milk products(butter, cheese, yoghurt, lactic bacterium beverage) 40.2% >canned foods 22.6% >meat products(ham, sausage, bacon) 20.9% > jelled fish 16.3%. Most of housewives wanted to have the knowledge and information for food additives at the highest point(4.11). The interest about food additives of housewives was shown to the order of synthetic preservative 3.86 >chemical seasoning 3.74 >synthetic sweetener 3.59 >synthetic coloring agent 3.53. Also, it was shown to be thought that the hazard factors effected on food safety were agricultural chemical residue(30.9%), environmental pollution(25.2, food additives(23.0%), and microorganism contamination(20.9%).
