Biological Control of Fusarium Wilt of Carnation Plants by Antagonistic Microorganism in Greenhouse (카네이션의 시설재배에서 길항성 세균을 이용한 Fusarium Wilt 의 생물학적 방제)

  • This study was carried out to screen and select the effects of antifungal bacterial strains which inhibit the growth of plant pathogen, Fusarium oxysporum(fusarium wilt) occurred in carnation plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it's identification. Twenty bacterial strains which strongly inhibited Fusarium oxysporum were isolated from roots of carnation plants and the soil in greenhouse, and the best antifungal bacteria designated as C121, was finally selected. Antagonistic bacterial strain, C121 was identified to be the genus Bacillus sp. based on the morphological, biochemical and cultural characterizations. The Bacillus sp. C121 showed 58.1% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the cultural broth and the heat bacterialization culture filtrate of it, Bacillus sp. C121 was shown 92.1% and 21.0% of antifungal activity, respectively.

  • This study was conducted to screen the antagonistic bacteria which inhibit the growth of plant pathogen, fusarium wilt(Fusarium oxysporum) occurred in tomato plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it’s identification. Ten bacterial strains which strongly inhibited Fusarium oxysporum were isolated from the nature, and the best antagonistic bacterial strain designated as KC175, was selected. The antagonistic strain KC175 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. The Bacillus sp. KC175 showed 58.2% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the culture broth and the heat bacterialization culture filtrate of it, Bacillus sp. KC175 showed 91% and 18% of antifungal activity, respectively.

  • The purpose of this study was to evaluate the growth of lettuce last for 36 days. Lettuce planted on the same medium with different treatment. The first treatment watering using bio-fertilizer (farmer) and second watering using effective microorganism. Lettuce grown in greenhouse with temperature $18-32^{\circ}C$.Characteristic observed that growth in height and weight of the plants. From the observations that can be generated at high observation, treatment 1 had a higher than treatment 2. On the last day treatment plant with treatment 1 had a 99.8 mm high while treatment 2 has height of 90.7 mm. Observation on severe treatment 1 has a heavier weight than treatment 2. Plants with treatment 1 have a weight on the last day of 0.15665 grams while in plants with treatment 2 weights are 0.11365 grams.

Effects of Special Mouth Care with an Aroma Solution on Oral Status and Oral Cavity Microorganism Growth in Elderly Stroke Patients (아로마 용액을 이용한 구강간호가 뇌졸중 노인의 구강상태와 구강 세균 집락에 미치는 효과)

  • Purpose: This study was conducted to examine the effect of oral care with an aroma solution on oral status and oral cavity microorganism growth in elderly patients with stroke. Methods: A non-equivalent control group, with a pretest-posttest design was used in this study. The participants were assigned to the experimental group (n=30) that received oral care with an aroma solution or the control group (n=31) that received 0.9% saline solution. To identify the effect of the experimental treatments, objective/subjective assessments of oral status and oral cavity microorganism growth were performed using the oral assessment guide, oral perception guide, and oral swab culture. Data were analyzed using Chi-square test, Fisher's exact test, and t-test with the SPSS version 21.0 program. Results: The objective oral status was significantly lower in the experimental group than in the control group (t= -3.64, p<.001). There was no significant difference between the subjective oral status of the experimental group and control groups (t= -1.24, p=.109). Oral microorganism growth was significantly lower in the experimental group than in the control group (t= -7.39, p<.001). Conclusion: These findings indicate that special mouth care using an aroma solution could be an effective oral health nursing intervention for elderly patients with stroke.

Comparative Study on Growth of Spoilage Microorganisms in Mungbean and Soybean Sprout (숙주나물과 콩나물에 대한 부패성 미생물의 증식 양상 비교)

  • This study was conducted to investigate effect of mungbean sprout and soybean sprout against the growth of spoilage microorganism (Bacillus subtilis, Bacillus cereus, Pseudomonas fluorescens, and Listeria monocytogenes). After blanching at different conditions, the viable cells of the spoilage microorganisms were increased with storage time, but the viable cells were almost same after treating with either mungbean sprout or soybean sprout. The viable cell population in minimal broth treated with filtrate of mungbean sprout was higher than that of soybean sprout after 24 hr. However, the growth of spoilage microorganism in filtrate of mungbean sprout and soybean sprout was depending upon strain type. During incubation for 72 hr at $25^{\circ}C$, the color change of mungbean sprout was appeared moderately, but in soybean sprout it was appeared clearly at 24 hr. These results indicate that the effect of mungbean sprout and soybean sprout against growth of spoilage microorganism was not different.

Isolation of Microorganism with HIgh Productivity and Cultivation Optimization for Lactic Acid Production (고생산성 젖산생성균 분리 및 배양 최적화)

  • In order to screen microorganism producing lactic acid with high productivity from nature, we used a medium containing 100 g/l glucose and selected several microorganisms producing more than 80 g/l L-lactic acid. We investigated their physiological characteristics and compared them. The best microorganism was identified as Lactobacillus casei subsp. rhamnosus. The optimum pH for growth and production of lactic acid was 6.0 and this strain showed the highest growth rate at around 30$\circ$C , but the optimum temperature for lactic acid production was 45$\circ$C . The growth was inhibited proportionally from 50 g/l to 300 g/l of glucose and the maximal cell mass increased according to increasing the concentration of corn steep liquor (CSL) protein up to 30 g/l. In batch fermentation for lactic acid production, we produced 128 g/l L-lactic acid with 20 g/l CSL protein and 150 g/l glucose in 35 hours. In pH-stat fed-batch fermentation, we were able to produce 183 g/l L-lactic acid.

  • This study was carried out to investigate about the effect of organic fertilizers(Pig manure fermentation compost; P, Organic fertilizer; B, F, Compost; D, and microorganism; M) on the growth and yield of the chinese cabbage and lactuca. At the standard applicated-amounts plot of organic fertilizer have appeared the best yield, but another the excess amount plot came into existence damage of fertilizer. However, according to the co-application of microorganism appeared certainly decreasing damage of fertilizers. Also at the same fertilizer and applicated-amounts plots were the higher than an-application of microorganism on the growth and yield. In conclusion, the excess application of organic fertilizer should be occurred fertilizer damages and increased on the growth and yield by use of microorganism.

  • This research aims to utilize lapilli from the Mt. Baekdusan as environmently-friendly construction material. First of all, the neutralizing method for fabricating lapilli-cement-mixed bioblock was examined. And then, by use of the neuralized bioblock with microorganism for water purification, the growth inhibition effect against the pathogenic coliform bacillus was evaluated. The result regarding growth inhibition effect on pathogenic coliform bacillus indicates that the pretreatment condition, which is a concurrent procession with aqueous solution of 10% di-ammonium Phosphate after water curing, led to pH degraded below 10 which was the target value. Therefore it was concluded that the method was effective on bio-block neutralization. The microorganisms purifying water and di-ammonium phosphate were detected through the examination for microorganism existence on the bioblock, therefore it was concluded that the bioblock composed of lapilli and cement is able to be utilized in various structures as an environment friendly construction material.

Toxicity Evaluation of Hazardous Contaminants by Measuring Lag Periods and Specific Growth Rates of a Test Microorganism (미생물의 비성장속도와 지연기의 측정을 통한 유해오염물질의 독성검사)

  • Among 31 water-born microbial strains isolated from various sites in Korea, strain DJ-4 was selected as a test organism for toxicity measurements in that its growth was completely inhibited by the presence of 668.4 mg/L of chloroform and 297.5 mg/L of toluene in the liquid LB medium whereas others did not. It was observed that lag periods and specific growth rates of DJ-4 batch vial cultures were prolonged and decreased, respectively, by phenol, benzene, toluene, ethylbenzene, p-xylene, perchloroethylene, trichloroethylene, and chloroform at the concentrations between 3.6 and 417.8 mg/L. There changes were found to be linear with respect to the concentrations of the toxic compounds. From the first-order regression equations, 50% effective concentrations (EC50${\mu}$ for concentrations of toxic compounds causing 50% decrease of specific growth rates and EC50lag for 50% increase of length of lag periods) were calculated for each compounds. By comparing DJ-4 EC50${\mu}$ values with Daphnia LC50's from a literature for benzene, ethylbenzene, toluene, and trichloroethlyene, it was concluded that microbial specific growth could be a new, fast, and reliable parameter for toxicity tests.

Effects of Eco-Friendly Organic Fertilizer on Growth and Yield of Angelica gigas Nakai (친환경 유기질 비료 시용이 참당귀의 생육과 수량에 미치는 영향)

  • This experiment was carried out to investigate the effect of several organic compost on the growth and root yield of Angelica gigas Nakai with organic cultivation. After fertilizing the soil with organic fertilizer, the dry weight of liming fertilizer showed a slow change, while microorganism fertilizer decreased about 29% until 20 day after fertilizing. At 110 days after fertilizing, microorganism fertilizer decreased more than liming fertilizer. Liming fertilizer decomposed slowly, while microorganism fertilizer decomposed early on rapidly but gently after 20 days in decomposed rate of organic fertilizer. Dried root yields per 10a of A. gigas were not significance between 277.6kg in conventional fertilizer and 277.7kg, 280.5kg in N 1.5, N 2.0 times of microorganism fertilizer. Decursin and decursinol angelate contents in A. gigas were 9.08 ~ 9.07% from N 1.0 and N 1.5 times in liming fertilizer, and 7.94 ~ 8.12% from N 1.5 times and N 2.0 times in microorganism fertilizer, compared to 7.31% of conventional treatment.
