유체이송 기술은 마이크로 유체시스템 개발에서 핵심문제로 인식되고 있다. 최근 명(2014)은 외부동력을 사용하지 않고 액적을 이동시킬 수 있는 새로운 개념을 제안하고, 초기에 반원통형 형상을 가지는 가상의 2차원 액적에 대한 수치해석을 통해 이 개념이 성립함을 보였다. 또한 명과 권(2015)은 친수성/소수성 표면위에서 초기 3차원 반구 형상의 실제 물 액적이송의 메커니즘을 시간에 따른 액적형상과 액적 내부의 운동에너지, 중력에너지, 표면자유에너지 및 압력에너지의 수치해석 결과를 통해 규명하였다. 본 연구는 새로운 개념을 확립시키기 위해 초기 구형액적에 대한 3차원 수치해석을 수행하고, 액적이송의 메커니즘을 모세관력 힘의 불균형 관점에서 액적 형상과 다양한 에너지의 수치해석 결과를 통해 규명하였다.
중합효소 연쇄반응(PCR)을 수행하려면 세포 용해(cell lysis)와 DNA추출(DNA purification)과정이 포함된 시료 전처리 과정을 거쳐야 한다. 종래의 시료 전처리 과정은 계면활성제와 같은 세포용해 버퍼를 이용하거나 열 또는 전기적 방법으로 세포막 파열을 유도하여 세포벽을 깬 후에 잔여물 처리과정을 거쳐 DNA를 추출하게 된다. 본 연구에서는 마이크로 비드와 PDMS 기둥을 이용한 필터가 있는 시료 전처리용 바이오칩을 설계 및 제작하였다. 또한 제작된 바이오칩을 사용하여 $80^{\circ}C$에서 2분간 세포용해를 수행하고 DNA를 추출하였다. 칩에서 전처리과정을 거친 시료내의 DNA농도와 순도를 측정하고 DNA PCR과 겔 전기영동을 통해 시료 전처리용 바이오칩의 성능을 평가하였다.
This paper presents a paper-based concentration gradient chip to analyze colorimetric glucose assay. The paper-based concentration gradient chip was fabricated through a wax patterning technique that can design the fluidic channel by selectively printing hydrophobic and hydrophilic areas. Afterwards, glucose and dilution solutions were loaded into the inlet of a concentration gradient chip and each solution was then mixed sequentially at mixing channel. Finally, concentration gradient was formed at each outlet of the chip. To measure the glucose concentration of the solution in outlets, we conducted colorimetric glucose assay with fixed concentration of glucose solution (0, 5, 10, 15 and 20 mM) and obtained normalized intensity. Subsequently, glucose concentrations of the outlets were calculated by substituting the normalized intensity to linear regression function based on the normalized intensity of fixed glucose concentration. Finally, the concentration gradient of glucose was formed on the chip with the result of colorimetric assay. The concentration gradient paper chip has the potential to accurately analyze unknown glucose concentration.

Recently, three-dimensional (3D) printing of biological tissues and organ has become an attractive interdisciplinary research topic that combines a broad range of fields including engineering, biomaterials science, cell biology, physics, and medicine. The 3D bioprinting can be used to produce complex tissue engineering scaffolds based on computer designs obtained from patient-specific anatomical data. It is a powerful tool for building structures by printing cells together with matrix materials and biochemical factors in spatially predefined positions within confined 3D structures. In the field of the 3D bioprinting, three major categories of the 3D bioprinting include the stereolithography-based, inkjet-based, and dispensing-based bioprinting. Some of them have made significant process. Each technique has its own advantages and limitations. Compared with non-biological printing, the 3D bioprinting should consider additional complexities: biocompatibility, degradability of printing materials, cell types, cell growth, cell viability, and cell proliferation factors. Numerous 3D bioprinting technologies have been proposed, and some of them have been making great progress in printing several tissues including multilayered skin, cartilaginous structures, bone, vasculature even heart and liver. This review summarizes basic principles and key aspects of some frequently utilized printing technologies, and introduces current challenges, and prospects in the 3D bioprinting.
Droplet in miniaturized microfluidic systems have received much focused attention recently. In this work, electrical charging phenomenon of a conducting water droplet on the electrode under the dc electric field is studied and using this phenomenon droplet actuation method for microreactor applications is experimentally demonstrated. To find effects of key factors, the effects of electric field, medium viscosity, and droplet size are investigated. A scaling law of charging for the conducting droplet is derived from the experimental results. Unlike the case of a perfect conductor, the estimated amount of electrical charge ($Q_{est}$) of a water droplet is proportional to the 1.59 power of the droplet radius (R) and the 1.33 power of the electric field strength (E). (For a spherical perfect conductor, Q is proportional to R2 and E.) It is thought that the differences are mainly due to incomplete charging of a water droplet resulted from the combined effect of electrochemical reaction at electrode and the relatively low conductivity of water. Using this phenomenon, we demonstrate the transport of the charged droplet and fusion of two oppositely-charged droplets. When electric field is subjected sequentially on the electrode, the charged droplet is transported on the electrode. For the visualization of fusion of charged droplets, the precipitation reaction is used. When subjected to a DC voltage, two droplets charged are moving and merging toward each other due to the Coulombic force and chemical reaction is simultaneously occurred by coalescence of droplets. It may be due to the interchange effect of charge. It is shown that the droplet can be used for microreactor where transporting, merging etc. of reagents constitute unit operation.
본 연구에서는 계면활성제를 첨가한 사각 PDMS(polydimethylsiloxane) 마이크로 채널에서의 모세관 흐름에 의한 충전 길이를 예측하기 위한 모델들을 제안하였다. 일반적으로 PDMS 의 소수성 특성 때문에 모세관 힘만을 이용한 PDMS 마이크로 채널에서의 물의 이동에는 어려움이 따르게 된다. 따라서 본 연구에서는 계면활성제를 첨가하여 친수성을 가지는 PDMS 표면을 제작하고, 이 표면에서의 물의 접촉각 변화 측정 및 단순 모델을 수립하였다. 또한 친수성 PDMS 마이크로 채널에서의 모세관 힘에 따른 충전 길이를 예측하기 위해서 Washburn 방정식을 바탕으로 한 단순 모델을 수립하였다. 그 결과 유체의 충전 길이는 접촉각의 초기변화속도의 영향을 받는 것을 확인할 수 있었다. 이를 바탕으로 대표적인 세가지 경우에 대한 모델을 제안하였으며, 이들은 MIMIC(MIcroMolding In Capillaries)과 같은 마이크로 유체 기반의 생산공정의 설계와 개발에 유용하게 적용될 수 있을 것이다.
알지네이트 하이드로 젤은 해조류에서 추출되는 천연 고분자인 알지네이트가 칼슘 또는 마그네슘 양이온과 이온가교(Ioninc cross linking)를 형성할 때 알지네이트의 고분자 구조가 칼슘, 마그네슘 양이온을 감싸면서 형성되는 고분자이다. 알지네이트 하이드로 젤은 높은 생체적합성(Biocompatibility)으로 인해 세포 재생을 위한 조직공학 및 재생의학, 약물전달 등의 제약 관련 분야에 광범위하게 적용될 수 있는 물질로 많은 연구가 이루어지고 있다. 본 연구에서는 마이크로 플루이딕 칩을 이용하여 알지네이트 튜브를 제조하였다. 먼저 유동 포커싱 방식(flow focussing)을 유도할 수 있는 PDMS(Polydimethylsiloxane) 마이크로 플루이딕 칩을 제조하였다. 마이크로 플루이딕 칩은 CNC(Computer Numeric Control) milling machine을 이용한 template를 만들고 NOA mold를 이용하여 최종 PDMS 칩을 제작하였다. 튜브를 만들기 위한 마이크로 채널은 내부 채널 ($200{\times}200um$), 중간 채널 ($200{\times}200um$) 및 외부 채널 ($200{\times}200um$)로 구성되며 내부, 중간, 외부의 유체가 합류하는 수집채널은 폭 500 um, 깊이 200 um로 구성되었다. 운반체로는 5%의 acetic acid를 함유한 mineral oil를 이용하였으며 내부의 core flow는 $H_2O$로 하였다. 중간 유체인 2% 알지네이트 프리폴리머는 칼슘 이온의 존재 하에서 젤화 과정이 매우 빠르기 때문에 마이크로 채널 내부에서의 반응을 제어하고 막힘을 방지하기 위해 수용성 복합 칼슘-에틸렌 디아민 테트라 아세트산 (EDTA)을 사용하였다. 본 마이크로 플루이딕 칩에 각각의 유체를 이동시켰을 때, 운반체인 oil phase의 수소이온은 중간 유체인 알지네이트 프리폴리머와의 계면을 통해 확산되어 Ca-EDTA 복합체로부터 칼슘 양이온의 방출을 유발하게 된다. 방출된 칼슘 양이온은 알지네이트 고분자와의 이온 가교를 통해 알지네이트 하이드로 젤을 형성하여, 각 유체의 flow에 따라 알지네이트 튜브를 쉽고 빠르게 제조 가능하였다. 본 연구에서 제조된 알지네이트 튜브는 인체 내 장기간 약물 전달을 위한 나노섬유로 활용하거나 인공혈관을 구성하는 extracellular matrix로 활용될 잠재력을 가지고 있어 추후 활발한 연구개발이 진행될 예정이다.
Micro actuator의 동력원으로 박테리아 주화성을 기반으로 한 편모박테리아 운동은 널리 연구되고 있다. 본 연구에서는 마이크로입자 추적유속계($\mu$-PTV)를 이용하여 박테리아 주화성에 의해 추진되는 형광입자의 움직임을 분석하였다. 일반적으로 활발한 운동성을 지니고 있는 편모 박테리아 중 Serratia marcescens가 배양액속에서 형광 폴리스티렌 미세입자 표면에 자발적으로 붙게 된다. 박테리아가 부착된 미세입자를 고형화된 화학적 유인물질 L-aspartate가 담겨져 있는 유체 속으로 주입하고, 시간에 따라 입자들이 서서히 L-aspartate가 높은 농도를 가지는 구역으로 이동하는 것을 관찰하였다. 본 연구의 결과로 편모박테리아가 micro actuator의 효율적인 동력원 개발에 적용될 수 있는 가능성을 제시하였다.
Microfluidics can provide unique experimental tools to visualize the development of neural structures within a microscale device, which is followed by guidance of neurite growth in the axonal isolation compartment. We utilized microfluidics technology to monitor the differentiation and migration of neural cells derived from human embryonic stem cells (hESCs). We co-cultured hESCs with PA6 stromal cells, and isolated neural rosette-like structures, which subsequently formed neurospheres in suspension culture. Tuj1-positive neural cells, but not nestin-positive neural precursor cells (NPCs), were able to enter the microfluidics grooves (microchannels), suggesting that neural cell-migratory capacity was dependent upon neuronal differentiation stage. We also showed that bundles of axons formed and extended into the microchannels. Taken together, these results demonstrated that microfluidics technology can provide useful tools to study neurite outgrowth and axon guidance of neural cells, which are derived from human embryonic stem cells.
