Microcystin Detection Characteristics of Fluorescence Immunochromatography and High Performance Liquid Chromatography

  • Different detection characteristics of fluorescence immunochromatography method and high performance liquid chromatography (HPLC) method for the analysis of cyanobacterial toxins were studied. In particular, low and high limits of detection, detection time and reproducibility and detectable microcystin species were compared when fluorescence immunochromatography method and high performance liquid chromatography method were applied for the detection of microcystin (MC), a cyclic peptide toxin of the freshwater cyanobacterium Microcystis aeruginosa. A Fluorescence immunochromatography assay system has the unique advantages of short detection time and low detection limit, and high performance liquid chromatography detection method has the strong advantage of individual quantifications of several species of microcystins.

Degradation of Taste-and-Odor Compounds and Toxins in Water Supply Source Using Plasma (플라즈마를 이용한 상수원 이취미 및 독성물질 분해 연구)

  • This study investigated the degradation of taste-and-odor compounds and toxins using dielectric barrier discharge plasma. The degradation of taste-and-odor compounds was conducted on geosmin and 2-methyl isoborneol (2-MIB), and the toxins investigated were microcystin-LR (MC-LR), microcystin-RR (MC-RR), microcystin-YR (MC-YR) and anatoxin-a. Largely depending on the type of gas fed to the plasma reactor, the degradation efficiencies of the taste-and-odor compounds decreased in order of oxygen (100%) > dry air (96%) > nitrogen (5%) for geosmin and in order of oxygen (100%) > dry air (94%) > nitrogen (2%) for 2-MIB on the basis of 150 s reaction time. This result suggests that the oxidative reactive species generated during plasma treatment, especially long-lived ozone, are mainly responsible for the degradation of these compounds. When using oxygen as the feed gas, geosmin and 2-MIB were totally degraded within 150 s, microcystins within 10 s, and anatoxin-a within 30 s. It was found that the taste-and-odor compounds and toxins were degraded more rapidly in real lake water than in distilled water.

Characteristics of Cyanobacterial Occurrence and Concentration Distribution of Cyanotoxins in Hoeya Reservoir (회야호의 남조류 발생 특성과 남조류 독소의 농도분포특성)

  • Algae blooms have soared recently in the lakes across the nation due to eutrophication. Blue-green algae cause unpleasant scene, produce taste and odor problem, and hinder processes in drinking water treatment. Algae toxicity monitoring has been strengthened, because the damages of wild lives and livestocks by algal toxins have been reported. Investigation on the characteristics of cyanobacterial occurrence and concentration distribution of Cyanotoxins in Hoeya reservoir have been conducted. Physical and chemical influences of water environment on cyanobacterial occurrences have also been studied. Movements of four species of Microcystin and five species of Anatoxin-a among Cyanotoxins were observed by LC-MS/MS analysis. Microcystis spp. among the cyanobacteria have mainly dominated in the Hoeya reservoir during the investigating period. The density of cyanobacteria were positively correlated with temperature and pH of water. Highest concentrations of Microcystin-LR and Microcystin-RR were $0.424{\mu}g/L$ and $0.117{\mu}g/L$ at the sampling points. Total concentration of Cyanotoxins in water coming into the water treatment plant was $0.182{\mu}g/L$, and they were not detected in treated water.

Effect of Zooplankton Exposures on the Biomass and Intracellular Microcystin in Microcystis aeruginosa and Planktothrix agadhii (동물플랑크톤 노출 강도가 유해남조 Microcystis aeruginosa와 Planktothrix agardhii의 생체량 및 세포내 microcystin함량변화에 미치는 영향)

  • This study was to evaluate microcystin production by two strains of cyanobacteria (Microcystis aeruginosa and Planktothrix agardhii) in response to three different levels of direct (0,4,8 inds.) or indirect (0,25, 50% of zooplankton culture media filtrate) exposures to zooplankton (Daphnia magna and Moina macrocopa). The cell biomass and intracellular microcystin (MC) were measured everyday. The survival rates of zooplankton were evaluated for daily intervals for the direct exposure. The intracellular MC produced peaked on the day 3 or 4, and then decreased over the both exposure experiment. In the direct experiment, the MC values were significantly different among the control and zooplankton treatments (ZT; repeated measures-ANOVA: P< 0.039). The MC contents of P. agardhii strain (No.204) were significantly higher (Tukey test, P< 0.082) in ZT2 (8 inds.) than in ZT2 (4 inds.). On the peak day, the intracellular MC exposed to both zooplanktons was significantly higher than the control (One-way ANOVA, P< 0.021). Higher zooplankton survivals were observed in the M. aeruginosa strain (No. 111) rather than in high toxic P. agardhii strain. In the indirect experiment, the intracellular MC of the M. aeruginosa strain was significantly different among the control and zooplankton culture media filtrate (ZCMF)treatments (rm-ANOVA: P<0.004), The MC exposed ZCMF2 (50%) were significantly higher than in ZCMFI (25%: Tukey test, P< 0.025) for both strains. This study strongly supports the induced-defensive MC production of potentially toxic cyanobacteria in response to the presence of zooplankton.

Inhibition of Growth and Microcystin Toxicity, and Characterization of Algicidal Substances from Lactobacillus graminis against Microcystis aeruginosa (Microcystis aeruginosa에 대한 Lactobacillus graminis의 성장 억제능, microcystin 분해 및 살조 물질의 특성)

  • For several decades, lactic acid bacterium (Lactobacillus graminis: LAB) has been generally recognized as safe. To develop the pan-environmental bio-control agent, algicidal activity of the live LAB cell and its culture filtrate (CF) was examined against Microcystis aeruginosa. LAB cells perfectly lysed M. aeruginosa within 3 days, while the CF had a less effect than the live cells, approximately 78% inhibition of algal growth during a same culture period. The concentration of microcystin in alone culture of M. aeruginosa was $7.1{\mu}gL^{-1}$, but gradually increased and leach $158.5{\mu}gL^{-1}$ on 10 days. However, LAB cells clearly decreased the microcystin by $10.3{\mu}gL^{-1}$ in the same period, approximately 93.5%. CF of LAB showed a strong algicidal activity over 75% between pH 2-7, 91.3% by the treatment of proteinase K, 87.8% by below 3 kDa in particle size, and 75.3% by heat treatment, respectively. Of five solvents, fractions of CF passed through solvents diethyl ether and ethyl acetate showed an obvious algicidal activity in the algal-lawn test. Among 5 fractions purified by silica-gel TLC plate, two spots showed a most strong removal activity on M. aeruginosa. Another analysis of GC indicate that CF contained six representative fatty acids. Even though most of these substance have been known as an anti-algal substance against M. aeruginosa, oleic acid is the most effective. These results suggested that the culture filtrate or specific substances, like a fatty acids, in comparison with live L. graminis can be a successful and eco-friendly agent to control Microcystis bloom.

Effects of Biological Control Agent Algicidal Bacterium on the Phytoplankton Community and Microcystin-LR Contents in a Mesocosm Experiment (살조세균 적용이 식물플랑크톤 군집과 조류독소 분포에 미치는 영향)

  • Biological control agents (BCA; algicidal bacterium Xantobacter autotrophycus) plus casitone media, strongly changed physicochemical variables, standing crops of phytoplankton and microcystin-LR phytoplankton in 100-L mesocosm constructed in a small hexagonal pond (3.5 m ${\times}$ 5 m). No M. aeruginosa showed by 8 days, and 60% of total standing crops of phytoplanktons were decreased by the BCA treatment. BCA treatment also induced a strong decline of cellular extracted microcystin-LR (MCLR) and a remarkable increase of dissolved MCLR with the decrement in standing crops of cyanobacteria. In addition, BCA strongly increased all nutrients, but new outbreak of phytoplanktons hardly showed in the experimental mesocosm. The field application of BCA to controling the cyanobacterial bloom in large lakes and reservoirs is not relevant due to high concentration of nutrients and toxins. Thus, a further study is needed to diminish the adverse effects after BCA treatment for water quality preservation.

Seasonal Variations of Cyanobacterial Toxins (microcystins) in Yeongchun Reservoir (영천호에서 남조류 독소(microcystins)의 계절적 변동)

  • Seasonal variations of cyanobacterial toxins (microcystins) in Yeongehun reservoir were studied from March to December, 2007. High level of microcystins production was shown during the late autumn and winter seasons. Total microcystins concentration increased sharply when the dominant species changed from Anabaena sp. to Microcystis aeruginosa. Microcystins-RR and -YR were the main components of the microcystins, whereas microcystin-LR was detected in small quantities. Especially, large amounts of microcystin-LR were detected when standing crops of M. aeruginosa increased exponentially. Total microcystins concentration showed a negative correlation with water temperature. However, total microcystins were lowly correlated with other environmental factors except for water temperature. As a result, this study clearly demonstrated that M. aeruginosa was the main producer of microcystins in Yeongchun reservoir.

Analysis of Sequence Diversity of mcyA Gene Involved in Microcystin Synthesis in Korean Reservoirs (국내 호수에서 Microcystins의 생합성에 관여하는 mcyA 유전자의 염기서열 다양성 분석)

  • The sequence diversity of mcyA gene involved in synthesis of microcystins was analyzed in Microcystis spp. isolated from the Korean reservoirs and in the environmental samples taken from the Daechung, Chungju, Yongdam, Soyang, and Euam Reservoirs at the cyanobacterial blooming season. It was estimated that the sequences of mcyA gene in the isolated Microcystis spp. were much conserved when compared with those in GenBank database. A few kinds of clones were dominant in the investigated environmental samples, occupying 87 to 100% of total clones. No mcyA sequences originated from Anabaena spp. or Planktothrix spp. was found. These results indicated that microcystins are produced mainly by Microcystis spp. and the sequences of mcyA genes are much conserved in the investigated Korean reservoirs.

Changes of Toxicological Factors in Rats Short-Term Treated with Two Feeding Methods at Low Level of Microcystin-LR (저농도의 Microcystin-LR를 단기간 처리한 흰쥐에서 투여방법에 따른 독성 지표의 변화)

  • To investigate the effects of low dosed microcystin -LR (MCLR) on the initial stage of subacute hepatotoxicity in vivo, male Sprague-Dawley rats after weanling were divided in four groups. The orally treated group (OG) was administered orally by 1 $\mu\textrm{g}$/kg B.W. at an interval of three days. The free group (FG) has taken the drinking water including 1 $\mu\textrm{g}$/L freely and the control group (CC) was only treated with 0.9% saline solution All groups were treated for a period of 3 weeks. There was a significant correlation in body growth rate between OG and FC and especially, a deterioration of the growth of spleen was observed in the FG after 5 days. The protein levels were also decreased in OG and FG after 9 days. Level of total fat was increased to the 9th day but again decreased up to the initial level. High hemolysis of the isolated erythrocytes occurred only in OG. Activities of ${\gamma}$-G7 of 0G and FG were higher twice-fold than CG, but the values of OG were already higher at the first treatment day. No significant change in aspartate aminotransferase (AST) activity was shown in all groups, but the activity of alanine aminotransferase (ALT) was slightly increased at the beginning state. There were much similarities in the results of OG and FG. except the growth inhibition of spleen in FG. It may be concluded that long -term effects of the low doses of mycrocystins in animals including human being can lead to serious health problems, especially to liver and spleen.
