김밥 조리조건에 따른 미생물 품질 평가와 중요관리점의 관찰 (Microbiological Quality Assessment of Kimbap According to Preparation and Cooking Condition and Identification of Critical Control Points in the Processes)

  • 본 연구에서는 보통의 조리조건과 위생적 조리조건에서 김밥을 조리하였을 때에, 각 조리조건하에서의 식자재(원재료),조리자의 손, 조리환경(조리기구 및 조리실),그리고 완성된 김밥의 미생물 품질을 살펴보고 이를 토대로 김밥조리과정 중의 중요관리점(CCPs)을 관찰하였다. 김밥과 그 원재료,조리자의 손, 그리고 조리기구(칼, 도마 및 김발)의 표준평판균, 대장균군, 분변성대장균군, 대장균, 황색포도상구균, 살모넬라균 등의 미생물을 식품공전에 준하여 측정하였으며 조리실의 미생물 오염도를 평가하였다. 모든 시료에서 대장균, 황색포도상구균 및 살모넬라균은 검출되지 않았으며 표준평판균, 대장균군 및 분변성대장균군은 보통의 조리조건에서보다 위생적 조건에서 현저하게 낮았다 김밥 원재료 중 비가열재료에서는 가열재료보다 표준평판균 및 대장균군수가 많이 검출되었다. 두 조건에서 조리실의 공중낙하균은 모두 양호한 수준이었다. 위생적 조리조건에서 만든 김밥은 보통의 조리조건에서 만든 김밥 에 비하여 표준평판균과 대장균군이 1/100수준이었으나 아직 분변성대장균군이 검출되었다. 따라서 비가열재료, 조리기구 및 조리자의 손의 미생물 오염, 그리고 조리 중의 교차오염이 김밥 미생물 오염의 주요 근원으로 지적되며, 비가열재료의 취급, 조리기구의 세척과 소독 및 조리자의 손씻기 단계가 CCPs로 제시된다.

이산화염소 가스 훈증 처리에 따른 곶감의 미생물학적 변화 및 품질특성 (Microbiological changes and quality characteristics of dried persimmon by chlorine dioxide gas fumigation treatment)

  • 본 연구는 저장 및 유통과정 중 곶감의 품질 변화를 방지하기 위한 방법으로 기존에 사용되어 왔던 유황 훈증 대체 물질로 이산화염소 훈증 방법의 효과를 확인하고자 하였다. 곶감을 이산화염소 가스 훈증 농도(0, 15, 30, 45 ppm) 및 시간(0, 15, 30, 45 min)에 따라 처리한 후 상온에서 보관하면서 미생물학적 변화, 물성 및 색도를 관찰하였다. 이산화염소 가스 훈증 처리 후 총 호기성 세균과 효모 및 곰팡이 수는 대조구와 비교하였을 때 미생물 수가 감소하였으며, 총 호기성 세균의 저해 효과는 저장기간 동안 지속되었고, 효모 및 곰팡이 수의 경우에도 총 호기성 세균과 유사한 경향을 나타내었다. 곶감의 물성 및 색도는 이산화염소 가스 훈증 농도 및 시간에 의해 영향을 받지 않는 것으로 나타났다. 이산화염소 가스 훈증 처리는 곶감의 저장 중 미생물학적 안전성 확보를 위한 처리 기술로 활용될 수 있을 것으로 기대된다.

절임방법에 따른 깍두기의 관능적 및 미생물학적 특성 (Effects of Salting Methods on the Sensory and Microbiological Properties of Kakdugi)

  • The effects of salting methods on sensory and microbiological properties of Kakdugi were evaluated during fermentation at 10$\^{C}$ for up to 52 days. Kakdugi samples were prepared by 4 different salting methods at the final salt concentration about 1.5%, which was appropriate for organoleptic quality. The salting methods for radish cubes(2cm size) of Kakdugi included; 1) Treatment S-1: spraying dry salt uniformly onto the radish cubes, at the concentration of about 1.5%(w/w) and cured for 1 hr, 2) Treatment S-5: spraying dry salt uniformly onto the radish cubes, at the concentration of about 1.2%(w/w) and cured for 5 hr, 3) Treatment B-1: brining radish cubes in a 8.5%(w/v) salt solution and cured for 1 hr, 4) Treatment B-5: brining radish cubes in a 4.0%(w/v) salt solution and cured for 5 hr. During the early stage of fermentation, sensory test showed higher scores in the overall acceptability of Kakdugi prepared with salting methods S-1 and S-5 than those with B-1 and B-5. However, the trend of acceptability has been reversed by the treatments B-1 and, more notably, by B-5 at the later stage of fermentation. The counts of lactic acid bacteria increased remarkably and then decreased gradually after the optimum ripening period. The major lactic acid bacteria isolated and identified from Kakdugi were Pediococcus, Streptococcus, Leuconostoc, and Lactobacillus.

Effect of Fat Level and the Ripening Time on Quality Traits of Fermented Sausages

  • The objective of this study was to investigate the effect of the fat reduction on the physicochemical and microbiological characteristics of fermented sausages during ripening and drying. Low fat fermented sausages were produced with different fat levels (30%, 20%, 10%, and 5%) under ripening conditions and fermented process. Samples from each treatment were taken for physicochemical and microbiological analyses on the 0, 1, 2, 3, 4, 5, 7, 10, 14, and 21st day of ripening. In proximate analysis, the fat reduction in sausages produced an increase in moisture, protein and ash contents during ripening and drying (p<0.05). The weight losses were significantly higher in high fat formulations during the first 4 days, whereas those were higher in low fat ones after 10 days of storage (p<0.05). Fat reduction was responsible for an increase in shear force values after 3 days of storage. The volatile basic nitrogen (VBN) value of the low fat samples was significantly higher (p<0.05). Low fat sausages reduced the extent of lipid oxidation. The lower fat level produced redder sausages. Total plate bacteria and Pseudomonas counts of sausages showed no significant differences. Production of low fat sausages resulted in the physicochemical and microbiological attributes equal to or better than the high fat sausages without negative effects, except only a higher VBN and weight loss.

Effect of Chemical Treatment with Citric Acid or Ozonated Water on Microbial Growth and Polyphenoloxidase Activity in Lettuce and Cabbage

  • Effects of chemical treatment with a citric acid solution or ozonated water on microbiological changes in lettuce and cabbage during storage were studied. Fresh lettuce and cabbage samples were cut into small pieces and treated by soaking in either ozonated water or a citric acid solution. After treatment, populations of total bacteria, yeast and mold, and E. coli were determined. Numbers of microorganisms increased during storage, but ozonated water and citric acid treatments retarded the increase in microbial growth. Among treatments, 1 % citric acid treatment was the most effective in terms of microbiological change and inhibition of polyphenoloxidase (PPO). For lettuce, citric acid treatment decreased the microbial growth overall by 1.5 log CFU/g and inhibited the PPO activity by 80%. These results indicate that chemical-treated lettuce and cabbage retained a better quality than those of the control during storage.

염장미역의 미생물적, 관능적 품질에 대한 감마선 조사의 영향 (Effect of Gamma Irradiation on the Microbiological and Organoleptic Qualities of Salted Sea Mustard (Undaria pinnatifida))

  • 염장미역은 염농도 감소와 저장조건의 완화를 위한 연구의 일환으로서 2~4kGy의 감마선을 조사하고 ${30{\pm}2^{\circ}C와\;10{\pm}1^{\circ}C$에 저장하면서 미생물적, 관능적 품질을 평가한 결과는 다음과 같다. 염장미역의 변질에 관련된 원인 미생물은 호염성 세균과 효모로 확인되었으며, 2~4kGy의 감마선 조사는 초기의 미생물 수준을 1~2 log orders 감소시켰으나 저장 기간 중 증식에는 뚜렷한 영향이 없었다. 관능적 품질에서도 감마선 조사에 의한 미역조직의 연화현상으로 비조사군보다 낮은 기호도를 나타내어 저장 중 품질 안정성 향상에 별 효과가 없는 것으로 나타났다.

초등학교 급식소에서의 HACCP 적용을 위한 계절별 환경미생물학적 위해분석 (The Seasonal Microbiological Quality Assessment for Application of HACCP System to the Elementary School Food Service)

  • Foodservice at elementary schools has been provided nation-wide. It is predictable that foodborne diseases would increase continuously. Formation of a counterplan is urgently needed. This study was designed to identify the stage which contains the critical control points (CCPs) for the microbiological management of HACCP (Hazard Analysis Critical Control Point) at the foodservice provided at elementary schools. Foodservice places at four elementary schools in Seoul were sampled and the overall hygiene of cooking, utensils and equipment, employees, and environment by season were examined. The results showed that the number of bacteria in overall samples was increased and that E. coli, Salmonella and Staphylococcus by biochemical test emerged in more diverse samples in summer than in spring. Particularly, the number of aerial bacteria in summer was three-fold greater than that in either spring or winter. E. coli 0157 was not detected, although Salmonella was identified by PCR analysis in the meat knives, chopping boards, waste bins and meat dish at elementary school foodservice. According to this data, cross-contamination should be managed in the stage of mixing up the ingredients with improper equipments and insanitary treatments. Thus, the establishment of SSOP (Sanitation Standard Operating Procedures) and GMP (Good Manufacturing Practice) at elementary school foodservice is stringently required, along with sanitation education for workers and employees as CCPs.

The Effect of Vacuum Films on Physicochemical and Microbiological Characteristics of Hanwoo (Korean Native Cattle)

  • The objective of this study was to investigate the physicochemical and microbiological characteristics of round of Hanwoo by vacuum packaging film materials, polyvinylidene chloride (PVDC) and ethylene vinyl alcohol (EVOH). The packaged beef samples were stored in refrigerated conditions (2±1℃) for 12 weeks. Physicochemical analysis with pH, surface color, thiobarbituric acid reactive substances (TBARS) values, and volatile basic nitrogen (VBN) values and microbiological analysis with aerobic plate count (APC) and metagenomic analysis of packaged beef samples were performed. The pH and surface color did not change substantially during the 12 weeks and EVOH-packaged beef tended to be lower than those of PVDC-packaged beef. PVDC- and EVOH-packaged samples showed low TBARS and VBN values below standard limits. APC did not exceed 7 Log CFU/g for both samples during storage. In metagenomic analysis, Firmicutes and Lactobacillaceae were dominant phylum and family of the PVDC- and EVOH-packaged beef. In both packaged samples, Dellaglioa algida was the dominant species during storage, with the notable difference being the presence of Lactococcus piscium. Therefore, this study provided the information on the quality of vacuum-packaged beef according to different vacuum films for long-term refrigerated storage.
