Effects of 13 organic compounds including glucose, fructose, xylose, glutamate, succinate, malate, glycine, lactate, acetate, pyruvate, citrate, formate and cis-aconitate on the oxidation of thiosulfate and the availability of these compounds as the substrate for the respiration by Thiobacillus ocncretivorus, which is known to be an obligated autotroph, were studied. Malate nad glycine at 0.5 per cent concentration nearly doubled the thiosulfate oxidation compared to the control. No other organic substances enhanced the thiosulfate oxidation compared to the control. No other organic substances enhanced the thiosulfate oxidation. Moreover, some 30 to 40 per cent decrease was recorded by fructose, sulfate-salts medium, some 30 to 40 per cent decrease was recorded by fructose, citrate, xylose, malate, flucose, glutamate and succinate. No respiration could occur when formate and pyruvate were supplied as the substrate for respiration. But it was obvious that flucose, fructose, xylose, glutamate, malate, citrate and succinate could be used as the substrate for respiration to some extent, regarding the fact that some increase in respiration rates could be recorded compared to the result from the salts medium, where neither thiosulfate nor orgnic compounds were added. Thus, it was postulated that this organism could possibly be converted into mixotroph or hetrotroph if appropriate conditions could be prepared.
This work was designed to illustrate physiological effects on the elimination of sulfur and its compounds in petroleum using sulfur-reducing bacteria. Deulfurizing bacteria were collected from sewage and soil at several areas in Soul and Ulsan, Korea. Seven supecies of sulfur-reducing microbes isolated were identified as : Pseudomonas marginata, Ps.effusa, Ps. putrefaciens, Ps.pseudcmcnelli, Ps. xanthochlora, Ps.bowlesiae, and Ps.aeruginosa. And some experiments were performed to define the growing characteristics of the Pseudomonads and the results obtained are as follows : 1) Shaking culture method was more effective for the growth of the cells than stagnant culture. 2) Beef peptone medium was better for the growth than other media. 3) Cuprous chloride of 50 ppm and cupper sulfate of 300 ppm treated, respectively, in the medium were effective for the growth. 4) Benzene of toluene of 5,000 ppm and petroleum ether of 50,000 ppm did not show remarkable inhibitory effects on the growth.
In order to improve the removal ability of phosphate, Spheroplast fusions were performed among auxotrophic mutants of Aeromonas hydrophila isolated from waste water, named A13 and A14, Aci37 auxotrophic mutant of Acinetobactercalcoaceticus, and auxotrophic E. coli HR262/pCE27 carring pit gene. Eight fusants obtained from this experiment showed different biochemical characteristics. When the rate of phosphate uptake among fusants (F1-F8) was investigated in Phosphate Uptake Medium (PUM), F8 strain showed the highest rate for phosphate removal, 7 times as much as control after two hours incubation. The role of cations ($Mg^{++}$ ,$Ca^{++}$ , $K^{+}$ in phosphate uptade by F8 was also investigated in PUM without each salt. $K^{+}$ seemed to be crucial. Being compared with phosphate untake rate in PUM, that in PUM without $K^{+}$ was reduced 1.5 times. Therefore, by applying F8 strain and $K^{+}$ in practical environmental system, the increased efficiency in phosphate removal can be derived.
An appropriate amount of samples, collected from three each paddy forest, field and riverside soil near Taegu city, was suspended in sterile water and then diluted in order to isolation of antagonistic to oral cancer. The diluted samples were inoculated on separating medium in the routing spreading method. So, seven hundred and eighteen strains were isolated on HV agar and 220 strains were on methanol medium from soil samples. So, during the screening of anti-oral cancer activity from soil, we isolated microorganisms showing powerful antagonistic activity. Among them, No. 78 strain exhibited the most strongly anti-oral cancer activity. Microbiological properties were investigated by the methods described in the Bergey's Manual of Systematic Bacteriology and experimental methods of identification of actinomycetes by Hamada et al. As a result, a methylotrophic actinomycetes strain No. 79 was estimated as Amycolatopsis sp. based on taxonomic studies.
Forty seven strains of yeast were isolated from traditional Meju and were identified as Saccharomyces spp. (7 strains), Zygosaccharomyces spp. (7 strains), Kluyveromyces spp. and Hansenula spp. (each 5 strains), Rhodotorular spp. (8 strains), Candida spp. (12 strains), Pichia spp. and Debaryomyces spp. from results of their microbiological characteristics. The optimal medium for growth of all the yeasts was YM media and the optimal initial pH of the medium was 6.0. The optimum temperature for growth was 30$circ$ and among them, Sacch. exiguus OE-5, Sacch. cerevisiae OE-16, Sacch. kluyveri C-1 strains were thermotolerant yeasts which could grow at 40$circ$C.
Irradiation with high doses of gamma rays induced the reduction of mycelial weight and anaylase activity, and increased relative amylase activity in surface cultures. Biphase in growth curves was shown in aeration-agitation cultures but the behavior of the first phase of growth could be eliminated by replacing the amylasehydrolysed starch substrates, so that enzyme production was shortened ca. 40 hours and relative amylase activity was increased about 3 times higher before onset of autolysis. In the effect of gibberellin on amylase production, the positive stimulation was appeared to only surface culturs of the liquid medium and the negative effect to shake-cultures in a mutant. Trials of various continuous culture were resulted not only the approalch to the value of amylase activity in surface cultures of liquid medium, but also higher productivity than in batch cultures. The culture-degeneration was observed in two-stage continuous culture, but did not appear in continuous elevation culture.
Twenty one strains of Rhodopseudomonas palustris from 7 different regions in Korea have been isolated and identified on the basis of Bergey's Manual in order to study their regional variation. They can use fructose as carbon source (or hydrogen donor). Capacity to produce molecular hydrogen during photosynthesis was tested using liquid medium, modified by omission of $NH_4Cl$ and addition of L0glutamic acid. As nitrogen source nitrate can be used, and isopropanol can be used as hydrogen donor. their pH ranges are 5.9 - 9.1. Their growth are inhibited in the medium that contains 100 units of penicillin G/ml. as for the growing test in dark aerobic condition, the period needed for adptation is longer than that of Korea strain of Rhodopseudomonas gelatinosa.
In order to increase the productivity of cellulolytic enzymes, medium composition and culture conditions were studied. When cellulose powder (Avicel) supplemented with rice straw was used as carbon source, productivity of ${\beta}-glucosidase$ was increased by about 3 times compared with the runs with only cellulose powder as a carbon source. In this case no negative effects on the production of CMC enzyme activity and filter paper activity was found. For the production of celulolytic enzymes using T. reesei QM 9414, casitone was found to be a good nitrogen source compared with other sources studied, such as peptone, yeast extract, tryptone, and casein. The highest cellulase activity was attained when 0.3% glucose and 0.01% Tween 80 were supplemented to the standard medium of Rese. An adequate oxygen transfer rate was also found to be important to the cellulase fermentation and about 50 mmole of oxygen/liter/hour supported good cellulase biosynthesis during cellulase fermentation.
Putrescine, spermidine, and spermine were found to persent in Methylobacterium extorquens AM1 growing on methanol, succinate, glucose, or nutrient broth as an energy source. Spermidine was found to be a major polyamine in cells growing on methanol or succinate, while putrescine to be the one in nutrient broth-grown cells. The overall content of polyamines in cells growing on glucose was less than that in cells growing on other substrates. Spermine was the most abundant polyamine in glucose-grown cells. Accumulation of polyamines in M. extorquens AM1 was maximal at the mid-exponential or early stationary phase during growth on each substrate. The effect of polyamines added into the medium on the polyamine composition in M. extorquens AM1 was variable. Each polyamine added into the nutrient broth medium was found to increase the amount of the respective polyamine in the cell. Exogeneously added polyamines had no effect on the growth of M. extorquens AM1.
Yeast-form cells of cadmium ion-tolerant Hansenula anomala B-7 were changed to mycelial cells in medium containing more than $400\mu$g/ml of cadmium. Moreover, the mycelial cells were exchanged into clumped cells in a medium containing more than $1,000\mu$g/ml of cadmium. Optimal conditions of mycelial cell formation were achieved in the presence of .$1,000\mu$g/ml of cadmium with shaking cultivation for 7 days. Glucan and mannan contents of the yeast cell wall frown with $1,000\mu$g/ml of cadmium decreased by 10% compared with those grown without cadmium. However, protein and lipid contents increased about 20% respectively. By cadmium, no significant findings in specific amino acid contents were discovered.
