A strain able to grow up m-toluate minimal medium has been isolated after selective enrichment and given the name T81X, which was later identified as pseudomonase putida according to its morphological and biochemical characteristics. After treatment with plasmied specific curing agent, mitomycin C, followed by replica plating on m-toluate and xylene minimal agar plate, T81Xstrain has been shown to harbour a curable plasmid relating to the m-toluate and xylene metabolism. Spontaneous curing frquency of this plasmid was also greatly enhanced by growing on benzoate minimal medium. After then, it was also xylene metabogrowing on benzoate minimal medium. After then, it was found to be conjugally nontransmissible. From the comparative investigation of catechol 1,2-oxygenase and catechol 2,3-oxygenase activities in wild type and cured strain on various growth substrate, it appeared that T81X strain has both of these two enzymes while cured strain has catechol 1,2-oxygenase only. Growing on m-toluate minimal medium T81X strain should carry the genetic information necessary for coding the catechol 1,2-oxygense induced by m-toluate or benzoate, on that curable plasmid.
Mycoplasma pneumoniae was examined for growth characteristics and morphology when cultivated in several media supplemented with a variety of sera and under different atmospheric conditions. different formula of the medium as well as different sources of lot numbers of the serum in the same medium exhibited varying effects on growth rate and adherence. When the organisms were cultivated in SSR-2 medium in a normal atmospheric environment or under a facultative anaerobic condition provided with carbon dioxide, they developed filamentous cells with heavy growth, whereas mainly round-shaped cells were produced under strict anaerobic conditions of hydrogen and carbon dioxide. Both morphologies of the organism were transformed by switching the incubation environments. An inverted phasecontrast microscopy using modified petri dishes was excellent to observe single cells and useful to follow the development of the cells. Growth, turbidity, and pneumoniae colonies developed on a solid medium and produced clear when overlaid with sheep blood agar.
The cultural conditions of the osmophilic red color yeasts (Strain L${1$, $L_2$, $L_3$ and $L_4$) isolated and identified in the previous report were examined and the results obtained were as follows ; 1. The optimum medium for growth of these osmophilic red color yeasts was soy sauce medium. 2. These strains were grown exceedingly well on the medium containing 3 percent of NaCl but somewhat restrained on the medium containing 6% or more. 3. The optimum temperautre for growth of these strains was $25^{\circ}C$ and their lethal temperature was $68^{\circ}C$(treatment for 5 minutes). 4. The optimum pH for growth of these strains was 6.0.
In solid culture of Endomycopsis fibuliger No.55, Eudomuopsis javanensis No.112 and Candida tropicalis No.340, the conditions of enzyme (protease, anylase and cellulase) production and the influence of addition of $(NH_4)_2;SO_4$ were examined, and the results obtained were as follows. 10 Wheat bran medium is found to be the best on the enzyme production in case of simple material. The optimum conditions ; are water content added 100 to 120%, temperature 25 to $80^{\circ}C$ and incubation times 2 to 3 days. 2) The cellulase production was scarely produced in the case of Endomyopsis fibuliger No.55, as well as, the amylase production was scarely producted in the case of Endomycopsis javanensis No.112 and Candida tropicalis No.340. 3) The enzyme production was remarkably increased when 5% of$(NH_4)_2;SO_4$ as inorganic nitrogen sources was admixed to wheat bran. 4) When 5% of $(NH_4)_2;SO_4$ was admixed to medium, the ratio of protein increase was 10.2 to 17.7% in wheat bran medium and 10.6 to 17.9% in sweet potato cake medium.
Selected microbiological and biochemical characteristics of the steeping process for the production of yukwa, a traditional Korean oil-puffed snack made of waxy rice, were investigated during steeping of waxy rice in water for 15 days. The lengthy steeping process was largely predominated by lactic acid bacteria (LAB), particularly, Lactobacillus and Leuconostoc. The predominat type of bacterium isolated was the Y26 strain tentatively identified as Lactobacillus plantarum. The titratable acidity of the steeping medium increased from 0.01 to 1.13%, in parallel with the decrease in pH ranging from 6.3 to 4.2 as the steeping period increased from 0 to 15 days. A high amount of lactic acid and to a much lesser extent, butyric acid, acetic acid, propionic acid, and succinic acid were detected during the steeping process. The amount of reducing sugars in the steeping medium increased from 0.61 to 10.43 mg/mL, whereas sucrose decreased from 0.46 mg% to an undetectable level. Starch degradation products including glucose, maltose and oligosaccharides ranging G3-G7 were not initially noticed, but their content increased during the steeping process until completion. However, no oligosaccharides larger than G8 were detected in the steeping medium. The activities of $\alpha$-amylase, $\beta$-amylase and protease in the steeping medium of waxy rice tended to rise increase with time during the steeping process. From these results, the lengthy steeping process in yukwa production can be characterized as the spontaneous fermentation, dominated by lactic acid bacteria, which is a necessary process for inducing biochemical modification of waxy rice.

The influence of feedstock amino acids, salt, carbon and nitrogen sources on glutathione production by Saccharomyces cerevisiae FF -8 was investigated. Glucose, yeast extract, KH$_2$PO$_4$, and L-cysteine were found to be suitable feedstock. Highest glutathione production was obtained after cultivation with shaking for 72 h in a medium containing glucose 3.0% (w/v), yeast extract 3.0%, KH$_2$PO$_4$ 0.06% and L-cysteine 0.06%. The glutathione concentration achieved using this medium increased 2.27-fold to 204 mg/l compared to YM basal medium.

The present study has been perromed to investigate the optimal conditions for protoplast formation and regeneration of oleandomycin-producing Streptomyces antibioticus (S. antibioticus) KCTC 1081. Mycelia were grown in YME medium containing 0.2% (w/v) glycine and converted into the protoplast by incubating at 35.deg.C for 60 minutes in protoplast buffer (P buffer) containing 4 mg/ml lysozyme. The reversion of protoplasts to the normal filamentous state was examined by the growth on various synthetic agar media. A high reversion rate was obtained by incubating the protoplasts on a hypertonic agar medium containing 20 mM $Mg^{++}$, 5 mM $Ca^{++}$ and 0.3 M sucrose at 28.deg.C for 5 days. From these experiments, we established the improved regeneration medium and a protocol which supports higher and more consistent levels of regeneration of S. antibioticus protoplasts.
