Biogenic Amines Formation and Content in Fermented Soybean Paste (Cheonggukjang) (청국장 중 biogenic amine의 함량 및 생성원인)

  • The potential to produce biogenic amines was investigated in microbial strains isolated from fermented soybean paste, cheonggukjang. The typical levels of 11 biogenic amines, including putrescine, histamine, and tryptamine, were analyzed in commercial cheonggukjang. The tyramine and histamine levels in the samples ranged from 4.2 to 483.1 mg $kg^{-1}$ and 0.2 to $70.3mg\;kg^{-l}$, respectively. A total of 4 microbial strains of Bacillus sp. were isolated from cheonggukjang. The Bacillus sp. were identified as B. amyloliquefaciens, B. subtilis, and B. licheniformis based on phenotypic characteristics, which included using the VITEK system. The screening plate method for detecting amino acid decarboxylase positive microorganisms was performed. The results fer amino decarboxylation were positive, and biogenic amine formation was evaluated by the confirmation of amine-forming capacity.

Anaerobic digestion for food wastewater using HADS Pilot Plant and analysis of microbial community in the digester (HADS Pilot Plant를 이용한 음폐수의 혐기성 소화 및 미생물 군집 변화 분석)

  • We(Hansol EME Co. Ltd.) proceeded anaerobic digestion test for domestic food wastewater applying to two operating method for increasing of OLR. The methods are as follows. One was the rapidity operating method which was increasing the OLR continuously and rapidly and the other was the terraced operating method which was increasing the OLR having adaptation period for each step. As a result of this tests, the ratio of VFA/Alkalinity of the process was very unstable under the rapidity operating method then the volume of produced biogas was dramatically decreased. However the process was shown stable performance under the terraced operating method maintaining the ratio of VFA/Alkalinity less than 0.4. Also, the process was performing the biogas recovery of $0.8Nm^3/kgVS_{rem}/d$ and the VS removal ratio of 85%. T-RFLP analysis about the community of bacteria and methanogen is also conducted to check the change of the microbial community according to the methods of OLR increasing operation. The microbial community was changed by the methods of OLR increasing operation according to the result of T-RFLP analysis. Although the anaerobic digestion test was executed by same pilot plant, the reactivity and the tolerance of microbial community for surrounding environment could be considerably changed by the operating method for the process.

Microbial community analysis of an eco-friendly recirculating aquaculture system for olive flounder (Paralichthys olivaceus) using complex microbial probiotics (복합미생물 프로바이오틱을 이용한 환경친화적 넙치 순환여과양식시스템에서의 미생물군집 분석)

  • This study was conducted to evaluate effects of dietary microbial probiotics on the growth and disease resistance of olive flounder (Paralichthys olivaceus) in a recirculating aquaculture system (RAS), and the effects of the probiotic bioaugmentation on the microbial community structure and water quality. For the analysis, 80 juvenile fish (average weight, $25.7{\pm}7.6g$; average length, $15.2{\pm}1.7cm$) were fed a basal diet containing a commercial microbial product CES-AQ1 (CES; $1{\times}10^9\;CFU/kg$ diet) in an RAS for 8 weeks. Weight gain, the specific growth rate, feed efficiency, and protein efficiency ratio of the fish fed the CES diet in the RAS were 1.5~2.5 times higher than those of fish fed the basal diet alone, or the basal diet containing oxytetracycline (OTC), yeast plus bacterium, or Bacillus subtilis in a still water system. There was no significant difference in the pathogen challenge test between fish fed the OTC diet and fish fed the CES diet in the RAS, suggesting the CES-AQ1 probiotic used in the RAS as a potential replacement for antibiotics. The RAS biofilter maintained the highest microbial diversity and appeared to harbor microbial communities with ammonium oxidation, denitrification, and fish pathogen suppression functions. Ammonia, which is hazardous to fish, was significantly decreased to < 0.5 mg/L in 19 days, indicating the effectiveness of probiotic supplementation to maintain good water quality in RAS. These results suggest that the intestinal microbial communities of fish are stabilized by a probiotic-containing diet (CES) and that bioaugmentation with probiotics may be an eco-friendly and economical supplement for aquaculture of olive flounder, promoting both good water quality and fish health in an RAS.

A Study on the Quantification and Chracterization of Endocrine Disruptor Bisphenoi-A Leaching from Epoxy Resin (에폭시 수지 용출물질에서의 내분비계장애물질 Bisphenol-A의 정량과 용출특성 파악)

  • Bisphenol-A (BPA), a known endocrine disruptor, is a main building block of epoxy resin which is widely used as a coating agent in residential water storage tanks. Therefore, BPA leaching from the epoxy resin may have adverse effects on human health. The possibility of BPA leaching from three epoxy resins were tested with a modified KS D 8502 method at 20, 50, 75 and $100^{\circ}C$ in deionized water and the specified test water, respectively. BPA leached to the test water was identified using GC-MS and quantified with GC-FID after a sequential extraction and concentration. The results showed that BPA leaching has occurred in all three samples tested. The quantify of BPA leaching from unit area of epoxy resin coating was in the range of $10.677{\sim}273.120{\mu}g/m^2$ for sample A, 29.737~1734.045 for sample B and 52.857~548.778 for sample C depending on the test temperature, respectively. In general, the amount of BPA leaching increased as the water temperature increases. This result implies a higher risk of BPA leaching to drinking water during a hot summer season. In addition, microbial growth, measured by colony forming units, in epoxy coated water tanks was higher than that in a stainless steel tank suggesting that compounds leaching from epoxy resin may support the growth of microorganisms in a residential water holding tank.

  • Various methods (OECD 301B, ISO 9439 and ASTM 5864) for biodegradability test of lubricants were reviewed, and a standard procedure was developed. Most lubrication products are released in rivers or sea then is degraded by microbial action in aerobic condition. Most international method are based on $CO_2$ evolution test. Inoculum obtained from a sewage disposal plant and test compound are cultivated in a mineral medium. Organic carbon of the test compound is degraded and oxidized through the enzymatic actions of inoculum, and ultimately mineralized to carbon dioxide. Biodegradability test conditions of lubricant oils were optimized. The highest biodegradability was achieved when the same medium as in ASTM 5864 and inoculum concentration of $10^4{\sim}10^5$ cell/L were used. The optimum standard materials were selected as aniline and sodium acetate. Additionally the effects of inoculum type on microbial growth and biodegradability were examined. Finally the standard operating procedure (SOP) for biodegradability test method was proposed.

Study on the Performances and Microbial Community in the Biofilm Process for Treating Nonpoint Source Pollutants (비점오염물질 처리를 위한 생물막 공정의 운전 및 미생물 군집의 특성)

  • In this study, biofilm process was introduced for treating nonpoint source pollutants. The ceramic media were provided for biofilm growth in the reactors. The packing ratio of ceramic media was 5% and 15(v/v)%, respectively. Thereafter, the reactors were operated intermittently with the different interevent periods such as 0, 5, 10 and 15 days, respectively. The removal efficiencies of COD and NH$_4{^+}$-N were investigated at the different operating conditions such as media packing ratio, temperature, and interevent period. Additionally, Polymerase chain reaction(PCR)-denaturing gel gradient electrophoresis(DGGE) and INT-dehydrogenase activity(DHA) test were conducted to observe the microbial community and activity in the biofilm. Consequently, the interevent period seemed to have no significant influence on the COD removal efficiency. COD was removed within 6$\sim$8 hours at 25$^{\circ}C$ and about 15 hours at 10$^{\circ}C$. DGGE profiles showed that the initial species of microorganisms were changed from seeded activated sludge into the microorganisms detected in sediments. INT-DHA test also showed that the activities of microorgnaisms were not decreased even in the 15 days of interevent period.

Cosmetic Application Using Skin Whitening and Anti-microbial effects of Ethyl Acetate and n-Butanol Fractions from Eruca sativa (Eruca sativa 에칠아세테이트와 부탄올 분획물의 미백 및 항균효과를 이용한 화장품 응용연구)

  • Eruca sativa, called arugula, is a perennial plant in the Brassicaceae family, an edible plant commonly used in Italian cuisine. To study as a cosmetic material application E. sativa was extracted with 70% ethanol (ES). Then ES was fractionated using n-hexane, chloroform, ethyl acetate, n-butyl alcohol and water (EHex, EEA, ECHCl3, EBuOH and EDW). EEA showed mushroom tyrosinase inhibitory activity. ES, EEA and EBuOH showed inhibition of tyrosinase activity. As a result, ES is expected to have skin whitening efficacy. ES was applied to 0.05, 0.1% the toner and emulsion formulation to test the stability. The anti-microbial activity of eight bacteria and fungi including Staphylococcus aureus and Propionibacterium acnes which cause dermatitis and acne was evaluated. EEA showed effects in all of microorganisms. The toner and emulsion containing ES with 0.05, 0.1% were passed in the challenge test. At -20, 4, 25, 55 ℃ and daylight, there was no significant change on pH, viscosity for 4 months. However, emulsions had phase separation phenomenon at 55 ℃, so the base formulation needs improvement. In addition, through the skin penetration test, EEA penetrated 0.058% in 6 hr, predicting the clinical efficacy. This means that E. sativa can contribute whitening agent and the synergistic effect of preservatives.

A Study on the Effects of Fumigation on Excavated Costumes (출토복식유물에 대한 훈증소독의 영향에 관한 연구)

  • This study examines the effects and influences of fumigation on the antimicrobial treatment of excavated dresses in two different museum settings using chemical composite of Methyl Bromide and Ethylene Oxide. The microbial test indicated that the prewashed samples contained Micrococcus, Bacillus, Staphylococcus, and some unidentified fungi and that the total number of microbes decreased after washing. While it was observed that most germs were removed immediately after fumigation, Bacillus was still found and its number even increased. After 5 months, the number of microbes found in Museum A was smaller than Museum B, which shows that Museum A equipped with better environment control facility was superior to Museum. B in terms of exhibition and preservation of dresses against organic damages. In addition, the result indicates that fumigation is an effective measure against the growth and reproduction of microorganisms, considering that the number of microorganisms decreased after fumigation. The color of test sample showed immediate change after fumigation but color difference became stabilized and decreased with the passage of time. The test samples from both museums showed similar patterns.

Studies on the Biodegradation Test Method of Sufactant (계면활성제 생분해도 측정방법에 관한 연구)

  • The biodegradability of some anionic surfactants were investigated using biological oxygen consumption measurement at different temperatures. As test surfactants, soap, alkyl sulfate (AS), alpha olefin sulfonate (AOS), alkyl polyoxyethylene sulfate (AES), linear alkylbezene sulfonate(LAS), microbial surfactants such as sophorose lipid (sopholipid) and spiculisporic acid (S-acid), were used. The test solution were incubated at $5^{\circ}C$, $18^{\circ}C$ and $32^{\circ}C$, respectively. The comparative rates of biodegradation were in accordance with the results obtained from the surface tension measurement and methylene blue method. The results of comparative blodegradabilities of the surfactants were as follows; soap, AS>AES>AOS>LAS at $18^{\circ}C$ and $32^{\circ}C$. However, at$ 5^{\circ}C$, the biodegradation rate of soap was better than other surfactants. Considering the actual environment of the river, it was concluded that the biological oxygen consumption rate method at lower temperature was more practical than the current method such as methylene blue assay with adapted shaking flask culture at $25^{\circ}C$

  • The available period of oral pharmaceutical liquid preparations was decided according to the study of the stability of unopened preparations. But if one reuses the drug after opening the sealed cap, the major components of the drug could change in quality. In addition, there isn`t any accurate information about the available period of opened oral pharmaceutical liquid preparations. In this study, a long term test, an accelerated test and a microbial limit test are run with C (pseudoephedrine and triprolidine), D (ibuprofen) that are marketed and used frequently. (omitted)
