전기장하 토양 내에서 미생물 이동은 주로 전기영동과 전기삼투에 의해 일어나며 미생물의 이동속도와 유속에 대한 전기영동의 공헌도가 전기삼투보다 대체로 높게 나타났다. Pentadecane-오염토양에 대해 동전기 생물학적복원을 실시한 결과 토양내 미생물 농도는 전기영동과 전기삼투가 함께 작용하여 양극과 음극의 인접 토양에서 동시에 증가하였으며 초반 공정이후에는 미생물의 표면전하특성과 양극의 산소발생에 의하여 미생물 농도가 양극, 중간, 음극의 순서로 나타났다. 하지만 미생물의 양방향 이동으로 토양의 모든 위치에서 오염물이 균일하게 제거될 수 있었다. 미생물의 전기적 이동을 이용한 동전기 생물학적복원은 기존의 생물학적복원의 단점인 늦은 분해속도와 낮은 제거효율의 단점을 극복할 수 있었다.

Barley stripe mosaic virus (BSMV) induces massive actin filament thickening at the infection front of infected Nicotiana benthamiana leaves. To determine the mechanisms leading to actin remodeling, fluorescent protein fusions of the BSMV triple gene block (TGB) proteins were coexpressed in cells with the actin marker DsRed: Talin. TGB ectopic expression experiments revealed that TGB3 is a major elicitor of filament thickening, that TGB2 resulted in formation of intermediate DsRed:Talin filaments, and that TGB1 alone had no obvious effects on actin filament structure. Latrunculin B (LatB) treat-ments retarded BSMV cell-to-cell movement, disrupted actin filament organization, and dramatically decreased the proportion of paired TGB3 foci appearing at the cell wall (CW). BSMV infection of transgenic plants tagged with GFP-KDEL exhibited membrane proliferation and vesicle formation that were especially evident around the nucleus. Similar membrane proliferation occurred in plants expressing TGB2 and/or TGB3, and DsRed: Talin fluorescence in these plants colocalized with the ER vesicles. TGB3 also associated with the Golgi apparatus and overlapped with cortical vesicles appearing at the cell periphery. Brefeldin A treatments disrupted Golgi and also altered vesicles at the CW, but failed to interfere with TGB CW localization. Our results indicate that actin cytoskeleton interactions are important in BSMV cell-to-cell movement and for CW localization of TGB3.
The purpose of this study is to estimate the movement of microbial contaminant caused by bio-attack using bio-agent such as bacillus anthracis for preventing contaminant diffusion. multizone simulation was carried out in the case of three types of bio-attack scenario in the government building. Simulation results show that severe contaminant diffusion is brought about in all cases of bio-attack scenario in one hour, though pollution boundaries have different mode according to bio-attack scenarios. Simulation results also show that immune building technology such as filter and UVGI technology gives us powerful alternatives to meet the emergent situation caused by unexpected bio-attack.
In this study, the TPH(Total Petroleum Hydrocarbon) contamination and microbial ecological characteristics in petroleum-contaminated site were investigated through the correlation among the vertical TPH contamination distribution of the site, the geochemical characteristics, and the indigenous microbial ecology. The high TPH concentration showed in the vicinity of 3~4 m or less which is thought to be affected by vertical movement due to the impervious clay layer. In addition, the TPH concentration was found to have a positive correlation with Fe2+, TOC concentration, and the number of petroleum-degrading bacteria, and a negative correlation with the microbial community diversity. The microbial community according to the vertical distribution of TPH showed that Proteobacteria and Firmicutes at the phylum level were dominant in this study area as a whole, and they competed with each other. In particular, it was confirmed that the difference in the microbial community was different due to the difference in the degree of vertical TPH contamination. In addition, the genera Acidovorax, Leptolinea, Rugoshibacter, and Smithella appeared dominant in the samples in which TPH was detected, which is considered to be the microorganisms involved in the degradation of TPH in this study area. It is expected that this study can be used as an important data to understand the contamination characteristics and biogeochemical and microbial characteristics of these TPH-contaminated sites.
The electrokinetic bioremediation employing electrolyte circulation method was carried out for the cleanup of phenanthrene-contaminated kaolinite, and microorganism used in the biodegradation of phenanthrene was Sphingomonas sp. 3Y. The electrolyte circulation method supplied ionic nutrientsand the microorganism into soil, and inhibited the significant pH change of soil by increasing the soil buffering capacity by providing phosphate buffer compounds. When the remediation process was conducted without surfactant, the removal efficiency of phenanthrene, at the initial concentration of 200 ppm, was 69% for only 7 days. Higher microbial population and lower phenanthrene concentration were observed in the anode and middle regions of soil specimen than in the cathode region. The higher density of microorganism was because the microbial movement was in the direction of the anode part due to the negative surface charge. When Triton X-100 and APG of 20 g/1 were used to improve the bioavailability of phenanthrene strongly adsorbed onto soil surface, about 90 and 39% of phenanthrene removal were obtained. Consequently, it was confirmed that the microorganism preferred APC to phenanthrene as carbon source and so the removal efficiency with APG decreased less than that without APG.
Interest in application of ultraviolet light technology for primary disinfection of potable water in drinking water treatment plants has increased significantly in recent years. The efficacy of disinfection processes in water purification systems is governed by several key factors, including reactor hydraulics, disinfectant chemistry, and microbial inactivation kinetics. The objective of this work was to develop a computational fluid dynamics(CFD) model to predict velocity fields, mass transport, chlorine decay, and microbial inactivation in a continuous flow reactor. In this paper, It describe the how to design optimal UV disinfection device for ground water, BWT and rainwater. Spring shape instrument silver coated located in inner side of disinfection chamber. It make lead the active flowing movement target water and maximize disinfection performance. To search the optimal design method, it was performed computer simulation with 3D-CFD discrete ordinates model and manufactured prototype. Using proposed design method, performed simulation and proved satisfied performance.
물속에 우라닐이온(${UO_2}^{2+}$) 형태로 존재하는 산화우라늄을 철환원세균인 스와넬라균(Shewanella p.)을 이용하여 제거하는 실험을 수행하였다. 용액상의 우라늄 초기농도는 $50{\mu}M$ 이었으며 미생물과의 반응에 의해 점차 그 농도가 감소하였고, 약 2주 후에 거의 대부분의 우라늄이 제거되었다. 우라늄이 제거된 기작은 대부분 미생물 표면에 대한 흡착, 침전 및 광물화에 의한 것이었다. 투과전자현미경으로 관찰한 결과로는 미생물 표면에 흡착되어 점차 결정화되어가는 우라늄이 큰 광물로 성장하고 여러 미생물개체 및 유기분비물과의 결합을 통해 그 크기가 수 ${\mu}m$ 이상으로 커져가는 것을 확인하였다. 이러한 미생물에 의한 우라늄 광물의 성장 및 결합은 방사성폐기물처분장의 우라늄 거동에 큰 영향을 끼칠 수 있으며, 특별히 본 실험에서 관찰한 생지화학적인 금속환원미생물의 역할에 의해 지하 우라늄의 이동이 상당히 지연되는 효과를 거둘 수 있을 것으로 사료된다.
부산시 영도구의 혁신지구의 인공해수천은 높아진 하상과 조류의 특성으로 인해 물이 순환되지 않고 더구나 주위의 오수가 유입되고 있어서 수질이 나빠지고 악취를 발생하고 있다. 이 문제를 해결하기 위한 방안으로 가장 오염되고 조류이동을 감안한 하천의 지점에 생물증강법을 적용하여 친환경적, 효율적으로 하천을 정화하고자 하였다. 현장에서 활성화된 복합미생물을 가장 오염된 지점(Site 2)에 7~10일 간격으로 투입하여, 수질의 pH, 온도, DO, ORP, SS, COD, T-N, 및 T-P를 측정하였고 또한 하상퇴적토의 COD 및 미생물군집을 분석하였다. 13개월 후 Site 2의 수질 SS, COD, T-N 및 COD (퇴적토)의 처리효율은 각각 51%, 58%, 27% 및 35%으로 나타났으나 T-P는 오히려 47% 증가를 보였다. 대부분의 측정지점에서 황산염환원세균(sulfate reducing bacteria)그룹 (Desulfobacteraceae_uc_s, Desulfobacterales_uc_s, Desulfuromonadaceae_uc_s, Desulfuromonas_g1_uc and Desulfobacter postgatei)과 Anaerolinaeles의 밀도는 대체적으로 생물증강에 의한 정화가 진행될수록 감소하였으며, 반면에 Gamma-proteobacteria (NOR5-6B_s and NOR5-6A_s), Bacteroidales_uc_s, 및 Flavobacteriales_uc_s의 밀도는 증가하는 경향이었다. 상대적으로 COD가 낮고 DO가 높은 청정지점인 St. 4에서는 호기성미생물인 Flavobacteriaceae_uc_s가 우점하였다. 이러한 미생물군은 하천의 정화과정을 추적할 수 있는 지표미생물로 활용될 수 있을 것으로 판단되었다. 생물증강 시행 후의 대표적 시점 퇴적토시료의 미생물군집 alpha diversity 지수(OTUs, Chao1 및 Shannon 지수)는 시행 전에 비해 감소하는 경향을 보였으며, 또한 beta diversity 분석기법(fast unifrac 분석)으로 분석한 결과 정화 전이나 초기에 비해서 정화가 진행될수록 전반적으로 시료에 무관하게 미생물군집의 유사성을 보여 생물증강이 현장 토착 미생물의 군집구조를 변화시키고 있음을 확인하였다. 이러한 사실로 보아 본 복합미생물에 의한 현장 생물증강법은 brine water 및 담수로 이루어진 오염된 하천을 환경친화적, 경제적으로 정화할 수 있는 대안으로 판단이 되었다.
Chemotaxis is the directed movement of cells in gradients of signaling molecules, an essential biological process that underlies morhpogenesis during development, and the recruitment of immune cells to sites of infection. Especially, bacterial chemotaxis has utilized as an important prelude to study metabolism, prey-predator relationship, symbiosis, other ecological interactions in microbial communities. Recently, novel analytical formats integrated with microfluidics were introduced to investigate the chemotaxis of the cells with the precise control of chemical gradient and small volume of cells. In this study, we present a method to detect bacterial chemotaxis by direct fluidic contacting. The developed fluidic-handling method is driven by capillary force, hydrophobic barrier and a cohesion force between fluids. We have investigated the chemotactic response of E Coli. and Pseudomonas aeruginosa to three kinds of chemoeffectors such as HEPES buffer, peptone and chloroform.
The clinical application of aligners is accompanied by the ageing of the polymer appliances and the attachments used, which may result in inefficiency in reaching the predicted range of tooth movement, and release of compounds and microplastics in the oral cavity as a result of the friction, wear and attrition of the aligner and composite attachment. The purpose of this review is to present the mechanism and effects of in vivo ageing; describe the hydrolytic degradation of aligners and enzymatic degradation of composite attachments; examine the ageing pattern of aligners in vivo, under actual clinical scenarios; and identify a link to the discrepancy between predicted and actual clinical outcome. Lastly, strategies to deal with three potentially critical issues associated with the use of aligners, namely the necessity of weekly renewal, the dissimilar mechanical properties of aligner and attachment resulting in wear and plastic deformation of the aligner, and the development of integuments and biofilms with microbial colonization of the appliance, are discussed.
