The impact of slash and burning on microbial biomass C, N and P in soils of semi-evergreen tropical deciduous forest were studied from February 1999 to January 2000. The experimental sites were located near Moreh town in the Chandel district of Manipur state (India) along the Indo-Myanmar border between 23° 49' N-24°28'N latitude and 93°45'E-94°16'E longitude. Microbial biomass C ranged from 319.50 ㎍ g/sup -1/ 905.50㎍ g/sup -1/ in the slash and burnt site and from 209.50 ㎍ g/sup -1/ to 708.80 ㎍ g/sup -1/ soil in the forest site. Microbial N ranged from 19.30 ㎍ g/sup -1/ to 99.45 ㎍ g/sup -1/ in the slash and burnt site and from 16.08㎍ g/sup -1/ to 88.90 ㎍ g/sup -1/ in the forest site. Microbial P varied from 10.90 ㎍ g/sup -1/ to 32.21 ㎍ g/sup -1/ in the slash and burnt site and from 2.50 ㎍ g/sup -1/ to 17.60 ㎍ g/sup -1/ in the forest site in different months throughout the year. Microbial biomass C, N and P were recorded to be higher in the slash and burnt site compared to the forest site The conversion of forest into slash and burnt site for agriculture - the traditional shilling cultivation practiced by tribal people in the north- eastern India leads to addition of large amount of organic matter in the soil thereby exhibiting higher values of microbial biomass C, N and P in the recent slash and burnt site than that of the forest site. Relationship between the soil moisture, soil organic C and microbial biomass C, N and P were found to be correlated significantly in both the sites.
최근 10년간 미생물생태분석 기반의 연구는 차세대염기서열분석법이 개발된 이래로 지속적으로 증가하고 있다. 장내미생물생태와 건강의 연관성은 미생물 생태학 분야에 있어서 중요한 결과로 여겨지고 있다. 미생물 군집 분석은 주로 16S rRNA 유전자 가변 영역의 염기서열을 통해 분석되지만 이는 미생물의 활성 정보를 제공하지 않는다. 본 연구에서는 cDNA 기반의 미생물 생태분석을 수행하고 DNA 및 cDNA기반의 미생물생태분석 결과를 비교하였다. 두 가지의 서로 다른 접근법이 Butyrate producer와 probiotics와 같이 장내 대사과정에서 중요한 미생물의 abundance 뿐만 아니라 비만 지표로 알려진 Firmicutes 와 Bacteroidetes의 비율에 있어서 차이가 있음을 나타내었다. 따라서, cDNA 기반 미생물 군집은 이전에 수행된 DNA 기반 미생물 군집 분석과 비교하여 장내미생물생태의 역할과 관련된 또 다른 분석 방향성을 제공한다.
본 연구에서는 기능적 특성이 우수한 우유 단백질 소재를 개발하여 식품산업에 응용하고자 우유 ${\alpha}$-La의 영양생리학적 특성에 대하여 분석하였다. 우유 ${\alpha}$-La의 기능성을 향상 시키려는 시도로써 MTGase의 첨가가 이 단백질의 이화학적 및 면역학적 특성에 미치는 영향을 조사하고, 수종의 암세포주의 생육에 대한 억제 효과를 탐색하였으며, ${\alpha}$-La의 구조적 상태에 따라 항암활성의 관련성을 검토하였다. ${\alpha}$-La에 반응한 MTGase에 의해 생성된 중합체는 인체 내 가수분해효소에 의해 분해가 용이한 것으로 확인되었다. 위암 세포주에 ${\alpha}$-La을 10 mg/mL 농도로 처리한 경우 높은 세포독성을 나타냈다. ${\alpha}$-La과 올레산의 복합체인 BAMLET은 간암 세포주에서 ${\alpha}$-La에 비해 더 강한 활성을 보였다. BAMLET은 apo 상태로 상당량의 이차구조를 지니고 있지만, 소수성 중심부는 느슨하게 형성된 MG와 같은 형태를 지니고 있을 것으로 추정되었다. NMR 분석 결과, BAMLET은 1.2 ppm과 0.8 ppm에서 지방산 보조인자인 올레산과 상호 작용하는 것으로 분석되었다.
생균력 안정성이 보장되고 품질이 규격화된 농용 미생물제공급을 위하여 캡슐형 미생물제의 캡슐화 소재로 열수 추출법과 알칼리 추출법을 이용하여 다시마로부터 Na-alginate를 직접 추출하여 겔 형성능과 생균력을 검토하였다. 열수추출 alginate(HWEA)의 경우 5%의 고농도에서 겔 형성이 성공적으로 이루어진데 반해 알칼리추출 alginate(ASA)는 3% 농도에서 $992.1\;{\pm}\;0.2\;g/cm^2$의 강한 겔이 형성되는 특성을 보여 시판되고 있는 alginate(CA) 1.5% 농도의 겔과 유사한 겔 형성능을 나타내었다. 또한 ASA의 경우 추출 수율이 20%로 HWEA 보다 2배 이상 높게 나타났으며 현재 시판되고 있는 Na-alginate(CA)에 비해 비용이 11배 이상 저렴한 것으로 산출되었다. 이상의 결과로부터 ASA를 사용할 경우 값싼 비용으로 추출이 용이하며 저농도에서 겔형성능이 우수하여 최적의 농용 미생물 캡슐소재로 평가되었다. ASA 캡슐제의 생균력을 조사한 결과 81%의 생균력을 나타내어 고가의 CA 캡슐제와 동일한 생균력을 보장 할 수 있음이 입증되었다. 미세 캡슐 내 미생물 생존력을 보다 안정적으로 유지하기 위해 캡슐막의 효과를 나타낼 수 있는 보조제로 starch와 zeolite를 이용하여 생균력 증진효과를 검토한 결과 단일소재 ASA만으로 제조된 캡슐제보다 보조제를 혼합한 캡슐제 경우 세균과 효모는 생균수가 크게 증가되는 효과를 볼 수 있었다. 미생물 혼합 배양액과 상기의 최적 복합 캡슐소재를 혼합하여 캡슐화한 미생물제의 생균력을 측정한 결과 세균은 93%의 높은 생균력을 나타내었고 유산균과 효모의 경우 70% 이상의 생균력을 나타내어 본 연구를 통해 다시마로부터 직접 추출한 ASA가 고가의 시판품 alginate를 대체할 수 있는 농용 미생물 캡슐화 소재로 이용가능할 것으로 판단되었다.
The decomposition of litters of Pinus densiflora and the growth of microbial populations in a calcareous region, Youngwal were compared with those in a noncalcareous region, Sinlim. The decay rate of litter in Pinus densiflora in Youngwal was 0.128 and that in Sinlim was 0.096. The differences in the populations of soil bacteria and total microorganisms between the two regions were signficant at the 5% level, but that of fungi and actinomycetes was not at that level. The differences in the content of calcium and pH value of soil between the two regions were significant at the 1% level. The excessive content of calcium became to increase pH value, in turn the high pH decreased the content of available phosphorus in soil. The vertical distribution of the content of available phsophorus was consistent with that of the populations of fungi and actinomycetes in Youngwal.
The relationships between microbial activity and disturbance level of soil were investigated from 15 parks in Seoul and undisturbed area. The physico-chemical characteristics of soil and dehydrogenase activity(DHA) as an index of soil microbial activity were analysed. There were ranges of 3.84~7.37 in pH, 9.63~40.33% in moisture content, 3.41~21.49% in organic matter, 0.36~0.79g/g in water holding capacity and 0.03~0.53% in total nitrogen investigated sites. DHA values of soil were 8.64~$146.76{\mu}g/g$ in park soil and 545.14~$1, 198.80{\mu}g/g$ in undisurbed area. DHA of park soil with high traffic density and contamination source from human activities was much lower than that of undisturbed area. DHA was positively correlated with moisture content, organic matter, water holding capacity and total nitrogen.
The relationships among the contents of Pb, Zn, Cd, Cu and microbial biomass and activity of soil were investigated in Kyongbu, Yongdong, and Chungbu highways. The heavy metal concentrations were the highest in Kyongbu highway with high traffic density. The levels of these metals in soil in three highway sites were much higher than Mt. Kwank as control site. The highest concentrations of heavy metals were found in the upper layer of soil adjacent to the roadside. Dehydrogenase activity (DHA) and adenosine tri-phosphate (ATP) contents were generally lowest at distance of 0.5 m from the roadside. Simple regression analysis indicated that DHA and ATP contents were highly negatively correlated with Pb and Zn concentrations.
The enzymology of an activated sludge system for a petroleum wastewater purification process was investigated. Leucine-aminopeptidase (L-AMP), ${\beta}$-glucosidase (${\beta}-GLC$), and lipase (LIP) were selected for the study. It was found that more than 81.7% of enzymatic activity was associated with microbial cells in the activated sludge floc. The metabolic response of a mixed microbial population to increased phenol concentration showed that L-AMP activity increased in the activated sludge, whereas activities of ${\beta}-GLC$ and LIP decreased, due to the inhibitory effect of the phenol which varied from 100 mg/l to 500 mg/l.
Global temperatures are steadily increasing, leading to significant changes in microbial diversity and ecology. In the present study, we isolated high-temperature-growing fungi and fungi-like group (Oomycota) strains from freshwater environments of Korea and identified them based on cultural, morphological, and multilocus phylogenetic analyses. As a result, we introduce Saksenaea (Fungi) isolates as a new species, Saksenaea longicolla sp. nov. and record Phytophthora chlamydospora and P. lagoariana (Oomycota) new to Korea. In the growth experiments, they exhibited high-temperature tolerance, which can grow at 35-40 ℃ but become inactive at 4 ℃ and below. This study confirms the presence of high-temperature-tolerant fungi and oomycetes in Korea and suggests that the Korean climate conditions are changing in favor of these species. This indicates that climate warming is altering microbial distributions in freshwater environments.

The past decades have been a golden era during which great tasks were accomplished in the field of microbiology, including food microbiology. In the past, culture-dependent methods have been the primary choice to investigate bacterial diversity. However, using culturein-dependent high-throughput sequencing of 16S rRNA genes has greatly facilitated studies exploring the microbial compositions and dynamics associated with health and diseases. These culture-independent DNA-based studies generate large-scale data sets that describe the microbial composition of a certain niche. Consequently, understanding microbial diversity becomes of greater importance when investigating the composition, function, and dynamics of the microbiota associated with health and diseases. Even though there is no general agreement on which diversity index is the best to use, diversity indices have been used to compare the diversity among samples and between treatments with controls. Tools such as the Shannon-Weaver index and Simpson index can be used to describe population diversity in samples. The purpose of this review is to explain the principles of diversity indices, such as Shannon-Weaver and Simpson, to aid general microbiologists in better understanding bacterial communities. In this review, important questions concerning microbial diversity are addressed. Information from this review should facilitate evidence-based strategies to explore microbial communities.
