Application of Bootstrap Method to Primary Model of Microbial Food Quality Change

  • Bootstrap method, a computer-intensive statistical technique to estimate the distribution of a statistic was applied to deal with uncertainty and variability of the experimental data in stochastic prediction modeling of microbial growth on a chill-stored food. Three different bootstrapping methods for the curve-fitting to the microbial count data were compared in determining the parameters of Baranyi and Roberts growth model: nonlinear regression to static version function with resampling residuals onto all the experimental microbial count data; static version regression onto mean counts at sampling times; dynamic version fitting of differential equations onto the bootstrapped mean counts. All the methods outputted almost same mean values of the parameters with difference in their distribution. Parameter search according to the dynamic form of differential equations resulted in the largest distribution of the model parameters but produced the confidence interval of the predicted microbial count close to those of nonlinear regression of static equation.

The Microbiological Safety Evaluation of Foodservice Facilities and Side Dishes in Elementary Schools and Universities in the Jeolla-do Region

  • The microbiological safety of foodservice facilities and side dishes in 6 elementary schools and 6 universities in the Jeolla-do region was evaluated. The micro-aerosol evaluation of foodservice facilities including the kitchen, refrigerators, and freezers showed a comparatively high level of microbial contamination. In the microbial safety evaluation of cooking utensils and appliances, the total plate counts of serving tables, cutting boards, and food plates were comparatively high, but did not reveal significant counts of coliforms, Staphylococcus aureus, and Escherichia coli were both below the general limit of microbial contamination. The microbiological safety of the cooking utensils and appliances were satisfactory. In the microbiological safety evaluation of side dishes served at these foodservice facilities, microbial counts were generally below the limit of microbial contamination.

  • The purpose of this study was to estimate the microbial and physicochemical quality of some raw vegetables and suggest safer methods of sanitization and storage for foodservice operations. Three sanitization methods were utilized during pre-preparation (tap water, chlorine water and electrolyzed water). Leek and Chicory were monitored as ingredient, with different storage temperatures $(3,\;10^{\circ}C)$ and periods (1, 2, 3, 4 and 7 days). The largest reduction in the microbial counts was shown with the electrolyzed water and for the case before immersion in chlorine water, performing a first washing was more effective in reducing the microbial counts than with no washing. The results showed that the storage temperature, pH, moisture content and microbial loads were important factors affecting the quality of vegetables.

Hygienic effect of modified atmosphere film packaging on ginseng sprout for microbial safety

  • This study evaluates the microbial safety of ginseng sprouts packaged in moss and a modified atmosphere (MA) film within Styrofoam boxes. Ginseng sprout samples were stored at 4℃ for seven days, and the total fungi and aerobic bacteria counts, relative humidity, and moisture content were measured at 0, 1, 3, 5, and 7 days. During the storage period, both packaging treatments caused an increase in the total fungi and aerobic bacteria counts. However, by the seventh day, the ginseng sprouts packaged in the MA film demonstrated significantly lower counts of total fungi (3.03 log CFU/g) and aerobic bacteria (7.32 log CFU/g) than those in moss (3.66 and 7.63 log CFU/g, respectively). Moss packaging alone resulted in the total fungi count reaching up to 3.36 log CFU/g, with the aerobic bacteria count consistently exceeding 7 log CFU/g, highlighting the importance of hygienic management. Moreover, no significant differences were observed in the moisture content and relative humidity between the MA-film- and moss-packaged groups throughout storage. These findings indicate that the functional MA film is a more hygienic packaging solution for ginseng sprouts than moss.

Microbiological Changes of Marinated Broiler Drumsticks Treated with the Lactoperoxidase System and with or without Thermal Treatment

  • The objective of this study was to evaluate the combined effects of lactoperoxidase system (LPS), thermal treatment and storage time on total microflora and psychrotrophs counts of the marinated broiler drumsticks. A marinade that contained acetic acid (1%) and salt (3%) with pH adjusted to 4 was developed as a standardized marinade. Drumsticks were marinated with various LPS levels, combined with thermal treatment (4 or $58^{\circ}C$ for 2 min), and then stored at $4^{\circ}C$ for 18 h. The microbial counts of the samples were measured after 0, 2, 4 and 7 days of storage for drumsticks held at $4^{\circ}C$. The results indicate that adding LPS at the level of 1 unit ($1{\mu}g/ml$ LP, 5.9 mM KSCN, and 2.5 mM $H_2O_2$) significantly (p<0.05) decreased the total microflora and psychrotrophs counts of the marinated broiler drumsticks. In addition, samples treated with a thermal treatment ($58^{\circ}C$ for 2 min) had significantly (p<0.05) lower microbial counts when compared with the control.

로즈마리를 첨가만 유지 코팅 생약제 환의 품질안정성 (Quality Stability of the Herb Pill Coated with Edible Oils Containing Rosemary Essential Oil)

  • Quality stability of the herb pill coated with edible oils containing rosemary was investigated. Herb pills were made of herb powders such as Panax ginseng, Cinnamomum cassia, Lycium chinense, Zyzyphus jujuba and Zingiber officinale. Rapeseed oil and lubriol were used as edible coating oil. After herb pills coated with edible oils with or without rosemary were stored at $40^{\circ}C$ for 180 days, the microbial viable cell counts and peroxide values(POV) of the herb pill were investigated. After 180 day storage, POVs of herb pills with only rapeseed oil or lubriol were 0.51 and 0.49 meq/kg, respectively. However, when rosemary was added in herb pills the POVs were decreased to 0.30 and 0.39 meq/kg, respectively. The addition of rosemary to the rapeseed oil and lubriol tended to decrease the microbial viable cell counts of the herb pill. The microbial viable cell counts of rapeseed oil and lubriol were 940 and 820CFU/g, respectively after 180 days of storage. However, these levels were suppressed to 720 and 640CFU/g by the resemary addition. On the other hand, the ginseng saponin content of herb pills was not affected by the rosemary addition during storage.

한국형 우육포의 제조공정 중 연화제가 육포품질 및 미생물학적 안전성에 미치는 영향 (Effect of Tenderizer on Physical Quality and Microbial Safety during Korean Beef Jerky Production)

  • The physical quality and microbial safety of Korean beef jerky was evaluated at various steps during its preparation. Microbial counts in raw beef demonstrated mesophillic bacteria at 4.20 Log CFU/g, psychrotrophic bacteria at 3.85 Log CFU/g, anaerobic bacteria at 4.90 Log CFU/g, and yeast and molds at 1.92 Log CFU/g. Spore-forming bacteria and coliforms were not detected in raw beef samples. Spices and spiced meats showed similar trends in microbial counts, demonstrating minimal microbial contamination during these stages of preparation. The final beef jerky product exhibited counts of mesophillic bacteria at 1.15-1.66 Log CFU/g, psychrotrophic bacteria at 1.15-1.66 Log CFU/g, and anaerobic bacteria at 0.81-1.72 Log CFU/g. Spore-forming bacteria, yeast and molds, and coliforms were not detected in beef jerky. Significant differences from added ingredients occurred for instron textural profile analysis traits for hardness. In general, Korean beef jerky with humectant and tenderizer had lower hardness than control (without humectant and tenderizer). Also, the sample added with 0.01% protease from Streptomyces griseus had lower hardness than all samples. All samples had 0.7l to 0.72 water activities, and the color and pH were not shown in significant changes of all samples.

어린잎채소의 생산·가공 공정 중 미생물 오염도 분석 및 총균수 예측모델 개발 (Analysis of Microbial Contamination in Microgreen from Harvesting and Processing Steps and the Development of the Predictive Model for Total Viable Counts)

  • This study was performed to assess the microbiological quality and safety of microgreen sampled from harvesting farms and food processing plant in Korea. The samples were analyzed for total viable counts, coliforms, Enterobacteriaceae, Escherichia coli, Salmonella spp., Listeria monocytogenes, Vibrio parahaemolyticus, Bacillus cereus, and Staphylococcus aureus. Total viable counts were highly contaminated in samples collected from farms (7.7~8.2 log CFU/g) and the final products (5.8~7.8 log CFU/g), respectively. B. cereus was detected less than 100 CFU/g, which was satisfied with Korean standards (<1,000 CFU/g) of fresh-cut produce. A predictive model was developed for the changes of total viable counts in microgreens during storage at 5~35℃. The predictive models were developed using the Baranyi model for the primary model and the square root model for the secondary model. The results obtained in this study can be useful to develop the safety management options along the food chain, including fresh-cut produce storage and distribution.

  • We investigated the microbial populations and viable cell counts of Codonopsis lanceolata from uncultivated and cultivated soil in the suing. The microbial populations and viable cell counts from both types of soils were also investigated simultaneously. It had existed 10 more kinds of microorganisms in uncultivated than those in cultivated. The total viable cell counts of C. laneolata from uncultivated soil, especially in the upper zone, were 9.7$\times$10$^{6}$ CFU/g. However, the C. lanceolata from cultivated soil was 4.2$\times$10$^{6}$ CFU/g. (omitted)

하수에서의 대장균수 확률분포 특성 분석 (Statistics and Probability Distribution of Total Coliforms in Wastewater)

  • Probability distribution of microbes in wastewater is a crucial factor to be determined for microbial risk assessment associated with its reuse. The objective of this study was to investigate probability distribution of an indicator microorganism in wastewater. Daily total coliform counts measured from nationwide wastewater treatment plants in 2010 by the Ministry of Environment were used for statistical analysis. Basic statistics and probability distributions were estimated in the three different spatial scales using the MS Excel software and FARD2006 model. Overall, wastewater from manure and livestock treatment plants demonstrated greater median coliform counts than from sewage and village treatment plants. Generalized logistic (GLO) and 2-parameter Weibull (WBU2) appeared to be the two probability distributions that fitted best for total coliform numbers in wastewater. The study results of microbial statistics and probability distributions would provide useful data for quantitative assessment of microbial risk from agricultural wastewater reuse.
