The objective of this study was to investigate biosorption of organic matter on EPS(Extracellular Polymeric Substances) at different SRT(Sludge Retention Time) in a SBR(Sequencing Batch Reactor) process, which was operated with the following operation steps : Fill-React-Settle-Decant-Idle. The hydraulic retention time was set to be 24 hours. The results obtained from this study showed that the organic removal efficiency per unit microbial biomass decreased with increasing SRT, and the corresponding EPS amount also did. The percent removal of organic by biosorption increased with SRT, and it reached to 53.2% at SRT of 30 days. However, the highest biosorption per microbial biomass(48.6 mgCOD/gVSS) was found at SRT of 2 days. The EPS analysis was performed by measuring TSS, TCOD$_{Cr}$, and TKN. The EPS production per unit microbial biomass was observed to be high at a low SRT. Due to the above result, the floc formation was hindered and therefore poor settlement of sludge resulted in decreasing the COD removal efficiency. It was therefore concluded that the consideration of the system design should include the characteristic of EPS as well as other factors such as SRT, MLSS, and organic loading.
Sustainable farming systems have remarkably increased in Korea due to the increase of environmental concerns caused by the conventional systems. This study was performed on eight-year-old 'Niitaka' pear tree s to investigate the effects of different organic fertilizers on soil chemical and microbial properties and leaf and bud developments. Treatment and applications included 10 kg of rice bran (RB), 10 kg of coffee bran compost (CBC), 10 kg of Chitin incubated compost (Micro keeper$keeper^{(R)}$, CIC), and 30 kg of the mixture of the RB, CBC, and CIC (RCC). Control received 60 g of NPK (16-11-12) chemical fertilizer. The organic fertilizers increased organic matter, nutrient concentrations such as P, K, Ca, and Mg, and microbial biomass in soils compared to the control. RCC had the greatest specific leaf weight (SLW), and the other treatments did not have a significant difference for the SL W. Leaf P and K concentrations were different among the treatments, but they were not affected by the amounts of the soil nutrients. No differences were observed in the leaf Ca and Mg concentrations in the treatments. Organic fertilizers had greater chlorophyll contents than the control but had similar flower bud characteristics to the control.

Environmental baseline information is necessary in order to assess the potential environmental impact of future manganese-nodule mining on the deep-seabed ecosystem. Total ATP (T-ATP), dissolved ATP (D-ATP) and particulate ATP (P-ATP) were measured to estimate total microbial biomass and to elucidate their vertical distribution patterns in the seabed of KODOS (Korea Deep Ocean Study) area, northeast equatorial Pacific Ocean. Within the upper 6 cm depth of sediment, the concentrations of T-ATP, D-ATP and P-ATP ranged from 4.4 to 40.6, from 0.6 to 16.1, and from 3.0 to 29.2 ng/g dry sediment, respectively. Approximately 84% of T-ATP, 81% of D-ATP, and 74% of P-ATP were present within the topmost 2 cm depth of sediment, and the distributions of ATP were well correlated with water content in the sediment. These results indicate that the distribution of total microbial biomass was largely determined by the supply of organic matter from surface water column. Fine-scale vertical variations of ATP were detected within 1-cm thick veneer of the sediment samples collected by multiple corer, while no apparent vertical changes were observed in the box-cored samples. It is evident that the box-core samples were disturbed extensively during sampling, which suggests that the multiple corer is a more appropriate sampling gear for measuring fine-scale vertical distribution pattern of ATP within thin sediment veneer. Overall results suggest that the concentrations of ATP, given their clear changes in vertical distribution pattern within 6 cm depth of sediment, are a suitable environmental baseline parameter in evaluating the variations of benthic microbial biomass that are likely to be caused by deep-seabed mining operation.

To investigate the seasonal distribution and grazing impacts of benthic protozoa in mud flat, their abundance, biomass and grazing rates of benthic protozoa were evaluated at interval of two or three month in Gangwha Island from April, 2002 to April, 2004. Heterotrophic flagellates and ciliates accounted for an average 98% of benthic protozoa biomass. Abundance and carbon biomass of heterotrophic flagellates ranged from $0.2{\times}10^5$ to $5.9{\times}10^5\;cells\;cm{-3}$ and from 0.02 to $9.2\;{\mu}gC\;cm^{-3}$, respectively. Biomass of heterotrophic flagellates was high in spring and fall, and showed no differences among stations. Abundance and biomass of heterotrophic flagellates decreased with the depth and were high within the surface 2.5 m sediment layer. The majority of heterotrophic flagellates were less than $10\;{\mu}m$ in length, and few euglenoid flagellates were larger than $20\;{\mu}m$. Abundance and carbon biomass of ciliates ranged from $0.1{\times}10^3$ to $17.8{\times}10^3\;cells\;cm^{-3}$ and from 0.02 to $9.1\;{\mu}gC\;cm^{-3}$, respectively, and those of ciliates were high in spring and fall. Biomass of ciliates was high within the surface 2.5 mm sediment layer and was higher at st. J2 and st. J3 than st. J1. Among the revealed benthic ciliates, the hypotrichs were the most important group in terms of abundance and biomass. During the sampling periods, an average 66% of benthic protozoa biomass was covered by ciliates. The seasonal distribution of benthic protozoa showed an almost similar fluctuation pattern to that of chlorophyll-a. The results suggest that the biomass of benthic protozoa were mainly controlled by prey abundance, for example, diatoms. Based on ingestion rates, benthic protozoa removed from 13.4 to 40.7% of bacterial production and from 20.1 to 36.4% of primary production. Ingestion rates of benthic protozoa on bacteria and microphytobenthos were high in April. Benthic protozoa in this study area may play a pivotal role in the carbon flow of the benthic microbial food web during spring.
Statistical analysis between sewage plant operating parameters and the effluent quality was performed. We extracted two factors from principal component analysis of operating parameters and effluent quality from each plant. The total variance of 84.7%, 79.2% was explained by the two factors at SB plant and SC plant, respectively. The factors were identified at SB plant in the following order 1) the oxidation of organic material by aeration basin microbe,2) biomass in aeration basin and at SC plant 1) the oxidation of organic material by aeration basin microbe, 2) thickening of acti-vated sludge. These results suggested that the control of microbial composition might be critical on the improvement of the effluent quality and plant operating efficiency because most of the factors were related with microbes.
A microbial consortium, TMC7, was enriched for the degradation of natural lignocellulosic materials under high temperature. TMC7 degraded 79.7% of rice straw during 15 days of incubation at $65^{\circ}C$. Extracellular xylanase was effectively secreted and hemicellulose was mainly degraded in the early stage (first 3 days), whereas primary decomposition of cellulose was observed as of day 3. The optimal temperature and initial pH for extracellular xylanase activity and lignocellulose degradation were $65^{\circ}C$ and between 7.0 and 9.0, respectively. Extracellular xylanase activity was maintained above 80% and 85% over a wide range of temperature ($50-75^{\circ}C$) and pH values (6.0-11.0), respectively. Clostridium likely had the largest contribution to lignocellulose conversion in TMC7 initially, and Geobacillus, Aeribacillus, and Thermoanaerobacterium might have also been involved in the later phase. These results demonstrate the potential practical application of TMC7 for lignocellulosic biomass utilization in the biotechnological industry under hot and alkaline conditions.
Sediment microbial fuel cells (SMFCs) illustrated great potential for powering environmental sensors and bioremediation of sediments. In the present study, array anodes for SMFCs were fabricated with graphite rods as anode material and stainless steel plate as electric current collector to make it inconvenient to in situ settle down and not feasible for large-scale application. The results demonstrated that maximum power of 89.4 ㎼ was obtained from three graphite rods, twice of 43.3 ㎼ for two graphite rods. Electrochemical impedance spectroscopy revealed that three graphite rods resulted in anodic resistance of 61.2 Ω, relative to 76.0 Ω of two graphite rods. It was probably caused by the parallel connection of the graphite rods, as well as more biomass which could reduce the charge transfer resistance of the biofilm anode. The presently designed array configuration possesses the advantages of easy to enlarge the surface area, decrease in anodic resistance because of the parallel connection of each graphite rod, and convenience to berry into sediment by gravity. Therefore, the as prepared array node would be an effective method to fabricate large-scale SMFC and make it easy to in situ applicate in natural sediments.

In a recently developed strategy for in-situ treatment of polychlorinated biphenyls (PCB), bioaugmentation was used in conjunction with a surfactant, sorbitan trioleate, as a carbon source for the degrader bacteria, along with the monoterpene, carvone, and salicylic acid as inducing substrates. Two bacteria were used for soil inoculants, including Arthrobacter sp. st. B1B and Ralstonia eutrophus H850. This methodology achieved 60% degradation of PCBs in Aroclor 1242 after 18 weeks in soils receiving 34 repeated applications of the degrader bacteria. However, an obvious limitation was the requirement for soil mixing after every soil inoculation. In the research reported here, bioaugmentation and biostimulation treatment strategies were modified by using the earthworm, Pheretima hawayana, as a vector for dispersal and mixing of surface-applied PCB-degrading bacteria and soil chemical amendments. Changes in microbial biomass and microbial community structure due to earthworm effects were examined using DNA extraction and PCR-DGGE of 16S rDNA. Results showed that earthworms effectively promoted biodegradation of PCBs in bioaugmented soils to the same extent previously achieved using physical soil mixing, and had a lesser, but significant effect in promoting PCB biodegradation in biostimulated soils treated with carvone and salicylic acid. The effects of earthworms were speculated to involve many interacting factors including increased bacterial transport to lower soil depths, improved soil aeration, and enhanced microbial activity and diversity.
To investigate the effect of long term application of organic matter in upland soils, plots for treatments of NPK, NPK+pig manure compost, rape seed cake, rice straw compost, and green manure were set up. Populations of Bacillus and Gram negative bacteria were high in the plot treated with green manure application, but microbial biomass was increased with chemical fertilizer or pig manure compost in upland soils. Activities of phosphomonoesterase and dehydrogenase were high with organic matter application comparing to control. Cluster patterns analysed using phospholipid fatty acid of plots treated with rice straw and or pig manure compost were clearly different comparing with other treatments. Dominant bacteria in upland soils were Bacillus flexus, B. subtilis and B. megaterium. And the strains isolated from upland soils had amylase, protease and lipase activities.

This study was conducted to understand the growth of attached microbial community in small and medium streams of Gyeonggi-do and the removal efficiency of fluoride with changes in growth. The sites monitoring were carried out for 12 weeks at the P1 and P2(P1 is the point where the discharge water flows. P2 is the downstream point of about 2 km flow), and investigated the water quality and the biomass analysis of the attached microbial community. The growth of the attached microbial community increased up to the 7th investigation, and it was observed that detachment phenomenon occurred. Influencing factors about growth of the attached microbial community were flow rate, velocity, and organic material(T-N & T-P). Meanwhile, fluoride content of attached microbial community also tended to increase until the 7th investigation, and decrease from the 8th. It is assumed that fluoride content has also be reduced with the detachment phenomenon of the attached microbial community. It is expected that this will contribute to the evaluation and management of the use of attached microbial community as a means of stream management. The application of techniques using the attached microbial community should include basic investigation of factors that may affect the growth of the attached microbial community and replacement of the attachment plate according to the time of removal.
