The modern era of microbial genome analysis began in earnest in the 2000s with the generalization of metagenomics and gene sequencing techniques. Studying complex microbial community such as oral cavity and colon by a pure culture is considerably ineffective in terms of cost and time. Therefore, various techniques for genomic analysis have been developed to overcome the limitation of the culture method and to explore microbial communities existing in the natural environment at the gene level. Among these, DNA fingerprinting analysis and microarray chip have been used extensively; however, the most recent method of analysis is metagenomics. The study summarily examined the overview of metagenomics analysis techniques, as well as domestic and foreign studies on disease genomics and cluster analysis related to oral metagenome. The composition of oral bacteria also varies across different individuals, and it would become possible to analyze what change occurs in the human body depending on the activity of bacteria living in the oral cavity and what causality it has with diseases. Identification, isolation, metabolism, and presence of functional genes of microorganisms are being identified for correlation analysis based on oral microbial genome sequencing. For precise diagnosis and treatment of diseases based on microbiome, greater effort is needed for finding not only the causative microorganisms, but also indicators at gene level. Up to now, oral microbial studies have mostly involved metagenomics, but if metatranscriptomic, metaproteomic, and metabolomic approaches can be taken together for assessment of microbial genes and proteins that are expressed under specific conditions, then doing so can be more helpful for gaining comprehensive understanding.
Probability distribution of microbes in wastewater is a crucial factor to be determined for microbial risk assessment associated with its reuse. The objective of this study was to investigate probability distribution of an indicator microorganism in wastewater. Daily total coliform counts measured from nationwide wastewater treatment plants in 2010 by the Ministry of Environment were used for statistical analysis. Basic statistics and probability distributions were estimated in the three different spatial scales using the MS Excel software and FARD2006 model. Overall, wastewater from manure and livestock treatment plants demonstrated greater median coliform counts than from sewage and village treatment plants. Generalized logistic (GLO) and 2-parameter Weibull (WBU2) appeared to be the two probability distributions that fitted best for total coliform numbers in wastewater. The study results of microbial statistics and probability distributions would provide useful data for quantitative assessment of microbial risk from agricultural wastewater reuse.
It was confirmed that malodor connected with an air-conditioner in an automobile is caused by microbial volatile organic compounds (MVOCs) produced by microorganisms and through microorganisms coexisting with each other to form a biofilm on the evaporator surface. A bacterium, Methylobacterium aquaticum, can form a biofilm on the evaporator surface. The biofilm was composed of 45.79% C (Carbon), 42.36% O (Oxygen), 1.85% Na (Sodium), 5.42% Al (Aluminum), 1.39% P (Phosphorus), 0.74% Cl (Chlorine) and 2.45% K (Potassium). This result matches the composition of the biofilm formed on the surface of the used evaporator. It was determined that sulfur compounds (Hydrogen sulfide, Dimethyl sulfide) and organic acids (n-Butyric acid, n-Valeric acid, iso-Valeric acid) in the air which was blown into the automobile were generated by Methylobacterium aquaticum and Aspergillus versicolor, respectively. On the other hand, volatile organic compounds (Toluene, Xylene, 2-Ethylhexanol, 2-Phenyl- 2-propanol, Ethylbenzene) were not found. It is estimated that the reason is due to the low concentration of generated MVOCs or is caused by the change of some MVOCs depending on the nutrients (medium).
Allyl isothiocyanate (AITC), as a fumigant, plays an important role in soil control of nematodes, soil-borne pathogens, and weeds, but its effects on soil microorganisms are unclear. In this study, the effects of AITC on microbial diversity and community composition of Capsicum annuum L. soil were investigated through Illumina high-throughput sequencing. The results showed that microbial diversity and community structure were significantly influenced by AITC. AITC reduced the diversity of soil bacteria, stimulated the diversity of the soil fungal community, and significantly changed the structure of fungal community. AITC decreased the relative abundance of dominant bacteria Planctomycetes, Acinetobacter, Pseudodeganella, and RB41, but increased that of Lysobacter, Sphingomonas, Pseudomonas, Luteimonas, Pseudoxanthomonas, and Bacillus at the genera level, while for fungi, Trichoderma, Neurospora, and Lasiodiplodia decreased significantly and Aspergillus, Cladosporium, Fusarium, Penicillium, and Saccharomyces were higher than the control. The correlation analysis suggested cellulase had a significant correlation with fungal operational taxonomic units and there was a significant correlation between cellulase and fungal diversity, while catalase, cellulose, sucrase, and urease were the major contributors in the shift of the community structure. Our results will provide useful information for the use of AITC in the assessment of environmental and ecological security.
The effect of soil damage on the physicochemical characteristics and activity of the soil microbial community is not well known. This study investigates this relationship by analyzing 11 soil samples collected from various points of soil damage across Gyeonggi-do. Soil damage resulted from forest fires, landslides, and development areas, with their impacts most severe on the topsoil layer (0-30 cm). Dehydrogenase and β-glucosidase activities were notably higher at locations damaged by forest fires compared to other sites. While enzyme activities in soils influenced by landslides and development areas were relatively low, sites with a pollution history exhibited elevated dehydrogenase activity, likely due to past microbial response to the pollution. Additionally, an assessment of carbon substrate usability by soil microorganisms indicated higher substrate availability in areas impacted by forest fires, contrasting with lower availability in landslide and development sites. Statistical analysis revealed a positive correlation between organic content of sand and clay and microbial activity. These findings provide valuable insights into soil damage and associated restoration research, as well as management strategies.
본 연구는 국내에서의 가정내 냉장고 온도에 대한 조사를 수행하여, 현 시점에서의 냉장고에서의 식품보관 온도분포를 추정하였고, 이를 MRA(미생물 위해평가: Microbial risk assessment)의 입력변수로 활용할 수 있도록 적정 확률분포 모델을 제시하였다. 일반적으로 가정내 냉장고에서의 식품 보관온도는 식중독 발생 등에서 있어 중요한 위해인자로 작용하는 것으로 알려져 있다. 조사대상 가구는 총 139가구이었으며, 조사기간은 2009년 5월에서부터 9월까지 data logger를 이용하여 측정하였다. 조사된 냉장고 온도의 평균은 $3.53{\pm}2.96^{\circ}C$로, $5^{\circ}C$ 이상은 23.6%로 나타났다. 수집된 온도자료는 @RISK를 이용, 적합성 검정(GOF: K-S와 AD test)을 수행하여 적정 확률분포모델에 대해 추정하였고, 이중 LogLogistic(-10.407, 13.616, 8.6107)분포 모델이 가장 적절한 국내에서의 가정내 냉장고 식품보관 온도분포 모델로 나타났다. 이 확률분포 모델은 MRA적용에 있어 노출평가에서 입력변수로서 직접적 활용이 가능하다고 할 수 있겠다.
