우리나라 산림의 대표적 인 침엽수인 소나무(Pinus densiflora) 생육 토양의 미생물 군집에 미치는 $CO_2$와 납의 영향을 파악하기 위해, 군집 수준 기질 이용도를 평가하는 CLPP (community level rhysiological profiles) 방법과 165 rDNA PCR-denaturing gradient gel electrophoresis (DGGE) 방법을 활용하여 토양 미생물군집 특성을 조사하였다. 납 오염 토양(500 mg/kg-soil)과 비오염 토양에 2년생 소나무를 식재한 후, $CO_2$ 농도를 380 ppmv 혹은 760 ppmv으로 조절한 배양기에서 3개월간 생육시킨 후 6종류의 토앙 시료의 미생물 군집을 비교 분석하였다. 3개월 후 비오염 토양(CA-3M vs EA-3M)의 기질 이용도는 $CO_2$에 의해 크게 영향을 받지 않았으나, 납오염 토양(OB-3M vs EB-3M)의 경우에는 $CO_2$를 760 ppmv로 높인 토양 시료(EB-3M)의 기질 이용도가 높았다. 각 시료간의 기질 이용도를 이용하여 PCA를 수행한 결과, 각 토양시료의 미생물 군집은 납의 존재 유무에 따라 그룹화 되었다. 비오염 토양(CA-3M vs EA-3M))사이의 DGGE fingerprint 유사성은 56.3%, 납 오염 토양(CB-3M vs EB-3M) 사이의 DGGE fingerprint 유사성은 71.4%였다. 동일 $CO_2$ 농도 시료인 CA-3M과 CB-3M사이의 유사성은 53.3%, EA-3M과 EB-3M사이의 유사성은 35.8%이었다. 이러한 결과는 소나무를 식재한 토양의 세균 군집 구조는 $CO_2$ 농도보다는 납 오염 여부에 의해 더 민감하게 특성화됨을 의미한다.
Lake Shiwha, an artificial lake located near metropolitan Seoul, offers a unique water environment and has been suspected to have high levels of chemical and microbiological contaminations. Lake Shiwha was originally connected to the sea but currently has four major surface water inputs from agricultural, municipal, industrial areas and in addition an occasional inflow from the sea. The objectives of this study are to investigate the relative contribution of microbial contaminants from each of the inflowing surface waters and to identify appropriate microbial indicator organisms in this unique water environment. We measured the levels of microbial contaminations in the four inflowing surface waters. A number of microbial indicator organisms including total coliform (TC), fecal coliform (FC), E. coli, Enterococci, somatic and male-specific coliphages were analyzed. Bacterial indicator microorganisms were detected and quantified by the $Colilert^{(R)},\;Enterolert^{(R)}$ kit. Surface water (50 l) was sampled by $ViroCap^{TM}\;5"$ cartridge filters and analyzed by the single agar layer method for detecting coliphages. The concentrations of TC, FC, E. coli, and Enterococci were 1543 CFU/100 ml${\sim}1.99{\times}10^6$ CFU/100 ml, 0 CFU/100 ml${\sim}202$ CFU/100ml, 0 CFU/100 ml${\sim}1.80{\sim}10^5$ CFU/100ml, 74 CFU/100 ml${\sim}3408$ CFU/100 ml, respectively. The male-specific and somatic coliphages were detected in three different inflowing surface waters. Isolated E. coli and Enterococci strains were further analyzed by 16s rDNA amplification and subsequent phylogenetic analysis from Jungwang-chun, Ansan-chun, Banwol-chun and penstock of inflowing surface water. Our results indicated that the concentrations of different fecal indicator microorganisms might not be highly correlated with each other. Multiple microbial indicator organisms should be used for monitoring microbial contamination and microbial source tracking methods.
BACKGROUND: Although antibiotics have contributed to treatment of bacterial infection, the antibiotic abuse can lead to antibiotic resistant bacteria. Impact of human activities on distribution of antibiotic resistance has been intensively issued and occurrence of antibiotic resistant bacteria in contaminated environments would not be a surprise. Nonetheless, anthropogenic contamination with the dissemination of antibiotic resistance along uncontaminated environments has been less considered. The aim of this study is to investigate antibiotic resistant bacteria across Ulleungdo, known as antibiotic resistance free and anthropogenic pollution free environment in Rep. of Korea. METHODS AND RESULTS: Antibiotic resistant bacteria in coastal seawater of Ulleungdo were investigated in July 2021. Antibiotic susceptibility test using the disk diffusion method was applied with six drugs according to the Clinical and Laboratory Standards Institute (CLSI) guideline. Total 43 bacterial isolates were tested and 20 isolates among of them showed multidrug resistance. Particularly, the number and ratio of resistant bacteria were relatively high in a densely populated area of Ulleungdo. The bacterial communities were investigated using 16S rRNA gene metabarcoding approach in the coastal seawater and soils of Ulleungdo. In the bacterial communities, Firmicutes were selectively distributed only in seawater, suggesting the possibility of anthropogenic contamination in coastal seawater of Ulleungdo. CONCLUSION(S): We found antibiotic resistant bacteria in a populated area of Ulleungdo. The occurrence of antibiotic resistant bacteria in Ulleungdo seems to result from the recent anthropogenic impact. Consistent monitoring of antibiotic resistant bacteria in the uncontaminated environment needs to considered for future risk assessment of antibiotics.

Mycopasma contamination of tissue culture cells easily evades detection and, thus, represents a continous therat to cell biologists. In case where infected cell can not simply be replaced, attempts have to be made to eradicate mycoplacma from the tissue culture cells. A variety of anti-microbial agents have been shown to be toxic to mycoplasma strains ; however, cell associated mycoplasma are often protected from antibiotics at concentrations shown to be effective in vitro. Antibiotic concentrations high enough to be lethal to cell as sociated mycoplasmas frequently are also detrimentrations to the host cells, while moderately increased antibiotic levels tolerated by the host cells often lead to only temporary growth suppression and/or to the emergence of mycoplasma strains resistanct even to high concentrations of the antibiotis applied. Hare, a genetic approach for the elimination of mycoplasma from tissue culture cells that overcomes thens limitations is described. By expression of a selection marker conferring resistance to an otherwise toxic agent, Acholeplasma laidlawii infected BHK-21 cells used as the model system were enabled to temporarily tolerate antibiotic concentrations high enough to be lethal to cell associated mycopalsma while leaving the host cells unharmed. Upon successful mycoplasma eradicated, cultvation of the cured host cells in the absence of the selective agent yielded revertant cell clones that had regained susceptibillity to the toxic agent. Cressation of the selection marker expression was shown to result from the loss of the selection marker DNA, which is a consequence of the fact that the stable and permanent integration of foreign DNA in eucaryotic cell chrosomes is highly inefficient. Thus, the cells were cured from mycoplasma yet remained biochemically unaltered.
본 실험은 경기 북부 일부 지역의 계란에서 미생물 오염도를 측정하여 계란의 위생 상태를 파악하고자 하였다. 조사 자료로서는 대형 마트에 유통 중인 계란 중에서 서로 브랜드가 다른 계란을 대상으로 하였다. 계란의 난각에 존재하는 일반 세균수를 측정하였다. 그 결과 $3.4{\times}10^4cfu/m{\ell}$로 상당히 높은 일반 세균수가 검출되었다. 다음으로는 식중독 지수의 중요한 지표가 되는 대장균 및 계란으로 인한 식중독에서 주요 원인 균으로 작용하는 살모넬라균의 오염 여부를 파악한 결과, 한 브랜드의 계란에서 대장균이 검출되었고, 살모넬라균은 검출되지 않았다. 또한 각 브랜드에 계란에서 검출된 세균을 분리 동정 한 결과, Klebsiella pneumoniae, Pseudomonas mendocina, Alcaligenes xylosoxidans, Alcaligenes faecalis, Enterobacter cloacae 등이 분리되었다. 이 세균들 중 일부는 사람에게 병원성을 나타내거나, 면역력이 약한 노약자나 어린이에게 기회감염 또는 병원 내 감염을 일으키기도 하는 것으로 알려졌다. 계란의 난각에서 분리된 대장균의 병원성 여부를 판정하기 위하여, 대장균의 DNA를 분리하여 PCR을 수행한 결과, 병원성 대장균들에서 특이적으로 나타나는 유전자형을 가지고 있지 않는 것으로 밝혀졌다. 위의 결과들을 종합하여 보면, 시중에 유통중인 계란의 난각에서 비록 살모넬라균과 병원성 대장균은 검출되지 않았지만, 사람에게 감염을 일으킬 수 있는 여러 병원성 세균들이 검출됨으로써 계란의 유통 및 판매에 더욱 철저한 위생관리가 필요할 것으로 사료된다.
수산 가공 부산물인 자숙액을 식품 및 공중보건 소재로 이용하기 위해 자숙액의 위생화를 위한 감마선 조사기술의 이용가능성을 검토하였다. 톳, 참치 및 굴 자숙액을 시료로 하여 1-5 kGy의 감마선을 조사하여 미생물 생존율을 측정하였고 생존미생물을 동정하였다. 굴 자숙액에서는 호기성 미생물이 검출되지 않았으나, 톳 및 참치 자숙액에서는 약 6.4 및 3.1 log CFU/g의 호기성 미생물이 검출되었고, 저장기간이 증가함에 따른 미생물 수에는 유의적인 차이가 없었다. 또한 감마선 조사선량이 증가함에 따라 생균수의 감소와 자숙액에 오염된 미생물의 살균효과를 확인하였으나, 자숙액 종류별로 미생물의 감마선 저항성이 상이하였다. 수산 자숙액에 존재하는 모든 미생물을 사멸시키기 위해서는 톳 자숙액의 경우 10 kGy, 참치자숙액의 경우 20 kGy 이상의 감마선 조사가 필요하다고 생각된다. 자숙액에 오염된 미생물을 동정한 결과 L. plantarum, L. pentosus, Paenibacillus sp., B. subtilis, B. gelatini 등을 포함하여 총 20종의 오염 미생물을 확인할 수 있었다. 톳 자숙액에서는 주로 Lactobacillus 속 들이 주로 검출되었고, 참치 자숙액에서는 Bacillus 속이 주로 검출되었다.
