  • This study was conducted to determine effects of whole and processed corns on attachment characteristics of rumen microbes in Hanwoo. Whole corn(WC) was processed into four different types; ground corn(GC), cracked corn(CC), flaked corn(FC), and soaked corn(SC). After each processed corns was incubated in the rumen of Hanwoo for 12 h and 24 h, attachment characteristics of rumen microbes and morphological changes of feed substrates was examined by utilizing scanning electron microscopy(SEM). In this study, the treatment group of GC and CC showed the highest in the attachment of rumen microbes. On the other hand, microbial attachments to the starch particle of WC and SC were hardly detected. However, with the increase of incubation time the surface of WC and SC was covered by many microbes, resulting in enhancing the surface degradation.

Deproteinized Mulberry Leaf Juice - A New Media for Growth of Microorganisms

  • Mulberry being a foliage crop is grown extensively for feeding of silkworms and are also used for cattle feeding. These loaves are highly nutritious, which contain various mineral elements and bio-molecules such as carbohydrates, proteins, lipids and other essential amino acids, etc. In the present study, deproteinized mulberry leaf juice was used for preparation of the medium for cultivation of various types of microbes. Results revealed that deproteinized mulberry leaf juice medium is best for isolation of fungi, bacteria and actinomycetes and this medium can be substituted with synthetic media, which are haying the costly ingredients for isolation and identification of bacteria, fungi and actinomycetes. Further, this deproteinized mulberry juice medium can also be used in mass multiplication of useful/beneficial microbes to enhance soil microflora to improve soil fertility and to avoid root diseases. Perspective enterprises can take up the mass multiplication/large-scale production of useful microbes such as Trichoderma, Rhizobium, Pseudomonas and Bacillus to use in mulberry and in other agricultural crops using deproteinized mulberry leaf juice.

  • Twenty isolates resistant to seven quinolones were isolated from major rivers in Korea. All isolates had three mutations, Ser83$\rightarrow$Leu and Asp87$\rightarrow$Asn in GyrA and Ser80$\rightarrow$Ile or Ser80$\rightarrow$Arg in ParC and three isolates had an additional mutation Glu84$\rightarrow$Gly or Glu84$\rightarrow$Val in ParC. In addition, a clonal spread was not found in these isolates.

  • Methanol extracts of Perilla and Mugwort were stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activities and inhibitory effect on growth of oral tumor cell of each extract were investigated. Each extracts of Perilla and Mugwort were investigated to anti-microbial effects on oral microbes by means of agar diffusion method and MIC. These results suggest that the hexane extracts of Perilla and Mugwort have antimicrobial activities against S. mutans and potent inhibitory Effect to KB cell growth.

Isolation and Identification of Microbes from AquaMats® for the Treatment of Leachate Originated with the Samsan Dong Landfill in Ulsan Metropolitan City (울산광역시 삼산동 매립장 침출수 처리를 위한 AquaMats®상에서 미생물의 분리 및 동정)

  • The high surface area polymer, AquaMats$^{circledR}$ was used for the leachate purification process originated from the Samsan Dong Landfill in Ulsan Metropolitan City. And then, three species of dominant microbes were isolated and identified from AquaMats$^{circledR}$. Gram staining revealed these microbes to be Gram-negative rod strains: They were identified as Agrobacterium radiobacter, Pseudomonas cepacia, Flavobacterium indologenes. All they showed no growth on media in which the leachate was added alone, but a rapid proliferation rate on media with yeast extract as nutrient requirements.

  • Mycotoxins are secondary metabolites produced by fungi. These toxins pose serious health concerns to animals as well as human beings. Biodegradation of these mycotoxins has been considered as one of the best strategies to decontaminate food and feedstuffs. Biodegradation employs the application of microbes or enzymes to contaminated food and feedstuffs. Ruminants are considered to be resistant to the adverse effects of mycotoxins presumably due to the biodegrading ability of rumen microbes compared to mono-gastric animals. Therefore, rumen microbial source or microbial enzyme could be a great asset in biological detoxification of mycotoxins. Isolation and characterization of pure culture of rumen microorganisms or isolation and cloning of genes encoding mycotoxin-degrading potential would prove to have overall beneficial impact in the food and feed industry.

A Novel Method of Removing Mn(II) Ions from Water by a Combination of New Symbiotic Microbes

  • Typically, manganese (II) ions are incompletely removed from water as $MnO_2$ on increasing the pH of the water to 10. The water then has to be neutralized before it can be used. We propose a new and effective method for removing Mn (II) from water using a new combination of symbiotic microbes consisting of manganese-oxidizing bacteria and filamentous algae. The microbes rapidly oxidize Mn(II) to Mn (IV) at a neutral pH with no organic matter required as a nutrient and $MnO_2$is precipitated immediately. This differs from the use of heterotrophic manganese-oxidizing bacteria where organic nutrients are required. Our results suggest that this method will be useful in developing new systems for removal of manganese(II) ions from industrial and mining wastewater and drinking water. In addition, there are other possibilities such as recycling of dry batteries which are presently discarded without treatment

Mass Spectrometry Imaging of Microbes

  • Microbes influence many aspects of human life from the environment to health, yet evaluating their biological processes at the chemical level can be problematic. Mass spectrometry imaging (MSI) enables direct evaluation of microbial chemical processes at the atomic to molecular levels without destruction of valuable two-dimensional information. MSI is a label-free method that allows multiplex spatiotemporal visualization of atomic- or molecular-level information of microbial and microberelated samples. As a result, microbial MSI has become an important field for both mass spectrometrists and microbiologists. In this review, basic techniques for microbial MSI, such as ionization methods and analyzers, are explored. In addition, we discuss practical applications of microbial MSI and various data-processing techniques.

Fundamental Study on the Removal Properties of Polyethylene Glycols by Mesh Filtration Batch Bio-reactor (메쉬 침지여과분리형 회분식 생물반응조를 이용한 PEG제거의 기초 연구)

  • The removal properties of Polyethylene glycols (PEGs) known as the important group of synthetic polymers of ethylene oxide were examined by the bio-reactor equipped with a mesh filter module. PEG-1000 and PEG-2000 were fairly removed on the basis of TOC, which were 75.1% and 51.6%, respectively. In the case of PEG-20000, the removal efficiency of TOC was less than 15.2% and the favorable acclimation of microbes was not obtained. It was suggested that this system could effectively maintain microbes for the biodegradation of low molecular weight of PEG and TOC removal was significantly influenced by PEG molecular weight.

  • 16 chicken isolates and four clinical isolates of VanB-vanA incongruent vancomycinresistant Enterococcus faecium strains without vanS were isolated in 1999. Pulsed-field gel electrophoresis revealed only a peripheral relationship between the chicken isolates and clinical isolates, but suggested clonal spread in the chicken isolates.
