Growth Profile and Toxigenicity of Bacillus cereus in Ready-to-eat Food Products of Animal Origin

  • The growth profile of Bacillus cereus in ready-to-eat (RTE) food products of animal origin was examined under different temperature and incubation conditions. In sandwiches and Kimbab, B. cereus did not grow or exhibited only minimal growth at 4 and $10^{\circ}C$, but it grew rapidly at ambient temperature. In sandwiches, B. cereus did not grow efficiently at $25^{\circ}C$, however, in ham, the main ingredient of sandwiches, B. cereus growth was observed at the same temperature, with bacterial levels reaching 7.94 Log CFU/g after incubation for 24 h at $25^{\circ}C$. Toxigenicity of B. cereus was observed only at temperatures above $25^{\circ}C$. In Kimbab, B. cereus produced toxin after 9 h at $30^{\circ}C$ and after 12 h at $25^{\circ}C$. Ingredients of sandwiches and Kimbab were collected from 3 different Korean food-processing companies to investigate the source of contamination by B. cereus. Among the 13 tested food items, 6 items including ham were found to be contaminated with B. cereus. Of these ingredients, B. cereus isolates from 3 items produced enterotoxins. None of these isolates harbored the emetic toxin-producing gene. The findings of the present study can be used for risk assessments of food products, including ham and cheese, contaminated with B. cereus.

ANTIMICROBIAL EFFECT OF ESSENTIAL OILS ON ORAL BACTERIA (구강 내 세균에 대한 Essential oil의 항균효과에 관한 연구)

  • Essential oils are mixture of volatile, lipophilic compounds originating from plants. Essential oils have potential biological effects, i.e., antibacterial, antifungal, spasmolytic and antiplasmodial activities and insect-repellent property. In this study, five essential oils, namely R, LG, FR, O, and NM, extracted from various aromatic plants were used to test their antimicrobial activity against the oral microorganisms. The effects of essential oils were investigated against eight important bacteria, Streptococcus mutans (S. mutans), Staphylococcus aureus (S. aureus), Streptococcus sanguis (S. sanguis), Streptococcus anginosus (S. anginosus), Actinobacillus actinomycetemcomitans (A. actinomycetemcomitans), Streptococcus sobrinus (S. sobrinus), Staphylococcus epidermidis (S. epidermidis), and Escherichia coli (E. coli). Essential oils, except NM, effectively inhibited the growth of tested oral pathogenic microorganisms dose-dependently. However, the essential oils didn't show a significant inhibitory effect against E. coli and S. epidermidis. Consequently, these results represented that essential oil-mediated anti-microbial activity was prominent against the oral pathogenic bacteria. For example, minimum bactericidal concentration(MBC) of R, LG, FR oil against A. actinomycetemcomitans was very low as 0.078 mg/mL. In addition, minimum inhibitory concentration (MIC) of R, LG, FR, O oil against S. mutans was low as 0.156 mg/mL in vitro.

Incidence and Control of Coliform Bacteria in the Manufacturing of Commercial Kimchi (상업용 김치 생산과정에서 대장균유사세균의 발생과 억제)

  • As consumption of kimchi has increased, factories have begun to produce this traditional Korean fermented vegetable dish on a large scale. Following the rise in manufacturing, the hygienic conditions under which commercial kimchi is being made have become an issue. We isolated 17 coliform bacteria from commercial kimchi that had not been fully fermented. These bacteria were partially identified as one of seven different species from three genera by 16S rDNA sequence analysis as follows: Enterobacter intermedius, Ent. cloacae, Ent. amnigenus, Klebsiella terrigena, K. ornithinolytica, K. oxytoca, and Hafnia alvei. Lactobacillus paraplantarum KNUC25 has been isolated from over-fermented Chinese cabbage kimchi and its antimicrobial activity reported in the literature. In our study, the KNUC25 strain showed antibacterial activity against isolated coliform bacteria and some pathogenic coliform bacteria through spot-on-the-lawn tests and viable cell tests. Through development and use of a cell-free supernatant of L. paraplantarum KNUC25, we effectively controlled coliform bacteria in commercial kimchi.

Clinical Analysis Of Ventricular Septal Defect (심실중격결손증의 외과적 고찰)

  • Two hundred one patients of ventricular septal defect, which were operated at Seoul National University Hospital, were analysed on clinical background during the period from January, 1975 to December, 1980. The results were as follows: 1. Of the 201 patients, 118 patients were male [58.7%] and 83 patients were female [41.3%]. Their age ranged from 15 months to 40 years, and the mean age was 8.7 years. 40% of the patients were between 4 and 8 years. 2. The most common symptoms showed frequent U RI and exertional dyspnea. 3. On Kirklin`s anatomical classification, type I constituted 26.9%, type II 58.2%, type III 12.4%, and type IV 1.0%. We showed marked increased incidence of type I VSD as compared to Caucasians* 4. 46 cases were associated with other congenital cardiac diseases. They were PDA [13 cases], AI[11 ], ASD[6], PS[10], MI[4], and Double aortic arch [1]. 5. In 128 patients, who had complete hemodynamic data and were not associated with other congenital cardiac diseases, an attempt was made to correlate the EKG findings with the hemodynamic data, and defect size with the hemodynamic data. The children had variable distribution of PA syst. pr. and Rp/Rs. But most of adults had $R_P$/$R_S$of 0.15 or less. As $P_P$/$P_S$increased, the rate of operative complication increased also. 6. When a normal EKG pattern was present, $Q_P$/$Q_S$and $R_P$/$R_S$and $P_P$/$P_S$were relatively low. When EKG findings were LVH pattern, there was diastolic volume overload to left ventricle. As RVH, there was systolic pressure overload to right ventricle. And as BVH, there was mixed pattern of diastolic volume overload to left ventricle and systolic pressure overload to right ventricle. 7. Among patients in defect was less than 1 $cm^2$ per $M^2$ of BSA, $Q_P$/$Q_S$was less than 2:1, and $R_P$/$R_S$less than 0.25, and PAsyst. pr. less than 50 mmHg, and $P_P$/$P_S$was less than 0.5. But patients with the defect greater than 1 $cm^2$ per $M^2$ of BSA had no correlationship between $Q_P$/$Q_S$, $R_P$/$R_S$, PAsyst. pr. and defect size in each other. Most of patients with the defect greater than 2 $cm^2$/$M^2$ BSA, $R_P$/$R_S$was greater than 0.5. 8. Operative mortality rate was 9.5% [19 cases] among 201 patients. And complication rate including mortality rate was 22.9% [46 cases].

Feasibility of near-infrared spectroscopic observation for traditional fermented soybean production (전통 메주 제조과정에 있어서 근적외 모니터링 가능성 조사)

  • In this study, near infrared (NIR) spectroscopy known as a non-destructive analysis technique was applied to investigate peptide cleavage and consequent release of amino acids in soybean lumps as affected by its moisture content and incubation time during fermentation at 25 for 3 weeks. The NIR spectra of the soybean lump semi-dried and soaked in saline water showed that absorption intensity around 1,400 nm originating from hydrogen bonds of water decreased and absorption band shifted to 1,430 nm as moisture content decreased during incubation at 25 for 3 weeks. In addition, absorption around 2,050 nm which was assigned to amino groups increased as incubation time increased. NIR spectra data from 1,000 to 2,250 nm showed higher accuracy in the discriminant analysis between outside and inside parts of fermented soybean lumps than visible spectra result. NIR spectroscopy for the amino acid and moisture contents in traditional fermented soybean lumps showed relatively good accuracy with the multiple correlation coefficient ($R^2$) of 0.91 and 0.81, respectively, and root mean square error of cross validation (RMSECv) of 0.23 and 0.83%, respectively, in partial least square regression (PLSR). These results indicate that NIR spectral observations could be applicable to control the fermentation process for preparation of soybean products.

Analysis of microplastics released from textiles according to filter pore size and fabric weight during washing (세탁 중 세탁물 중량과 여과 기공 크기에 따른 미세플라스틱 분석)

  • This study observed the release of microplastics according to washing weights and filtering conditions, measured microplastic generation rates, fiber lengths, and fiber diameters. This study attempted to present data for the development of filters that decrease microplastic generation. For test samples, polyester piled knit fabric (cut-pile) was selected, which currently has the highest amount of consumption in the clothing industry, but can easily cause marine pollution because of its low biodegradability. For test equipment, a drum washer was used and microplastics were collected using two filter pore sizes, 5 ㎛ and 20-25 ㎛. Microplastic fibers weights and lengths were measured. The results of the experiment showed the following: 1) The release of microplastics differed according to the fabric weights and washing process; 2) washing fabric weights showed a differences in the collection amount according to the filter pore size (5 ㎛, 20-25 ㎛); 3) observations of differences in the lengths of the microplastics that occur during the washing process by filter pore size were made. Fibers with shorter lengths appeared with filter pore sizes of 5㎛ in comparison to filter pore sizes of 20-25㎛. The results from this study on microplastic generation by fabric during washing, demonstrated the following conclusions that can be used to reduce the release of microplastics. First, the release of microplastics according to fabric weights and washing courses are affected by physical force. Therefore, it is necessary to reduce the amount of physical force due to water flow, increase the fabric weight, or wash the material in low temperatures. Second, in the manufacturing of washing machines, microplastic filtration can be promoted or legislatation supporting microplastic filtration can be introduced.

Production and Quality Properties of Capsule Type Meju Prepared with Rhizopus oligosporus (Rhizopus oligosporus를 이용한 캡슐형 메주의 제조 및 품질특성)

  • In order to improve some problems such as contamination of undesirable mold, mycotoxin production and excessive drying on the surface of traditional meju. Control meju without koji and capsule type meju(CM) coated with soybean mixture containing 0.5%, 1% and 2%(w/w) R. oligosporus koji were dried at room temperature (10∼15$^{\circ}C$) for 3 days. Control meju I was fermented in outdoor for 27 days. Control meju IIand CMs were fermented in Korean yellow clay room at 25$^{\circ}C$ for 7 days under 80% relative humidity as first step, and then fermented in outdoor (average temp. 2.7$^{\circ}C$, December) for 20 days as second step. The moisture content of CMs were higher than that of control meju I to the range of 2.88∼7.55%(w/w). pH and titratable acidity in CMs were similar to control group. Amino type nitrogen content in CMs(800.80, 816.0, 901.60 mg%) were 2.2∼2.6 times higher than that in control meju I (347.2 mg%). Reducing sugar content in CMs(2.78∼3.13%) was similar to control meiu I (2.10%) and control meju H(2.31%). Lightness(L) value of control meju I was higher than that of control meju IIand CMs.

  • Eight-membered ring spiro orthocarbonate (C$_{25}H_{20}O_4$, M$_r$ = 384) is monoclinic, space group C2/c, with a = 15.319(4), b = 9.057(3), c = 13.168(3)${\AA}$, ${\beta}$ = 98.53(3)$^{\circ}$, Z = 4, F(000) = 808, T = 290 K, ${\mu}$(Mo-K${\alpha}$) = 0.55 cm$_1$, D$_c$ = 1.36 g/cm$^3$ and D$_m$ = 1.40 g/cm$^3$. The intensity data were collected with Mo-K${\alpha}$ radiation (${\lambda}$ = 0.7107 ${\AA}$) on an automatic four-circle diffractometer with a graphite monochromater. The structure was solved by direct methods and refined by full matrix least-squares methods. The final R value was 0.052 for 1412 observed reflections. The molecule has C$_2$point symmetry. The eight-membered ring has a chair conformation with pseudo-C$_s$ symmetry. The naphthyl ring is planar with the C-C bond lengths being in the range of 1.352∼1.444${\AA}$ and bond angles of 117.2∼123.5$^{\circ}$. The bond lengths of C(1)-C(9), C(8)-C(9) and C(9)-C(10) are somewhat longer than those of the other C-C bonds.

Development of New BNR Process Using Fixed-Biofilm to Retrofit the Existing Sewage Treatment Plant (고정생물막을 이용한 기존 하수처리장의 생물학적 영양염 제커 신공정개발)

  • The object of this study was to develop new BNR process using fixed-biofilm which could be applied to retrofit the existing wastewater treatment plant or to introduce as tertiary treatment plant. To achieve complete denitrification from typical raw sewage in Korea, external carbon source must be supplied because $SCOD_{cr}/T-N(NH_4{^+}-N+NOx-N)$of raw sewage was lower than other countries. In this study, the ratio of $SCOD_{cr}/NH_4{^+}-N$ was 2.49 and the influent $NH_4{^+}$-N concentration during the experimental period was varied from 25 to 37 mg/L. To enhance nitrogen removal from the sewage, the two processes using fixed biofilm were adopted as R-Hanoxic/mid.settler/aerobic/anoxic/ aerobic) and R-2(aerobic/mid.settlerlanoxic/anoxic/aerobic), respectively. In the comparison of $NH_4{^+}$-N, T-N effluent quality and T-N removal efficiency in both processes without external carbon source, R-1 process was better than R-2 process for nitrogen removal from raw sewage. With respect to $SCOD_{cr}$/NOx-N ratio and total nitrogen removal in each anoxic reactor of two processes, R-1's was more effective than R-2's for distributing organic matters of raw sewage. In the both processes using fixed biofilm, the amount of required alkalinity to remove unit $NH_4{^+}$-N were 5.18 and 5.76($g{\cdot}CaCO_3/g{\cdot}NH_4{^+}-N_{removed}$), respectively and were lower than activated sludge BNR process(7.14).

Modelling Pasture-based Automatic Milking System Herds: The Impact of Large Herd on Milk Yield and Economics

  • The aim of this modelling study was to investigate the effect of large herd size (and land areas) on walking distances and milking interval (MI), and their impact on milk yield and economic penalties when 50% of the total diets were provided from home grown feed either as pasture or grazeable complementary forage rotation (CFR) in an automatic milking system (AMS). Twelve scenarios consisting of 3 AMS herds (400, 600, 800 cows), 2 levels of pasture utilisation (current AMS utilisation of 15.0 t dry matter [DM]/ha, termed as 'moderate'; optimum pasture utilisation of 19.7 t DM/ha, termed as 'high') and 2 rates of incorporation of grazeable complementary forage system (CFS: 0, 30%; CFS = 65% farm is CFR and 35% of farm is pasture) were investigated. Walking distances, energy loss due to walking, MI, reduction in milk yield and income loss were calculated for each treatment based on information available in the literature. With moderate pasture utilisation and 0% CFR, increasing the herd size from 400 to 800 cows resulted in an increase in total walking distances between the parlour and the paddock from 3.5 to 6.3 km. Consequently, MI increased from 15.2 to 16.4 h with increased herd size from 400 to 800 cows. High pasture utilisation (allowing for an increased stocking density) reduced the total walking distances up to 1 km, thus reduced the MI by up to 0.5 h compared to the moderate pasture, 800 cow herd combination. The high pasture utilisation combined with 30% of the farm in CFR in the farm reduced the total walking distances by up to 1.7 km and MI by up to 0.8 h compared to the moderate pasture and 800 cow herd combination. For moderate pasture utilisation, increasing the herd size from 400 to 800 cows resulted in more dramatic milk yield penalty as yield increasing from c.f. 2.6 and 5.1 kg/cow/d respectively, which incurred a loss of up to $AU 1.9/cow/d. Milk yield losses of 0.61 kg and 0.25 kg for every km increase in total walking distance (voluntary return trip from parlour to paddock) and every one hour increase in MI, respectively. The high pasture utilisation combined with 30% of the farm in CFR in the farm increased milk yield by up to 1.5 kg/cow/d, thereby reducing loss by up to $0.5/cow/d (c.f. the moderate pasture and 800 cow herd scenario). Thus, it was concluded that the successful integration of grazeable CFS with pasture has the potential to improve financial performance compared to the pasture only, large herd, AMS.
