  • The purpose of this study was to compare the degree of microleakage of Glass-Ionomer root canal sealer possessed several enviable properties with that of the other sealers and to evaluate clinical performance. One hundred twenty single-rooted teeth were used in this experiment. The teeth were cleaned mechanically and immersed for 24 hours in 5.25% sodium hypochlorite and clinical crowns then were removed. After the root canals were instrumented using a step-back technique. one hundred twenty single-rooted teeth were divided into five groups of 24 in each. Group 1 : Tubli-Seal(Kerr Co., MI, U.S.A/ZOE-based), lateral condensation Group 2 : Sealapex(Kerr/Sybron, Romulus, MI/ $Ca(OH)_2$-based), lateral condensation Group 3 : AH 26(De Trey Co., Zurich Switzerland/Resin-based), lateral condensation Group 4 : Ketac-Endo(ESPE GMBH & CO. KG Seefeld:oberbay. Germany/ Glass Ionomer Cement-based), lateral condensation Control group : no sealer. lateral condensation And then. the root canals were obturated by lateral condensation technique with gutta-percha and experimental sealers. The control group were obturated without sealer. The teeth were placed in a vacuum chamber for 15 minutes and immersed 2% methylene blue under vacuum for 15 minutes. The teeth were passively stained for 1 week and 2 weeks and were cleared and evaluated for linear dye leakage using Tool maker's microscope(${\times}200$). The results were as follows: 1. There were statistically significant differences in the degree of dye penetration between the control group and experimental groups(p<0.05). 2. In the experimental groups, Sealapex($1.2871{\pm}0.9180mm$) exhibited the lowest mean value of dye penetration, followed by Ketac-Endo($1.4432{\pm}0.8082mm$), AH 26($1.5030{\pm}0.7752mm$) and Tubli-Seal($1.6458{\pm}1.0292mm$)(p>0.05). 3. There were statistically significant differences in the variation of microleakage between 1 week and 2weeks in Tubli-Seal and Seal apex groups (p<0.05). 4. The degree of dye penetration of all groups were increased as the time elapsed and AH 26 showed the lowest variation(+0.11) and Tubli-Seal(+ 1.03) showed the highest variation (p<0.001).

  • The purpose of this study was to evaluate the microleakage of 5 current dentin bonding systems which are composed of 2 multi-bottle systems(Scotchbond Multi-Purpose, All Bond2) and 3 one-bottle systems(Single bond, One-Step, Prime & Bond). In this in vitro study, class V cavities were prepared on buccal and lingual surfaces of sixty extracted human premolars and molars on cementum margin. The experimental teeth were randomly divided into six groups of 10 samples (20 surfaces) each, Group 1 : Scotchbond Multi-Purpose ; Group 2 : All Bond 2 ; Group 3 : Single Bond ; Group 4 : One-Step ; Group 5 : Prime & Bond ; Group 6 : no bonding agent(control). The bonding agent and composite resin were applied for each group following the manufacturer's instructions. After 500 thermocycling between $5^{\circ}C$ and $55^{\circ}C$, the 60 teeth were placed in 2% Methylene blue dye for 24 hours, then rinsed with tab water. The specimen were embedded in clear resin, then sectioned buccolingually through the center of restoration with a low speed diamond saw. The dye penetration on each of the specimen were then observed with a stereomicroscope at ${\times}20$. The results of study were statistically analyzed using the Student-Newmann-Keul's Methods and the Mann-Whitney Rank Sum Test. The resin/dentin interfaces were examined under Scanning Electron Microscopy. The results of this study were as follows. 1. None of the dentin bonding systems used in this study showed significant difference in leakage values at both the enamel and the dentin margins (P>0.05). 2. In all groups except the control, leakage value seen at the enamel margin was significantly lower than that seen at the dentin margin (P<0.05). 3. Compared to the control group, all the groups treated with dentin bonding systems showed significantly lower leakage value at both enamel and dentin margins (P<0.05). 4. In the SEM view, gaps were observed in the composite resin / dentin interface in group 6 where no dentin bonding agent was used, and in all the other groups (group 1, 2, 3, 4, 5) composite resin, hybrid layer, and dentin were seen to be closely adhering to each other where there were no leakages. Well-developed resin tags 3~100${\mu}m$ in length infiltrated dentinal tubules past the hybrid layer and a hybrid layer 1~5${\mu}m$ thick had developed between the dentinal surface and the composite resin surface.

  • The purpose of this study was to evaluate the effect of heated spreader on the sealing ability of lateral condensation, compared with regular cold spreader. Forty two extracted human teeth with single canal were randomly placed into 3 experimental groups, and four additional teeth were used as positive and negative controls. Each group was prepared with Ni-Ti Profile #40 using step-down technique and obturated with standardized colored gutta-percha cone by standard(cold) lateral condensation technique, warm lateral condensation technique with Endotec and hot spreader soaked in glass bead sterilizer, each with Sealapex sealer. Control groups were not obturated, but prepared. After 2 days in 2% methylene blue, the teeth were invested and made into transparent resin blocks. And then, each block was sectioned horizontally with microtome at 1, 2, 3, 4, 5 mm levels from the apex. The linear extent of dye penetration was examined with stereomicroscope at ${\times}$20 magnification. At each of 5 levels, ratio of the area of gutta-percha was obtained by calculating the area of gutta-percha to the total area of the canal. The data collected were then analyzed statistically using an analysis of variance(ANOVA) and Scheffe test. The results were as follows ; 1. All experimental groups produced the apical microleakage. 2. The mean leakage was 1.57${\pm}$0.76mm for cold spreader group, 0.86${\pm}$0.95mm for Endotec spreader group, and 0.64${\pm}$0.93mm for hot spreader group. The difference between hot spreader group and cold spreader group was statistically significant(p<0.05). 1. At the 1 mm level, the mean ratio of area of gutta-percha was 74.58${\pm}$13.15(%) for cold spreader group, 65.42${\pm}$14.62(%) for Endotec spreader group, and 80.72${\pm}$14.63(%) for hot spreader group. There was statistically significant difference between hot spreader group and Endotec spreader group(p<0.05). 2. At the 2mm level, the mean ratio of area of gutta-percha was 87.86${\pm}$11.22(%) for cold spreader group, 66.55${\pm}$14.02(%) for Endotec spreader group, and 92.93${\pm}$7.24(%) for hot spreader group. There was statistically significant difference between Endotec spreader group and other two spreader groups(p<0.05). 3. At the level 3, 4, 5 mm, there was no statistically significant difference between each group. Within the limits of the results of this experiment, warm lateral condensation technique with hot spreader soaked in a glass bead sterilizer demonstrated favorable apical sealing effect and improved density of gutta-percha mass. Thus, it is thought that this obturation technique is effective for clinical use and beneficial to reduce condensation forces, also economical and easy. • From August to October of 1991, a bacterial disease occurred in cultured flounder, of 0 age Paralichthys olivaceus, in Cheju-do. Typical symptoms of the diseased fish were the hemorrage of the opercle, discoloration of the body, exophthalmus and hemorrhage of the eyes. The causative organism isolated from diseased fish was identified as a $\beta$-hemolytic Streptococcus. This bacterium was not identical with any known Streptococcus species listed in Bergey's Manual of Determinative Bacteriology 8th edition. However there was a close similarity between this species and Streptococcus sp. isolated from flounder and reported by Tashio (1983). The isolated bacteria streptococcus sp. showed very high sensitivity to the SM and EM, however, it resisted to the high concentration of Penicillin. In the pathagenicity analysis, about $1.1{\times}10^9$, $1.1{\times}10^7$, $1.1{\times}10^5$, $1.1{\times}10^3$, $1.1{\times}10^1cells$/100g B.W of isolated bacteria were injected percutaneouly. The results showed that this inoculation size $1.1{\times}10^3cells$/100g B.W was the threshold for the induction of mortality in pathogenicity analysis.

  • This study aimed to investigate the relaxation effects and its underlying mechanisms of Rubus coreanus(RC) extract in contracted rabbit corpus cavernous tissues by phenylephrine(PE) $1{\mu}M$. In order to define the relaxation effects of RC, rabbit corpus cavernous tissues were prepared in $2{\times}2{\times}6mm$ sized strip. The dose-dependent relaxation responses of RC at 0.01-3.0 $mg/m{\ell}$ in contracted strips induced by PE were measured and also observed after endothelial denudation. To analyze the underlying mechanisms of RC-induced relaxation, indomethacin(IM), tetraethylammonium chloride(TEA), $N{\omega}$-nitro-L-arginine (L-NNA), methylene blue(MB) were treated before RC extract infused into precontracted strips induced by PE. To study the effect of RC extract on influx of extracellular $Ca^{2+}$ in corpus cavernous strips, calcium chloride(Ca) 1 mM infused into precontracted strips induced by PE after pretreatment of RC extract in $Ca^{2+}$-free krebs-ringer solution. To investigate cytotoxic activity and nitric oxide(NO) concentration of RC extract on human umbilical vein endothelial cell(HUVEC), cell viability on HUVEC was measured by MTT assay, and NO concentration was measured by Griess reagent system. The cavernous strips were significantly relaxed by RC extract at 1.0 $mg/m{\ell}$, 3.0 $mg/m{\ell}$ and the relaxation responses to RC were inhibited significantly by endothelial disruption. The pretreatment of IM, TEA didn't affect RC extract-induced endothelium-dependent relaxation, but the pretreatment of L-NNA, MB reduced RC extract-induced endothelium-dependent relaxation. When $Ca^{2+}$ was supplied the cavernous strips which were precontracted by PE in a $Ca^{2+}$-free krebs-ringer solution, contraction of strips was increased, but pretreatment of RC inhibited contractile response to $Ca^{2+}$. When RC extract was applicated on HUVEC, NO concentration was increased. Our findings show that RC extract exerts a relaxing effect on corpus cavernosum in part by suppressing influx of extracellular $Ca^{2+}$ through activating the NO-cGMP system.

  • The purpose of this study was to measure the polymerization shrinkage and hygroscopic expansion of resin-based temporary filling materials and to evaluate microleakage at the interface between the materials and cavity wall. Five resin-based temporary filing materials were investigated: Fermit (Vivadent), Quicks (Dentkist), Provifil (Promedica), Spacer (Vericom), Clip (Voco). Caviton (GC) was also included for comparison. Polymerization shrinkage of five resin-based temporary filling materials was measured using the bonded disc method. For the measurement of hygroscopic expansion, the discs of six cured temporary filling materials were immersed in saline and a LVDT displacement sensor was used to measure the expansion for 7 days. For estimating of microleakage, Class I cavities were prepared on 120 extracted human molars and randomly assigned to 6 groups of 20 each. The cavities in each group were filled with six temporary filling materials. All specimens were submitted to 1000 thermocycles, with temperature varying from $5^{\circ}C/55^{\circ}C$. Microleakage was determined using a dye penetration test. The results were as follows: 1. Fermit had significantly less polymerization shrinkage than the other resin-based temporary fill ing materials. Fermit (0.22%) < Spacer (0.38%) < Quicks (0.64%), Provifil (0.67%), Clip (0.67%) 2. Resin-based temporary filling materials showed 0.43-1.1% expansion in 7 days. 3. Fermit showed the greatest leakage, while Quicks exhibited the least leakage. 4. There are no correlation between polymerization shrinkage or hygroscopic expansion and microleakage of resin-based temporary filling materials.

  • In this study, we determined the changes in microbial numbers in solar salts according to the manufacturing process and storage duration. The salt samples were harvested from salt farms in Shinan (area 2) and Yeonggwang (area 1). They were serially diluted ten-fold and then placed on 4 kinds of cultivable media (mannitol salt agar, eosin methylene blue, plate count agar, and trypticase soy agar). After incubation, we obtained 62 halophilic isolates from the salt samples. Coliform and general bacteria were not detected in all salt samples. By 16S rRNA sequencing analysis, we found 12 kinds of halophilic bacteria belonging to the genera Halobacillus, Halomonas, Bacillus, Idiomarina, Marinobacter, Pseudoalteromonas, Vibrio, Salinivibrio, Virgibacillus, Alteromonas, Staphylococcus and some un-known stains. In our study, we discovered two novel species that have a 16S rDNA sequence similarity below 97%.

  • The present study investigated the effects of aging on Leydig cells of Sprague Dawley rats. Rats of 3, 6, 12 and 18 months of age were used. Testes of rat were fixed by whole body perfusion using a fixative containing 2.5% glutaraldehyde in cacodylate buffer, processed and embedded in epon-araldite. Using $1{\mu}m$ sections stained with methylene blue, qualitative and quantitative morphological studies were performed. Testis incubations were used to determine luteinizing hormone (LH; 100 ng/ml) stimulated testosterone secretory capacity per testis in vitro. Testosterone levels in the incubation medium, and testosterone and luteinizing hormone levels in serum of these four groups of rats were determined by radioimmunoassay. Morphological studies revealed that Leydig cells were more abundant in the testis interstitium at 6, 12 and 18 months when compared with 3 months. The volumes of Leydig cells per testis was significantly higher, at 6, 12 and 18 months of age than those at 3 months. The number of Leydig cells per testis was doubled at 6, 12 and 18 months of age compared with 3 months. The average volume of a Leydig cell was not significantly different between 3 and 6 months of age, however, at 12 and 18 months a significantly lower value was observed. LH-stimulated testosterone production per testis in vitro was reduced by 45% at 6 months of age compared with 3 months; a further significant reduction was observed at 12 and 18 months. Serum testosterone and LH levels were not significantly different between 3 and 6 months of age but at 12 and 18 months a significantly lower value was observed in both groups for these hormones. These results showed that signs of aging are apparent in Leydig cells of Sprague Dawley rats at 12 months of age.

  • There are accumulating evidences that high levels of dietary iron may play a role in colon carcinogenesis. Elevated iron status has been associated with oxidative stress. Phytic acid (PA) functions as an antioxidant by chelating divalent cations and prevents formation of reactive oxygen species responsible for cell injury and carcinogenesis. The protective effect of PA was investigated on formation of aberrant crypt foci (ACF) induced by azoxymethane (AOM) in iron-overloaded male F344 rats. After acclimation with AIN-93G purified diet (35 ppm Fe, normal control diet) for one week, animals were fed iron-overloaded diet (350 ppm Fe) and PA (0.5% or 2% PA in water) for 8 weeks. Animals received two (1st and 2nd week) injections of AOM (15 mg/kg b.w.) to induce colonic ACF. The colonic mucosa was examined for the total numbers of aberrant crypt (AC) and ACF after staining with methylene blue. The blood and serum were analyzed with a blood cell differential counter and an automatic serum analyzer. Iron-overloaded diet increased the concentration of iron in liver of the rats. But iron-related parameters in blood were not changed among experimental groups. The numbers of ACF per colon and AC per colon were $178.8{\pm}33.2$ and $448.4{\pm}110.2$ in the iron-overloaded F344 rats. The total AC was significantly increased, compared with normal-diet AOM control group (p < 0.05). The treatments of PA at the dose of 0.5% slightly decreased the number of ACF and AC per colon to $153.6{\pm}29.5$ and $396.3{\pm}107.5$. However, there were no significant differences in the total numbers of ACF and AC between the AOM control group and PA (0.5% or 2%)-treated groups. These results suggest that PA may not affect the formation of ACF or AC induced by AOM in ironoverloaded F344 rats.

  • Granular bamboo-derived active carbons (AC) were impregnated (or coated) with $TiO_2$ nano crystalline powders. The photocatalytic activity of the $TiO_2$-impregnated active carbons ($TiO_2$/AC) were determined on the basis of the degradation rate of methylene-blue aqueous solution under UV irradiation. The active compounds of $TiO_2$ were impregnated onto the AC under moderate hydrothermal conditions (${\leq}200^{\circ}C$, pH 11). The mean size of $TiO_2$ particles calculated from BET surface area were found to be as 50 nm. The $TiO_2$ precipitates were coated on the cavities or pores on the surfaces of highly activated carbons. Since the hydrothermal process led to a lowering of the on-set temperature of the anatase-to-rutile transition of $TiO_2$ as low as $200^{\circ}C$, $TiO_2$ crystallites of a pure anatase or a mixed form with rutile were successfully coated on the AC depending on the synthesis temperatures.
