Comparative Anatomy of the Korean Native Goat 2. Mesenteric mast cell (한국 재래 산양의 비교해부학적 연구 2. 장간막 비만세포에 대하여)

  • This study was carried out to investigate on the morphology, distribution and stainability of the mast cells in the Korean Native goat. For the study, the experimental animals were anesthetized with pentobarbital sodium and opened the anterior abdomianl wall to remove immediately the specimens with a minimum of mechanical effects. The mesenteries were fixed in 10% neutral formalin, 4% basic lead acetate, absolute alcohol and ethlene glycol monoethyl ether. Following 24 hours of fixation, the toto preparation stained with 0.4% toluidine blue, 1% methylene blue, 1.5% bismark brown, saturated thionine and thionlne-methylene blue complex solution. The preparation were observed from 10 microscopic field with 450 magnification. The results were as follows: 1. The form of the mesenteric mast cell was found 2 types. One was spindle form in larger number around vessels, the other was ovoid or spherical form in connective tissue far from blood vessels. 2. The average size was $18.63{\pm}5.75{\mu}m$ in length, $10.61{\pm}3.39{\mu}m$ in width and number was $105.50{\pm}18.45$. 3. Ethylene glycol monoethyl ether was particularly useful in preserving the mast cell granules. 4. Thione-methylene blue complex solution might be recommended to stain of granules.

  • Effect of sodium taurodeoxycholate (TDC) on the pharmacokinetics of methylene blue (MB) was investigated in the rats of experimental hepatic failure induced by $CCI_4$. Intravenous infusion of TDC increased the distribution volume of central compartment ($Vd_1$) and the total body clearance ($CL_t$) of MB. Increased lipophilicity through ion-pair formation with TDC seemed to be the probable cause of increased $Vd_1$ and $CL_t$.

  • Spectrophotometric and spectrofluorimetric methods of analysis were conducted in dilute aqueous solutions of methylene blue and tetraphenylborate. The formation of double ion-pair was confirmed and its overall formation constant, ${\Delta}H^{\circ}$, and ${\Delta}S^{\circ}$ were obtained. The irradiation with wavelengths over visible region resulted in bleaching the mixed solution. The bleaching reaction was also proceeded at $55^{\circ}C$. The reactions of the double ion-pair were briefly discussed.

  • Spectroscopic studies have been carried out on the metachromatic behavior of methylene blue(MB) and acridine orange(AO) in the presence of chondroitin sulfate A(CSA) and chondroitin sulfate C(CSC). The characteristic changes of the meta-band with the changes of P/D value are discussed in terms of the stacking theory. Quantitative studies on the stacking effect are made to calculate the number of bound molecules of dye per unit molecule of the polyanion. The result shows that MB has stronger stacking effect than AO. A stacking model and the dimension of the bound dyes on the surface of the polyanion are proposed, on the basis of the dimer model of planar aromatic dyes and the most stable conformation of the CSA chain. The model is found to be reasonable in accordance with the experimental results.

  • Electrochemical characteristics of nitrite ion were investigated at a poly(methylene blue)-modified glassy carbon electrode by cyclic voltammetry and differential pulse voltammetry. The poly(methylene blue)-modified glassy carbon electrode exhibited enhanced anodic signals for nitrite. The effects of key parameters on the detection of nitrite were evaluated at the modified electrode, such as pH, accumulation time, and scan rate. Under optimum condition, the chemically modified electrode can detect nitrite in the concentration range $2.0{\times}10^{-6}$ to $5.0{\times}10^{-4}$ M with the detection limit of $2.0{\times}10^{-6}$ M and a correlation coefficient of 0.999. The detection of nitrite using the chemically modified electrode was not affected by common ions such as $Na^+$, $K^+$, $Ca^{2+}$, $Cl^-$, $HPO_4^{2-}$ and $H_2PO_4^- $. The modified electrode showed good stability and reproducibility. The practical application of the present method was successfully applied to the determination of nitrite ion in cabbage samples.

Spectroscopic Study on Three States of Water in the Reverse Micelle Using Methylene Blue as a Probe (Methylene Blue를 이용한 역미셀에서 물의 세 가지 상태에 대한 분광학적 연구)

  • In order to find out the microscopic environmental information on the nonionic reverse micelle of Triton X-100/n-hexanol/water in cyclohexane, an absorption and fluorescence spectroscopic study has been conducted using a methylene blue(MB). The information on the microscopic states of water in the polar core of the reverse micelle has been found by investigating complex formation and solvatochromic behavior between MB and Triton X-100. As a result, it was found that there exist three states in the polar core of the reverse micelle. The measured values of $W(=[H_2O]/[Surf])$ for the three states of water are 0.71, 4.98, and 7.26, and the corresponding lifetimes of MB are $15.45 ns{\pm}0.56$, $12.27 ns{\pm}0.79$, and $8.28 ns{\pm}0.82$, respectively.

  • The aim of this review is to introduce about the issue of benzocaine and methemoglobinemia. Through blocking the pain during dental treatment, fear and anxiety of patients will be reduced. Thus, anesthetic agent containing benzocaine is commonly used while controlling the pain of patients during treatment. However, on May 28, 2018, the Ministry of Food and Drug Safety reported a medication safety report about restricting the use of benzocaine-containing agents in infants under 24 months and children. Also, they recommended a cautious use to adolescents and adults to prevent methemoglobinemia (MHb). This report was published due to an advice from Food and Drug Administration (FDA) on May 23, 2018. When using agents containing benzocaine, dentists must consider the probability of MHb and prepare for early diagnosis and appropriate action. Since 1930s, methylene blue is known to cure MHb patients. Therefore, the proper use of methylene blue for emergencies and diagnosis methods for early diagnosis of MHb should be familiar to dentists planning for the use of topical anesthetic agents. Dentists should be trained for emergency situation of MHb caused by the use of benzocaine.

  • Spectroscopic studies have been carried out on the metachromatic behavior of methylene blue(MB) and acridine orange(AO) in the presence of polyvinylsulfate(PVS) and polystyrenesulfonate(PSS) The characteristic changes of meta-band with the change of P/D value are discussed in terms of stacking theory. It has been found that the stacking effect in the PVS-dye system is stronger than that in the PSS-dye system and that MB shows stronger stacking effect than AO. A stacking model and dimension of bound dyes on the surface of polymer chain is proposed on the basis of the previously suggested model of dimer found in the aqueous solution of planar aromatic dyes. The proposed model is found to be reasonable in accordance with the experimental results obtained by various workers.

On Improvement of the Adsorption by Chemical Treatment on Yeong-Il Bentonite (영일산 Bentonite의 화학적 처리에 의한 흡착력 개선)

  • The adsorption of Methylene Blue on Yeong-Il bentonite which was treated by aqueous NaOH, $Na_2SO_4 or NaHSO_4 solution respectively, varying concentrations, temperature and time, was studied. In case of treatment with NaHSO_4 solution, slight improvement of the adsorption of Methylene Blue on bentonite was observed. With Na_2SO_4$ solution, the best results obtained when bentonite was treated with 1 N of the solution for 2hr at $100^{\circ}C$, and the adsorption capacity of the product was 3 times better than that of original bentonite. At the higher concentration and the higher temperature than above, faujasite was formed. With NaOH solution, the best condition was in 1N solution for 1hr at $100^{\circ}C$ and the adsorption capacity of the product was 3.3 times better than that of original bentonite. At the higher concentration of the treating agent and at the higher temperature than above, hydroxysodalite was formed.
