Physicochemical Characteristics of Antidementia Acetylcholinesterase Inhibitor-containing Methanol Extract from Sorghum bicolor and Industrial Application (항치매성 Acetylcholinesterase저해물질을 함유하고 있는 수수 메탄올 추출물의 특성 및 산업적 응용)

  • Alzheimer's disease is charaterized by the acetylcholine depletion, amyloid b-protein aggregation and neurofibrillary tangles. The prevention of the breakdown of acetylcholine by acetylcholinesterase (AChE) inhibitor has the best clinically therapeutic efficacy for Alzheimer's disease patients. To develop new antidementia alternative drugs or nutraceuticals, methanol extracts of Sorghum bicolor was screened from various extracts of cereals and legumes as a potent AChE inhibitor-containing extract in previous paper. In this paper, physicochemical properties of the methanol extracts was investigated. The methanol extracts was soluble by water, methanol and DMSO and had 215 nm and 282nm of maximum absorption spectra. It was also stable at 20-$100^{\circ}C$ and pH 2.0-10.0 for 1 hr. Test product was prepared by using methanol extracts from Sorghum bicolor and changes of its quality during storge at $20^{\circ}C$ and $40^{\circ}C$ were investigated. It was very stable for 8 weeks at $40^{\circ}C$.

In Vitro Assay on Antioxidant Activity and Cytotoxicity of Methanol Extracts from Young Sprouts of Several Korean Salad Plants

  • Antioxidant activity, total phenolics level and cytotoxicity of the methanol extracts from the young sprouts of 5 Korean woody salad plants were determined. Methanol extracts of Kalopanax pictus had the highest DPPH radical scavenging activity, with an $IC_{50}$ value of 23.5mg $100g^{-1}$, and followed by Valeriana fauriei(43.1mg $100g^{-1}$), and Morus alba(>100mg $100g^{-1}$). Total phenolic content showed the highest amount in methanol extracts from Kalopanax pictus(23.7mg $100g^{-1}$), and followed by Valeriana fauriei(22.7mg $100g^{-1}$), Aralia elata(16.8mg $100g^{-1}$) and Morus alba(14.2mg $100g^{-1}$). In a MTT assay, methanol extracts of Aralia elata with $IC_{50}$ values of 151.0 and $140.7{\mu}g\;mL^{-1}$ showed the most potent cytotoxicity on Calu-6 and MCF-7, respectively. On the other hand, methanol extracts of Kalopanax pictus($IC_{50}=96.5{\mu}g\;mL^{-1}$) showed the highest activity against HCT-116, and followed by those of Aralia elata($123.3{\mu}g\;mL^{-1}$), and Actinidia arguta($162.0{\mu}g\;mL^{-1}$). Total phenolic content of the tested plant extracts was correlated with the DPPH radical scavenging activity, suggesting the phenolic compounds may contribute to the antioxidant properties of Korean salad plants.

Biological Activities of Methanol Extract of Angelica gigas Nakai (참당귀 Methanol 추출물의 생리활성)

  • The biological activities of methanol extracts of Angelica gigas Nakai, such as antioxidation, anticancer and immuno-activity, were investigated in relation to development of functional foods. Anti-oxidation activity in the methanol extracts were assessed by hydrogen donating activity, reducing power and hydroxyl radical scavenging activity. Activities were dose-dependent over concentrations of 0.1, 0.5 and 1 mg/mL, with thehydrogen donating activity being over 50% at 1 mg/mL concentration. The methanol extracts inhibited the proliferation of SW480 cells in a dose-dependent manner, and chromatin condensation and apoptotic bodies were observed by fluorescence microcopy in the cells treated with the extracts for 24 hr. Caspase-3 activity was also increased in a dose-dependent manner in cells treated with the extracts relative to control cells. The extracts did not induce the proliferation of mouse spleen cells or NO production in macrophage cells (RAW 264.7). These results show that the methanol extract had slight anti-oxidative activity and did not increase immuno-activity, but inhibited proliferation of SW480 through apoptosis via a caspase dependent pathway.

In vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and inhibition of nitric oxide production activities of methanol and hot water extracts of Russula rosacea mushroom

  • Russula rosacea, a mycorrhizal fungus, has been used for edible and medicinal purposes. This study was conducted to evaluate the in vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and nitric oxide inhibitory effects of the fruiting bodies from R. rosacea extracted with methanol, and hot water. The 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activities of the methanol and hot water extracts (2.0 mg/ml) of R. rosacea were comparable with BHT, the positive control. The chelating effects of the mushroom and hot water extracts were significantly higher than that of BHT. The reducing power of methanol and hot water extract (6 mg/ml) were significantly lower than that of BHT. Seven phenolic compounds were detected from acetonitrile and hydrochloric acid solvent extract of the mushroom. alpha-amylase and alpha-glucosidase inhibitory activities of methanol and hot water extracts were lower than that of acarbose, the positive control. The acetylcholinesterase and butyrylcholinesterase inhibitory effects were moderate compared with galanthamine, the standard drug. Nitric oxide (NO) production in lipopolysaccharide (LPS) induced RAW 264.7 cells were inhibited significantly by the mushroom extracts in a concentration dependent manner. Therefore, we demonstrated that fruiting bodies of R. rosacea possess in vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and NO production inhibitory activities. The experimental results suggest that the fruiting bodies of R. rosacea are good natural antioxidant, anti-hyperglycemic, anti-cholinesterase, and anti-inflammatory sources.

Hepatoprotective activity of methanol extracts of Berberis tinctoria

  • The methanol extracts of the roots, root bark and stem of Berberis tinctoria, were investigated for their hepatoprotective activity against carbon tetrachloride $(CCl_4)$ induced toxicity in freshly isolated rat hepatocytes, HEp-G2 cells and animal models. The methanol extracts were able to significantly normalise the levels of aspartate amino transferase, alanine aminotransferase, alkaline phosphatase, triglycerides, total proteins, albumin, total bilirubin and direct bilirubin, which were altered due to $CCl_4$ intoxication in freshly isolated rat hepatocytes and also in animal models. The anti-hepatotoxic effect of the methanol extracts in vitro were observed at $600\;-\;1,000\;{\mu}g/ml$ concentrations. A dose dependent increase in the percentage viability was observed when $CCl_4$ exposed HEp-G2 cells were treated with different concentrations of the methanol extracts. The highest percentage viability of HEp-G2 was observed at a concentration of $1,000\;{\mu}g/ml$. The results from the present investigations also indicate good correlation between the in vivo and in vitro studies.

Protective Effects of Ulva lactuca Methanol Extracts against the Ultraviolet B-induced DNA Damage (자외선 B에 의해 유도되는 DNA 상해에 대한 참갈파래 메탄올 추출물의 보호 효과)

  • In this study, we investigated the protective effects of Ulva lactuca methanol extracts against ultraviolet B (UVB)-induced DNA damage in HaCaT cells. First, the contents of general and antioxidative nutrient contents of Ulva lactuca were measured. The moisture, carbohydrate, crude protein, crude fat and ash were 14.01%, 44.80%, 23.19%, 3.10% and 14.90%, respectively. Magnesium that acts as DNA repair enzyme cofactor was the most abundant mineral followed by Ca, P and Fe. The total phenolic and anthocyanoside contents of Ulva lactuca were 2.69 mg/g and 0.13 mg/g, respectively. Cells treated with Ulva lactuca methanol extracts for 24 hours post UVB exposure increased cell viability in a concentration-dependent manner compared to the non-treated control. Also, Ulva lactuca methanol extracts decreased the levels of UVB-induced DNA damage such as cyclobutane pyrimidine dimer and DNA damage response (DDR) proteins such as p-p53 and p21. These results suggest that Ulva lactuca methanol extracts comprising physiological active substances such as Mg, polyphenols and anthocyanosides promote DNA repair by regulating genes related with DDR.

The Nitrite-Scavenging Effects by Component of Green Tea Extracts (녹차 추출물의 아질산염 분해작용)

  • This paper was investigated to elucidate the nitrite-scavenging action as one of biological actions of green tea extracts. Green tea extracts exhibited remarkable nitrite-scavenging action. Among the green tea extracts, steamed green tea showed higher nitrite-scavenging action than roasted green tea. Nitrite-savenging actions of gren tea extracts were hardly influenced until three times extraction with hot water. Furthermore , the nitirte-scavenging action of green tea extracts showed ph dependent , highest at pH 1.2 and lowest at pH 6.0. The nitrite -scavenging rate in sample extracts /amine /nitrite systems exhibited faster than that in amine/nitrite systems. In order to screen the nitirte-scavenging factors, samples extracts were fractionated with methanol. in both samples, methanol-soluble fractions possessed greater nitrite-scavenging action than methanol-insoluble fractions .The nitrite-scavenging action of methanol-soluble fractions increased with proportion to absorbance at 420nm and at 280nm, while that of green tea extracts increased with the contents of total phenols and absorbance at 280nm.

Antimicrobial Activity and Bactericidal Activity of Caesalpinia sappan L. Extract (소목 추출물의 항균력 및 살균소독력)

  • Antimicrobial activity and bactericidal activity of Caesalpinia sappan L. extracts were investigated against five food-borne pathogens, E. coli, S. aureus, S. typhimurium, B. cereus and L. monocytogenes. Methanol extract of Caesalpinia sappan L. revealed antimicrobial activities against five pathogens. In particular, by paper disc diffusion the highest activity was shown against L. monocytogenes. Antimicrobial activities of methanol extracts showed the most potent activities, but hexane fraction had no activity. Fractions of ethyl acetate and butanol turned out to have higher antimicrobial activities against Gram(+) bacteria than Gram(-) bacteria. The minimum inhibitory concentration against five food-borne pathogens was 1.563 mg/ml on Gram(+) bacteria and 3.125 mg/ml on Gram(-) bacteria. The result of antimicrobial activity in a shaking flask method showed that bacterial growth rate fell by more than 99.999% at 3.125 mg/ml of methanol extract. The highest rate of viable reduction (99.998%) was shown at 0.781 mg/ml of methanol extract against L. monocytogenes. After five minutes of reaction between test strains and methanol extracts, the growth rates of five kinds of bacteria were reduced by more than 99.999% at a concentration of 100 mg/ml. Therefore, it is suggested that methanol extracts of Caesalpinia sappan L. can be developed as a natural sanitizer or disinfectant.

Negative Effects of Water Extracts from Pinus densiflora Sawdust on Mycelium Growth of the Shiitake Mushroom Lentinula edodes

  • Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.

A Study on the Effects of Rhodiola rosea Root on the Immune System (홍경천(紅景天)의 면역증진(免疫增進) 효능(效能)에 대한 연구(硏究))

  • Objectives: Effects of Rhodiola rosea root on immunity were examined. Methods: Water extracts and methanol extracts of Rhodiola rosea root were treated on the increase of immunity cells and the activation of cytokines were examined. Results: 1. Water extracts and methanol extracts of Rhodiola rosea root had positive effects on the increase of B-cells, T-cells, and NK cells, and on the facilitation of cytokines such as IL-6 and TNF-${\alpha}$. 2. Water extracts and methanol extracts of Rhodiola rosea root activated NK cells, and induced the generation of NO- of NK cells and macrophages. Conclusions: Rhodiola rosea root can be used to increase immunity.
