갈화 및 초두구를 주재로 하는 알코올 해독제 (Ethanol Detoxicant Composed of Pueraria thunbergiana and Alpinia katsumadai)

  • Alpinia katsumadai is used in traditional oriental medicine as an antiemetic and for treatment of stomach disorders. This study was to investigate the effect of the 50% methanol extract of Alpinia katsumadai and Puerariae thunbergiana on ethanol metabolism in rat. The administration of 50% methanol extract of Alpinia katsumadai significantly decrease the ethanol concentration in rat. The AUC and Cmax of Alpinia katsumadai was 3.1 and 2.4 times less than the control group, respectively. It was more effective than Pueraria thunbergiana is known to as ethanol detoxicant. Furthermore, we tested on its ethanol metabolizing effect with the preparation containing this herb (Haeju pill) in beagle. The AUC of Haeju pill was about 1.9 times less than the control group. It was estimated that the extract of Alpinia katsumadai had feasibility as a ethanol detoxication agent.

Protective Effects of Alpinia katsumadai Extract Against Oxidative Stress

  • In the present study, total methanol extracts prepared from Alpinia katsumadai showed significant protective effects against the oxidative stress induced by hydrogen peroxide, UV-C or ${\gamma}$-ray irradiation. These protective effects were substantially increased by treatment with 20~100 ${\mu}g$/ml of the extract. The A. katsumadai total methanol preparation was further fractionated into n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these five fractions, the ethylacetate and butanol fractions of A. katsumadai showed the strongest protective effects against oxidative stress induced by UV-C and ${\gamma}$-ray irradiation. These fractions also showed high DPPH radical scavenging and lipid peroxidation inhibitory activities. In addition, both fractions displayed cell proliferation activation effects, as evidenced by significant increases in colony formation. Our current data thus suggest that the mechanisms underlying the protective effects of A. katsumadai against oxidative damage may include radical scavenging, protection against cell membrane damage and stimulation of cell proliferation.

천연약용식물의 활성산조종에 의한 산화적 손상의 보호 효과 (Protective Effect of Natural Medicinal Plants against Oxidative Damage Induced by Reactive Oxygen Species)

  • In order to evaluate anti -oxidant activities and protective effect against oxidatve damage, DPPH radical scavenging activity and lipid peroxidation inhibitory activity were measured among methanol extracts prepared from natural medicinal plants. Fourteen natural medicinal plants which were reported to have anti -oxidative or anti-inflammatory effects were selected based on our previous report. In addition to the total methanol extracts, n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions were prepared from each total extract. DPPH radical scavenging assay was performed against 14 total extracts and all samples showed dose-dependent activities in various extent. Among those, 6 samples, methanol extracts of Euryale ferox, paeonia suffruticosa, Areca catechu var. dulcissima, Cinnamomun cassia, Alpinia katsumadai and Betula platyphlla var. japonica showed IC$\sub$50/ value lower than 6.0 $\mu\textrm{g}$/ml. The highest DPPH radical scavenging activity was found in ethylacetate fraction of paeonia suffruticosa with IC$\sub$50/ value of 1.1 $\mu\textrm{g}$/ml. Analysis of lipid peroxidation inhibitory activity on hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast (V79-4) cells revealed that the highest inhibitory effect was observed in methanol extract of Betula platyhpylla var. japonica. Lipid peroxidation inhibitory activity was observed as a dose-dependent manner in all samples used in this study. Among fraction samples, ethylacetate fraction of Alpinia katsumadai had the strongest inhibitory activity with IC$\sub$50/ value of 0.9 $\mu\textrm{g}$/ml.

  • ${\alpha}$-glucosidase 활성을 저해하는 초두구 메탄올 추출물의 항바이러스 활성을 연구하였다. Newcastle disease virus (NDV) 감염된 baby hamster kidney (BHK) 세포에서 Syncytium (합포체) 형성은 세포막 표면으로의 수송된 바이러스 당단백질 hemagglutinin-neuramidase (HN)에 의해 일어난다. 초두구 추출물$IC_{50}$ $25{\mu}g/mL$) 처리된 세포에서 바이러스 당단백질의 수송과 함께 합포체 형성이 저해되었다. 또한 $IC_{50}$ 농도에서는 세포 내에서 당단백질 생합성은 저해되지 않으며 당단백질의 수송을 저해하는 것으로 추론된다.

소목추출물의 젖소 유방염 원인균에 대한 항균효과 (Antibacterial Effect of Caesalpinia sappan Extract against Mastitis Pathogens from Dairy Cows)

  • To find alternative agents for bovine mastitis, the extracts of 30 selected herbal medicine were screened for their antibacterial activity against mastitis pathogens. The extracts of 9 herbal medicine, Terminaliae chebula, Cinnamonum loureirii, Paeonia japonica, Inula britannica, Caesalpinia sappan(C. sappan), Salix babylonica, Alpinia katsumadai, Polygonum aviculare and Coptidis japonica showed strong antibacterial activity. Among of them, C. sappan extract was found to be the most active against all bacteria in this experiment. Methanol extract of C. sappan and brazilin, the main component of C. sappan, had inhibitory activity against a variety of pathogens, however they had less of an inhibitory effect on gram negative bacteria. Minimal inhibitory concentrations of brazilin against gram positive pathogens was lower than that of methanol extract. These results showed that the extract of C. sappan has a potential as a therapeutic agent for bovine mastitis.
