The role of immunomodulatory metabolites in shaping the inflammatory response of macrophages

  • Macrophage activation has long been implicated in a myriad of human pathophysiology, particularly in the context of the dysregulated capacities of an unleashing intracellular or/and extracellular inflammatory response. A growing number of studies have functionally coupled the macrophages' inflammatory capacities with dynamic metabolic reprogramming which occurs during activation, albeit the results have been mostly interpreted through classic metabolism point of view; macrophages take advantage of the rewired metabolism as a source of energy and for biosynthetic precursors. However, a specific subset of metabolic products, namely immune-modulatory metabolites, has recently emerged as significant regulatory signals which control inflammatory responses in macrophages and the relevant extracellular milieu. In this review, we introduce recently highlighted immuno-modulatory metabolites, with the aim of understanding their physiological and pathological relevance in the macrophage inflammatory response.

Is FDG -PET-CT A Valuable Tool in Prediction of Persistent Disease in Head and Neck Cancer

  • Objectives: To evaluate accuracy of FDG-PET CT in prediction of persistent disease in head and neck cancer cases and to determine prognostic value of metabolic tumor response. Materials and Methods: Between 2009 and 2011, 46 patients with squamous cell carcinoma of head and neck receiving PET-CT were treated with definitive radiotherapy, with or without chemotherapy. There were 29 nasopharyngeal, 11 hypopharyngeal, 3 oropharyngeal and 3 laryngeal cancer patients, with a median age of 50.5 years (range 16-84), 32 males and 14 females. All patients were evaluated with PET-CT median 3-5 months (2.4-9.4) after completion of radiotherapy. Results: After a median 20 months of follow up, complete metabolic response was observed in 63% of patients. Suspicious residual uptake was present in 10.9% and residual metabolic uptake in 26.0% of patients. The overall sensitivity, specificity, positive predictive value and negative predictive value of FDG-PET-CT for detection of residual disease was 91% and 81%, 64% and 96% respectively. Two year LRC was 95% in complete responders while it was 34% in non-complete responders. Conclusions: FDG PET CT is a valuable tool for assessment of treatment response, especially in patients at high risk of local recurrence, and also as an indicator of prognosis. Definitely more precise criteria are required for assessment of response, there being no clear cut uptake value indicating residual disease. Futhermore, repair processes of normal tissue may consume glucose which appear as increased uptake in control FDG PET CT.

Proteome analysis of sorghum (Sorghum bicolor L.) leaf in response to waterlogging stress

  • Growth related to morphological and proteome response under waterlogging stress in sorghum has not yet been elucidated. Understanding how plants respond to waterlogging, the present study was conducted in seedlings leaf of the Nam-pung chal cultivar. Regarding 3-leaf stage of sorghum, stem length and plant height were slightly decreased in the treatments during ten days of waterlogging, and chlorophyll contents were also significantly different from 7 days of waterlogging treatment. The results observed from the present study were considered to be influenced by the waterlogging stress more in the $5^{th}$ leaf stage of the growth period of the sorghum, and as the waterlogging treatment progressed, the waterlogging stress gradually influenced the growth difference between the control and the treatment respectively. Using 2-DE method, a total of 74 differentially expressed protein spots were analyzed using LTQ-FT-ICR MS. Of these proteins, 45 proteins were up-regulated in the treatment group, and 32 proteins were down-regulated. Analysis of LTQ-FI-ICR MS showed that about 50% of the proteins involved in carbohydrate metabolic process, metabolic process, and cellular metabolic compound salvage were affected by stress. Malate dehydrogenase protein and Glyceraldehyde-3-phosphate dehydrogenase protein related to carbohydrate metabolic process increased the level of protein expression in both 3 and 5-leaf stage under waterlogging stress. The increased abundance of these proteins may play an active role in response to waterlogging stress. These results provide new insights into the morphological alteration and modulation of differentially expressed proteins in sorghum cultivar.

Application of metabolic profiling for biomarker discovery

  • An important potential of metabolomics-based approach is the possibility to develop fingerprints of diseases or cellular responses to classes of compounds with known common biological effect. Such fingerprints have the potential to allow classification of disease states or compounds, to provide mechanistic information on cellular perturbations and pathways and to identify biomarkers specific for disease severity and drug efficacy. Metabolic profiles of biological fluids contain a vast array of endogenous metabolites. Changes in those profiles resulting from perturbations of the system can be observed using analytical techniques, such as NMR and MS. $^1H$ NMR was used to generate a molecular fingerprint of serum or urinary sample, and then pattern recognition technique was applied to identity molecular signatures associated with the specific diseases or drug efficiency. Several metabolites that differentiate disease samples from the control were thoroughly characterized by NMR spectroscopy. We investigated the metabolic changes in human normal and clinical samples using $^1H$ NMR. Spectral data were applied to targeted profiling and spectral binning method, and then multivariate statistical data analysis (MVDA) was used to examine in detail the modulation of small molecule candidate biomarkers. We show that targeted profiling produces robust models, generates accurate metabolite concentration data, and provides data that can be used to help understand metabolic differences between healthy and disease population. Such metabolic signatures could provide diagnostic markers for a disease state or biomarkers for drug response phenotypes.

우유 섭취와 대사증후군의 관련성 - 2007~2010년 국민건강영양조사 자료를 이용하여 (Milk Intake is Associated with Metabolic Syndrome - Using Data from the Korea National Health and Nutrition Examination Survey 2007~2010)

  • This study aimed to examine associations between milk intake and metabolic syndrome. The subjects included 1,928 males and 3,103 females, aged 19 to 64 years, from the data of 'The Korean National Health and Nutrition Survey 2007-2010'. Daily intake of milk and dairy products was obtained by a 24 hour dietary recall method and divided into two categories by equivalent weight of one serving. The average individual intakes of milk and dairy products were 59.4 g and 74.1 g per day respectively. Milk intake was inversely associated with metabolic syndrome (OR: 0.69, 95% CI: 0.54~0.89), central obesity (OR: 0.75, 95% CI: 0.62~0.91), and hypertriglyceridemia (OR: 0.73, 95% CI: 0.59~0.90). The total intake of dairy products was also inversely associated with metabolic syndrome (OR: 0.74, 95% CI 0.60~0.92), central obesity (OR: 0.73, 95% CI: 0.62~0.86), hypertension (OR: 0.80, 95% CI: 0.65~0.99). The association between intakes of milk and dairy products and metabolic syndrome was significant in women, but not in men. These results indicate that increased consumption of milk and its products is associated with a reduced likelihood of metabolic syndrome and metabolic syndrome risk factors. Further research on causal relationship and dose-response association between milk intake and metabolic syndrome risk is necessary prior to applying the observed results in nutrition policies and programs to prevent the metabolic syndrome.

Tumor bioenergetics: An emerging avenue for cancer metabolism targeted therapy

  • Cell proliferation is a delicately regulated process that couples growth signals and metabolic demands to produce daughter cells. Interestingly, the proliferation of tumor cells immensely depends on glycolysis, the Warburg effect, to ensure a sufficient amount of metabolic flux and bioenergetics for macromolecule synthesis and cell division. This unique metabolic derangement would provide an opportunity for developing cancer therapeutic strategy, particularly when other diverse anti-cancer treatments have been proved ineffective in achieving durable response, largely due to the emergence of resistance. Recent advances in deeper understanding of cancer metabolism usher in new horizons of the next generation strategy for cancer therapy. Here, we discuss the focused review of cancer energy metabolism, and the therapeutic exploitation of glycolysis and OXPHOS as a novel anti-cancer strategy, with particular emphasis on the promise of this approach, among other cancer metabolism targeted therapies that reveal unexpected complexity and context-dependent metabolic adaptability, complicating the development of effective strategies.

허베이 스피리트호 원유유출시 방제작업과 대사증후군의 연관성 (Association between Metabolic Syndrome and Participation in Clean-up Work at the Hebei Spirit Oil Spill)

  • Objectives: We aimed to assess the risk of metabolic syndrome one year after the 2007 Hebei Spirit oil spill in Taean, Korea among people exposed to spilt oil during clean-up work. Methods: A total of 6,923 adults, including 3,019 males and 3,904 females, participated in the study. Health examinations and blood tests (total cholesterol, triglycerides, HDL, fasting blood sugar) were performed. A logistic regression model adjusting for age, gender, smoking history, drinking history, income, education, and marital status was used to estimate the risk of metabolic syndrome associated with the level of oil spill exposure. Results: The prevalence of metabolic syndrome was 24.9% among males and 18% among females. Compared with people living within 20 km of the coastline, the risk of metabolic syndrome among people living within 0.8 km of the coastline was significantly higher (male OR=1.696, 95% CI=1.320-2.178, female OR=1.992, 95% CI=1.549-2.561), including a significant dose-response relationship for distance from early contaminate coastline (p<0.001). The risk of metabolic syndrome was higher according to the increase of duration of cleaning work. The risk of metabolic syndrome among people who participated in the clean-up work for more than 116 days, compared with people who participated in the cleaning work for or less 14 days, was significantly higher (male OR=1.845, 95% CI=1.448-2.353, female OR=1.752, CI=1.378-2.228), with a significant dose-response relationship for days of clean-up work (p<0.001). Conclusion: This study showed that there is a significant association between exposure to the oil spill during the clean-up work, distance from early contaminate coastline and the risk of metabolic syndrome in a doseresponse manner.

비만 중년여성의 복합운동이 대사증후군 지표와 건강체력에 미치는 영향 (The effects of participation in a combined exercise program on the metabolic syndrome indices and physical fitness in the obese middle-aged women)

  • 본 연구는 12주간 복합운동프로그램이 비만 중년여성들의 대사증후군 지표와 건강 체력에 미치는 영향을 알아보기 위한 것으로 최근 6개월 이내에 규칙적으로 운동에 참여하지 않는 비만 중년여성 22명을 임의로 선정하였다. 본 연구의 자료분석을 위한 통계프로그램은 SPSS 18.0을 이용하여 모든 종속변인의 평균값과 표준편차를 산출하였으며, 12주 전 후 집단 내 변인들의 변화를 알아보기 위해 윌콕슨의 부호순위 검정을 실시하였고, 각 분석에서의 통계적인 유의수준은 p<.05로 설정하였다. 본 연구에서 실시한 12주간 복합운동프로그램은 비만 중년여성의 대사증후군 지표와 건강체력에 긍정적인 효과를 미친 것으로 나타났다. 따라서 향후 비만 중년여성의 건강관리와 체력향상을 위하여 신체적 여건, 체력수준, 사회 경제적 상황을 충분히 고려한 다양한 수준의 운동프로그램 개발에 노력해야 할 것이다.

대사증후군 개선을 위한 뽕잎, 오디, 누에 분말의 혼합 비율 최적화 (Optimization of Mixing Ratio of Mulberry Leaf, Mulberry Fruit, and Silkworm for Amelioration of Metabolic Syndrome)

  • Objectives: The aim of this study was optimized mixing ratio of mulberry leaf, mulberry fruit, and silkworm for amelioration of the metabolic syndrome by using response surface method (RSM). Methods: Antioxidant, antidiabetic and antihypertensive activities of fifteen mixed powder of mulberry leaf, mulberry fruit, and silkworm by RSM were measured as indicators of metabolic syndrome. Results: The optimal mixing ratio of mulberry leaves, mulberry fruits, and silkworm with the greatest antioxidant, antidiabetic and antihypertensive activities was as follows: 2.5890 of mulberry leaf (A), 0.1222 of mulberry fruit (B), 2.9999 of silkworm (C). At this time, it was predicted that the total polyphenol content was estimated to be 185.51 tannic acid equivalent mg/g, 1, 1-diphenyl-2-picrylhy drazyl radical scavenging activity 84.77%, 1-deoxynojirimycin content 415.66 mg/100 g, ${\alpha}-glucosidase$ inhibitory activity 64.31%, ${\gamma}-aminobutyric$ acid content 267.77 mg/100 g, potassium content 1,899.11 mg/100 g, and angiotensin-converting-enzyme inhibitory activity was estimated to be 73.78%. Conclusions: It was concluded that the significant effect of the mulberry leaf, mulberry fruit, and silkworm on the metabolic syndrome-related biological activity indices (antioxidant activity, antidiabetic activity and antihypertensive activity) was as follows: 2.5890 of mulberry leaf (A), 0.1222 of mulberry fruit (B), 2.9999 of silkworm (C).

Serum vitamin D status and metabolic syndrome: a systematic review and dose-response meta-analysis

  • BACKGROUD/OBJECTIVES: Evidence has suggested an association between serum vitamin D and metabolic syndrome (MetS), but prospective studies are very limited. The objective was to assess the dose-response association between serum vitamin D concentration and MetS risk using a systematic review and meta-analysis of updated observational studies. MATERIALS/METHODS: Using MEDLINE, PubMed, and Embase, a systematic literature search was conducted through February 2020 and the references of relevant articles were reviewed. A random-effects model was used to estimate the summary odds ratio/relative risk and 95% confidence interval (CI). Heterogeneity among studies was evaluated with I2 statistic. In total, 23 observational studies (19 cross-sectional studies, and four cohort studies) were included in the meta-analysis. RESULTS: The pooled estimates (95% CI) for MetS per 25-nmol/L increment in serum vitamin D concentration were 0.80 (95% CI, 0.76-0.84; I2 = 53.5) in cross-sectional studies, and 0.85 (95% CI, 0.72-0.98; I2 = 85.8) in cohort studies. Similar results were observed, irrespectively of age of study population, study location, MetS criteria, and adjustment factors. There was no publication bias for the dose-response meta-analysis of serum vitamin D concentrations and MetS. CONCLUSIONS: Dose-response meta-analysis demonstrated that a 25-nmol/L increment in the serum vitamin D concentration was associated with 20% and 15% lower risks of MetS in cross-sectional studies and cohort studies, respectively.
