Effects of $\omega$6 and $\omega$3 Fatty Acid Diets on the Fatty Acid Composition of the Mesenteric and Subcutaneous Fat of Lactating Rats

  • Long chain polyunsaturated fatty acids (LCPUFA) are important components of brain phospholipds and play important role (s) in brain function. In rats, the maximum brain growth occurs during the period of lactation even though it happens during the third trimester of gestation in human. Since milk contained docosahexaenoic acid (DHA) even through the maternal diet had no DHA and/or a very small amount of its precursor, $\alpha$-linolenic acid ($\alpha$-LnA), an emphasis was given to maternal adipose tissue as a reservoir of this fatty acid. We, therefore, investigated the mesenteric and subcutaneous adipose tissues for their fatty acid composition in dams reared with different fat diets. Diets containing various amounts of $\omega$6 and $\omega$3 fatty acids were given to adult female rats (200-250g) throughout the pregnancy and lactation periods. Diets were composed of 10% (wt/wt) corn oil (CO), soybean oil (SO), perilla seed oil (PO) containing about 60% $\alpha$-LnA, or fish oil (FO) rich in eicosapentaenoic acid (EPA) and DHA. The fatty acid ompositions of mesenteric and subcutaneous fat were measured and evaluated at Day-2 and Day-15 after parturition. In general, major characteristics of dietary fatty acid composition was reflected on the fatty acid composition of adipose tissues. Dietary fatty acid composition was reflected more on mesenteric fat as compared to subcutaneous fat. Mesenteric fat was found to contain less arachidonic acid (AA) and mesenteric fats of CO, SO and PO groups contained less DHA than did the subcutaneous fat. The P/M/S ratios of adipose tissues were similar between experimental groups while dietary P/M/S ratios differed significantly. It was noticeable that a small proportion of DHA was found in the adipose tissues of animals of CO, SO and PO groups (Day-2) and in SO and PO groups (Day-15), the groups which do not contain DHA in their diets. The percentage of DHA in mesenteric fat o CO, SO and PO groups decreased as lactation continues, while the proportion of DHA in FO group increased. Adipose tissues of FO group had higher DHA/EPA ratio as compared to the diet. Considering the fact that the body contains a large amount of adipose tissues, our present finding suggests that the adipose tissue can serve as a reservoir of DHA for pregnant and lactating rats.

Vitamin C Inhibits Visceral Adipocyte Hypertrophy and Lowers Blood Glucose Levels in High-Fat-Diet-Induced Obese C57BL/6J Mice

  • Vitamin C (ascorbic acid) supplementation has been suggested to negatively correlate with obesity in humans and other animals. Previous studies, including ours, have demonstrated that a high-fat diet (HFD) induces obesity and related diseases such as hyperlipidemia, hyperglycemia, insulin resistance, and nonalcoholic fatty liver disease. Here, we investigated the effects of vitamin C on visceral adipocyte hypertrophy and glucose intolerance in C57BL/6J mice. Mice received a low-fat diet (LFD, 10% kcal fat), HFD (45% kcal fat), or the same HFD supplemented with vitamin C (HFD-VC, 1% w/w) for 15 weeks. Visceral adiposity and glucose intolerance were examined using metabolic measurements, histology, and gene expression analyses. Mice in the HFD-VC supplementation group had reduced body weight, mesenteric fat mass, and mesenteric adipocyte size compared with HFD-fed mice. Vitamin C intake in obese mice also decreased the mRNA levels of lipogenesis-related genes (i.e., stearoyl-CoA desaturase 1 and sterol regulatory element-binding protein 1c) in mesenteric adipose tissues, inhibited hyperglycemia, and improved glucose tolerance. In addition, vitamin C attenuated the HFD-induced increase in the size of pancreatic islets. These results suggest that vitamin C suppresses HFD-induced visceral adipocyte hypertrophy and glucose intolerance in part by decreasing the visceral adipose expression of genes involved in lipogenesis.

Mesenteric Panniculitis in a Thirteen-Year-Old Korean Boy Treated with Prednisolone: A Case Report

  • Pediatric mesenteric panniculitis is an extremely rare disease of unknown etiology characterized by chronic inflammation, fat necrosis, and fibrosis in the mesenteric adipose tissue. A previously healthy 13-year-old boy was admitted because of right upper abdominal pain. An abdominal computed tomography scan revealed increased attenuation and enhancement in the left upper abdominal omental fat and anterior peritoneal wall thickening. A laparoscopic biopsy showed mesenteric panniculitis with chronic inflammation, adiponecrosis, and septal fibrosis. Serological tests for autoimmune diseases, nested polymerase chain reaction for Mycobacterium tuberculosis, and special immunohistochemical stains for malignancy were all negative. Symptomatic improvement and improved abnormal findings were achieved after an 8-month treatment with prednisolone according to a follow-up abdominal computed tomography scan. Here, we report a case of pediatric mesenteric panniculitis treated with prednisolone.

PARTITIONING OF LIPID IN THE BODY OF FAT-TAILED LAMBS AS INFLUENCED BY DOCKING AND SEX

  • Twelve docked and 12 intact Najdi lambs of equal numbers of males and females were slaughtered at 40 kg shorn shrunk body weight, Lipid in all empty body fat components; namely, subcutaneous, intermuscular, intramuscular, omental, mesenteric, channel, perirenal, pericardial, tail, viscera, bone and hide, were determined. Except for ram lambs, which had a lower percentage of lipid in intermuscular partition in loin cut, sex did not influence the relative proportion of subcutaneous or intermuscular fat in each wholesale cut. The data also showed that docking did not change the distribution of lipid in intermuscular and intramuscular fat partitions in each wholesale cut. Docked lambs tended to accumulate lower proportions of the lipid in subcutaneous fat component in the cuts located along the dorsal line than intact lambs. The total amount of lipid deposited in the empty body of ewe lamb was heavier in weight than that of ram lamb. Docking had no effect on the distribution of total lipid in the empty body, except for subcutaneous fat component, being greater in docked lambs than did intact ones. Generally, the highest proportion of fat was associated with subcutaneous depot followed, in order, by intermuscular, mesenteric, tail, intramuscular and omental fat partitions.

  • Primary mesenteric liposarcoma is rare. It is difficult to make an accurate preoperative diagnosis of the myxoid type of liposarcoma by using imaging such as ultrasound or computed tomography (CT) due to the very small amount of fat that is located in the tumor. We report a case of primary myxoid liposarcoma of the mesentery which was difficult to differentiate from other solid mesenteric tumors with a myxoid component such as low grade fibromyxoid sarcoma, myxoid leiomyosarcoma or myxoma. Use of chemical shift magnetic resonance (MR) imaging to detect small fat components and its cystic appearance with solid components on the MR images can be useful to differentiate myxoid liposarcoma from the other mesenteric tumors with a myxoid component.

복부의 T2강조 영상에서 지방소거기법의최적의 평가 (The Evaluation of Optimized Inversion-Recovery Fat-Suppression Techniques for T2-Weighted Abdominal MR Imaging : Preliminary report)

  • To test the real image quality of a spectral attenuated inversion-recovery (SPAIR) fat-suppression (FS) techniquein clinical abdominal MRI by comparison to turbo spin echo inversion-recovery (TSEIR) fat-suppression (FS) technique. 3.0T MRI studies of the abdomen were performed in 30 patients with liver lesions (hemangiomas n: 15; HCC n: 15). T2W sequences were acquired using SPAIR TSEIR. Measurements included retroperitoneal and mesenteric fat signal-to-noise (SNR) to evaluate FS; liver lesion contrast-to-noise (CNR) to evaluate bulk water signal recovery effects; and bowel wall delineation to evaluate susceptibility and physiological motion effects. SPAIR-TSEIR images produce significantly improved FS and liver lesion CNR. The mean SNR of the retroperitoneal and mesenteric fat for SPAIR were 20.5, 10.2 and TSEIR were 43.2, 24.1 (P<0.05). SPAIR-TSEIR images produced higher CNR for both hemangiomas CNR 164.88 vs 126.83 (P<0.05) and metastasis CNR 75.27 vs 53.19 (P<0.05). Bowel wall visualization was significantly improved using in both SPAIR-TSEIR (P< 0.05). The real image quality of SPAIR was better than over conventional TSEIR FS on clinical abdominal MRI scans.

Troglitazone Regulates white Adipose Tissue Metabolism by Activating Genes Involved in Fatty Acid ${\beta}$-Oxidation in High Fat Diet-fed C57BL/6J Mice

  • This study aimed to determine whether troglitazone stimulates genes related to fatty acid ${\beta}$-oxidation, leading to modulation of white adipose tissue (WAT) metabolism in high fat diet-fed mice. Female C57BL/6J mice were randomly divided into two groups (n=10/group). After they received either a high fat diet or the same high fat diet supplemented with troglitazone for 4 weeks, the effects of troglitazone on gene expression and physiology of WAT were measured using Northern, histological and serological analyses. Administration of troglitazone induced the expression of genes involved in mitochondrial and peroxisomal fatty acid ${\beta}$-oxidation in mesenteric WAT. Troglitazone also significantly increased uncoupling protein 2 mRNA levels. The changes in WAT gene expression were accompanied by reductions in circulating levels of free fatty acids and triglycerides as well as glucose and insulin. Histological studies showed that troglitazone treatment decreased the average size of adipocytes in mesenteric WAT. These results suggest that troglitazone-stimulated WAT expression of genes associated with fatty acid ${\beta}$-oxidation regulates WAT metabolism of high fat diet-fed mice, contributing to improvement of insulin sensitivity.

  • 4, 919 Samples of the slaughtered cattle(female) were investigated the abattoir in southern Kangwon to reveal the incidence rates of abdominal fat necrosis from June to December 1993. The results obtained were summarized as follows ; 1. The incidence rate of abdominal fat necrosis investigated from 4, 919 samples was 4.39%. 2. It revealed that the incidence rate of abdominal fat necrosis increased gradually with the advance in the age, 2.5% in below 5 years, 6.0% in 6 years, 7.2% in 7 years, 8.5% in 7 years, 8.9% in older than 9 years and that incidence rate of the sites of lesion, pericolonic fat 84.7%, perirectumic fat 48.6%, perirenal fat 37.9%. mesenteric fat 24.0%, others 7.8%. 3. The size of necrotic fat were $2{\times}3cm{\sim}10{\sim}18{\times}15{\sim}25cm$ in average and color was yellowish white or milk white.

Regulation of Lipoprotein Lipase by Fasting in Epididymal and Mesenteric Adipocytes of Rats

  • There are marked variations in the activity of lipoprotein lipase (LPL) among adipose depots. The aim of this study was to compare the mechanisms of 24 h of fasting on LPL regulation between epididymal (EPI) adipocytes and mesenteric (MES) adipocytes in rats. 1-Day fasting consistently decreased activities of heparin-releasable LPL, total extractable LPL and cellular LPL markedly in both EPI and MES fat pads. LPL activity in MES fat pads was relatively lower than in the EPI fat pads. Consistent with data on LPL activity, the levels of expression of LPL mRNA in both nutritional states were lower in MES than EPI adipose tissue and isolated adipocytes. The decreased LPL activity after 1 day of fasting in MES adipocytes was explained mainly by a 50% decrease in the relative abundance of LPL mRNA level and a parallel 50% decrease in relative rate of LPL synthesis. In contrast, fasting of 1 day in EPI adipocytes decreased total LPL activity by 47% but did not affect LPL mRNA level or relative rate of LPL synthesis. A decrease in overall protein synthesis contributed to the decreased LPL activity after 1 day fasting both in EPI and MES adipocytes. In MES adipocytes the decrease in LPL activity, LPL mRNA and LPL synthesis were comparable, but in EPI adipocytes the changes in LPL activity were substantially larger than the changes in LPL mRNA level and LPL synthesis. Therefore, fasting decreased fat cell size, LPL activity, LPL mRNA level and relative rate of LPL synthesis in rats, and these effects were more marked in the MES adipocytes. These results clearly demonstrate the regional variations in the metabolic response of adipose tissue and LPL functions to fasting.

삼채가 고지방식이를 급여한 흰쥐의 고지혈증 개선 효과에 미치는 영향 (The Hypolipidemic Effect of Allium Hookeri in Rats Fed with a High Fat Diet)

  • This study evaluated the effect of Allium hookeri roots on lipid metabolism of the serum, liver, and adipose tissues induced by a high-fat diet in male Sprague-Dawley rats. The rats were divided into four groups with 8 rats per group for 4 weeks: the normal-diet group (N), the high-fat diet (HFD) group, the HFD containing 3% Allium hookeri (HFD-A3) group, and the HFD containing 5% Allium hookeri (HFD-A5) group. The results showed that the body weight gain and food intake of rats in the HFC-A3 and HFC-A5 groups were significantly decreased compared with those in the HFD group. The epididymal adipose tissue weight in the HFD-A5 group was significantly decreased compared with those in the HFD group, and adipose tissue weights of liver and mesenteric adipose tissues in the HFD-A3 and HFD-A5 groups were significantly decreased compared with those in the HFD group. Serum triglyceride, total cholesterol and LDL cholesterol concentrations, atherogenic index, and cardiac risk factor were significantly lower in the HFD-A3 and HFD-A5 groups than in the HFD group. Serum lipid profiles, as well as ALT and AST activities did not show any difference in all groups. Serum ALP and LDH activities were suppressed in the HFD-A5 group compared with those in the HFD group. The hepatic triglyceride and total cholesterol levels of rats in the HFD-A5 group was significantly lower than those in the HFD group. Moreover, triglyceride and total cholesterol in the epididymal and mesenteric adipose tissues were significantly lower in the HFD-A5 group than in the HFD group. These results demonstrated that the intake of Allium hookeri showed a hypolipidemic effect, changing the lipid metabolsim of a high-fat diet induced rats.
