The Roles of the SNARE Protein Sed5 in Autophagy in Saccharomyces cerevisiae

  • Autophagy is a degradation pathway in eukaryotic cells in which aging proteins and organelles are sequestered into double-membrane vesicles, termed autophagosomes, which fuse with vacuoles to hydrolyze cargo. The key step in autophagy is the formation of autophagosomes, which requires different kinds of vesicles, including COPII vesicles and Atg9-containing vesicles, to transport lipid double-membranes to the phagophore assembly site (PAS). In yeast, the cis-Golgi localized t-SNARE protein Sed5 plays a role in endoplasmic reticulum (ER)-Golgi and intra-Golgi vesicular transport. We report that during autophagy, sed5-1 mutant cells could not properly transport Atg8 to the PAS, resulting in multiple Atg8 dots being dispersed into the cytoplasm. Some dots were trapped in the Golgi apparatus. Sed5 regulates the anterograde trafficking of Atg9-containing vesicles to the PAS by participating in the localization of Atg23 and Atg27 to the Golgi apparatus. Furthermore, we found that overexpression of SFT1 or SFT2 (suppressor of sed5 ts) rescued the autophagy defects in sed5-1 mutant cells. Our data suggest that Sed5 plays a novel role in autophagy, by regulating the formation of Atg9-containing vesicles in the Golgi apparatus, and the genetic interaction between Sft1/2 and Sed5 is essential for autophagy.

Identification of Genetic Variations in CBL, SORBS1, CRK, and RHOQ, Key Modulators in the CAP/TC10 Pathway of Insulin Signal Transduction, and Their Association with Type 2 Diabetes Mellitus in the Korean Population

  • Recent evidence has strongly suggested that the CAP/TC10 pathway is involved in the trafficking, docking, and fusion of vesicles containing the insulin-responsive glucose transporter Glut4 to the plasma membrane. However, little is known about how the genes employed in the CAP/TC10 pathway are associated with the development of type 2 diabetes mellitus. In this study, we sequenced 4 genes of the CAP/TC10 pathway [SORBS1, CBL, CRK, and RHOQ] in 24 individuals to identify genetic variations in these loci. A total of 48 sequence variants were identified, including 23 novel variations. To investigate the possible association with type 2 diabetes mellitus, 3 single nucleotide polymorphisms from SORBS1, 3 from CBL, and 4 from RHOQ were genotyped in 1122 Korean type 2 diabetic patients and 1138 nondiabetic controls. Using logistic regression analysis, 1 significant association between SNP rs1376405 in RHOQ and type 2 diabetes mellitus [OR = 8.714 (C.I. 1.714-44.29), p = 0.009] was found in the recessive model. Our data demonstrate a positive association of the RHOQ gene in the CAP/TC10 pathway with T2DM in the Korean population.

RPK118, a PX Domain-containing Protein, Interacts with Peroxiredoxin-3 through Pseudo-Kinase Domains

  • RPK118 is a sphingosine kinase-1-binding protein that has been implicated in sphingosine 1 phosphate-mediated signaling. It contains a PX (phox homology) domain and two pseudo-kinase domains, and co-localizes with sphingosine kinase-1 on early endosomes. In this study we identified a novel RPK118-binding protein, PRDX3 (peroxiredoxin-3), by yeast two-hybrid screening. The interaction between these proteins was confirmed by pull-down assays and co-immunoprecipitation experiments. Deletion studies showed that RPK118 interacted with PRDX3 through its pseudokinase domains, and with early endosomes through its PX domain. Double immunofluorescence experiments demonstrated that PRDX3 co-localized with RPK118 on early endosomes in COS7 cells. PRDX3 is a member of the antioxidant family of proteins synthesized in the cytoplasm and functioning in mitochondria. Our findings indicate that RPK118 is a PRDX3-binding protein that may be involved in transporting PRDX3 from the cytoplasm to its mitochondrial site of function or to other membrane structures via endosome trafficking.

Surface expression of TTYH2 is attenuated by direct interaction with β-COP

  • TTYH2 is a calcium-activated, inwardly rectifying anion channel that has been shown to be related to renal cancer and colon cancer. Based on the topological prediction, TTYH2 protein has five transmembrane domains with the extracellular N-terminus and the cytoplasmic C-terminus. In the present study, we identified a vesicle transport protein, ${\beta}$-COP, as a novel specific binding partner of TTYH2 by yeast two-hybrid screening using a human brain cDNA library with the C-terminal region of TTYH2 (TTYH2-C) as a bait. Using in vitro and in vivo binding assays, we confirmed the protein-protein interactions between TTYH2 and ${\beta}$-COP. We also found that the surface expression and activity of TTYH2 were decreased by co-expression with ${\beta}$-COP in the heterologous expression system. In addition, ${\beta}$-COP associated with TTYH2 in a native condition at a human colon cancer cell line, LoVo cells. The over-expression of ${\beta}$-COP in the LoVo cells led to a dramatic decrease in the surface expression and activity of endogenous TTYH2. Collectively, these data suggested that ${\beta}$-COP plays a critical role in the trafficking of the TTYH2 channel to the plasma membrane.

Peptides derived from high voltage-gated calcium channel β subunit reduce blood pressure in rats

  • The β subunits of high voltage-gated calcium channels (HGCCs) are essential for optimal channel functions such as channel gating, activation-inactivation kinetics, and trafficking to the membrane. In this study, we report for the first time the potent blood pressure-reducing effects of peptide fragments derived from the β subunits in anesthetized and non-anesthetized rats. Intravenous administration of 16-mer peptide fragments derived from the interacting regions of the β1 [cacb1(344-359)], β2 [cacb2(392-407)], β3 [cacb3(292-307)], and β4 [cacb4(333-348)] subunits with the main α-subunit of HGCC decreased arterial blood pressure in a dose-dependent manner for 5-8 min in anesthetized rats. In contrast, the peptides had no effect on the peak amplitudes of voltage-activated Ca2+ current upon their intracellular application into the acutely isolated trigeminal ganglion neurons. Further, a single mutated peptide of cacb1(344-359)-cacb1(344-359)K357R-showed consistent and potent effects and was crippled by a two-amino acid-truncation at the N-terminal or C-terminal end. By conjugating palmitic acid with the second amino acid (lysine) of cacb1(344-359)K357R (named K2-palm), we extended the blood pressure reduction to several hours without losing potency. This prolonged effect on the arterial blood pressure was also observed in non-anesthetized rats. On the other hand, the intrathecal administration of acetylated and amidated cacb1(344-359)K357R peptide did not change acute nociceptive responses induced by the intradermal formalin injection in the plantar surface of rat hindpaw. Overall, these findings will be useful for developing antihypertensives.

Purinergic regulation of calcium signaling and exocytosis in rat prostate neuroendocrine cells

  • Prostate gland contains neuroendocrine cells (PNECs) are playing important roles in physiological and pathophysiological processes of the prostate gland. Here, we investigated the role of purinoceptors in PNECs freshly isolated from rat ventral prostate (RPNECs) that show immunoreactivity to chromogranin A. Fura-2 ratiometry revealed that ATP evokes both fast Ca$\^$2+/ influx and store Ca$\^$2+/ release in RPNECs. A whole-cell patch clamp study demonstrated fast inactivating cationic current activated by ATP or by ${\alpha}$,${\beta}$-MeATP, which was blocked by ATP-TNP. The activation of P2X inward current was tightly associated with a sharp increase in [Ca$\^$2+/]$\sub$c/. The presence of P2X1/3 subtypes were proved by RT-PCR analysis. For the stored Ca$\^$2+/ release, ATP and UTP showed similar effects, suggesting the dominant role or P2Y2 subtypes, also confirmed by RT-PCR. Both P2X (${\alpha}$,${\beta}$-MeATP) and P2Y (UTP) stimulation induced changes in the cell morphology (initial shrinkage and blob formation on the surface) reversibly. Exocytotic membrane trafficking events were monitored with the membrane-bound fluorescent dye, FM1-43 using confocal microscopy. In spite of the similar Ca$\^$2+/ responses, UTP was far less effective in triggering exocytosis than ${\alpha}$,${\beta}$ -MeATP. Since serotonin is reportedly stored in the secretory granule of PNECs, we directly examined whether the aforementioned agonists elicit release of serotonin using carbon fiber electrode-amperometry. In accordance with the results of FM1 -43 experiments, ${\alpha}$,${\beta}$-MeATP efficiently evoke serotonin secretion while not with UTP. In summary, the P2X-mediated Ca$\^$2+/ influx plays crucial roles in the exocytosis of RPNECs. Although a global increase in [Ca$\^$2+]$\sub$c/ might be related with the morphological changes, a sharp rise of [Ca$\^$2+/]$\sub$c/ in the putative sub-plasmalemmal ‘microdomains’ might be a decisive factor for the exocytosis.

Molecular Characterization and Expression Analysis of Clathrin-Associated Adaptor Protein 3-δ Subunit 2 (AP3S2) in Chicken

  • 닭의 clathrin-associated adaptor protein $3-{\delta}$ subunit 2(AP3S2)는 clathrin-coated vesicle를 가진 표적 세포막으로 암 배양 단백질 수송에 관여한다. AP3S2는 C형 간염 바이러스 감염으로 간 섬유화를 매개하고, 2형 당뇨병과 관련이 있는 것으로 알려져 있다. 또한, AP3S2는 clathrin-dependent endocytosis를 통해 숙주 세포로의 바이러스 유입에 관련된 역할을 하는 것으로 알려져 있다. 본 연구는 기존 연구에서 닭 신장조직에서 차별 발현 유전자로 발굴된 닭 AP3S2 유전자의 분자유전학적 특성을 구명하고, 닭의 조직에서의 유전자 발현 양상을 조사하며, 톨-유사수용체 3 (Toll-like receptor 3; TLR3) 자극에 의한 전사 조절을 연구하였다. 닭 AP3S2 유전자가 코딩하는 단백질의 구조는 다른 종과 매우 보존적이고 진화적으로 제브라 피쉬와 가장 가깝고, 포유류와 가장 먼 것으로 추정되었다. 닭의 다양한 조직에서 닭 AP3S2 유전자의 전사 수준을 조사한 결과, 폐에서 가장 높게 발현되었으며, 그 다음은 비장 순이었다. 닭의 배아 섬유아세포 주인 DF-1세포에서 조사한 결과, AP3S2 유전자의 발현은 TLR3 신호자극에 의해 감소하였다. 전사조절인자인 $NF{\kappa}B$나 AP-1의 억제제를 이용하여 조사한 결과, $NF{\kappa}B$나 AP-1의 억제에 의해 유전자 발현이 영향을 받지 않았다. 이 결과는 DF-1 세포에서 닭 AP3S2 유전자의 발현은 적어도 이 두 전사조절인자와는 독립적인 경로에 의해 조절됨을 시사한다. 본 연구의 결과는 닭 AP3S2가 바이러스 감염에 역할을 하고, TLR3 신호에 관여함을 제시한다. 추가연구를 통해 닭 AP3S2의 전사 조절과 바이러스 침입 메커니즘을 구명할 필요가 있다고 사료된다.

인체 폐암조직에서 Phospholipase C 동위효소의 발현양상 (Expression of Phospholipase C Isozymes in Human Lung Cancer Tissues)

  • 배경 : Phospholipase C는 세포내 신호 전달과정의 초기 단계에 있어서, 매우 중요한 역할을 하는 효소로 알려져 있으며, 세포막에 존재하는 인지질 가운데 중요한 부분을 차지하는 $PIP_2$를 분해하여 제2전령물질인 DAG와 IP3를 생성한다. 이들 제2전령물질은 각각 PKC를 활성화시키고, 세포내의 $Ca^{2+}$농도를 증가 시켜서 세포내 여러 단백질 효소들을 활성화시키는 동시에, PLC 효소는 자체적으로 갖고 있는 고유의 SH2, SH3 및 PH domains 등의 기능 영역을 통해, 다른 신호 전달 인자들과 상호 작용하고, 세포막의 재구성이나, 세포분열, 발안과정에 관여 하게 된다. 이에 저자 등은 이전의 연구에서, 인체 정상 폐조직에서 PLC-${\beta}1$, -${\beta}3$, -${\gamma}1$ 및 -${\delta}1$ 동위효소가 존재함을 보고하였으며, bovine lung 내에서 PLC-${\gamma}1$ 동위 효소를 활성화 시키는 AHNAK 단백을 분리, 정제 및 클로닝 하였고, 이 AHNAK 단백이 인체 폐암조직내에서 정상조직에 비해 증가 되어 있음을 보고하여, 폐암의 발암과정에 있어서 칼슘-inositol 신호전달체계의 이상이 연관 되어있음을 제시하였다. 하지만 아직 인체 폐암조직이나 다른 종류의 폐질환에 대해서는, PLC 동위효소의 발현양상에 대한 보고가 없었으므로, 이에 저자 등은 수술로 적출한 인체 폐암 조직 내에서 PLC 동위 효소의 발현을 연구하여, 폐암의 발암과정에서 이들 효과가 갖는 역할을 규명하고자 하였다. 대상 및 방법 : 아주 대학교 병원에 내원하여 원발성 폐암으로 수술적 절제술을 받은 환자중에서, 신선냉동상태의 암조직과 동일환자의 정상폐조직이 확보가, 가능했던 37예의 환자를 대상으로 하였다. 이들조직을 대상으로 PLC-${\beta}1$, -${\beta}3$, -${\gamma}1$ 및 -${\delta}1$ 동위효소에 대해 Western blot 분석을 시행하였고, 대표적인 표본에 대해서는 PLC-${\gamma}1$에 대한 면역조직화학검사를 시행하였다. 결과 : 연구의 대상이었던 15예의 선암조직 모두에서, PLC-${\gamma}1$ 동위효소의 과발현을 관찰할 수 있었으며, 편평상피세포암 19예 중 16예에서 PLC-${\gamma}1$의 발현이 정상조직에 비해 증가하였음이 확인되었다. PLC-${\delta}1$ 동위효소의 경우, 대부분의 폐암조직에 감소되어 있었다. 하지만 이와는 반대로, 일부 선암 및 편평상피 암 조식(각3예)에서는, 현저한 증가를 보이기도 했다. 또한, 비록 그 증례수가 적기는 하였지만, 소세포 폐암 4예에서는, 모두에서 정상 폐조직보다, PLC-${\delta}1$ 효소의 발현이 현저히 감소되어 있음을 관찰하였다. 결론 : 이상의 결과로 미루어 폐암 조직 내에서 PLC-${\gamma}1$ 동위효소의 발현이 증가되어 있었는데, 이는 저자등이, 이미 보고한바 있는, PLC-${\gamma}1$의 활성화 AHNAK의 과발현과 함께, 폐암의 발암과정에, 칼슘-inositol 신호전달 채계의 이상이 관여할것이라는 실험적인 증거가 될수 있다고 하겠다. 하지만, PLC-${\delta}1$ 동위효소의 감소에 대해서는, 좀더 구체적인 기전의 규명 및 PLC-${\delta}1$ 효소의 역할에 대한 보다 많은 연구가 필요할것으로 사료된다.
