In order to know the antifungal characteristics of saturated fatty acids having 6 to 12 carbons, their minimum inhibitory concentrations (MICs) and minimum fungicidal concentrations (MFCs) were estimated against Saccharomyces cerevisiae. Fatty acids from C6 to C11 exhibited increasing activity with chain length, but C12 fatty acid did not show activity at all. In relation to antifungal modes of actions, fatty acids investigated showed on inhibitory activity toward the plasma membrane H+-ATPase of Saccharomyces cerevisiae. Their inhibitions to the glucose-induced acidification and ATP hydrolysis caused by the proton pump were found to be in common wiht antifungal activities. At the test concentration of 1mM, hexanoic acid (C6) showed the lowest inhibition of about 30%, while undecanoic acid(C11) showed the strongest inhibition of over 90%. In addition, as seen with antifungal activity, the inhibitory activity of dodecanoic acid (C12) was suddenly reduced to less than 50%.
This study was conducted to evaluate dietary fat intake and its effect on the plasma lipids and fatty acids composition in plasma and red blood cells(RBC) in 96 healthy Korean female college student. Three-day food intakes were recorded, and fasting blood samples were collected and analyzed for plasma total cholesterol and triglyceride. Fatty acid compositions were determined in plasma and RBC membrane. Oleic acid was the most abundant in diet, followed by palmitic and linoleic acids. Mean daily intake of cholesterol was 219$\pm$127mg, mean plasma cholesterol was 160$\pm$24mg/이 and mean plasma triglyceride was 68$\pm$25mg/dl. Plasma fatty acids were mostly composed of linoleic, palmitic and oleic acids, while palmitic, stearic and arachidonic acids were high in RBC membrane. Plasma triglyceride showed positive correlation with BMI. Among dietary fatty acids, arachidonic acid, EPA and DHA showed negative correlation with plasma total cholesterol. Plasma triglyceride levels were negatively correlated with dietary arachidonic acid, plasma n-6 fatty acids and plasma polyunsaturated fatty acids. Dietary EPA and DHA levels were positively correlated with plasma EPA, dietary n-3/n-6 ratio were positively correlated with plasma n-3 fatty acids and n-3/n-6 ratio. Highly significant correlations were shown between the levels in plasma and RBC for several fatty acids.

Lipid composition and fatty acid composition were characterized in the membrane of a marine ciliate protozoan (Uronema marinum). Phospholipids accounted for 70% of total lipid, and the remainder was neutral lipids. Total phospholipids were separated as phosphatidylcholine $(24.26\%)$, phosphatidylethanolamine $(22.21\%)$, phosphatidylinositol $(6.14\%)$, phosphatidylserne $(5.11\%)$, cardiolipin $(3.07\%)$ and unidentified phospholipids $(28.72\%)$ through high performance liquid chromatography (HPLC). Fatty acid composition of neutral lipids and phospholipids was determined by gas chromatography (GC), based solely on comparision of retention times. In neutral lipids, the most abundant fatty acid group was monounsaturated fatty acid $(48.3\% of total fatty acids)$ with oleic acid (18:1) and nervonic acid (24:1). Saturated fatty acids comprised $29.6\%$ of total fatty acids, with palmitic acid (16:0), stearic acid (18:0) ane myristic acid (14:0), and polyunsaturated fatty acid accounted for $33.0\%$ with $Di-homo-\gamma-linolenic$ acid (20:3) and linoleic acid (18:2). Wherease phospholipids predominantly contained the fatty acid group in the following order: polyunsaturated fatty acids $(52.7\%\;of\;total\;fatty\;acids)$ with linoleic acid (18:2) and $\gamma-linolenic$ acid (18:3) > monounsaturated fatty acids $(28.5\%\;of\;total\;fatty\;acids)$ with oleic acid (18:1) and palmitoleic acid (16:1) > saturated fatty acids $(25.5\%\;of\;total\;fatty\;acids)$ with palmitic acid (16:0), stearic acid (18:0) and myristic acid (14:0).
휘발성 지방산의 생성 및 회수를 위한 분리막의 유효성을 실험적으로 검토한 결과, 분리막의 적용에 의해 발효조 내의 부유고형물 농도, 유기산 생성균수 및 유기산 농도가 증가하였다. 혐기성 발효액의 고액분리 및 발효 효율향상을 위한 분리막의 적용은 발효조 내의 관련 미생물 농도를 증가시키고 따라서 분리막을 적용하지 않은 경우에 비해 유기산 생성효율이 훨씬 증가하였다. 분리막이 결합된 산 발효조의 적용은 유기성 슬러지로부터 휘발성 지방산의 회수 효율증대에 효과적인 적용기술이라 판단된다.
To analyze the effects of surfactants on the biosynthesis of galactolipid and the composition of fatty acids, the chloroplast envelope and thylakoid membrane were cultivated in medium treated with anionic surfactants, such as linear alkylbenzene sulfonate (0.002%, LAS), a-olefin sulfonate (O.01%, AOS), and sodium lauryl ether sulfate (0.08%, SLES), respectively. During the cultivation, the chloroplast envelope and thylakoid membrane were isolated from the cells collected at the early and middle phase of the culture and the contents of their fatty acid composition were compared with the control. When treated with surfactants, the contents of total lipid MDGD methylesters, and DGDG methylesters decreased significantly when compared with the control. It was also confirmed that more unsaturated fatty acids were involved in the biosynthesis of galactolipid. The fatty acids utilized in the biosynthesis of MGDG were in the chloroplast envelope and in the control, and linoleic acid in LAS, linolenic acid and oleic acid in AOS, and linolenic acid and oleic acid in SLES. The fatty acids in the biosynthesis of DGDG were linolenic acid and oleic acid in the control linolenic acid and stearic acid in LAS, oleic acid and linolenic acid in AOS, oleic acid and linolenic acid in SLES. In the thylakoid membrane, the major fatty acids in the biosynthesis of MGDG were linolenic acid and oleic acid in the control, oleic acid and linolenic acid in LAS, linolenic acid and linoleic acid in AOS, linolenic acid and palmitoleic acid in SLES. The fatty acids in the biosynthesis of DGDG were linolenic acid and oleic acid in the control, oleic acid and linolenic acid in LAS, linolenic acid and linoleic acid in AOS, palmitoleic acid and oleic acid in SLES.
Path of a small hydrophobic molecule through the aqueous cytoplasma is not linear. Partition may favor membrane binding by several orders of magnitude : thus significant membrane association will markedly decrease the cytosolic transport rate. The presence of high concentration of soluble binding proteins for these hydrophobic molecules would compete with membrane association and thereby increase transport rate. For long chain fatty acid molecules, a family of cytosolic binding proteins collectively known as the fatty acid binding proteins(FABP), are thought to act as intracellular transport proteins. This paper examines the mechanism of transfer of fluorescent antyroyloxy-labeled fatty acids(AOFA) from purified FABPs to phosholipid membranes. With the exception of the liver FABP, AOFA is transferred from FABP by collisional interaction of the protein with a acceptor membrane. The rate of transfer increased markedly when membranes contain anionic phospholipids. This suggests that positively charged residues on the surface of the FABP may interact with the membranes. Neutralization of the surface lysine residues of adipocyte FABP decreased fatty acid transfer rate, and transfer was found to proceed via aqueous diffusion rather than collisional interaction. Site specific mutagenesis has further shown that the helix-turn-helix domain of the FABP is critical for interaction with anionic acceptor membranes. Thus cytosolic FABP may function in intracellular transport of fatty acid to decrease their membranes association as well as to target fatty acid to specific subcellular sites of utilization.
Modifications in dietary fatty acid intake might lead to a modification in membrane phospholipid fatty acid composition. The purpose of this study was to investigate relationship between different type of oil consumption and leukocyte membrane phospholipid composition. This study was carried out in subjects utilizing butter (n = 15), margarine (n = 15), fluid oil (n = 15) and mixed types of oils (n = 15) in total 60 subjects. Leukocytes were separated from total blood by dextran sedimentation method. Membrane lipids and proteins were isolated following the cell disruption. Fatty acids of membrane phospholipids were isolated by hydrolysation with phospholipase B under ultrasonic dismembranator. Free fatty acids were identified with gas chromatography at chloroform phase. The results obtained were compared with data obtained by chromatograms of the standards. Results more prominent values of arachidic, dihomo-$\gamma$-linolenic and palmitoleic acids were found in butter-or mixed oil-user groups; eicosadienoic, eicosamonoenoic, dihomo-$\gamma$-linolenic and behenic acids in fluid oil; heptanoic, valeric, eicosadienoic and linolenic acids in margarine groups. The fatty acid composition of mixed oil was similar to butter, while other two oils were so different. From this study, it was concluded that the type of oil consumption might have an influence on phospholipid components of plasma membranes.
This study was conducted to asses the $\omega$-3 fatty acid intake levels and to evaluate the relationship between the levels of $\omega$-3 fatty acids in plasma and erythrocyte membrane and the dietary intake of these fatty acids over the period of two months in 56 young Korean females. Dietary survey was conducted to obtain 7-day weiged records and six 24-hour recalls. Fasting blood sample was collected from each subject after the dietary survey period. Mean daily intakes of energy, fat, and cholesterol were 1,569㎉, 41.8g, and 217mg, respectively. Fat suppled 24% of total energy intake. Mean daily intake levels of eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA), and total $\omega$-3 fatty acids were 0.04g, 0.06g, and 0.48g, respectively. Plasma cholesterol levels of most of the subjects were within normal range, and there was no significant correlation between plasma cholesterol levels and intake levels of any specific fatty acid. Levels of EPA, DHA, EPA+DHA in plasma and erythrocyte membrane, but not the levels of $\alpha$-linolenic acid(LNA), were significantly correlated with dietary in take of respective fatty acids. Such a correlation, however, was not observed in a previous study where dietary intake was assessed for 3 days. The results of this study show that dietary intakes of $\omega$-3 fatty acids are low in the subject and that about two-month period is required to assess dietary intake levels of $\omega$-3 fatty acids with a reasonable accuracy.
Recent research results regarding the very long chain transmembrane ${\alpha},{\omega}-dicarboxylic$ components in the membrane of extremophilic eubacteria, such as Sarcina ventriculi, Thennotoga maritima, and Thermoanaerobacter ethanolicus have raised interesting questions concerning the physical and biochemical function on these components in the membrane. In order to understand the dynamic characteristics of these acids which reside in the bilayer membrane, 580 ps molecular dynamic simulations at 300 K were performed for two model systems. These systems were the bilayer with regular chain (C16:0 or C18:1) fatty acid methyl esters and the fatty acid bilayer containing very long chain transmembrane dicarboxylic acid methyl esters (${\alpha},{\omega}-15,16-dimethyltriacotane-dioate$ dimethyl ester; C32:0). Our analyses indicate that very long chain transmembrane dicarboxylic acids have a noticeable influence on the bilayer dynamics at a sub-nanosecond time scale. The center-ofmass mean-squared-displacement (MSD) of regular chain fatty acids adjacent to the very long chain transmembrane dicarboxylic acids decreased, the long-axis order parameter increased, and the reorientational motions of methylene groups were slowed along the hydrocarbon chains. These results indicate that the very long chain transmembrane dicarboxylic acids reduce the molecular order of the whole bilayer membrane.
The cellular fatty acid composition is important for metabolic plasticity in Rhodobacter sphaeroides. We explored the effects of changing the cellular ratio of unsaturated fatty acids (UFAs) to saturated fatty acids (SFAs) in R. sphaeroides by overexpressing several key fatty acid biosynthetic enzymes through the use of expression plasmid pRK415. Bacteria containing the plasmid pRKfabI1 with the fabI1 gene that encodes enoyl-acyl carrier protein (ACP) reductase showed a reduction in the cellular UFA to SFA ratio from 4 (80% UFA) to 2 (65% UFA) and had decreased membrane fluidity and reduced cell growth. Additionally, the ratio of UFA to SFA of the chromatophore vesicles from pRKfabI1-containing cells was similarly lowered, and the cell had decreased levels of light-harvesting complexes, but no change in intracytoplasmic membrane (ICM) content or photosynthetic (PS) gene expression. Both inhibition of enoyl-ACP reductase with diazaborine and addition of exogenous UFA restored membrane fluidity, cell growth, and the UFA to SFA ratio to wild-type levels in this strain. R. sphaeroides containing the pRKfabB plasmid with the fabB gene that encodes the enzyme β-ketoacyl-ACP synthase I exhibited an increased UFA to SFA ratio from 4 (80% UFA) to 9 (90% UFA), but showed no change in membrane fluidity or growth rate relative to control cells. Thus, membrane fluidity in R. sphaeroides remains fairly unchanged when membrane UFA levels are between 80% and 90%, whereas membrane fluidity, cell growth, and cellular composition are affected when UFA levels are below 80%.
