Aspect of Incidence of the Major Citrus Diseases Recently (최근 주요 감귤 병의 발생 양상)

  • The percent of diseased fruit by Diaporthe citri was the most 16.8% in 2010 and the least 3.8% in 2005, and the average was 8.8% from 2003 to 2012. The degrees of disease incidence were relatively high, 17.0, 22.6, 19.2 and 18.9%, in 2003, 2004, 2007 and 2010, respectively. The percent of diseased fruit by Elsinoe fawcettii was the most 0.72% in 2004, and then gradually decreased to 0.08% in 2008, and the average was from 0.28% from 2004 to 2011. In case of canker by Xanthomonas citri subsp. citri, the percent of diseased fruit was the most 2.19% in 2004, and gradually decreased to 0.08 and 0.10% in 2009 and 2010, respectively. The average was 0.81% from 2004 to 2012. The melanose by D. citri was most in south-east area of Jeju Island, and east and west in case of canker. The melanose symptoms were initially appeared from mid June, mainly peaked late of July to late August. The canker symptoms were initially produced on leaves of natsudaidai and grapefruit cultivated in open field middle of June in 2010 and 2011 and late of May in 2012. The scab disease was firstly appeared approx. early or mid May and then rapidly increased late of May to middle of June. The inoculum was mainly produced from dead twigs late June to early of August.

Sporulation and Dissemination of Pycnidiospores of Diaporthe citri in Yuzu Tree (Citrus junos Sieb) in Jeonnam Area (전남지역 유자과원의 검은점무늬병균 포자 형성과 비산)

  • Several time-course experiments were carried out to understand seasonal development of melanose on yuzu trees at koehung area, Jeonnam province, during May to October. The occurrence of dead twigs, known as a source of infection, was much more in older trees, and from June to August, mostly in July. In the experiment of pycnidia development on dead twigs seasonally collected, the number of developed pycnidia was highest on July-collected dead twigs especially with the diameter of 1.1~1.5 cm. In the collection survey of disseminated pycnidiospores, although the collected number of pycnidiospores was affected with amount of precipitation, the number of observed pycnidiospores in rainwater was relatively high from June to August, with highest in early August in 1997 and late July in 1998. In the inoculation tests on 3-year-old trees and fruits in natural condition, disease occurrences were mostly affected on twigs by inocula treatment in June, and on fruits by inocula treatment in July, respectively.

Chemical Resistance of Diaporthe citri against Systemic Fungicides on Citrus

  • Citrus melanose, caused by Diaporthe citri, has been one of the serious diseases, and chemical fungicides were used for protection in many citrus orchards of Jeju Island. Establishing a disinfectant resistance management system and reducing pesticide usage would be important for contributing to safe agricultural production. In this study, monitoring of chemical resistance was performed with 40 representative D. citri isolates from many citrus orchards in Jeju Island. Four different fungicides, kresoxim-methyl, benomyl, fluazinam, and prochloraz manganese, with seven different concentrations were tested in vitro by growing the mycelium of the fungal isolates on the artificial medium potato dextrose agar. Among the 40 fungal isolates, 12 isolates were investigated as resistant to kresoxim-methyl which could not inhibit the mycelium growth to more than 50%. Especially isolate NEL21-2 was also resistant against benomyl, whose hyphae grew well even on the highest chemical concentration. However, any chemical resistance of fungal isolates was found against neither fluazinam nor prochloraz manganese. On the other hand, in vivo bio-testing of some resistant isolates was performed against both kresoxim-methyl and benomyl on young citrus leaves. Typical melanose symptoms developed on the citrus leaves pre-treated with both agrochemicals after inoculation with the resistant isolates. However, no or less symptoms were observed when the susceptible isolates were inoculated. Based on these results, it was suggested that some resistant isolates of D. citri occurred against both systemic fungicides, which may be valuable to build a strategy for protecting citrus disease.

Current Status of the Occurrence of Mandarins in Korea (우리나라 감귤 주요 병의 최근 발생 동향)

  • 본 연구는 우리 나라 감귤에 발생하는 주요 병의 최근 발생 동향을 파악하기 위하여 수행되었다. 1995년과 1996년 조사 포장에서 12종의 병해가 확인되었는데 그 중에서 검은점무늬병, 더뎅이병 및 잿빛곰팡이병이 감귤 재배에서 주요 병해로 밝혀졌다. 1995년과 1996년 무방제 포장에서 조사한 더뎅이병의 이병과율은 89.0∼100%, 검은점무늬병의 이병과율은 99.0∼99.5%인 반면에 잿빛곰팡이병의 이병과율은 23.0∼24.0%였다. 한편 더뎅이병과 검은점무늬병은 발병율이 높음에도 불구하고 약제 방제가 비교적 잘 되지만 잿빛곰팡이병은 노지에 비해 비닐하우스에서 발생이 많고 약제 방제가 잘 되지 않음을 알 수 있었다. 과거 주요 병해였던 궤양병은 지금은 경미한 피해를 주는 병해로 전환되어 온주밀감 수출단지가 아닌 일부 재배지에서만 경미하게 발생하고 있음이 확인되었다.

Effects of Dryness, Moisture Interruption, and Temperature on Germination of Diaporthe citri Pycnidiospores on Yuzu (건조, 습윤중단 및 온도가 유자 검은점무늬병원균 Diaporthe citri 포자 발아에 미치는 영향)

  • Melanose caused by Diaporthe citri is a serious disease on yuzu fruits. Moisture is the most important to infect after the pycnidiospores are released. In order to understand how dryness and moisture interruption affect on germination, the conidial suspensions on slide glass were dried for 0-48 h. For the moisture interruption experiment, moisture was supplied for 10 h then interrupted for 0-6 h depending on the treatments. Germinations on the treatments with longer than 15 h of dryness were less than 10%. And those with longer than 3 h of moisture interruption were less than 30%. Compared with other fungal phytopathogens, D. citri was xero-sensitive. Germination on yuzu feel medium was measured under 15, 20, 25, 30, $35^{\circ}C$ with 3, 6, 9, 12, 15, 18, and 24 h of wetness. The minimum wetness periods achieving 10% and 50% germinations were 4.5 and 13 h, and the optimum temperatures were $29.1^{\circ}C$ and $29.2^{\circ}C$, respectively. From the yuzu medium study, the regression equations to be required wetness period achieving 10% and 50% germinations on various temperatures were $Wh_{50%}=0.1093{\times}T^2-6.3762{\times}+106.08$ and $Wh_{10%}=0.082{\times}T^2-4.8025{\times}T+74.861$.

Induction of pathogenicity mutants from Elsinoe fawcettii, the causal fungus of citrus scab by genetic transformation

  • Genetic transformation carried out to induce the pathogenicity mutants from the two isolates, Elsinoe fawcettii R-34 and MUD of citrus scab fungus to hygromycin resistant by transferring plasmides (pUCATPH) that contain hygB gene. We produced protoplast for transformation by using of combinations of available enzymes including ${\beta}$-D-glucanase, ${\beta}$ -glucuronidase, Iyticase and driselase. The protoplasts regenerated at 64 $\mu\textrm{g}$/ml of hygromycin B but not 128 $\mu\textrm{g}$ in sensitivity test to identify the concentration of useful marker for the selection of transformants. Approximately 1200 and 67 hygromycin resistant isolates from strain R-34 and strain MUD, respectively, were isolated on PDA added with 200 $\mu\textrm{g}$ /ml of hygromycun B. Fifty seven and 4 of hygromycin resistant isolates from strain R-34 and MUD, respectively, did not produce necrotic lesions on the leaf in detached-leaf assay. Finally, 9 isolates were isolated from strain R-34, and these Isolates produced non or very few symptoms on seedlings of citrus in greenhouse pathogenicity test. And it's very interesting that some isolates produced melanose-like symptom on very young leaves which it was not typical symptom and somtimes produced on only expanded leaf.

A New Early Maturing Satsuma Mandarin Cultivar, 'Haryejosaeng' (온주밀감 신품종 '하례조생' 육성)

  • A new early maturing satsuma mandarin (Citrus unshiu Marc.) 'Haryejosaeng' was developed as a nucellar seedling selection of 'Tachima Wase' (C. unshiu Marc.) that was crossed with C. natsudaidai Hayata at the National Institute of Subtropical Agriculture in Jeju island in 1992. The 'Jegam ga No. 2', a first selection from the seedlings fruited in 2001 was finally named "Haryejosaeng" through field evaluation trials at three locations in Jeju island from 2003 to 2004. 'Haryejosaeng' produces seedless fruit maturing in early November, and has higher soluble solids and lower acidity than 'Miyagawa Wase', the leading early-maturing satsuma mandarin cultivar in Jeju island. Fruit weight is moderate at about 80~90 g and its shape is compressed-oblate globose with a light orange color. The rind thickness of about 2 mm provides easy peeling. The flesh shows light orange colored and contains 10 to 11 Brix and 1 to 1.1% acidity when mature. Tree shows vigorous growth and spreading thornless twigs. Alternate bearing is similar to 'Miyagawa Wase'. 'Haryejosaeng' is susceptible to citrus scab disease and melanose, but resistant to citrus canker.

Effective Usage of Copper Fungicides for Environment-friendly Control of Citrus Diseases (친환경적 감귤 병 방제를 위한 구리제의 효율적 사용)

  • This studies were carried out to elucidate the control effect of copper fungicides against some citrus diseases, assess copper spray injury on leaf and fruit of citrus by spraying of copper fungicides according to growth stages and spray seasons and by tank mixing with paraffin oil in field experiments for 3 years (2003-2005 season). As results, the control value was approximately $75\%$ against scab disease in all tested copper fungicides, $85-88\%$ to canker and $75-86\%$ to melanose. In case of leaf, the copper injury was the severest in early stage of shooting, and the injury was gradually weakened according to growth of leaf. In fruit, the copper injury was more severe in midsummer (late of July and August) than early of summer and spring season (June and ealy of July). Among copper fungicides, the bordeaux mixture which made by level of farm induced more injury compared to other copper fungicides commercialized. Less copper spray injury was observed on treatments in which copper fungicide was sprayed as tank mixtures with paraffin oil when compared to treatment which sprayed with only copper fungicide.
