Malignant melanoma in a grey pony, Korea (회색 조랑말에서 발생한 악성 흑색종 증례)

  • A 16 year-old female grey pony was presented to Animal and Plant Quarantine agency for diagnosis in Feb 2, 2015. At necropsy, multiple pigmented masses, likely melanomas, were detected peri-anally and under the tail. Further metastatic spread to the spleen, liver, lung and lymph nodes was also identified. Histopathologically, anaplastic and pleomorphic melanocytes were observed in the mass. By immunohistochemistry, PNL2, S100 and PGP 9.5 protein were detected, but Melan A was not expressed in the neoplastic melanocytes. Based on the pathological and immunohistochemical examination, we diagnosed this case as malignant melanoma in a grey pony. To the authors' knowledge, this is the first report of equine malignant melanoma in republic of Korea.

  • A 8-year-old, intact male Yorkshire Terrier Dog was presented with dark-black mass on the third digits of the left forelimb. Three months earlier, the dog had experienced an episode of digit amputation because of growing mass with ulcerated nodule of the same area. According to the owner, the recurrence of the mass appeared suddenly and was growing rapidly from the amputation site. The mass was more infiltrative than the first one and measured 1.5 cm in diameter. The clinical signs were anorexia, coughing, respiratory distress, exercise intolerance, cardiac murmurs, and cyanosis on the oral mucous membrane. Plain radiographic findings revealed multiple, various-sized(0.5 to 7 cm in diameter), slightly firm-nodules on the thoracic region but digital bone lysis was not seen. These lesions on the thoracic cavity were considered likely to be metastatic from the digit and the dog was naturally died after 3 weeks from the time. Histologically, the digital mass confirmed the diagnosis of metastatic malignant melanoma that was composed of round melanocytic neoplastic, anaplastic, and melanin-containing cells. This report records clinical information and gross and light microscopic features of metastatic malignant melanoma in a dog.

Inhibitory Effect of Ligularia Stenocephala on the Cancer Metastasis

  • Ligularia stenocephala has been used as a traditional medicine for the treatment of asthma, arthritis, jaundice, and hyperpiesia. In this study, we investigated the anti-metastatic and hypnotic effects of the methanolic extract of L. stenocephala (MLS). Gelatin zymographic analysis revealed that MLS suppresses matrix metalloproteinase-2 (MMP-2) and MMP-9 activities in B16F10 cells. The gene expressions of MMPs were also down-regulated by MLS treatment in a dose-dependent manner. In addition, cancer cell invasion and migration were attenuated by MLS via suppression of NF-${\kappa}B$ activation. The in vivo lung metastasis of B16F10 melanoma cells was also inhibited by the treatment of MLS. These findings show that MLS has anti-metastatic properties, and, therefore, it might be applicable as a valuable anti-metastatic agent.

  • Metastases to the liver can be found in various malignancies, most commonly originating from the colon, rectum, pancreas, stomach, esophagus, breast, lung, and melanoma. Surgical resection of liver metastasis is generally considered to be the definitive therapy fore cure. However, many patients are unable to undergo surgical resection due to medical comorbidities or multifocal extent of malignant disease affecting the liver. Among patients not eligible for surgery, other therapies exist for treatment in order to down stage the disease for surgical resection or for palliation. Radioembolization of hepatic metastases has shown to improve outcomes among patients with variety of malignancies including more common malignancies such as colorectal cancer. Yttrium-90 (Y-90) radioembolization has been successfully used in the management of hepatic metastases. A small series of metastatic sarcoma to the liver treated with radioembolization showed a promising response. We report a case of metastatic gastric leiomyosarcoma to the liver treated with Y-90 glass microspheres therapy using the radiation segmentectomy approach, previously described for hepatocellular carcinoma.

  • There are various types of malignancy in eyelid, such as squamous cell carcinoma, melanoma, and sarcoma. These malignant tumors have potential of metastasis by regional lymph node drainage. The lymph node around parotid gland has been known as a common site of regional lymph node metastasis. The rarity of malignant tumors in the periorbital area makes it difficult to determine the optimal extent of treatment. We report a case of parotid metastasis after eyelid cancer operation in a 60-year-old man.

Extracranial systemic antitumor response through the abscopal effect induced by brain radiation in a patient with metastatic melanoma

  • The abscopal effect is a term that has been used to describe the phenomenon in which localized radiation therapy treatment of a tumor lesion triggers a spontaneous regression of metastatic lesion(s) at a non-irradiated distant site(s). Radiation therapy induced abscopal effects are believed to be mediated by activation and stimulation of the immune system. However, due to the brain's distinctive immune microenvironment, extracranial abscopal responses following cranial radiation therapy have rarely been reported. In this report, we describe the case of 42-year-old female patient with metastatic melanoma who experienced an abscopal response following her cranial radiation therapy for her brain metastasis. The patient initially presented with a stage III melanoma of the right upper skin of her back. Approximately 5 years after her diagnosis, the patient developed a large metastatic lesion in her upper right pectoral region of her chest wall and axilla. Since the patient's tumor was positive for BRAF and MEK, targeted therapy with dabrafenib and trametinib was initiated. However, the patient experienced central nervous system (CNS) symptoms of headache and disequilibrium and developed brain metastases prior to the start of targeted therapy. The patient received radiation therapy to a dose of 30 Gy delivered in 15 fractions to her brain lesions while the patient was on dabrafenib and trametinib therapy. The patient's CNS metastases improved significantly within weeks of her therapy. The patient's non-irradiated large extracranial chest mass and axilla mass also shrank substantially demonstrating the abscopal effect during her CNS radiation therapy. Following radiation therapy of her residual chest lesions, the patient was disease free clinically and her CNS lesions had regressed. However, when the radiation therapy ended and the patient continued her targeted therapy alone, recurrence outside of her previously treated fields was noted. The disease recurrence could be due to the possibility of developing BRAF resistance clones to the BRAF targeted therapy. The patient died eventually due to wide spread systemic disease recurrence despite targeted therapy.

Magnetic Resonance Findings of a Canine Benign Uveal Melanocytoma

  • A 13-year-old spayed female Beagle dog was referred with high intraocular pressure, hyperemia, and exophthalmos of the left eye and underwent ultrasound, which revealed a mass in the ciliary body of the left eye. Magnetic resonance (MR) imaging was ordered to evaluate invasion of surrounding structures and metastasis to the brain via the optic nerve. On MR imaging, a single, well-defined, smoothly marginated, triangular-to-oval-shaped mass was found. The mass was hyperintense on T1-weighted images and hypointense on T2-weighted images, similar to a previous case of ocular melanoma. The mass originated from the ciliary body and extended from the anterior chamber to the posterior chamber. Slight enhancement was observed in the mass. There was no evidence of invasion into surrounding structures or the optic nerve, and no sign of metastasis to the brain. The mass was histopathologically diagnosed as benign uveal melanocytoma.

A Further Study on the Inhibition of Tumor Growth and Metastasis by Red Ginseng Acidic Polysaccharide (RGAP)

  • We have recently reported that red ginseng acidic polysaccharide (RGAP), isolated from Korean red ginseng (Panax ginseng C. A. Meyer), showed immunomodulatory antitumor activities, mainly mediated by nitric oxide (NO) production by macrophage. In this study, we examined the effect of RGAP on anticancer activity using lung carcinoma 3LL, sarcoma 180 and adenocarcinoma JC tumor cells transplanted into mice as well as antimetastatic activity using B16-F10 melanoma. When RGAP (300 mg/kg) were treated to mice implanted with one of the three kinds of tumor cells, the tumor weight significantly decreased compared with control mice. Tumor inhibition ratios of RGAP (300 mg/kg) in mice transplanted with lung carcinoma 3LL, sarcoma 180 and adenocarcinoma JC cells were 26.8%, 29.3% and 31.6%, respectively. Hundred mg/kg of RGAP did not cause a significant decrease in tumor weight compared with control group. When RGAP was administered i.p. with the dose of 100 and 300 mg/kg in B16-F10 melanoma-bearing mice, lung metastasis were reduced significantly in mice. Corrected phagocytic index was also remarkably increased by RGAP. These results suggest that stimulation of phagocytic activity of macrophages may be a mechanism for in vivo anticancer and antimetastasis activities of RGAP.

The Effect of Anti-Cancer and Immune Response Improvement of ELP(Ecliptae Herba) Herbal-Acupuncture into Chung-wan(CV12) (백서(白鼠)의 B16-F10 Melanoma에 대한 한련초약침(旱蓮草藥鍼)의 항암(抗癌) 및 면역증강효과(免疫增强效果))

  • Objective & Methods : To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Eclipta prostrata diffusae herba infusion solution(ELP-HAS), we injected ELP-HAS into Chung-wan(CV12) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. We have reached the following conclusion through the effect on the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$cells in mouse PBMCs, the effect on the pulmonary colony formation number, and the effect on MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. Results : The results were obtained as follows : 1. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the spleen cell proliferation in BALB/c mouse was significantly increased compared with control group. 2. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$cells in C57BL/6 mouse PBMCs was increased compared with control group. 3. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the pulmonary colony formation number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with control group. 4. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with control group. Conclusions : ELP Herbal acupuncture injection into Chung-wan(CV12) of C57BL/6 mice is considered to be effective in anti-cancer, anti-metastasis and immune response improvement.

Inhibitory effect of EtOAc Subfraction-2 of Spatholobus Suberectus Dunn. on tumor Metastasis (계혈등 EtOAc subfraction-2가 종양전이 억제효과에 관한 연구)

  • Spatholobus Suberectus dunn. has been applied to blood stasis in oriental medicine. We selected one potent ethyl acetate subfraction-2 from Spatholobus Suberectus Dunn.(SSD) from anti-metastasis screening. It exerted the cytotoxicity against HT1080 and B16BL6 with the IC50 of 60 ug/ml and also significantly inhibited tumor cell induced platelet aggregation (TCIPA). It effectively didn't inhibit cell adhesion of HT1080 to matrigel coated wells, while it inhibited the cell invasion of HT1080 at the doses of 10, 20, 40 μg/ml by Boyden chamber assay. It effectively suppressed lung metastasis by B16BL6 melanoma in C57BL6 mice. These results indicate that the EtOAc subfraction-2 of Spatholobus Suberectus Dunn. can be applied to caner treatment with anti-metastatic activity.
