Acquired pigmentary skin diseases such as abnormal melanogenesis, vitiligo, chloasma and inflammatory pigmentation are related to regulate the melanin production. The aim of this study was to investigate the effect of Codonopsis lanceolata on the melanogenesis of HM3KO human melanoma cells biologically. The cells were treated for 5 days with Codonopsis lanceolata at several concentrations. Treatment with Codonopsis lanceolata suppressed melanin contents as a dose dependent manner without cytotoxicity and morphological change. And the extract of Codonopsis lanceolata also inhibited tyrosinase, a key enzyme forming melanin, in a dose-dependent manner. And it did not affect the DOPAchrome tautomerase(TRP-2) activity. These results suggest that Codonopsis lanceolata is a candidate for an efficient whitening agent which supresses melanogenesis by a Raper-Mason pathway.
Optimization study of the hot water extraction for enhancing antioxidative activity from Auricularia auricula was performed by Taguchi approach using orthogonal matrix $L_9(3^4)$ method. The correlation between DPPH radical scavenging activity and the components of samples extracted from different extraction conditions were also analyzed. The correlation coefficient between DPPH radical scavenging activity and melanin content of A. auricula were 0.93, indicating 'good correlation'. The optimum extraction conditions was obtained at the extraction time of 1 hr. temperature of $85^{\circ}C$, solid: water ratio of 1: 40(w/v) and frequency of 2 times. Under these conditions, values of maximum DPPH free radical scavenging activity and melanin contents of A. auricula were $67.21{\pm}2.17$ and $52.94{\pm}2.10 mg/g$, respectively. Melanin content of 1.6 times and DPPH free radical scavenging effect of 130% were enhanced by optimization.

Diarylpropionitrile (DPN), a selective agonist for estrogen receptor β (ERβ), has been reported to regulate various hormonal responses through activation of ERβ in tissues including the mammary gland and brain. However, the effect of DPN on melanogenesis independent of ERβ has not been studied. The aim of this study is to examine the possibility of anti-melanogenic effect of DPN and its underlying mechanism. Melanin contents and cellular tyrosinase activity assay indicated that DPN inhibited melanin biosynthesis in alpha-melanocyte stimulating hormone-stimulated B16F10 melanoma cell line. However, DPN had no direct influence on in vitro tyrosinase catalytic activity. On the other hand, 17β-estradiol had no effect on inhibition of melanogenesis, suggesting that the DPN-mediated suppression of melanin production was not related with estrogen signaling pathway. Immunoblotting analysis showed that DPN down-regulated the expression of microphthalmia-associated transcription factor (MITF), a central transcription factor of melanogenesis and its down-stream genes including tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2. Also, DPN attenuated the phosphorylation of protein kinase A (PKA) and cAMP-response element-binding protein (CREB). Additionally, DPN suppressed the melanin synthesis in UVB-irradiated HaCaT conditioned media culture system suggesting that DPN has potential as an anti-melanogenic activity in physiological conditions. Collectively, our data show that DPN inhibits melanogenesis via downregulation of PKA/CREB/MITF signaling pathway.
The aim of this study was to investigate the effect of Adenophorae Radix (AR) on melanogenesis of B16F10 mouse melanoma cells. The cells were treated for 5 days with AR at several concentrations. Treatment with AR suppressed melanin contents as a dose dependent manner without cytotoxicity and morphological change. And the extract of AR also inhibited tyrosinase activity, a key enzyme forming melanin, in a dose-dependent manner, Treatment of the cells with Adenophorae Radix also suppressed the increase of α-MSH (10 nM)-induced melanin contents and tyrosinase activity. These results suggest that AR inhibits melanogenesis and abrogates α-MSH-induced melanogenesis in B16 melanoma cells.
본 연구에서는 낙과 배 물 추출물에 존재하는 미백기능 유효성분으로써 arbutin을 선정하여 HPLC를 통해 가장 높은 함량을 보인 낙과 배 물 추출물로 미백 효과를 알아보았다. 낙과 배 물 추출물의 미백 효과를 알아보기 위하여 B16F10 melanoma 세포를 이용하여 세포 내 tyrosinase 저해 효과, melanin 생성 저해 효과, tyrosinase와 관련된 단백질 발현에 미치는 영향을 연구하였다. 그 결과 낙과 배 물 추출물은 tyrosinase 활성과 B16F10 melanoma 세포의 melanin 생성 저해 효과를 나타내었다. 또한, MiTF와 tyrosinase의 발현의 억제를 통해 미백 효과를 나타내는 것으로 확인되었으며, mushroom tyrosinase 결과를 통해 melanin 생성 과정에 있어 tyrosinase를 직접 저해하는 것이 아닌 세포 내에서 tyrosinase의 발현 조절을 통해 저해하는 것으로 확인하였다. 따라서 낙과 배 물 추출물은 천연 arbutin 미백 기능성 화장품 소재 개발로 이용 가능성이 높은 것으로 판단된다. 낙과 배를 이용한 새로운 뷰티케어 소재를 개발하고 이들의 피부 기능성을 밝혀냄으로써 낙과 배의 다양한 활용성이 증대될 것으로 기대되며, 이를 통하여 국내 과실 농가 및 관련 사업에 기여할 것으로 생각된다.
Objective: Recently the demands for the effective and safe depigmentative and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The purpose of this study is to investigate the MKG(Methanol Extract of Kaempferia galanga) and their dermal bioactivity properties related to cosmeceuticals such as depigmentation. Methods: We assessed inhibitory effects of MKG on melanin production in B16/F10 melanoma cells, on mushroom tyrosinase activity, effects of MKG on the expression tyrosinase, TRP-1, TRP-2, GSK-$3{\beta}$, CREB, MITF in B16/F10 melanoma cells without cytotoxicity range. Cell viability was measured by MTT assay and tyrosinase activity was assessed using by DOPA staining, western-blot analysis. We measured inhibition of melanin synthesis and tyrosinase activity by down-regulation of melanogenic enzyme expressions in ${\alpha}$-MSH induced melanogenesis B16/F10 melanoma cells. Results: MKG inhibited tyrosinase-activity, total melanin contents and dendrite out-growth. MKG inhibited melanogenesis by down-regulation of tyorsinase, TRP-1, TRP-2, CREB, and MITF in B16/F10 cells. The treatment with MKG at the 12.5, $25{\mu}g/ml$ level significantly inhibited the melanin synthesis induced ${\alpha}$-MSH in B16/F10 melanoma cells compared with untreated control. Conclusion: These results suggest that MKG inhibit melanin biosynthesis which is involved in hyper-pigmentation. So MKG is considered to be used as a whitening components reducing cytotoxicity.
본 연구는 산말 3종류 Desmarestia dudresnayi subsp. Tabacoides (담배잎산말), Desmarestia viridis (쇠꼬리산말), Desmarestia ligulate (참산말) 잎 추출물의 항산화 및 항 멜라닌 활성에 대한 기능적 연구를위해 수행하였다. 3종류의 산말 중 담배잎산말 추출물이 2.5 mg / mL의 농도에서 DPPH 및 ABTS 라디칼 소거활성이 각각 68.0 % ± 1.9 % 및 84.6 % ± 1.7 %로 강한 항산화력을 나타냈다. 질소 (NO) 라디칼 소거 활성은 91.6 % ± 1.1 %의 높은 항산화능을 보였다 산말 추출물을 이용한 B16F10세포의 세포독성은 100 ㎍/mL 농도에서 3종의 산말 추출물이 모두 85% 이상의 세포 생존률을 보였고, 100 ㎍ / mL농도에서 담배잎산말 및 쇠꼬리산말이 70% 이상의 멜라닌 억제효과를 보였다. 이러한 내용은 산말이 항산화 및 항멜라닌 활성과 같은 천연 화장품 소재로 사용될 수 있을 것으로 사료된다.
Objectives : Since hypopigmentation is known to increase the risk of skin cancer, melanogenesis in the skin needs to be regulated. Here, we evaluated the melanogenesis stimulatory effects of a modified Goagium herbal remedy (HR) and HR+ox bile (Bos taurus domesticus) extract (OBE) to address hypopigmentation disorders. Methods : B16F10 melanoma cells were treated with different dosages of HR and HR+OBE for 24 to 48 h after 1 h of 10 nM ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH). After the treatment, cell viability, tyrosinase activity, melanin synthesis and the expression of genes related to melanin synthesis were measured and the regulation of the ${\alpha}$-MSH signalling through cAMP responding element binding protein (CREB) was determined. Results : HR and HR+OBE with the ranges of $15{\sim}100{\mu}g/mL$ did not affect cell viability in melanoma cells. The 1 h treatment of HR+OBE (50 and $100{\mu}g/mL$) potentiated the phosphorylation of CREB by enhancing ${\alpha}$-MSH signaling and its 24 h treatment increased CREB expression. Consistent with CREB potentiation, their treatment for 24 h, the expression of microphthalmia-associated transcription factor (MIFT), tyrosinase, tyrosinase related protein (TRP)-1 and TRP-2 were increased in realtime PCR. Ultimately, the 48 h treatment of HR+OBE (50 and $100{\mu}g/mL$) increased tyrosniase activity and melanin contents in the melanoma cells in comparison to the control. Conclusions : HR+OBE (50 and $100{\mu}g/mL$) increases melanin synthesis in B16F10 melanoma cells via the stimulation of tyrosinase activity and expression of MIFT, tyrosinase, TRP-1 and TRP-2. HR+OBE can be used as the a possible treatment for hypopigmentation of the skin.
사람의 피부색에 결정적인 영향을 미치는 멜라닌은 표피 기저층의 멜라닌형성세포(Melanocyte)에 의해 생성되어 피부 전반에 분포하고 있으며, 표피의 가장 외각인 각질층에서도 멜라닌은 존재하고 있다. 이러한 각질세포 내 멜라닌과 피부색과의 관계를 알아보고자 각질세포 내 멜라닌의 선택적 염색(Fontana-Masson method) 및 영상분석을 통한 정량적 평가법을 확립하였고, 각질세포 내 멜라닌 양과 색차계를 통한 피부색과의 상관관계를 조사하였다. 71명의 여성 피시험자를 대상으로 색차계를 이용한 피부색과 동일부위의 각질세포에 함유된 멜라닌을 정량하여 비교하였을 때, 각질 세포 내 멜라닌이 차지하는 면적을 나타내는 MCA (Melanin covering area)값이 피부색의 $L^*$값, $ITA^{\circ}$ (Individual Typology Angle)값과 비교적 높은 상관관계를 보였다(각각 r = 0.6049, r = 6651). 이러한 결과를 통해 각질세포 내 멜라닌 양이 피부색 분류의 새로운 기준이 될 수 있는 가능성을 확인했으며, MCA값에 따라 새로이 피부색을 분류하는 기준을 제시할 수 있었다. 본 실험을 통해, 각질세포 내 멜라닌 정량법의 개발과 멜라닌 양에 따른 새로운 피부색 분류 기준을 세울 수 있었으며 개발된 각질세포 내 멜라닌 정량법은 향후 새로운 in vivo 미백 평가 및 피부 색상과 관련된 여러 분야에서 매우 유용하게 이용될 수 있을 것이다.
Melanin은 대부분의 식물과 동물에서 발견되는 자연 색소로, 피부와 모발의 색을 결정하는 게 관여한다. Melanogenesis는 일반적으로 피부를 자외선과 같은 외부 자극으로부터 보호하기 위한 반응으로 melanocyte에서 수행한다. Tyrosinase는 기질인 tyrosine을 melanin으로 생합성 하는데 관여한다. 그러나 melanin의 과다 생성은 melasma, blotch, hyperpigmentation, 그리고 피부암과 같은 피부질환으로 이어질 수 있다. Kojic acid 및 arbutin과 같은 미백 물질에 대한 연구가 수행되었지만, 부작용으로 인해 일부 국가에서는 사용을 금지하거나 제한하고 있다. 따라서 본 연구에서는 기능성화장품의 새로운 미백재료로서 잠재력을 연구하기 위해 겨자무(horseradish)를 선택하였다. 더 나아가 효율적인 추출을 위해 아임계수 추출을 사용하였다. 연구 결과, 아임계수 200℃ 추출물은 높은 DPPH 라디칼 소거능, 총 페놀 함량, tyrosinase 저해능 그리고 B16-F10 melanoma 세포주의 melanin 생성 억제 효과를 나타내었다. B16-F10 melanoma 세포주에 대한 세포 독성을 조사하기 위해 MTT reduction assay 및 형태학적 변화를 관찰하였으며, 그 결과, 겨자무 methanol 추출물 및 아임계수 200℃에서는 세포독성이 확인되지 않았다. 결론적으로, 이 연구는 화장품의 천연 미백 기능성소재로서 가능성을 제안하는 바이다.
