Investigation of the Condition of Acetic Acid Fermentation with High Concentration Ethanol Resistant Acetobacter sp. FM-10 (고농도 에탄올 내성균 Acetobacter sp. FM-10을 이용한 초산 발효조건 검토)

  • The fermenting conditions for acetic acid production with Acetobacter sp. FM-10 which could grow in the medium containing 10% ehtanol were investigated. Initial concentration of acetic acid in broth medium affected greatly to the fermentation speed. For example , the acetic acid production increased proportionally by the increasing of initial concentration was higher that 1.0%. When the cultivation was started with broth medium containing 5% ethanol, the additional adding ethanol during the fermentation was not significantly increased the acidity of the medium. The acidity of the medium containing 10% ethanol was reached to 8.3% after shaking than static cultivation by about 10 days with 150 rpm shaking speed. Acetic acid production with shaking cultivation was faster the static cultivation by abot 10 days under the same condition except shaking. In acetic acid fermentation with the batch style fermentor , the optimum fermentation condition was 700 rpm of agitation speed and 5L/min air flow rate in 3L culture medium .

  • Porcine oviduct epithelial cells (POEC) are widely used in co-culture experiments to improve early embryonic development, in vitro fertilization in embryo transfer programs for domestic animals and in vitro maturation of immature germ cells. POEC were mechanically isolated and cultured in tissue culture medium 199. Cells grew continuously, and confluent monolayers were formed after 7 days. After forming confluent monolayer of epithelial cells, supernatant was collected as the condition medium for maturing round spermatids in vitro. Round spermatids were also separated mechanically and cultured in the POEC condition medium. In this study we observed that $20\%$ of round spermatid cultured were matured into elongating spermatid after 24 h, and about $10\%$ of round spermatid cultured showed complete elongation (elongated spermatid) within $24\~48$ h of in vitro culture. No further development was observed within $50\~72$ h and transformed cells lost their viability after 72 h. These preliminary findings suggest that the condition medium from POEC may be possible to overcome the round spermatid block by improving the milieu of culture system.

  • Metabolic alterations of Cordyceps bassiana mycelium were investigated under the following culture medium and light conditions: dextrose agar supplemented with 0.5% yeast extract (SDAY) medium with light (SL), SDAY medium without light (SD), nut medium without light (ND), and iron-supplemented SDAY medium without light (FD). The levels of asparagine, aspartic acid, glutamic acid, glutamine, histidine, lysine, ornithine, and proline were significantly higher under SD and SL conditions. The levels of most of the alcohols, saturated fatty acids, unsaturated fatty acids, fatty acid esters, sterols, and terpenes were higher under the ND condition than in the other conditions, but beauvericin was not detectable under the ND condition. The FD condition was favorable for the enhanced production of aminomalonic acid, malic acid, mannonic acid, and erythritol. Thus, the metabolic characteristics of C. bassiana can be manipulated by varying the cultivation conditions, rendering this fungus potentially favorable as a nutraceutical and medicinal resource.

A Study on a Linear Phase Conjugate Oscillator with Photorefractive Medium (광굴절 매질로 구성된 선형 위상 공액 발진기의 발진 조건에 관한 연구)

  • Usng the P.Yeh's analytic method, we derive the condition for oscillation of a linear phase conjugate oscillator which consists of photorefractive medium and two conventional mirrors. From this general oscillation condition, we obtain the threshold oscillation conditon of a single phase conjugate resonator and the self-oscillation condition of photorefractive medium, then in special case (phase shift =90\ulcorner: no external dc electric field), oscillation conditions of the linear phase conjugate oscillator for any cavity length are derived. The results indicate that, unlike in a phase conjugate oscillator with Kerr-like medium, oscillation cannot occur at special cavity length for given couplinyg strengths.

  • This study was performed to investigated the application of mixed resins for corrugated container board. The corrugated container board yields a sandwich structure in which a linerboard material is glued to a corrugated medium . Now, manufacturing corrugated container boards don't provide sufficient strength, and result in box failure during shipping . Therefore improvement of box strength is necessary . In this study, we intend to improve box strength by improving corrugated medium strength with mixed resins and to find the optimum treatment condition of this resins. First, we tried to mixed resins as Starch+CMC, Starch_Irea, CMC+Urea, Second, investigated to applicability of this resins for corrugated medium , and the third, measured tensile index, burst index, and edgewise compression index on liner, medium paper, and single faced corrugated container board. In this test results, we obtained that the improvement ratios of tensile index in liner and medium paper were approximately 80-185%, 60-118% , respectively. The respecting improvement ratios of edgewise compression index of single faced corrugated container board was approximately 91-124%, relatively. In addition, we concluded that optimum condition in mixing ratio was 1 :3 with CMC + Urea and the ap[plication amounts was 9% on materials. Fro manufacture of corrugated container board, optimum condition in mixing ratio was 1 : 3 with 5% CMC +Urea , because of considering to improvement of strength on cost.

  • The nar promoter as an inducible promoter was characterized for the process development for the gene expression and the protein production under anaerobic condition. The LB medium was selected as a main culture medium showing the enzyme activity of 18,000 units/min/g cell in the flask cultivation. The optimum concentration of nitrate was 1%. Under anaerobic conditions, the gene expression was fully induced in the presence of nitrate.

On the Thermal Boundary Conditions at the Interface Between the Porous Medium and the Impermeable Wall (다공성 매질과 비투과성 벽면 사이의 경계면에 대한 열적 경계 조건)

  • The present work investigates a heat transfer phenomenon at the interface between a porous medium and an impermeable wall. In an effort to appropriately describe the heat transfer phenomenon at the interface, the heat transfer at the interface between the microchannel heat sink, which is an ideally organized porous medium, and the finite-thickness substrate is examined. From the examination, it is clarified that the he heat flux distribution at the interface is not uniform for the impermeable wall with finite thickness. On the other hand, the first approach, based on the energy balance for the representative elementary volume in the porous medium, is physically reason able. When the first approach is applied to the thermal boundary condition, and additional boundary condition based on the local thermal equilibrium assumption at the interface is used. This additional boundary condition is applicable except for the very th in impermeable wall. Hence, for practical situations, the first approach in combination with the local thermal equilibrium assumption at the interface is suggested as an appropriate thermal boundary condition. In order to confirm our suggestion, convective flows both in a microchannel heat sink and in a sintered porous channel subject to a constant heat flux condition are analyzed. The analytically obtained thermal resistance of the microchannel heat sink and the numerically obtained overall Nusselt number for the sintered porous channel are shown to be in close agreement with available experimental results when our suggestion for the thermal boundary conditions is applied.

  • Currently, the consumer pattern is directed to small quantity and high variety these days. World market is led by technological innovations and changes rapidly. To survive in this environment, the raising of small and medium enterprises is highly desirable. In the context of nationwide restructuring, competitive medium and small industries which are in good financial status should lead the nation's economy. But general ignorance of the safety issues could lead to a miss management of this industry in this field. In this study, actual condition and improvement of safety management are as follows; We have to 1) find out the factors which endangers this industry, and set a proper guideline for safety management. 2) encourage safety management of these industries by themselves, and advertise and provide technical support for using PC and internet. 3) set a higher standard of safety issue when accrediting a prospective medium and small industry, and have a firm control of this issue once accredited. 4) supply more governmental financial aids to terms of safety issues for medium and small industries.

Optimal Conditions for the Production of Immunostimulating Polysaccharides from the Suspension Culture of Angelica gigas Cells. (면역증강성 다당 생산을 위한 참당귀 세포배양의 최적조건)

  • An Immunostimulating polysaccharide was produced from the suspension culture of Angelica gigas H4, plant cells. In order to enhance the polysaccharide production by the A. gigas cell culture, medium composition and physical conditions were optimized. Schenk and Hildebrandt (SH) medium was selected as an optimal basal medium for the growth of A. gigas. The maximum cell and polysaccharide concentration obtained in SH medium were 15.8 g DCW/l and 0.85 g polysaccharide/l, respectively, at $25^{\circ}C$ under dark condition. For the enhanced polysaccharide production, a polysaccharide production medium (PPM) was established by modifying Gamborg B5 medium with optimized carbon sources, growth regulators, organic and inorganic elements. Optimal initial pH and temperature were 6.0-6.6 and $20^{\circ}C$, respectively, and the dark condition was better than the light condition. The maximum polysaccharide concentration of 1.5 g/l could be obtained through the optimization of the medium composition and physical conditions.

Effects of Low Dose $\gamma$ Radiation on Callus Growth of Lithospermum erythrorhizon S. (지치 (Lithospermum erythrorhizon S.) Callus 생장에 미치는 저선량 $\gamma$선의 효과)

  • The effect of low dose ${\gamma}$-radiation on the callus growth of Lithospermum erythrorhizon S. cultured on different medium and lighting condition was investigated. The 8 Gy irradiation stimulated callus growth on LS medium supplemented with BA 2 mg/L and NAA 2 mg/L, however, the growth of callus was more effective on LS medium supplemented with BA 1 mg/L and NAA 1 mg/L under 16 hrs day light. And on the LS medium containing IAA 0.2 mg/L, 16 Gy irradiation increased the callus growth by supplement with kinetin 2 mg/1 and the effect of kinetin was higher than BA at same concentration. The growth of callus was more vigorous on LS medium than MS medium in general. On M-9 medium, the growth of callus was poor regardless lighting conditions, however, by ${\gamma}$-ray irradiation of 16 Gy or 30 Gy, callus growth rates were increased by 30% or more than 30%, especially, under 16 hrs day light condition.
