Brassica와 Raphanus 간에 속간 교잡종을 육성하기 위해서 34개의 교배 조합을 설정한 다음 수분 12일이 경과된 배주를 BA와 GA 1.0 ㎎/L가 첨가된 MS배지에 접종하고 배의 발아율을 조사하였다. BA나 GA 첨가는 배의 발아에 큰 영향을 미치지 않았다. 무 속의 '내병총태'와 '중국청피'를 부계로 교잡한 조합에서 배의 발아율이 낮았으나 갓(Brassica juncea)을 부계로 교잡한 조합에서는 비교적 배의 발아율이 높았다. 이러한 결과는 배주배양시 배지에 첨가한 casein hydrolysate, malt extract, BA, kinetin 및 glutamine이 배의 발아에 미치는 영향보다는 부계나 모계의 교잡조합에 따라 배의 발아율에 상당한 차이가 있다는 것을 의미한다.
가시오갈피의 조직배양을 통한 대량증식방법체계 확립의 한 방법으로 미숙배 배성숙정도 및 배지의 종류가 callus 형성 및 식물체 분화에 미치는 영향을 구명코자 실시한 실험결과는 다음과 같다. 1. 미숙배배양으로부터 캘러스형성은 MS, WPM 배지보다 SH 배지가 더 효과적이었으며, SH 배지에 2,4-D 1mg$\iota$를 첨가시 83.3%의 캘러스형성율을 보였다. SH배지에 TDZ을 단독처리하였을 때 보다 양호한 신초 분화를 나타났고 배의 성숙정도가 C단계 (2mm이상)인 미숙배를 치상하였을 때가 미숙배의 크기가 2mm 이하인 A, B단계의 미숙배를 치상하였을 때보다 식물체 분하가 더 잘되었다. 2. SH배지를 기본으로 하여 2,4-D와 TDZ을 조합처리하였을 때 배의 성숙정도가 C(2mm이상)인 배를 2,4-D 0.1mg/$\iota$+TDZ 3mg/$\iota$ 처리시 양호한 신초 분화를 보였다. 3. WPM 배지를 기본으로 하여 2,4-D와 TDZ을 처리하였을 때 배의 성숙정도가 C(2mm이상)인 배를 TDZ 0.1mg/$\iota$ 처리시 4.7개의 신초로 양호한 결과를 보였다.
Petiole explant of Glehnia littoralis Fr. Schmidt was in vitro cultured MS plant medium(DUCHEFA co.) supplemented with various plant growth regulators and examined to find out their optimum combination and concentration for plantlet regeneration. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on MS plant medium with various kinds of plant growth regulators. Embryogenic calli and adventitious shoot formation were greatly influenced by plant growth regulators. Embryogenic calli induction showed a good response on MS medium supplemented with NAA and BA than others. Especially, combination of 1.0 mg/L NAA and 0.5 mg/L BA on MS medium led to the greatest frequency in adventitious shoot. The results suggest that plant regulator selection be important factor to achieve an efficient regeneration.
Somatic embryos were formed from calli obtained from axillary shoots (raised from nodal segments of glasshouse-grown plants under aseptic conditions), internodal segments (from in vitro-raised plants), and root and coty-ledonary leaf segments (from in vitro-raised seedlings) after 8 weeks of initial culture. Embryo formation was the highest (97.33%) from cotyledonary leaf callus on Mura-shige and Skoog's (MS) medium containing kinetin (KN) (3 mg/L). Somatic embryo induction was lesser with different combinations of auxins while it increased to 100% in internodal segment and cotyledonary leaf calli with 6-benzyladenine (BA) (2mg/L) along with 2,3,5-triiodobenzoic acid (TIBA) (2mg/L). The shoots were induced from somatic embryos raised from root, coty-ledonary leaf and internodal segment calli grown on MS medium containing BA in combination with indole-3-acetic acid (IAA). Maximum of 66.67% cultures formed shoots on MS medium containing BA (1mg/L) in combination with IAA (2mg/L). The shoots raised from somatic embryos were rooted on MS medium supplemented with indole-3-butyric acid (IBA) (2mg/L). The plantlets transferred to the field showed 70% survival rate after one year.
Objective: To investigate the clinical effects of whole brain radiotherapy concomitant with targeted therapy for brain metastasis in non-small cell lung cancer (NSCLC) patients with chemotherapy failure. Materials and Methods: Of the 157 NSCLC patients with chemotherapy failure followed by brain metastasis admitted in our hospital from January 2009 to August 2012, the combination group (65 cases) were treated with EGFR-TKI combined with whole brain radiotherapy while the radiotherapy group (92 cases) were given whole brain radiotherapy only. Short-term effects were evaluated based on the increased MRI in brain 1 month after whole brain radiotherapy. Intracranial hypertension responses, hematological toxicity reactions and clinical effects of both groups were observed. Results: There were more adverse reactions in the combination group than in radiotherapy group, but no significant differences were observed between the two groups in response rate (RR) and disease control rate (DCR) (P>0.05). Medium progression free survival (PFS), medium overall survival (OS) and 1-year survival rate in combination group were 6.0 months, 10.6 months and 42.3%, while in the radiotherapy group they were 3.4 months, 7.7 months and 28.0%, respectively, which indicated that there were significant differences in PFS and OS between the two groups (P<0.05). Additionally, RPA grading of each factor in the combination group was a risk factor closely related with survival, with medium PFS in EGFR and KRAS mutation patients being 8.2 months and 11.2 months, and OS being 3.6 months and 6.3 months, respectively. Conclusions: Whole brain radiotherapy concomitant with target therapy is favorable for adverse reaction tolerance and clinical effects, being superior in treating brain metastasis in NSCLC patients with chemotherapy failure and thus deserves to be widely applied in the clinic.
Hormonal requirements inducing different regeneration pathways with particular emphasis on somatic embryo-genesis in Alhagi graecorum were studied. While combination of 0.5 $\mu{M}$ 2,4-dichlorophenoxyacetic acid (2,4-D), 2.5 $\mu{M}$ 6-benzylaminopurine (BAP) and 5 $\mu{M}$ 1-naphthaleneacetic acid (NAA) in MS medium induced callus formation and callus maintenance from internodal explants, each alone or in combination with other induced distinct regeneration pathway. Adventitious bud formation was induced on MS medium supplemented with 2.5 $\mu{M}$ BAP. It was improved when 2.5 $\mu{M}$ BAP was used in combination with 5 $\mu{M}$ NAA. MS medium containing 0.5 $\mu{M}$ 2,4-D or 5 $\mu{M}$ NAA induced the formation of abnormal direct somatic embryos. While increase of 2,4-D concentration (1.125-9) resulted in the formation of viable embryogenic mass, increase of NAA did not change its effect. NAA should be used in combination with 2,4-D even at low concentration (0.5 $\mu{M}$) to form embryogenic mass. In A. gaecorum, the role of 2,4-D as trigger of somatic embryogenesis and BAP as trigger of adventitious bud formation was deduced, but for maximum yield certain auxin-cytokinin ratio should be applied. Embryogenic masses characterized by high water content, low peroxidase activity, and low number of peroxidase and glutamate oxaloacetate transaminase bands in comparison with calli obtained under conditions stimulating adventitious bud formation. The resulted differential gene expression, which could be detected by native-PAGE patterns, could be used as marker for organogenic pathway in A. graecorum.
The top-to-bottom compression strength of corrugated board box is the most important mode of loading during it's no, and it depends largely on the edgewise compression strength of the corrugated board in the cross-machine direction and to a considerable extent on the flexural stiffness in both principal directions (CD; cross-machine direction, MD; machine direction) of the corrugated board. Corrugated board is a sandwich structure with an orthotropic property. The purpose of this study was to elucidate the principal design parameters for board combination of corrugated board from the viewpoint of bending strength through the finite element analysis [FEA] fur the various corrugated board. In general, the flexural stiffness [FS] in the MD was 2-3 times larger than that in the CD, and the effect of liner for the FS of corrugated board was much bigger than that of corrugating medium. The flexural stiffness index [FSI] was high when the stiffness of liner was in the order of inner, outer, and middle liner in double-wall corrugated board [DW], and the effect of the stiffness arrangement or itself reinforcement of corrugating medium on the FSI was not high. In single-wall corrugated board [SW] with DW. the variation of FSI with itself stiffness reinforcement of liner was much bigger than that with stiffness arrangement of liner. The highest FSI was at the ratio of about 2:1:2 for basis weight distribution of outer, middle, and inner liner if the stiffness of liner and total basis weight of corrugated board were equal in DW Secondarily. basis weight was in the order of inner, outer, and middle liner. However, the variation of FSI with basis weight distribution between liner and corrugating medium was much bigger than that with itself basis weight distribution ratio of liner and corrugating medium respectively in both DW and SW. md the FSI was high as more total basis weight was divided into liner. These phenomena fur board combination of corrugated board based on the FEA were well verified by experimental investigation.
아시아틱백합 'Gran Paradiso'의 약유래 식물체를 획득하기 위해 auxin과 cytokinin을 다양하게 조합한 MS배지에 약배양을 실시하였다. 배양 약은 초기 및 후기 2핵기인 개화 3일 전의 약이 캘러스 유도에 가장 적합하였다. MS배지에 5.0mg/L 2,4-D 단용처리나 1.0mg/L 2,4-D와 1.0 mg/L kinetin이 혼용처리되었을 경우 기관분화성 캘러스가 발생되었으나 NAA와 BA 혼용처리가 2,4-D와 kinetin 혼용처리보다 기관분화를 통한 식물체 재생에 효과적이었다. 캘러스에서 shoot는 0.5mg/L NAA와 1.0mg/L BA혼용처리에서 배양 180경에 분화되었으며 3~4개의 잎이 부착된 다아체와 뿌리 및 소자구는 2.0mg/L NAA와 2.0mg/L BA혼용처리에서 배양 250일 후에 관찰되었다. 재생된 식물체의 근단조사 결과 2배체 (2n=2x=24)로 확인되었으며 식물체를 분에 옮긴 후 2년이 경과되었을 때 모두 개화되었다.
We micropropagated pear (Pyrus species) using shoot tips and nodal explants from three pear genotypes. The ability to establish shoot tip cultures, proliferate shoots, induce rooting, and acclimatize the resulting plantlets are all elements of in vitro micropropagation. Shoots were induced from shoot tips on Murashige and Skoog medium (MS) with five different plant growth regulator combinations. The highest shoot formation rates were achieved for the three genotypes using MS supplemented with 1.0 mg/L N6-benzyladenine (BA) and 0.1 mg/L gibberellic acid (GA3). The maximum shoot number and shoot length for the three cultivars were recorded with 2.0 mg/L BA and 0.2 mg/L indole-3-butyric acid (IBA) in multiplication medium using nodal explants produced from microshoots. Nodal explants with one or two axillary buds cultured for three weeks initiated roots on medium supplemented with various concentrations of 1-naphthaleneacetic acid (NAA) or/and IBA in half-strength MS medium for adventitious rooting. The highest rooting response was with the combination of 0.2 mg/L NAA and 0.2 mg/L IBA. A combination of NAA and IBA resulted in a significant increase in the rooting ratio over NAA or IBA alone. In this medium, the root formation rate according to ranged from 68.9% for the BaeYun No. 3 genotype to 51.8% for the Hwanggeum genotype. We also investigated the influence of the concentration the polyamine phloroglucinol in rooting medium. For all three genotypes, the highest rooting ratio, longest root length, and greatest root number were observed in the treatments with 75-150 mg/L phloroglucinol. Most rooted plants were acclimatized successfully.
본 연구는 지금까지 돼지 난포란 성숙을 위해서 많이 사용하고 있는 mTCM-199, mWaymouth MB 752/1 그리고 NCSU-23 성숙배지를 비교하고 액상정액을 이용한 체외수정 방법을 개발하고자 실시하였다. 미성숙 난포란은 0.5 $m\ell$의 성숙배지에 각 well 당 30~40개씩 적하하였고, 38.5$^{\circ}C$, 5% CO2, 95% 공기로 조절된 CO2 배양기에서 44시간 성숙 시켰다. 미성숙 난포란을 mTCM-199, mWaymouth MB 752/l 그리고 NCSU-23 성숙배지에서 44시간 배양한 결과 CVBD 발생율은 각각 95.6, 94.1 그리고 94.9%였으며, MH단계까지의 성숙율은 각각 92.5, 90.1 그리고 91.1%였다. 성숙배지별, GVBD 발생율과 MH까지의 성숙율간에 유의성은 인정되지 않았다. 액상정액의 제조용 정액은 90% 이상의 운동성을 가진 농후정자부분을 사용하였으며 정액 채취 후 2시간 동안에 22~24$^{\circ}C$의 실온까지 냉각시켰다.실온까지 냉각한 정액은 BTS 희석액으로 2$\times$$10^{8}$$m\ell$ 정자농도로 조정하여 100 $m\ell$ 플라스틱병에 30 $m\ell$씩 주입하여 17$^{\circ}C$에서 5일간 보관하였다. 5일 보관 후 운동성이 70% 이상인 정자를 체외수정에 이용하였다. 성숙 후 cumulus cell들이 제거된 성숙난포란은 0.5 $m\ell$의 mTCM-199 또는mTBM 수정배지에 30~40개씩 적하하고, 최종정자농도를 2$\times$$10^{6}$$m\ell$되도록하여 6시간 동안 수정시켰다. 체외수정시킨 수정란들은 0.5 $m\ell$의 NCSU-23 배양배지에서 수정 후 6시간 배양하여 정자침입율, 다정자침입율 그리고 웅성전핵형성율을 조사하였고, 수정 후 45시간 배양하여 난할율을 조사하였다. NC-SU-23 성숙배지와 mTBM 수정배지를 이용하였을때 웅성전핵형성율이 48.0%로써 mTCM-199 성숙배지와 수정배지 또는 mWaymouth MB 752/1 성숙배지와 mTCM-199 수정배지를 이용하였을 때보다 웅성 전핵 형성율이 높았다. 2~4세포기까지의 난할율은 mTCM-199 성숙, 수정 및 배양배지에서 24.1%, mWaymouth 752/1 성숙배지, mTCM-199 수정 및 배양배지에서 43.6%, 그리고 NCSU-23 성숙배지, mTBM 수정배지 및 WCSU-23 배양배지를 이용한 것이 71.2%였다. 이상의 결과를 종합하면 BTS 희석액으로 17$^{\circ}C$에서 5일 보존한 액상정액으로 체외수정이 가능함을 입증하였고, NCSU-23 성숙배지, mTBM 수정배지 및 NCSU-223 배지가 미성숙 난포란의 성숙, 수정 및 배양에 우수한 배지임을 입증하였다.
