In vitro Plant Regeneration from Apical Bud and Nodal Segments of Anthocepahalus Cadamba - An important sacred and medicinal tree

  • Multiple shoot induction and plant regeneration using apical bud and nodal explants of 100 year old tree of Anthocephalus cadamba, an important sacred and medicinal tree in India was achieved for the first time. Aseptic explants cultured in Murashige and Skoog (MS) medium augmented with different concentrations of BAP (0.1, 0.5, 1.0, 2.5, 5.0 and 10 mg/l), when maintained for 60 days, healthy shoots were induced in presence of BAP (1 mg/l). Lower concentrations of BAP (0.1 - 0.5 mg/l) induced only one shoot per explant. Increase in number of shoots per explant was observed in presence of higher concentrations of BAP (2.5, 5.0 and 10 mg/l). However, elongation of shoots was completely inhibited. Bud break and shoot regeneration was largely associated with seasonal factors. Apical buds cultured during June to August exhibited early bud break within two weeks of initial culture. In rest of the months, bud break and shoot regeneration was very slow irrespective of the various concentrations of BAP used in the medium. Explants sourced from three different maturity levels of shoots indicated that actively growing shoots from the mother plant with 1 - 2 nodal segments was more suitable for culture initiation than the explants collected from mature shoots at dormant stage. Regenerated shoots with 2 - 3 pairs of leaves when transferred to half strength MS medium fortified with IBA (1 mg/l), 60% of the shoots induced healthy roots, indicating the possibility of large scale micropropagation.

Chemical Variability of Leaf Cuticular Waxes According to Leaf Position in Tea Tree

  • Cuticular waxes on tea (Camellia sinensis L.) loaves consisted mainly of alkanes, fatty acids, primary alcohols, triterpenes, and a group of unknown compounds, dominated by primary alcohols and triterpenes. Tea tree accessions used in this study were M-1, M-2, Sakimidori, and Yabukita. For all accessions, the alkane, fatty acid, and primary alcohol constituents consisted of a homologues series, and the major constituents of primary alcohol class were the C28 and C30 homologues. Triterpenes consisted of friedelin, $\beta-amyrin$, and three unidentified ones and friedelin was the most abundant. Leaf area and the total amounts of cuticular waxes per leaf increased with lower leaf position from the apical bud in Yabukita variety. With different leaf position, total wax amount per unit leaf area on the youngest leaves of P1 (the uppermost leaf position) showed the largest amount $(12.80{\mu}g/cm^2)$, and on mature loaves of P2 to P6 ranged from 7.08 to $7.77{\mu}g/cm^2$, and then on the oldest loaves of P7 (the lowest leaf position) remained at an increased level $(17.53{\mu}g/cm^2)$. During leaf development (lower leaf position), the amount of primary alcohols decreased from P1 to P6 and increased at P7, whereas that of triterpenes increased from P1 to P7. The percentage of each wax class in the total wax amount occurred a decrease in primary alcohol and an increase in triterpene, with leaf age.

Anti-Inflammatory Effect of the Root extracts from Hibiscus syriacus in LPS-Stimulated RAW264.7 Cells

  • Hibiscus syriacus (H. syriacus) as the national flower of Korea has been used as the herbal medicine in Asia. In this study, we evaluated the anti-inflammatory effect of 70% ethanol extracts from the root of Hibiscus syriacus (RHS-E70) and elucidated the potential signaling pathway in LPS-stimulated RAW264.7 cells. RHS-E70 dose-dependently suppressed NO production by inhibiting iNOS and IL-${\beta}$ expression in LPS-stimulated RAW264.7 cells. RHS-E70 inhibited the phosphorylation and degradation of $I{\kappa}B-{\alpha}$, which contributed to the inhibition of p65 nuclear accumulation and NF-${\kappa}B$ activation. Furthermore, RHS-E70 suppressed the phosphorylation of ERK1/2 and p38, which results in the inhibition of ATF2 phosphorylation and subsequent nuclear accumulation. These results indicate that RHS-E70 may exert anti-inflammatory activity by inhibiting NF-${\kappa}B$ and MAPK/ATF2 signaling. From these findings, RHS-E70 has potential to be a candidate for the development of chemopreventive or therapeutic agents for the inflammatory diseases.

침엽수림과 혼효림에서 토양특성과 토양세균 군집이 산양삼 초기 생육특성에 미치는 영향 (Effect of Soil Properties and Soil Bacterial Community on Early Growth Characteristics of Wild-simulated Ginseng (Panax ginseng C. A. Meyer) in Coniferous and Mixed Forest)

  • Background: This study investigated the effect of soil properties and soil bacterial community on early growth characteristics of wild-simulated ginseng (Panax ginseng C. A. Meyer) in coniferous and mixed forest experimental fields. Methods and Results: The soil bacterial community was analyzed using a high throughput sequencing technique (Illumina MiSeq sequencing). The relationship between the soil bacterial community, soil properties, and growth characteristics of wild-simulated ginseng were analyzed using principal coordinate analysis (PCoA) and the Pearson's correlation analysis. Soil properties and soil bacterial community showed significant difference with forest physiognomy. Results of Pearson's correlation analysis and PCoA showed that the soil properties (soil pH, organic matter, total nitrogen, and cation exchange capacity) and soil bacterial community had significant correlation with tree species ratio and early growth characteristics of wild-simulated ginseng. Conclusions: This study clearly demonstrated the effect of soil properties and soil bacterial community on early growth characteristics of wild-simulated ginseng in coniferous and mixed forest. Moreover, these results will help in the selection of suitable cultivation sites for wild-simulated ginseng.

Inhibitory Effects of the Extract of Rhus verniciflua Stokes on the Reverse Transcriptase of AIDS

  • Four olefinic catechols, commonly referred to as urushiol were isolated from the sap of Korean Rhus verniciflua Stokes and had the stronger inhibitory effects on the reverse transcriptase of AIDS. The hexane extract with a inhibitory effects on reverse transcriptase was purified by silica and ODS gel column chromatography. The active compounds were identified by MS and $^1H-NMR$ as 3-[8' (Z), 11' (Z), 14' -pentadecatrienyl]catechol, 3-[8' (Z), 11' (Z)-pentadecadienyl]catechol, 3-[8' (Z)-penta- decenyl]catechol, and 3-pentadecylcatechol. All of these compounds showed strong inhibitory effects on reverse transcriptase of AIDS, in which 3-pentadecylcatechol exhibited the highest activity $(IC_{50}\;:\;10.87\;{\mu}g/ml)$.

  • 본 논문에서는 비동기 통신에 의해 웹페이지를 다시 읽지 않고 서버로부터 데이터를 가져올 수 있는 Ajax 기술과 경량의 크로스 브라우저 라이브러리인 jQuery를 이용하여 사용자 요구사항을 토대로 약용식물정보시스템을 구현했다. 또한 시스템 구현을 통해 체험한 응답 데이터 이용 방법을 코드로 제시하고 특징을 비교 분석하였다. 구현시스템은 서버에서 간단한 데이터를 응답 받아 클라이언트 측에서 처리하고 DOM 트리 조작에 의한 웹페이지 변경으로 사용자 상호작용 및 반응속도 향상과 사용자에게 자료 접근 편리성을 제공한다.

Development of HPLC method for differentiation of three parts of mulberry tree

  • The leaves (Mori Folium; MF), branches (Mori Ramulus; MR), and root bark (Mori Cortex Radicis; MCR) of the mulberry tree have been used as therapeutic herbs for centuries. Existing analytical methods were developed specifically for different parts of the tree and cannot be applied to samples containing a mixture of tree parts. Such method specialization is time-consuming and requires separate identification and quality control of each tree part. This report describes an HPLC method for the simultaneous quality control and discrimination of MF, MR, and MCR using four marker compounds: rutin, kuwanon G, oxyresveratrol, and morusin. An Optimapak $C_{18}$ column ($4.6{\times}250mm$, $5{\mu}m$) was used with a gradient elution of 0.1 % formic acid in water and acetonitrile. The flow rate was 1.0 mL/min and the detection wavelength was 270 nm. In quantitative analyses of the three parts, rutin (0.11 % w/w) was detected only in MF. The oxyresveratrol content (0.12 % w/w) was highest in MR. Kuwanon G (0.33 % w/w) and morusin (0.18 % w/w) were higher in MCR than in other parts. The HPLC method given herein can be used to simultaneously classify and quantify three herbal medicines from the mulberry tree.

Distribution and Traditional Practice on Neem in the Rural Homesteads of Chittagong Coastal Plain of Bangladesh

  • Neem tree (Azadirachta indica), native to the Indian sub-continent, has been known since the ancient time for its medicinal and insect repellent properties. In recent years, Neem has attracted global attention due to its potential as a source of natural drugs and also environment-friendly pesticides. In the households. The distribution and traditional practice on Neem can be important to the agriculturist, ethno-pharmaceutical developers and to the rural development practitioners in Bangladesh. An exploratory survey on the distribution and traditional practice on Neem tree in the rural homesteads of Chittagong coastal plain, Bangladesh, was conducted over a period of three months from September 2002 to November 2002. It was found that maximum, 64% households used seedling as planting material having 40% maximum sources Within the major tree species present in the homesteads, Neem accounted for 12% among the total individuals. It was found that the availability of Neem trees was found maximum, 31%, in the homesteads of medium sized households. Maximum, 42% individuals of Neem were found within the DBH of 10.1 to 20 cm; and 37% within the 3.1 to 6 m height classes. Neem trees were found to be used maximum, 88% for furniture. The findings of the study will be of immense importance for the rural development practitioners and ethno pharmacological developers in Bangladesh.

樹種, 接種量 및 接種方法이 茯笭 菌絲生長과 結笭에 미치는 영향 (Effect of Tree Species, Inoculation Amount and Inoculation Methods on Mycelium Growth and Sclerotia Formatino of Poria cocos Wolf)

  • When medicinal mushroom, Poria cocos, is cultured , inoculation method of spawn is cross slice inoculation of which the both sides of pine tree were peeled and spawn of P.cocos was inoculated. However, this method required lots of inoculation amount. This study was carried out to improve the culturing method of P. cocos. A good growth of P.cocos was observed in MCM(mushroom complete medium), showing proper mycelia growth and density. In inoculation amount, conventional method(cross slice inoculation) requires 20 bottles of spawn. In contrast, short log method required 8 bottles of spawn and drilling inoculation method 2~3 bottles, which could save by 60% and 85-90% respectively. In the selectrion of tree species, pine and larch had better condition for spawn culture and sclerotia formation condition.In terms of yield , pine was 33.7kg/3.3$m^2$. In the yield of pine, conventional method was 23.4kg/3.3$m^2$, drilling inoculation 29.4kg/3.3$m^2$, short log inoculation 31.7kg/3.3$m^2$, therefore drilling inoculation could increase by 25% and short log inoculation 35%, In addition, management cost was also saved.
