구지뽕 나무의 엽록체 TrnL-F 영역 염기서열 분석을 통한 특이적 SNP 분자마커의 확인 (Identification of specific SNP molecular marker from Cudrania tricuspidata using DNA sequences of chloroplast TrnL-F region)

  • 구지뽕 나무(Cudrania tricuspidata Bureau)는 일반적으로 이용되는 대표적인 다년생의 약용식물이다. 최근 국제적 추세에 따라 자국의 유전자원의 발굴, 보존 등이 강화됨에 따라 인접국가와 국내 자생 꾸지뽕 계통을 판별 할 수 있는 기준설정에 관한 연구의 필요성이 대두되고 있지만, 분자생물학적 판별 기술의 개발은 아직 미흡한 실정이다. 본 연구에서는 국내 토종과 중국 계통의 구지뽕 나무의 기원을 판별하기 위해 엽록체에 존재하는 trnL-trnF 유전자단편에서 SNP를 이용한 판별 프라이머를 확보하였으며 이를 보완하여 보다 신속하게 판별하기 위하여 ARMS-PCR기술을 이용한 판별마커와 그 조건을 확립하였다. 그러므로, 본 연구에서 개발된 SNP 마커는 다양한 지역에서 서식하는 구지뽕 계통들의 신속한 확인을 위해 매우 유용하게 이용될 것으로 생각된다.

Production, Purification, and Characterization of Antifungal Metabolite from Pseudomonas aeruginosa SD12, a New Strain Obtained from Tannery Waste Polluted Soil

  • A new strain, SD12, was isolated from tannery waste polluted soil and identified as Pseudomonas aeruginosa on the basis of phenotypic traits and by comparison of 16S rRNA sequences. This bacterium exhibited broad-spectrum antagonistic activity against phytopathogenic fungi. The strain produced phosphatases, cellulases, proteases, pectinases, and HCN and also retained its ability to produce hydroxamate-type siderophore. A bioactive metabolite was isolated from P. aeruginosa SD12 and was characterized as 1-hydroxyphenazine ((1-OH-PHZ) by nuclear magnetic resonance (NMR) spectral analysis. The strain was used as a biocontrol agent against root rot and wilt disease of pyrethrum caused by Rhizoctonia solani. The stain is also reported to increase the growth and biomass of Plantago ovata. The purified compound, 1-hydroxyphenazine, also showed broad-spectrum antagonistic activity towards a range of phytopathogenic fungi, which is the first report of its kind.

Anti-inflammatory Effect of Leaves Extracts from Aralia cordata through Inhibition of NF-κB and MAPKs Signaling in LPS-stimulated RAW264.7 Cells

  • Aralia cordata (A. cordata), which belongs to Araliaceae, is a perennial herb widely distributed in East Asia. We evaluated the anti-inflammatory effect of stems (AC-S), roots (AC-R) and leaves (AC-L) extracted with 100% methanol of A. cordata and elucidated the potential signaling pathway in LPS-stimulated RAW264.7 cells. The AC-L showed a strong anti-inflammatory activity through inhibition of NO production. AC-L dose-dependently inhibited NO production by suppressing iNOS, COX-2 and $IL-{\beta}$ expression in LPS-stimulated RAW264.7 cells. AC-L inhibited the degradation and phosphorylation of $I{\kappa}B-{\alpha}$, which donated to the inhibition of p65 nuclear accumulation and $NF-{\kappa}B$ activation. Furthermore, AC-L suppressed the phosphorylation of ERK1/2 and p38. These results suggested that AC-L may utilize anti-inflammatory activity by blocking $NF-{\kappa}B$ and MAPK signaling pathway and indicated that the AC-L can be used as a natural anti-inflammatory drugs.

Plant Regeneration from Anther Culture of Panax ginseng

  • The research concerned of the regeneration of plants from embryos obtained from anther cultures of ginseng (Panax ginseng C. A. Meyer). The aim was to determine the influence of the regeneration medium on the efficiency of the regeneration process. We conducted to determine the optimum conditions such as cold pretreatment, plant growth regulators and carbon sources on anther culture of P. ginseng. Highest callus formation rate was obtained when flower buds pretreated at $4^{\circ}C$ for 1 day. Among the treated growth regulators with various degrees of concentration in Murashige and Skoog's (MS) medium, 4.53 ${\mu}M$ of 2.4-dichlorophenoxyacetic acid and 4.44 ${\mu}M$ of 6-benzylaminopurine gives the most responsive callus with the frequency of 73.89% and 129.53 g of fresh weight. When we used 3-9% of sucrose and maltose among the different kinds and various concentrations of carbohydrates, callus was formed highest 67.29% in the medium with 3% of sucrose. Shoots induced from callus supplemented with 28.9 ${\mu}M$ of gibberellic acid and rooted in Gamborg's B5 medium supplemented with 14.7 ${\mu}M$ of indole-3-butyric acid.

Ecological and Ethnomedicinal Survey of Plants within Homesteads in Abia State, Nigeria

  • Ecological and ethnomedicinal survey of plants was conducted in one hundred and twenty homesteads in Mbala, Amuda, Umuaku, and Nneato communities of Nneochi Local Governement Area, Abia State-Nigeria. A total of ninety-one medicinal plant species belonging to seventy-eight genera and forty-eight families, used in the treatment of malaria, yellow fever, fibroid, hepatitis, convulsion, hypertension, diabetes, insomnia, ulcer, rashes, low sperm count, snake bite, among others, were documented. Plant remedies were prepared mostly as infusions or decoctions from different plant parts with mainly water, and palm wine/gin sometimes. The highest number of medicinal plant species (73) was recorded in Mbala, followed by Amuda (71), Umuaku (68) and Nneato (61). Medicinal plant species diversity was highest in Amuda (Simpson 1-D=0.9621;H=3.663), followed by Umuaku (Simpson 1-D=0.9481; H=3.471), Mbala (Simpson 1-D=0.9345; H=3.341), and Nneato (Simpson 1-D=0.9307; H=3.277), respectively. Similarity in medicinal plant species was highest between Umuaku and Nneato (76.71%), followed by Amuda and Umuaku (75.95%), Mbala and Amuda (71.43%), while Mbala and Nneato had the lowest similarity (59.52%). The results of the study showed that traditional medicine is pivotal in the treatment of ailments in the study area, and that the indigenous people of Nneochi have recognized the need to conserve medicinal plants of importance ex situ within homesteads due to threats from unsustainable exploitation and deforestation.

하우스 재배(栽培) 잇꽃의 생육(生育) 및 종실수양(種實收量) (Growth and Seed Yield of Safflower in Plastic house)

  • 홍화(紅花)의 안전다수확(安全多收穫) 재배기술(栽培技術) 확립(確立) 일환(一環)으로 무처리(無處理), 파종시(播種時), 개화시(開花時), 개화기(開花期), 개화후(開花後) 10일부터 수확기(收穫期)까지 비가림재배의 효과를 구명코자 시험한 결과를 요약하면 다음과 같다. 홍화(紅花)는 일찍 비가림 재배에 의하여 출아기(出芽期)와 개화기(開花期)가 빨랐으며 지상부 생육이 양호하였다. 10a당 종실(種實)은 파종부터 수확기까지 일찍 비가림한 것이 387kg으로 노지재배 198kg에 비해 95%증수 되어 비가림 적기로 판단되었다. 홍화 종실의 색도는 파종부터 수확기까지 비가림 한 것이 명도가 가장 높고 적색도와 총색도가 가장 낮아 거의 횐색 을 나타내었다. 비가림 시기에 따른 경제성(經濟性)은 파종부터 수확기까지 비가림이 가장 높았는데 노지재배 10a당 소득 3,675천원에 비해 파종부터 수확기까지 비가림은 6,705천원으로 82%정도 소득이 향상되었다.

Ginsenoside compound K protects human umbilical vein endothelial cells against oxidized low-density lipoprotein-induced injury via inhibition of nuclear factor-κB, p38, and JNK MAPK pathways

  • Background: Oxidized low-density lipoprotein (ox-LDL) causes vascular endothelial cell inflammatory response and apoptosis and plays an important role in the development and progression of atherosclerosis. Ginsenoside compound K (CK), a metabolite produced by the hydrolysis of ginsenoside Rb1, possesses strong anti-inflammatory effects. However, whether or not CK protects ox-LDL-damaged endothelial cells and the potential mechanisms have not been elucidated. Methods: In our study, cell viability was tested using a 3-(4, 5-dimethylthiazol-2yl-)-2,5-diphenyl tetrazolium bromide (MTT) assay. Expression levels of interleukin-6, monocyte chemoattractant protein-1, tumor necrosis factor-${\alpha}$, intercellular adhesion molecule-1, and vascular cell adhesion molecule-1 were determined by enzyme-linked immunosorbent assay and Western blotting. Mitochondrial membrane potential (${\Delta}{\Psi}m$) was detected using JC-1. The cell apoptotic percentage was measured by the Annexin V/ propidium iodide (PI) assay, lactate dehydrogenase, and caspase-3 expression. Apoptosis-related proteins, nuclear factor $(NF)-{\kappa}B$, and mitogen-activated protein kinases (MAPK) signaling pathways protein expression were quantified by Western blotting. Results: Our results demonstrated that CK could ameliorate ox-LDL-induced human umbilical vein endothelial cells (HUVECs) inflammation and apoptosis, $NF-{\kappa}B$ nuclear translocation, and the phosphorylation of p38 and c-Jun N-terminal kinase (JNK). Moreover, anisomycin, an activator of p38 and JNK, significantly abolished the anti-apoptotic effects of CK. Conclusion: These results demonstrate that CK prevents ox-LDL-induced HUVECs inflammation and apoptosis through inhibiting the $NF-{\kappa}B$, p38, and JNK MAPK signaling pathways. Thus, CK is a candidate drug for atherosclerosis treatment.

The root extract of Paeonia lactiflora Pall inhibits the oxidative damage via its anti-oxidant activity

  • Objectives : Reactive oxygen species (ROS) have been associated with pathogenic processes including carcinogenesis through direct effect on DNA directly and by acting as a tumor promoter. Therefore, it has been regarded that ROS may be a major target for cancer prevention. The root of Paeonia lactiflora pall (PL), a traditional Chinese herb, has been a component of effective prescriptions for treatment of liver disease. Also, there are some reports about the antioxidant activities of the extracts from PL. However, little has been known about the effects of PL against oxidative damage. This work aimed to elucidate the anti-oxidant effects of Paeonia lactiflora pall (PL) in the non-cellular system and cellular system. Methods : Antioxidant activities of PL were evaluated by hydroxyl radical scavenging assay and $Fe^{2+}$ chelating assay. Anti-oxidative effect of PL was evaluated by ${\varphi}X$-174 RF I plasmid DNA cleavage assay in non-cellular system. In addition, DNA migration assay, expression level of phospho-H2AX, MTT assay and lipid peroxidation assay were performed for evaluate the anti-oxidative effect of PL in cellular system. Results : PL had a dose-dependent hydroxyl radical scavenging and $Fe^{2+}$ chelating capacity. In addition, PL inhibited oxidative DNA and cell damage induced by hydroxyl radical in non-cellular system and cellular system. Conclusion : Taken together, P. lactiflora pall may be possible for the application to a potential drug for treating the oxidative diseases such as cancer.
