약용식물의 품질평가 현황과 전망 (Current Status and Prospects for the Quality Evaluation of Medicinal Plants)

  • This is a brief review and discussion for present status and prospects of quality evaluation in medicinal plants as oriental medicine materials (OMM). Quality in medicinal plants could be defined as the combination of origin, external appearance, effectiveness and safety, and be evaluated by plant taxonomic, morphological, physiochemical and biological methods. For high-quality improvement and standardization of OMM, medicinal plants should be produced through using proper species or good variety and standard cultivation method in suitable cultivation area. Standardizing of quality means to meet with minimum qualifying criteria of OMM in the market while improving of quality to be over standard quality of medicinal piano in production. For making new high quality and standardized products, we need to keep standard field plants, standard OMM, standard plant specimen, and standard compounds. Researchers and administration have to study and propose the quality factors and their evaluating techniques and criteria, so high-quality and standardized produce of medicinal plane could be produced and distributed under the control of the relevant regulations, and would be contributed to increasing of national health.

An efficient transformation method for a potato (Solanum tuberosum L. var. Atlantic)

  • We found that a long period of in vitro culture is a critical factor on the low transformation rate for a specific potato genotype, Solanum tuberosum L. var. Atlantic when phosphinothricin (PPT) was added to select putative transformants in a solid media. The fresh explants of the newly produced plants from a micro-tuber was able to increase the transformation rate significantly while the old explants prepared from a plant maintained for longer than 6 months in vitro by sub-culturing every 3 ~ 4 weeks resulted in a very low transformation frequency. However, Jowon cultivar was not so much influenced by the period of in vitro culture with high transformation rate (higher than 10.0%). Further research need to be explored for the reason why a particular potato genotype, Atlantic is more vulnerable than the Jowon cultivar during the regeneration stage resulting in the low transformation frequency.

Antimicrobial activity of Mongolian medicinal plants

  • The antimicrobial activity of seventy five ethanol extracts obtained from 67 different kinds of plant species of the Mongolian flora were evaluated by means of the disc diffusion method against five species of microorganisms, Escherichia coli, Enterococcus faecalis, Staphylococcus aureus, Micrococcus luteus and Pseudomonas aeruginosa. Among the plant extracts examined, 34 kinds of extracts demonstrated significant antibacterial activity against one or more species of microorganisms, respectively. Especially, the root extract of Paeonia anomala, the whole herb extract of Myricaria alopecuroides, the whole herb extract of comarum zalesovianum, the whole herb extract of Agrimonia pilosa and some other plant extracts demonstrated a particularly potent antimicrobial activity. The ethylacetate fractions obtained from the whole herb extract of Myricaria alopecuroides and from those of Sedum aizoon, Paeonia anomala, Sedum hybridum and Dasiphora fruticosa exhibited a particularly potent antibacterial activity especially against Staphylococcus aureus and Micrococcus luteus.

Anti-Inflammatory Effects of Ethyl Acetate Fraction from Cnidium officinale Makino on LPS-Stimulated RAW 264.7 and THP-1 Cells

  • This work aimed to elucidate the anti-inflammatory effects of ethyl acetate fraction from Cnidium officinale Makino with a cellular system of LPS-stimulated RAW 264.7 and THP-1 cells. Some key pro-inflammatory cytokines and mediators including NO, iNOS, $PGE_2$, COX-2, TNF-${\alpha}$, NF-${\kappa}B$ p50 and NF-${\kappa}B$ p65 were studied by sandwich ELISA and western blot analysis. Ethyl acetate fraction could significantly inhibit the production of NO, $PGE_2$, TNF-${\alpha}$, iNOS and COX-2 in LPS-stimulated cell than that of single LPS-stimulated. And ethyl acetate fraction suppresses the activation of NF-${\kappa}B$ p50 and NF-${\kappa}B$ p65. All the results showed that ethyl acetate fraction had a good anti-inflammatory effect on LPS-stimulated RAW264.7 and THP-1 cells. Taken together, the anti-inflammatory actions of ethyl acetate fraction from Cnidium officinale Makino might be due to the down-regulation of NO, $PGE_2$, TNF-${\alpha}$, iNOS and COX-2 via the suppression of NF-${\kappa}B$ activation.

Silymarin-Mediated Degradation of c-Myc Contributes to the Inhibition of Cell Proliferation in Human Colorectal Cancer Cells

  • In this study, we elucidated the molecular mechanism of silymarin by which silymarin may inhibits cell proliferation in human colorectal cancer cells in order to search the new potential anti-cancer target associated with the cell growth arrest. Silymarin reduced the level of c-Myc protein but not mRNA level indicating that silymarin-mediated downregulation of c-Myc may result from the proteasomal degradation. In the confirmation of silymarin-mediated c-Myc degradation, MG132 as a proteasome inhibitor attenuated c-Myc degradation by silymarin. In addition, silymarin phosphorylated the threonine-58 (Thr58) of c-Myc and the point mutation of Thr58 to alanine blocked its degradation by silymarin, which indicates that Thr58 phosphorylation may be an important modification for silymarin-mediated c-Myc degradation. We observed that the inhibition of ERK1/2, p38 and $GSK3{\beta}$ blocked the Thr58 phosphorylation and subsequent c-Myc degradation by silymarin. Finally, the point mutation of Thr58 to alanine attenuated silymarin-mediated inhibition of the cell growth. The results suggest that silymarin induces the cell growth arrest through c-Myc proteasomal degradation via ERK1/2, p38 and $GSK3{\beta}-dependent$ Thr58 phosphorylation.

Anti-inflammatory Activity of Cone from Red Pine (Pinus densiflora)

  • Pinus densiflora, the Korean Red Pine, is the predominant tree species of the cool, temperate forests of northeast Asia, occurring in pure stands across Korea, Japan, and parts of northern China and Russia. Pinus densiflora leaves, pollen, and bark have been widely used for traditional medicine, or edible purposes. However, pine cones contain many bioactive phytochemicals, but they are rarely used as natural raw materials. This study was conducted to evaluated the anti-inflammatory effect of pine cone extracts and its possbility of natural sources were evaluated. Pine cones were extracted with 80% methanol, concentrated and then partitioned with ethyl acetate, and the organic layer was used as a sample. The Pine cone Ethyl acetate Fraction (PEF) showed no toxicity to RAW 264.7 cells at a concentration of less than $50{\mu}g/ml$. PEF inhibited the production of nitric oxide (NO) in RAW 264.7 cells treated with lipopolysaccharide (LPS). Also, It suppressed the expression of cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS) and transcription of nuclear factor-kappa B (NF-${\kappa}B$). These results suggest that pine cones can be used as an effective natural material for anti-inflammatory agent.

Anticancer Activity of Taxillus yadoriki Parasitic to Neolitsea sericea against Non-Small Cell Lung Carcinoma

  • In this study, we evaluated the anti-cancer activity and potential molecular mechanism of 70% ethanol extracts of branches from Taxillus yadoriki parasitic to Neolitsea sericea (TN-NS-B) against human lung cancer cells, A549. TY-NS-B dose-dependently suppressed the growth of A549 cells. TY-NS-B decreased ${\beta}$-catenin protein level, but not mRNA level in A549 cells. The downregulation of ${\beta}$-catenin protein level by TY-NS-B was attenuated in the presence of MG132. Although TY-NS-B phosphorylated ${\beta}$-catenin protein, the inhibition of $GSK3{\beta}$ by LiCl did not blocked the reduction of ${\beta}$-catenin by TY-NS-B. In addition, TY-NS-B decreased ${\beta}$-catenin protein in A549 cells transfected with Flag-tagged wild type ${\beta}$-catenin or Flag-tagged S33/S37/T41 mutant ${\beta}$-catenin construct. Our results suggested that TN-NS-B may downregulate ${\beta}$-catenin protein level independent on GSK3${\beta}$-induced ${\beta}$-catenin phosphorylation. Based on these findings, TY-NS-B may be a potential candidate for the development of chemopreventive or therapeutic agents for human lung cancer.

Anticancer Activity of Sageretia thea Through β-catenin Proteasomal Degradation in Human Colorectal Cancer and Lung Cancer Cells

  • In this study, we evaluated the effect of branch (STB) and leave (STL) extracts from Sageretia thea on ${\beta}$-catenin level in human colorecal cancer cells, SW480 and lung cancer cells, A549. STB and STL dose-dependently suppressed the growth of SW480 and A549 cells. STB and STL decreased ${\beta}$-catenin level in both protein and mRNA level. MG132 decreased the downregulation of ${\beta}$-catenin protein level induced by STB and STL. However, the inhibition of $GSK3{\beta}$ by LiCl or ROS scavenging by NAC did not block the reduction of ${\beta}$-catenin protein by STB and STL. Our results suggested that STB and STL may downregulate ${\beta}$-catenin protein level independent on $GSK3{\beta}$ and ROS. Based on these findings, STB and STL may be a potential candidate for the development of chemopreventive or therapeutic agents for human colorectal cancer and lung cancer.

Anti-cancer Activity of the Leave Extracts of Rodgersia podophylla through β-catenin Proteasomal Degradation in Human Cancer Cells

  • In this study, we evaluated the effect of Rodgersia podophylla leave extracts (RPL) on ${\beta}$-catenin level in human cancer cells. RPL dose-dependently inhibited cell proliferation in SW480, A549, MDA-MB-231, PC-3 and AsPC-1 cells. RPL dramatically decreased ${\beta}$-catenin protein level in all cancer cells. However, decreased level of ${\beta}$-catenin mRNA expression was observed in A549 and AsPC-1 cells. In addition, RPL dramatically attenuated cyclin D1 mRNA expression in all cancer cells. MG132 decreased the downregulation of ${\beta}$-catenin protein level induced by RPL in all cancer cells, while RPL-induced downregulation of ${\beta}$-catenin was inhibited by the inhibition of $GSK-3{\beta}$ by LiCl in MDA-MB-231 cells. RPL phosphorylated ${\beta}$-catenin and $GSK-3{\beta}$. In addition, the inhibition of $GSK-3{\beta}$ by LiCl attenuated RPL-induced ${\beta}$-catenin phosphorylation. Based on these findings, RPL may be a potential candidate for the development of chemopreventive or therapeutic agents for human cancer.
