The First Report of Fusarium solani Causing Wilting in Cnidium officinale in Korea

  • Cnidium officinale is a perennial herb belonging to the family Umbelliferae. Its origin is China and is also distributed in Korea and Japan. In 2017, a phenomenon of browning and wilting of leaves was discovered in the cultivation field of the National Institute of Forest Science, Suwon, Korea. The pathogens isolated from plants were consistent with Fusarium solani as a result of morphological examination and molecular phylogenetic analysis. This is the first record of F. solani on C. officinale in Korea.

Fatty acid composition and antioxidant capacity of some medicinal mushrooms in Turkey

  • Turkey has a very rich fungal flora due to its phytogeographical position. The screening of chemical content and active substances of mushrooms becomes an important subject not only for Turkey but also for all over the world. In the last decade, Analyses on phytochemical and biological activity of fungi have gradually increased as a result of improvement in the number and quality of facilities. In the scope of the present research, four medicinal mushrooms; Morchella elata, Lactarius volemus, Cantharellus cibarius and Tricholoma terreum were analyzed for their fatty acid compositions and antioxidant capacities. The fungal species have been found with unsaturated fatty acid/saturated fatty acid ratio of 6.73 for Morchella elata, 4.12 for Lactarius volemus, 5.21 for Cantharellus cibarius, 3.73 for Tricholama terrum. In addition, the concentration of malondialdehyde which was an indicator of lipid peroxidation was also determined in these species. According to the results, free radical scavenging activity of Morchella elata and Lactarius volemus were found higher than the other species. Any of the mushroom species investigated were found having very high metal chelating activity. The results showed that the extract of Morchella elata and Lactarius volemus exhibited significant antioxidant activities. Hence, the mushrooms have a potential to be a natural antioxidant in food industries as antioxidant agent.

인삼 주요병에 대한 길항미생물 선발 (Isolation of Antagonistic Bacteria against Major Diseases in Panax ginseng C.A. Meyer)

  • 인삼의 안정적인 생산 및 합리적인 병해방제에 기여하기 위하여 연구를 실시한 결과 갈잎 퇴비에서 분리된 Streptomyces lauretii B8180과 Bacillus subtilis B8856, Burkholderia cepacia B7944 세 균주의 인삼병원균 5종에 대한 항균력이 인정되었으며, 특히 실내실험을 통하여 높은 역병 방제효과가 인정되었다. 그러나 이 실험에서와 같이 식물의 지상부에 길항균을 살포하는 방법을 통하여 역병 방제효과를 높일 수 있을 것으로 생각된다. 이 길항균들은 갈잎 퇴비에서 분리되었기 때문에, 갈잎 퇴비에서 증식시켜 인삼재배 예정지에 투입하면, 억제형 토양 (suppressive soil) 조성을 통한 주요병해 방제가능성도 클 것으로 생각된다.

Diversity and Bioactive Potential of Culturable Fungal Endophytes of Medicinal Shrub Berberis aristata DC.: A First Report

  • Bioactive natural compounds, isolated from fungal endophytes, play a promising role in the search for novel drugs. They are an inspiring source for researchers due to their enormous structural diversity and complexity. During the present study fungal endophytes were isolated from a well-known medicinal shrub, Berberis aristata DC. and were explored for their antagonistic and antioxidant potential. B. aristata, an important medicinal shrub with remarkable pharmacological properties, is native to Northern Himalayan region. A total of 131 endophytic fungal isolates belonging to eighteen species and nine genera were obtained from three hundred and thirty surface sterilized segments of different tissues of B. aristata. The isolated fungi were classified on the basis of morphological and molecular analysis. Diversity and species richness was found to be higher in leaf tissues as compared to root and stem. Antibacterial activity demonstrated that the crude ethyl acetate extract of 80% isolates exhibited significant results against one or more bacterial pathogens. Ethyl acetate extract of Alternaria macrospora was found to have potential antibacterial activity. Significant antioxidant activity was also found in crude ethyl acetate extracts of Alternaria alternata and Aspergillus flavus. Similarly, antagonistic activity of the fungal endophytes revealed that all antagonists possessed inhibition potential against more than one fungal pathogen. This study is an important step towards tapping endophytic fungal diversity for bioactive metabolites which could be a step forward towards development of novel therapeutic agents.

Characteristic for Growth and Fruit Body Polysaccharide of Caterpillar Fungi, Cordyceps sobolifera(Hill.) Berk. et Br.

  • Mycelial expansion and fruit body development of caterpillar fungi, Cordyceps sobolifera(Hill.) Berk. et Br. was examined on various carbon and nitrogen sources, some substrates with various supplements. Best temperature for the mycelial expansion was $25^{\circ}C$, while optimal pH was 7. Glucose and polypeptone were the best sources of carbon and nitrogen, respectively, as well as 1:1 was the best ratio of carbon and nitrogen. The unpolish rice and bean curd lees were used as core substrates combined individually with five various supplements. In substrates based, all the supplements with all combinations were found to exhibit the mycelial growth and fruit body formation. Likewise, yolk was found as a superior supplement and also clarified quite high polysaccharide content in the fruit body.

잔디 병해 방제를 위한 항균성 약용식물의 탐색 (Screening of Antifungal Medicinal Plants for Turfgrass Fungal Disease Control)

  • 잔디에 발생하는 주요 병해로 한지형 잔디에서 갈색잎마름병(Rhizoctonia solani AG1-1), 동전마름병(S. homoeocarpa), 피시움마름병(Pythium. spp.), 탄저병(C. graminicola)과 난지형 잔디에서 누른잎마름병(R. cerealis), 라이족토니아마름병 (R. solani AG2-2)이다. 본 연구에서는 이들 잔디 주요 병원균 6종에 대한 한약재 23종의 항진균 활성을 알아보았다. 디스크 확산법으로 항진균 활성이 있는 한약재를 선발하였고, 선발된 한약재를 대상으로 균사생육억제 효과를 실험하여 검정하였다. 항진균 활성 시험 결과, 한약재 23종 중고삼 외 11종의 시료에서 항진균 활성이 있었으며, 균사생육 억제효과를 통한 검정에서는 10 mg/10 ml(1000배) 추출물 첨가 배지에서 잔디 병원균 6종 중 1종이라도 균사 억제율이 80% 이상인 추출물은 고삼, 울금, 대황, 황련, 족두리풀로 나타났다. 특히 고삼 추출물은 실험 대상인 잔디 병원균 6종에 대해 모두 항진균 활성이 나타났고, 울금 추출물 10 mg/10 ml(1000배), 5 mg/10 ml(2000배), 2 mg/10 ml(5000배) 첨가 배지에서 S. homoeocarpa 에 대해 100% 균사 생육 억제 효과를 나타내었다. 그리고 Pythium spp. 균사 억제효과가 100%인 것은 고삼, 대황, 황련으로 나타났다.

Phytophthora sansomeana에 의한 큰꽃삽주 역병 발생 보고 (Occurrence of the Phytophthora Blight Caused by Phytophthora sansomeana in Atractylodes macrocephala Koidz.)

  • Background: In September 2017, wilting and rhizome rot symptoms were observed on Atractylodes macrocephala Koidz. in Jecheon-si and Eumseong-gun. This study was carried out to isolate hitherto unidentified pathogenic fungi from A. macrocephala and to test the pathogenicity of isolated fungi against Atractylodes spp. genus such as A. macrocephala, A. japonica, and their interspecific hybrids. Methods and Results: The diseased plants were washed with running tap water, and the boundary between the healthy area and the diseased area was cut while the pathogens were isolated by growing cultures from the diseased areas on Phytophthora semi-selective medium. The internal transcribed spacer (ITS) region of the isolates was used in this study for identification. Test plants were cultivated in the glasshouse at 20℃ - 30℃ for 4 months and then used for pathogenicity test. The pots with plants inoculated with mycelial plugs and zoospores were placed at 25℃ for 48 h in a dew chamber where relative humidity was above 95%, and then moved into the glasshouse at 20℃ - 30℃. The presence or absence of pathogenicity of the strains was determined by evaluating the symptom of plant wilting. The inoculation test was performed in three replicates with a non-treated control. Conclusions: On the basis of results of ITS sequence analysis, the strains isolated from the diseased plants was identified as Phytophthora sansomeana. Biological assay using test plants confirmed the pathogenicity of P. sansomeana against Atractylodes macrocephala. This is the first report of rhizome rot in A. macrocephala caused by P. sansomeana.
