Receptor activator of nuclear factor-kappa B ligand (RANKL) is a critical factor in osteoclastogenesis. It makes osteoclasts differentiate and multinucleate in bone remodeling. In the present study, RANKL was expressed as a soluble maltose binding protein (MBP)-fusion protein using the Escherichia coli maltose binding domain tag system (pMAL) expression vector system. The host cell E. coli DH5α was cultured and induced by isopropyl β-D-1-thiogalactopyranoside for rRANKL expression. Cells were disrupted by sonication to collect soluble MBP-fused rRANKL. The MBP-fusion rRANKL was purified with MBP Trap affinity chromatography and treated with Tobacco Etch Virus nuclear inclusion endopeptidase (TEV protease) to remove the MBP fusion protein. Dialysis was then carried out to remove binding maltose from the cleaved rRANKL solution. The cleaved rRANKL was purified with a second MBP Trap affinity chromatography to separate unsevered MBP-fusion rRANKL and cleaved MBP fusion protein. The purified rRANKL was shown to have biological activity by performing in vitro cell tests. In conclusion, biologically active rRANKL was successfully purified by a simple two-step chromatography purification process with one column.
Purpose: The purpose of this study was to verify the influence of health point system on the lifestyle among the patients with chronic diseases listed in iCDMS, a project of the Incheon Metropolitan Center for Chronic Disease Control and Prevention. Methods: The data were collected 1,000 persons among the patients listed in iCDMS from March 16, 2009 to December 21, 2010 by telephone survey. The data were analyzed by descriptive statistics, ${\chi}^2$-test, odds ratio and logistic regression with the SPSS 18.0 program. Results: The higher the percentage of the accumulation of the points of necessary medical examinations they have, the better the lifestyle the participants practice moderation in drinking, exercise, and diet (p<.05). Also the higher the percentage e accumulative points of education and visit, the stronger the intention to improve the lifestyle such as for example, receiving the guidance of no smoking, giving up drinking, or being conscious of nutrition and obesity (p<.05). Conclusion: This study suggests that the motivation through an incentive system can increase self-care make an effect on the care of patients with chronic diseases.
This Study was conducted to describe relatioship between health promoting behaviors and Self-efficacy of 427 Students during the period form June 1. 1994 to June 30. 1994. The research Scale used Kim and Choe's (994) that was modified Sheredr and Maddux's (1982) Self-efficacy measurement scale. Data was. analysed by SPSS-PC program. and reavealed to percentage. t-value. F-value and Pearson's correlation coefficency. 1. The general characteristics of subjects was that men $55\%$. average age 22.5years old. There are the most highest destribution in religion. abscence $52.2\%$. and in residence. own house $49.9\%$ There are 129 subjects $(30.2\%)$ regularity exerciseed now. and 46 subjects $(35.7\%)$ exercise time was above 30 mins below 1 hour. There are 78 subjects $(60.5\%)$ above 4 times per weeks the most highest distribution in exercise times. 2. Health promoting behaviors score of subjects was 94.287 (2.548) and self-efficacy was 967.63 (69.12) 3. There are statistically significant difference in health promoting behavior score according to sex. sibling number. residence place (p<.05). In the subconcept of health promotion lifestyle profile (HPLP). there was the more higher score in men than women for self actualization (t=2. 67. p=.008). exercise(t=5.92. p=.000). There are statistically significant difference in nutrition according to sibling number (F=3.05. p=0.01). resident place (F=2.93. p=0.02). and in interpersonal support according to religion (F=2.88. p=0.02). 4. In the Self-efficacy score. there was statistically significant difference according to sex (t= 5.88. p=.000). 5. There was postive correlation between health promoting behavior and self-efficacy (r=.43. p=.000). On the basis of this result. I hope that develp Korean type health promoting behavior scale to understand health promotion for people. and nursing intervention method to improve health promoting behavior through increasment of Self-efficacy.

Gestational diabetes mellitus (GDM), one of the common metabolic disorders of pregnancy, leads to functional alterations in various cells including stem cells as well as some abnormalities in fetal development. Perivascular stem cells (PVCs) have gained more attention in recent years, for the treatment of various diseases. However, the effect of GDM on PVC function has not been investigated. In our study, we isolated PVCs from umbilical cord of normal pregnant women and GDM patients and compared their phenotypes and function. There is no significant difference in phenotypic expression, response to bFGF exposure and adipogenic differentiation capacity between normal (N)-PVCs and GDM-PVCs. However, when compared with N-PVCs, early passage GDM-PVCs displayed decreased initial rates of cell yield and proliferation as well as a reduced ability to promote wound closure. These results suggest that maternal metabolic dysregulation during gestation can alter the function of endogenous multipotent stem cells, which may impact their therapeutic effectiveness.
Objectives: The purpose of this study is to investigate the effects of the M&L Trauma Psychotherapy Program on: Impact of Event Scale - Revised (IES-R-K); Euro QOL-5 Dimension (EQ5D); Five-facet Mindfulness Questionnaire (FFMQ); and psychological tests. Methods: Eight middle-aged women subjected to domestic violence participated in the two-day M&L Trauma Psychotherapy Program. The Program was executed 5 times 2 days for about 3 hours. We evaluated IES-R-K, EQ5D, FFMQ, Beck Depression Inventory (BDI), State-Trait Anxiety Inventory (STAI), State-Trait Anger Expression inventory (STAXI) and SUDS before the Program and for four weeks afterwards, to analyze the effects of the M&L Trauma Psychotherapy Program. Results: The scores of IES-R-K, BDI, STAI-S, STAI-T, Hwa ST and Hwa CT decreased significantly (p<0.05). EQ5D scores increased significantly (p<0.05) and overall physical condition scale (VAS) increased. In the FFMQ scores, all five facet scores increased slightly. Scores of SUDS decreased significantly right after the program, and remained decreased four weeks later, rather than before the Program (p<0.05). Conclusions: The results suggested that the M&L Trauma Psychotherapy Program improved post-traumatic stress, quality of life and mindfulness skills, and had positive responses to psychological problems - depression, anxiety, anger and distress.
Purpose: This study was done to investigate the relationship between depression, self-care activity and $HbA_1c$ in clients with type-2 diabetes mellitus (DM) clients. Method: This was a cross sectional descriptive study and data were collected between October 4 and 27, 2006 using a questionnaire. The participants were inpatients and outpatients with type-2 diabetes being seen in one of two hospitals. The questionnaire included general as well as disease related characteristics, a depression scale (CES-D), and the self-care activity scale developed by Kim (1996). $HbA_1c$ was obtained from medical records. Results: There were 50 men (41.7%) and 70 women (58.3%). The mean scores for depression and self-care activity were $18.22{\pm}10.03$ and 3.51, respectively. Almost half of the Participants (46.7%) had an $HbA_1c$ greater than 7%. Self-care activity was negatively correlated with depression (r=-.436, p=0.00) and $HbA_1c$ (r=-.269, p=0.03). Depression was positively correlated with $HbA_1c$ (r=-.325, p=0.00). Participants who had diabetes education showed a significantly lower level of $HbA_1c$ than the group who had never had diabetes education. Conclusions: It is Important to make ensure that patients with diabetes have education on diabetes mellitus and to screen and manage depression as part of the care for these patients.
Purpose: The purpose of this study was to compare sociodemographic characteristics of a normal cognitive group and mild cognitive impairment group, and establish prediction models of Mild Cognitive Impairment (MCI). Methods: This study was a secondary data analysis research using data from "the 4th Korea Longitudinal Study of Ageing" of the Korea Employment Information Service. A total of 6,405 individuals, including 1,329 individuals with MCI and 5,076 individuals with normal cognitive abilities, were part of the study. Based on the panel survey items, the research used 28 variables. The methods of analysis included a χ2-test, logistic regression analysis, decision tree analysis, predicted error rate, and an ROC curve calculated using SPSS 23.0 and SAS 13.2. Results: In the MCI group, the mean age was 71.4 and 65.8% of the participants was women. There were statistically significant differences in gender, age, and education in both groups. Predictors of MCI determined by using a logistic regression analysis were gender, age, education, instrumental activity of daily living (IADL), perceived health status, participation group, cultural activities, and life satisfaction. Decision tree analysis of predictors of MCI identified education, age, life satisfaction, and IADL as predictors. Conclusion: The accuracy of logistic regression model for MCI is slightly higher than that of decision tree model. The implementation of the prediction model for MCI established in this study may be utilized to identify middle-aged and elderly people with risks of MCI. Therefore, this study may contribute to the prevention and reduction of dementia.
Objective: Congenital muscular torticollis (CMT) is a disease with abnormal head and neck posture in infants. It affects the child's movement and development and can lead to complications. Therefore, this study aims to find out what factors influence the rehabilitation treatment duration of infants with CMT. Design: Cross-sectional study. Methods: The subjects were 63 infants under 90 days of age who were diagnosed with CMT. Age, thickness of the sternocleidomastoid muscle (SCM) on the affected and non-affected side, head tilt angle, and head rotation angle of the affected head in infants were collected. The ratio of muscle thickness was calculated from the thickness of the affected SCM and the thickness of the non-affected SCM (A/N ratio). All subjects underwent conservative physical therapy twice a week for 30 minutes, and the end of the treatment was when the angle of head tilt was normal or less than 5 degrees, and the treatment duration was calculated. Results: Age, thickness of affected SCM, and head tilt were significantly correlated with treatment duration (p<0.05). The thickness of the non-affected SCM, A/N ratio, and head rotation angle did not show any correlation with treatment duration. The factors affecting the treatment duration were head tilt and age, showing 21% explanatory power of adjusted R2. Conclusions: The main factors affecting the treatment duration of infants with CMT are head tilt and age. Therefore, more attention should be directed to the infant's head tilt and age for effective physical therapy of infants with CMT.
The purpose of this study was to investigate the effect of aqua exercise on women 50 years of age or older with knee degenerative arthritis, and then examining the therapeutic value of aqua exercise in degenerative arthritis of the knee. The subjects of the study were 115 residents 50 years of age or older who had been diagnosed as degenerative arthritis of the knee by doctors from medical institutions and applied for the aqua exercise program in the Public Health Care Center in Suseong-gu in 2006 and 2007. Experiments were conducted to all the subjects with the application of the 8-week aqua exercise program (2 times a week, 50 minutes a time), designed in the study, and the differences before and after the aqua exercise were compared through questionnaires for measuring the state of the body such as the knee extension angles, weight, body fat, and abdominal obesity. The data was analyzed by t-test and the Wilcoxon method through SPSS 14.0 and the statistical significance level ${\alpha}$ was set at 0.05 and 0.01. The results of the study are as the following. First, In comparison of before and after the aqua exercise, there was improvement in the extension angle of the right knee(p<0.01), while there was some improvement in the extension angle of the left knee, it was not significant. Second, In comparison of before and after the aqua exercise there were statistically significant differences in weight(p<0.05), body fat ratio, and abdominal obesity(p<0.01). The results of this study showed that the aqua exercise program to female patients with knee degenerative arthritis significantly increased the extension angle of the knee. Therefore it is thought that aqua exercise is an effective meditating method to improve their symptoms of female patients 50 years of age or older

Objective: Human milk bank is known as best alternative option for sick infants. However, only two human milk banks (HMBs) are working in Korea. In the present study, we evaluated the awareness on the HMB in female health care provider to find out the issues that would help facilitating HMB establishment in Korea. Methods: The survey questionnaire was developed by a team composed with neonatologists, obstetricians, nurses, nutritionists, and health care specialists. The survey was performed for female health care workers in Soon Chun Hyang Cheonan Hospital between March and June in 2016. Eighty-eight of 110 questionnaires were collected and the results were analyzed. Results: Most of subjects were found to have correct knowledge on the breast milk feeding. However, only 49% of subjects had awareness of the presence of HMB. Unfortunately, less than half of them could get information about HMB by official way including mass media. In addition, 76% of subjects were found not to want to use HMB. Most important reason was found as a concern on the safety of donor milk including life style of donors, infection controls, and possible nutritional loss of banked milk. Conclusion: The purpose and function of HMB was not widely educated and it seems to be one of most important reasons why women have negative concept about HMB in Korea. Thus, further study is warranted with more numbers of subjects after active public education about the HMB.
