A Consideration of Logit Transformation for Estimating the Dosage-Mortality Regression Equation (약량 반응곡선의 추정에 있어서 Logit 변환법의 이용)

  • With the current advances in insect toxicant bioassay, the need for easy methods of estimating the dosage-mortality regression equation has become vital. The Probit analysis seems to be not convenient for estimating the dosage-mortality regression equation and median lethal dose(LD50) because of its complexity in calculation. This study presents a comparision between Probit and Losit transformation for the estimation from bioassay results. Validation of the two methods is presented for the pathogenecity of nuclear polyhedrosis virus to the larva of fall web worm, Hyphantria cunea D.

Effect of Copper on Lethal Dose and Pupation of Aedes albopictus (Diptera: Culicidae) (흰줄숲모기, Aedes albopictus (Diptera: Culicidae)의 치사농도 및 용화에 대한 구리의 영향)

  • The mosquito, Aedes albopictus is important vector of humen phatogen such as dengue fever. And Aedes albopictus has widly distribution in oriental regions. This study was conducted to investigate the biological effects of cooper on the development of the Asian tiger mosquitoes, Aedes albopictus (Skuse). Aedes albopictus was affected with pupation ratios, mortalities, and wing lengths of the 3th and 4th instars by exposure to different concentration $CuCl_2$ (0.0, 2.5, 25.0 and 50.0 ppm) for 24 h and 48 h depending on dose and exposure period. The lowest $LC_{50}$ (median lethal concentration) value of $CuCl_2$ was 18.1 ppm at the 3rd instars for 48 h exposure. The pupation ratio of the larvae exposed at 50.0 ppm for 48 h was 14% which was much lower than those exposed to other treatment groups Changes of adult wing length showed significant decreased by Cu treatment. In conclusion, the most prominent effects of high concentration and longer exposure period were to reduce in the survival and pupation rates of larvae and wing length of Ae. albopictus.

  • Purpose: Anaplastic thyroid cancer (ATC) is a rare tumor with a lethal clinical course despite aggressive multimodal therapy. Intensity-modulated radiotherapy (IMRT) may achieve a good therapeutic outcome in ATC patients, and the role of IMRT should be assessed. We retrospectively reviewed outcomes for ATC treated with three-dimensional conformal radiotherapy (3D-CRT) or IMRT to determine the optimal treatment option and explore the role of radiotherapy (RT). Materials and Methods: Between December 2000 and December 2015, 41 patients with pathologically proven ATC received RT with a sufficient dose of ${\geq}40Gy$. Among them, 21 patients (51%) underwent surgery before RT. Twenty-eight patients received IMRT, and 13 received 3D-CRT. Overall survival (OS) and progression-free survival (PFS), patterns of failure, and toxicity were examined. Results: The median follow-up time for survivors was 38.0 months. The median and 1-year OS and PFS rates were 7.2 months and 29%, 4.5 months and 15%, respectively. Surgery significantly improved the prognosis (median OS: 10.7 vs. 3.9 months, p = 0.001; median PFS: 5.9 vs. 2.5 months, p = 0.007). IMRT showed significantly better PFS and OS than 3D-CRT, even in multivariate analysis (OS: hazard ratio [HR] = 0.30, p = 0.005; PFS: HR = 0.33, p = 0.005). Significantly higher radiation dose could be delivered with IMRT than 3D-CRT ($EQD2_{10}$ 66 vs. 60 Gy, p = 0.005). Only 2 patients had grade III dermatitis after IMRT. No other severe toxicity ${\geq}grade$ III occurred. Conclusion: Patients with ATC showed better prognosis through multimodal treatment. Furthermore, IMRT could achieve favorable survival rates by safely delivering higher dose than 3D-CRT.

Effects of Gamma-ray Irradiation on Radio Sensitivity in Oat (Avena sativa) (감마선 조사가 귀리(Avena sativa)의 감수성에 미치는 영향)

  • This study examined radiation damage and the optimal gamma-ray dose for mutation breeding in oat (Avena sativa L. cv. Samhan). The seed germination rate decreased as the dose increased over 500 Gy. The median lethal dose (LD50) was approximately 392 Gy. The median reduction dose (RD50) for plant height, tiller number, root length, and flash weight was 411, 403, 394, and 411 Gy, respectively. The optimal dose of gamma irradiation for inducing oat mutation appears to be in the range 300-400 Gy. We performed the comet assay to observe nuclear DNA damage induced by gamma-ray irradiation. This assay showed a clear difference with gamma-ray treatments. DNA damage increased temporarily 7 days after treatment depending on the dose, while no significant difference was identified in response to 300 Gy 30 days after the gamma-ray treatments. The growth characteristics of the M2 generation decreased as the dose increased over 400 Gy.

A Study on the Safety of Korean Domestic Fermented Meju (한국산 재래식 발효메주의 안전성에 관한 연구)

  • For this experiment frist the fermented Soybean Meju was prepared by domestic ordinary method of Korean and concentrated Meju suspensoidal extract was made for toxicological test sample on mice and rat. Even the oral dose of 10g extract per kg of mice had not any acute toxicity. So the median lethal dose(LOD50) is lilely to be much greatr or not to be. In comparison of the average weight gain rate for 4 weeks the control and test groups of 0.308g Ex/rat kg and 9.24g Ex/rat kg daily were the much the same as about 19% but 3.08g Ex/rat kg group waas exceptionally 2.7% higher than control For this period all test groups did not show any abnormality in appearance and anatomic findings. In serum GPT GOT alkaline phosphatase and blood urea nitrogen value there were no difference significantly between control and the test groups of 0.308g/rat kg and 3.08g/rat kg but 9.24g/rat kg group was some what higher than control. Especially in cholesterol alkaline phosphatase and blood urea nitrogen value 9.24g/rat kg group or 28days had 100mg/이 37.5u 32mg/이 respectively which were increased significantly. The pathological findings of all test groups especially 9.24g/kg group did not showed any characterized sign of acute or subacute liver damage or intestine lesion. So it is supposed that there is not any tocixants originated from some molds in fermented Korean domestic Meju which is prepared traditionally in winter.

Single Dose Acute Toxicity of Ssanghwa-tang in Crl : CD (SD) Rats (랫드에서 쌍화탕의 급성독성에 관한 연구)

  • Objectives : This study was conducted to evaluate the acute toxicity and safety of Ssanghwa-tang (Shuanhetang in Chinese, Sou-wa-to in Japanese) in Crl : CD Sprague-Dawley (SD) rat though the current regulatory guideline. Methods : In this study, 10 rats of each sex were randomly assigned to two groups of 5 rats each and were administrated singly by gavage at dose levels of 0 and 2000 mg/kg/day of ssanghwa-tang water extract (SHT). After single administration of SHT, mortalities, clinical signs, body weight changes, gross findings were observed for the 15-day period. Results : Acute toxicity tests revealed that a single oral administration of SHT at dose levels of 2000 mg/kg did not affect clinical signs, body weight, and gross findings, evaluating the safety of SHT. The SHT treatment did not result in any toxicologically significant changes in mortality, clinical signs, body weight changes. Conclusions : These results showed that the single oral administration of SHT did not cause any toxic effect at the dose levels of 2000 mg/kg/day in rats. In conclusion, the median lethal dose (LD50) of SHT was considered to be over 2000 mg/kg/day body for both sexes.

Toxic Components of Auricularia polytricha

  • To find biologically active components of the higher fungi of Korea, the carpophores of Auricularia polytricha, a well-known edible mushroom, were extracted with 0.14 M NaCl solution. The extract was successively fractionated by adding ammonium sulfate at various concentrations, and the respective precipitates were separated by centrifugation, then dialyzed and freeze-dried. When a does of 60 mg/kg of each was injected i.p. into ICR mice, the fraction which precipitated at 20% ammonium sulfate showed the highest toxicity, killing seven out of seven mice within two days. The fraction obtained at 40% ammonium sulfate showed the second highest toxicity. The two fractions were named auritoxin I and II after the genus name. However, they Nere shown to have nearly identical composition by physicochemical and 6.8% protein. The polysaccharide moiety was found to have 12.3% $\alpha$-linkage and 87.7% $\beta$-linkage and to be a heteromannoglucan consisting of 45.1% glucose, 435 mannose and 11.0% xylose. The protein moiety contained ten amino adids. The molecular weight of the toxin was $1.5\times10^6$ dalton by Sepharose CL-4B gel filtration. The modian lethal doses of auritoxin in mice were 56.4, 157.2 and 454.6 mg/kg by i.p., s.c. and p.o.administrations, respectively. The signs of intrxication were convulsion during the first 30 minutes after the injection, coma or sleeping within an hour, termor, lacrimation, nasal bleeding congestion, and death in 24 hours. Smong the various organs, the spleen was found to be enlarged remarkably. Human platelet aggregation was inhibited by the addition of auritoxin. The activity of malic dehydrogenase in vitro was inhibited by the toxin.

Radiosensitivity of Lentil Bean (Lens culinaris L.) to Gamma-irradiation (감마선 조사가 렌틸(Lens culinaris L.)의 감수성에 미치는 영향)

  • We examined damages from gamma-irradiaion and determined the optimal gamma-ray dose for mutation breeding in lentil (Lens culinaris L.) bean. Four individual lines (L-C, L-2, L-8 and L-9), that have remarkable adaptability in South Korea were gamma-irradiated at doses of 50, 70, 100, 200, 300, 400, and 500 Gy. The germination rate of seed decreased as the dose increased over 50 Gy in all lines. However, $LD_{50}$ and $RD_{50}$ were different among lines. The median lethal doses($LD_{50}$) were approximately 127 (L-C), 74 (L-2), 95 (L-8), and 144 (L-9) Gy. The median reduction doses($RD_{50}$) for plant height, number of leaves, root length, and flash weight were 156, 176, 150, and 180 Gy for L-C, 253, 198, 127, and 142 Gy for L-2, 188, 175, 200, and 190 Gy for L-8, and 162, 210, 224, and 184 for L-9, respectively. The growth characteristics of the $M_1$ generation decreased as the dose increased over 70 Gy. The optimal doses of gamma irradiation for mutation breeding of lentil were determined to be 70 Gy (L-2, L-8) and 100 Gy (L-C, L-9). We performed the comet assay to observe nuclear DNA damage induced by gamma-irradiation. In comet assay, a clear difference was identified over 100 Gy treatments. With increasing doses of gamma-ray in the range of 50 to 500 Gy, the rate of head DNA was decreased significantly from 97.5% to 81.6%. Tail length was consecutively increased from $1.9{\mu}m$ to $17.4{\mu}m$. Our result provides basic information for construction of mutant pools in lentils.

A Study on Acute Oral Toxicity of Pyungwi-san and Fermented Pyungwi-san in ICR Mice (ICR 마우스를 이용한 평위산과 발효평위산 급성독성 연구)

  • Objectives: This study was conducted to investigate the acute toxicity of Pyungwi-san(Pingwei-san in Chinese) in ICR mice, according to KFDA and OECD guideline. Methods: In the present study, 15 male and female ICR mice administrated singly by gavage at dose levels of 0 and 2000 mg/kg of Pyungwi-san. During the experimental period, no treatment-related death was observed. There were no adverse effects on clinical signs, body weight, and gross findings at all treatment groups. Results: These results showed that the single oral adminstration of Pyungwi-san (Pingwei-san) did not cause any toxic effect at the dose levels of 2000 mg/kg in rats. Conclusions: Taken together, the median lethal dose($LD_{50}$) of Pyungwi-san (Pingwei-san) was considered to be over 2000 mg/kg body for both sexes.

Study on the Acute and Sub-Acute Inhalation Toxicity of 1-Bromopropane in SD Rats (Rat를 이용 1-Bromopropane의 급성 및 아급성 흡입독성 연구)

  • The purpose of this study was to investigate the acute(4 hrs) and repeated-dose(6 hrs a day, 5 days a week, 8 weeks) toxic effects of 1-bromopropane(1-BP) on Sprague-Dawley (SD) rats which were treated by inhalation. The results were as follows ; 1. The median lethal concentration($LC_{50}$) was estimated 14,374 ppm(confidence limit 95% ; 13,624~15,596 ppm) in acute inhalation. Abnormal clinical signs related to the 1-BP were not observed with the acute inhalation dose. Gross findings of necropsy revealed no evidence of specific toxicity related to the 1-BP. 2. By sub-acute inhalation the body weights of male and female were significantly reduced(p<0.001) by the dose of 1,800 ppm compared with control group, while the relative weights of liver were significantly increased(p<0.001) in both sexes. However there were no significant variation in food consumption, urine biochemistry, hematology and blood biochemistry for the exposed rats compared with the control rats. Abnormal clinical signs and gross findings of necropsy related to the 1-BP were not shown. No toxicologic lesions were observed by the histopathological test.
