Nutritional Studies on Production of Antibacterial Activity by the Zebra Mussel Antagonist, Pseudomonas fluorescens CL0145A

  • Pseudomonas fluorescens strain CL0145A was discovered at the New York State Museum Field Research Laboratory as an effective agent against the environmentally destructive zebra mussel, which has contaminated US waters. Dried cells of the microbe are being commercialized as an environmentally friendly solution to the problem. We found that antibiotic activity against the Gram-positive bacterium Bacillus subtilis is produced and excreted by this strain. We have carried out studies to optimize production of the antibiotic. Studies were begun in a complex corn meal medium. Activity was found in both cells and culture supernates and was maximal after one day of fermentation. Static fermentation conditions were found to be superior to shaken culture. Production of extracellular antibiotic in complex medium was found to be dependent on the content of sucrose and enzyme-hydrolyzed casein. Indeed, production was greater in sucrose plus enzyme-hydrolyzed casein than in the complex medium. Of a large number of carbon sources studied as improvements over sucrose, the best was glycerol. An examination of nitrogen sources showed that production was improved by replacement of enzyme-hydrolyzed casein with soy hydrolysates. Production in the simple glycerol-Hy-Soy medium was not improved by addition of an inorganic salt mixture or by complex nitrogen sources, with the exception of malt extract. In an attempt to keep the medium more defined, we studied the effect of amino acids and vitamins as replacements for malt extract. Of 21 amino acids and 7 vitamins, we found tryptophan, glutamine, biotin, and riboflavin to be stimulatory. The final medium contained glycerol, Hy-Soy, tryptophan, glutamine, biotin, and riboflavin.

전북지역 초중고등학교 학교급식소의 내부시설 및 위생관리 실태 조사 (Survey on Internal Facilities and Sanitary Management of Elementary, Middle, and High School Foodservice in Chonbuk Area of Korea)

  • This study was conducted to investigate the situation of internal facilities and sanitary management of elementary, middle, and high school foodservice in Chonbuk area. Self administered questionnaires were collected from 252 nutrition teachers and school dietitians. Statistical data analysis was completed using a SPSS 11.5 program. The results are summarized as follows: Approximately 99.2% of the subjects were women and those who were married accounted for 76.6%. Those in their 30s to 40s accounted for over 87%, those with more than 11 years experience accounted for 56.8%, and those 69.0% were regular nutrition teachers. Among the 280 school foodservice systems, 51.6% of the schools were located in urban areas and 48.0% in rural areas. About 68% of the schools prepared meals the conventional way and 32.3% prepared them the commissary way. The number of employees accounted for in the schools was 37.7% with 1 to 3 employees, 27.8% with 4 to 6 employees, and 25.4% with 7 to 9 employees. About 54% of schools have an expectance of school meal service for more than 11 years. However 67.5% of their facilities had not been remodeled since the implementation of the foodservice. As a result of the conditions of the school foodservice facilities, there were significant differences between elementary and middle & high schools; preparation room (p<0.01), toilet (p<0.01), and boiler room (p<0.01). About 62% of the nutrition teachers responded that they were dissatisfied with the situation of the facilities (e.g. size and materials of the kitchen, heating and cooling systems). The washing and disinfection methods in elementary, middle and high schools were very different, because they had no sanitizers and disinfectant guidelines. Therefore the governmental regulating agencies must review and approve of the plans prior to new construction or extensive remodeling of the school foodservice facilities. In addition, we suggest that it is necessary to provide practical sanitizers and disinfectant guidelines and other useful education.

Effects of Extruded Full Fat Soybean in Early-Weaned Piglets

  • A total of 80 piglets ($5.18{\pm}0.61kg$ of BW; 16 d of age) were fed experimental diets to evaluate the effect of extruded full-fat soybean (EFS) on the growth of eatly weaned pigs. Pigs were allotted into five treatments based on body weight, in a completely randomized block design. Each treatment has 4 replicates of 4 animals each. Treatments consisted of diets representing substitutional ratios of EFS for soybean meal. 1) 100:0 (SBM), 2) 75:25 (EFS 25), 3) 50:50 (EFS 50), 4) 25:75 (EFS 75) and 5) 0:100 (EFS 100). During phase I (d 0 to 7), piglets were fed diets containing 3,340 kcal ME, 26% crude protein, 1.85% lysine, 1.0% Ca and 0.9% P. For phase II (d 8 to 21), piglets were fed diets supplying 3,340 kcal ME, 23% crude protein, 1.65% lysine, 0.9% Ca and 0.8% P. Urease activity (pH rise) of EFS (0.18) was three times higher than that of SBM (0.06) indicating that processing conditions were not efficient enough to inactivate urease activity. During the first week postweaning, pigs fed SBM had significantly greater average daily gain (ADG), average daily feed intake (ADFI) and better feed conversion ratio (FCR) compared to pigs fed FFS diets. Linear negative effect on growth rate was found as the inclusion rate of FFS increased. During d 8 - 21 postweaning, piglets fed EFS 50 diet showed the best ADG and FCR despite no significant difference between treatment SBM and EFS 25 have been observed. Overall, piglets fed diets up to 50% FFS inclusion rate exhibited similar weight gain. Only piglets fed EFS 100 diet showed a significantly decreased growth rate. No other significant effect was found in feed intake and feed conversion ratio. At d 7, dry matter digestibility was higher in pigs fed SBM diet than piglets fed EFS 75 diet (p<0.05) and crude protein digestibility was higher in piglets fed SBM diet than piglets fed EFS 50, EFS 75 and EFS 100 (p<0.05). At d 21, no difference other than in phosphorus digestibility was detected. This indicates that piglets at 21 d postweaning are capable of utilizing nutrients from FFS. No treatment effects were detected in blood metabolites. The data suggests that piglets at 16 d of age are not sufficiently mature to use extruded FFS in their diets. Nevertheless, FFS seemed to be able to replace upto 50% of SBM in weaned piglet diet.

Development of a Biofungicide Using a Mycoparasitic Fungus Simplicillium lamellicola BCP and Its Control Efficacy against Gray Mold Diseases of Tomato and Ginseng

  • To develop a commercial product using the mycoparasitic fungus Simplicillium lamellicola BCP, the scale-up of conidia production from a 5-l jar to a 5,000-l pilot bioreactor, optimization of the freeze-drying of the fermentation broth, and preparation of a wettable powder-type formulation were performed. Then, its disease control efficacy was evaluated against gray mold diseases of tomato and ginseng plants in field conditions. The final conidial yields of S. lamellicola BCP were $3.3{\times}10^9conidia/ml$ for a 5-l jar, $3.5{\times}10^9conidia/ml$ for a 500-l pilot vessel, and $3.1{\times}10^9conidia/ml$ for a 5,000-l pilot bioreactor. The conidial yield in the 5,000-l pilot bioreactor was comparable to that in the 5-l jar and 500-l pilot vessel. On the other hand, the highest conidial viability of 86% was obtained by the freeze-drying method using an additive combination of lactose, trehalose, soybean meal, and glycerin. Using the freeze-dried sample, a wettable powder-type formulation (active ingredient 10%; BCP-WP10) was prepared. A conidial viability of more than 50% was maintained in BCP-WP10 until 22 weeks for storage at $40^{\circ}C$. BCP-WP10 effectively suppressed the development of gray mold disease on tomato with control efficacies of 64.7% and 82.6% at 500- and 250-fold dilutions, respectively. It also reduced the incidence of gray mold on ginseng by 65.6% and 81.3% at 500- and 250-fold dilutions, respectively. The results indicated that the new microbial fungicide BCP-WP10 can be used widely to control gray mold diseases of various crops including tomato and ginseng.

가정간편식 용기용 바이오매스 기반 발포구조체의 특성에 관한 연구 (Characterization of Biomass-Based Foam Structures for Home-Meal-Replacement Containers)

  • 바이오매스가 포함된 발포구조체의 발포온도에 따른 SEM, 밀도, 공극률, 수분투과도 측정을 통해 특성 변화를 분석하였으며, 소비자가 해당 용기를 사용할 때의 열적 안정성을 확인하기 위해 MIL-STD규격에 따라 열충격 처리의 영향을 분석하였다. 측정 결과 발포온도가 증가할수록 대체로 기공의 크기는 증가하고 기공의 수는 감소하여 밀도는 통계적으로 유의한 차이가 없음을 확인하였다. 또한 공극률과 수분투과도 측정결과는 서로 다른 경향성을 보이고 있는데 이는 기공의 크기와 수, 기공 간의 구도가 변화하였기 때문인 것으로 사료되며, 이에 대한 추가적인 연구가 필요하다. 열충격 시험 결과, 열충격 반복횟수를 거듭할수록 기공의 크기가 감소하면서 밀도는 증가하고 공극률과 수분투과도는 감소하는 것을 확인할 수 있었다. 이는 시료에 고온을 가함으로써 시료가 연화되었고, 연화된 상태의 시료에 곧바로 저온을 가하여 시료가 수축되면서 기공의 크기와 구도가 변화하였기 때문인 것으로 판단된다. 하지만 밀도의 경우 통계적으로 유의한 차이가 없었으며 수분투과도의 경우 수치가 감소하는 경향을 확인할 수 있었으므로 해당 발포구조체로 제작된 HMR 용기는 열적 안정성이 있다고 판단할 수 있다.

초임계 CO2 유체 추출법을 이용한 탈지 유채박 중 표면활성물질 추출의 최적화 (Extractions of Surface-Active Substances from Defatted Rapeseed Meal (Brassica napus L.) by Supercritical Carbon Dioxide)

  • 본 실험에서는 탈지 유채박 중 표면활성물질의 추출을 위해 초임계 $CO_2$ 유체 추출법을 이용하였다. 추출 독립변수는 추출압력(150~350 bar), 온도($33{\sim}65^{\circ}C$), 보조용매량(ethanol, 150~250 g)으로 하였으며, D-optimal design에 의한 반응표면 분석을 통해 추출수율, 중성지질 분획, 인지질 분획, 당지질 분획 함량 등의 종속변수에 대한 최적 추출조건을 검토하였다. 그 결과, 압력, 온도, 그리고 보조용매량이 증가함에 따라 종속변수 즉, 추출수율, 인지질, 당지질 함량 등은 증가하였으나, 중성지질 함량은 감소하는 것으로 나타났으며, 그중 보조용매량이 각 종속변수에 가장 큰 영향을 주는 것으로 나타났다. 본 연구를 통해서 유도된 회귀식 모형은 실험을 통해 얻은 결과와 잘 일치하였으며, 초임계 $CO_2$ 유체 추출에 있어서 종속변수인 추출수율, 인지질, 당지질 함량 등을 최대로 하면서 중성지질 함량을 최소로 하는 최적화된 추출조건은 추출압력 350 bar, 추출온도 $65^{\circ}C$, 보조용매량 228.55 g으로 분석되었으며, 이 조건에서 추출수율은 5.98%, 추출물은 중성지질 3.0%, 인지질 69.43%, 당지질 17.46% 등의 조성을 나타낼 것으로 예측되었다.

서울 . 경기 일부지역의 전체급식소에 대한 실태조사 (A Survey on the Actual Conditions of Group Feeding in Seoul City and Kyongki Province.)

  • Recently this writer has conducted a comprehensive survey of 38 private businesses where group feeding is practiced, with a view to determining the condition of nutrition intake by the blue collar workers of these businesses firms as well as gaining an indication of their facilities management along with their sanitary precautions. The survey has revealed, among others, a number of interesting data such as shown below. 1. There are a total of 1,054 workers being fed in groups daily at each of the above business companies, with only one nutrition specialist regardless of the number of workers to be fed or the number of times they are fed daily. The average number of cooks comes to 2.7 persons per 100 workers, while 42.2% of the companies provide four meals a day including mid-night supper, with the result that the workers are fed 3.13 times a day on an average. 2. The average feeding cost amounts to 151 won per meal per person, of which 86 won is payed by the workers themselves and 115 won by the companies involved. 3. The average fbod intake has been found to be 490 grams which can be further broken down into 44grams of animal food and 446grams of vegetable food. These figures are higher than those revealed by the 1975 investigation of the nation's average nutrition intake. 4. The survey also shows the daily calorie consumption per person to be as high as 896 calories including 37 grams of protein which is further broken down into 11 grams of animal protein and 26grams of vegetable protein. With respect to other types of nutrients excepting Vitamin-A, the companies concerned are believed to prorids more than what the survey of the nation's nutrition has revealed or the amount recommended by Korea FAO assoc!ation as desirable. 5. As for facilities management, there is still much room for future improvement. Some 97.4% of the companies surveyed are not equipped with the steam table, which indicates lack of concern along this line as well as a very how degree of equipment utilizations. Distribution of the food in the dining room is generally depedent on self-service method. 6. Speaking of sanitary precautions taken by those businesses involved, some 76.3% are found to have their compounds haunted by rats and other insects. Some 73.7% of them are dependent on boiling method for disinfection of kitchen utensils, which is conducted by 50% of those industries only two or three times a week. It seems hardly feasible to conduct disinfection everyday.

참깨의 볶음조건이 참깨박 정유성분의 휘발성 성분 패턴에 미치는 영향 (The Effects of Sesame Seed Roasting Conditions on Volatile Component Patterns of Essential Oils Obtained from Sesame Meals)

  • 참깨로부터 참기름을 채유하고 부산물로 얻어진 참깨박을 이용하여 천연 향료의 원료를 얻기 위하여 참깨의 전처리 조건을 달리하여 각각 2종의 참기름과 참깨박을 얻었다. 참깨의 볶음온도를 200$^{\circ}C$, 220$^{\circ}C$로 차등화하여 얻어진 참기름 A, B는 각각 1,397.1 ppm, 1,518.8 ppm이었고, 참깨박 A, B는 각각 663.2 ppm, 775.3 ppm 및 참기름향은 16,507.9 ppm으로 차이를 보였다. 2종의 참기름에서 공통적으로 주요 휘발성 성분은 pentane, 2-butanone, pyrazine, methyl pyrazine, 2,5- 및 2,6-dimethyl pyrazine, furfuryl alcohol, guaiacol, 4-vinylguaiacol 이었다. 총 26종의 성분 중 pyrazine류는 각각 834.4 ppm, 816.4 ppm으로 전체의 59.72%, 53.75%를 차지하여 이 성분이 참기름의 향기를 결정해 주는 가장 큰 영향인자로 판단된다. 2종의 참깨박 A, B시료에서 주요 휘발성 성분은 2-butanone, hexanal, pyrazine, methyl pyrazine, 2,6-dimethyl pyrazine, furfuryl alcohol로 참기름과는 차이를 보였다. 상대적으로 참기름 향료에서는 총 16,507.9 ppm의 휘발성 성분이 검출되었고, 주요 성분은 2-butanone, guaiacol, 4-vinylguaiacol이 각각 1,000 ppm 이상으로 전체의 35.02%를 차지하였다.

뇨중 대사체 정량에 의한 capsaicinoid의 생화학적 섭취지표 개발 연구 (Capsaicin Intake Estimated by Urinary Metabolites as Biomarkers)

  • 최근 고추섭취량과 위암과의 양의상관도를 보이는 역학조사 결과가 발표되었는데 이런 자료를 신뢰하기 위해서는 향신료로 쓰이는 고추의 섭취량이 정확히 측정되어야 할 것이다. 이에 고추의 매운맛 성분 capsaicin의 인체대사산물을 확인하고 이를 응용하여 생리학적으로 의미있는 섭취량을 측정하려는 취지에서 실험을 계획하였다. 먼저 동물실험에서 관찰된 capsaicin의 대사체들을 소변에서 HPLC로 동시에 검출하는 방법을 고안하고 이들 대사체의 신속한 검색을 위해, 이전부터 사용하던 유기용매 추출을 지양하고 효소 glucuronidase 처리한 소변희석액을 바로 HPLC에 주입하는 방법을 사용하였다. 20대의 젊은 여성 5-6명을 대상으로, 이틀 간의 capsaicin 고갈식이 이후 capsaicin이 포함되지 않은 식사시 capsacin의 제공원으로서 고추장을 섭취하게 하였다. 60g씩 사홀 고추장을 섭취한 실험시 여섯명의 참가자 중 네명에게서 섭취를 시작한지 3일 째 4-hydroxy-3-methoxybenzoic acid가, 한 명에게서 3일 째 capsaicin이 검출되었다. 80g씩 나흘을 섭취한 2차 실험시는 세 명의 피실험자에게서 4-hydroxy-3-methoxybenzoic acid가 나흘째 소변에서 검출되었다. 2차 실험에서 나흘째 검출된 4-hydroxy-3-methoxybenzoic acid의 총량은 실험기간 중 섭취한 capsaicinoids 총량의 85% 이상이었다. 따라서 쥐 등을 이용한 capsaicinoid 대사 실험에서는 4-hydroxy-3-methoxybenyl alcohol이 주대사물질이었으나 인체에서는 그와 달리 4-hydroxy-3-methoxybenzoic acid가 주대사산물이라고 여겨진다. 그러나 1, 2차 실험식이를 충실히 따른 두명의 소변에서는 4-hydroxy-3-methoxybenzoic acid를 포함한 어떤 대사체도 찾을 수 없었기 때문에 개인간의 흡수 대사 차이가 크게 기여한다고 추정되며 생리적으로 의미 있는 capsaicin 섭취의 지표 물질은 인체를 대상으로 처음 얻어진 이 결과에 기반으로 한 계속 실험을 통해 선정되리라 기대한다.

일부 대학생의 흡연 실태와 구강건강에 관한 인식 및 행태 조사연구 (Research on the Smoking Conditions of College Students and the Behavior and Awareness of Oral Hygiene)

  • 본 연구는 일부 대학생들의 흡연행태와 흡연자와 비흡연자 간의 흡연에 대한 인식과 구강건강관리행태를 알아보고자 경상북도, 대구광역시, 강원도에 위치한 4년제 대학교에 재학 중인 대학생 260명을 대상으로 흡연에 대한 인식과 구강건강관리행태에 대해 조사한 결과 다음과 같은 결론을 얻었다. 1. 흡연자의 하루 평균 흡연량은 반 갑 미만이, 흡연기간은 1년~3년 미만이 가장 높았으며, 흡연을 시작하게 된 동기는 호기심과 주변 권유가 가장 높았고, 흡연이유는 스트레스 해소, 습관적 순으로 나타났다. 2. 흡연유무에 따른 구강건강관리행태 중 1회 잇솔질 시간, 정기적 치과 내원, 최근 치과 내원시기, 치과 방문 목적, 스켈링 및 구강보건교육 경험에서 유의한 차이가 있었다. 3. 흡연유무에 따른 구강건강에 대한 인식 중 주관적 구강건강상태에서 유의한 차이가 있었다. 4. 흡연유무에 따른 흡연에 대한 인식 중 담배, 한 두 개피 정도의 담배, 식후의 담배 및 간접흡연이 건강에 미치는 영향에 대한 인식에서 유의한 차이가 있었다. 이상의 결과로 미루어 볼 때 흡연자는 비흡연자에 비해 구강건강관리행태 및 구강건강에 대한 인식, 흡연에 대한 인식이 부족하므로, 대학생들을 위한 금연교육과 구강보건교육 프로그램 개발의 활성화가 필요하다고 사료된다.
