Analysis of Genetic Relation among Collected Landraces of Agrimonsa pilosa L. Using RAPD (RAPD를 이용한 짚신나물(Agrimonia pilosa Ledeb.) 수집종 유연관계 분석)

  • Agromonia pilosa Ledeb. has been used as a medicinal plant in traditional folk remedy. There are few reports on classification, physiology, ecology and morphological studies of Agromonia pilosa L. in Korea. Therefore, advanced approaches on study and development with this plant would be done urgently. Present stndy was carried out to gain basic information on genetic resources and variation with collected domestic landraces through RAPD analysis in Agromonia pilosa L. Forty two collections of Agromonia pilosa L. from nation-wide area including USA one were analyzed by RAPD test. Molecular marker size by amplified DNA band pattern ranged from 300 to 2,100bp. Among the collection, two landraces of Hadong and Cheonghak-dong showed close relation in genetic similarity. Minimum and maximum value by matrix of 1-F among 26 collected landraces were figured out as 0.365 and 0.827 showing mean value for 0.624, respectively. Those landraces were classified into two groups with cluster analysis by Nei and Li's formula from RAPD-analyzed values, and considerable genetic differences were recognized between two groups.

  • Salurnis marginella Guérin-Méneville, 1829 (Hemiptera: Flatidae) is an invasive species first reported in 2003 in Iksan, which is located in the mid-western region of South Korea, and subsequently found in the nearby regions in 2005. However, molecular-perspective reports on their invasive characteristics are not yet available. In this study, population genetic characteristics of Korean S. marginella were evaluated using the mitochondrial COI region and sequencing 124 individual samples collected in 11 Korean localities. A total of 12 haplotypes were identified with a maximum sequence divergence of 1.368% (9 bp). Haplotype diversity was relatively higher than that of other insect species invaded into Korea, providing 2-6 haplotypes per populations, indicating that introduction to Korea may have happened rather extensively and consistently. Nucleotide diversity (π) was the highest in Iksan but owing to the limited sample size (three individuals) from this locality, additional studies are required for drawing conclusive inference regarding the place of entry to Korea. Ulsan, the easternmost population in the present study, revealed nearly the lowest diversity estimates, such as the lowest H and the second-lowest π; a unique haplogroup with a higher frequency; and an independent genetic cluster, suggesting that the introduction of S. marginella to Ulsan was an independent event. Further collection in Korea and neighboring countries, including the original distributional range is necessary to elucidate the invasive dynamics of S. marginella

Variation in Content of Marker Compounds and Quality of Processed White Ginseng of Different Cultivation Ages from Different Regions (지역별, 연근별 가공백삼의 품질과 지표 성분의 변이)

  • Background: The ginsenosides Rb1 (G-Rb1) and Rg1 (G-Rg1) are used as marker compounds, and are the principal bioactive compounds assessed in the quality control of white ginseng. This study was conducted to analyze white ginseng samples of different and to obtain useful data for the quality control of white ginseng. Methods and Results: The variation in the content of G-Rb1 and G-Rg1 was evaluated among 35 samples of 4-, 5-, and 6-year-old white ginseng. The content of both G-Rb1 and G-Rg1 did not significantly differ among ages, and the relative ratio of the maximum to the minimum content of these within ginseng of the same ages was more than two. However, the ratio of G-Rb1 to G-Rg1 content in the 5- and 6-year-old ginseng was significantly higher than that in the 4-year-old one. According to the 'Ginseng industrial act', the standard (w/w, %) minimum $G-Rg_1$ and $G-Rb_1$ content is 0.10% and 0.20% or more, respectively. Among the 35 samples examined, the content of $G-Rg_1$ was found to be 0.124 - 0.399% with none being less than the standard level, while that of $G-Rb_1$, was 0.147 - 0.595%, with 4 samples (11.4%) failing to meet the standard levels. The content of $G-Rg_1$ and $G-Rb_1$ did not show a constant relationship with the size of ginseng. Conclusions: In our study, the content of both G-Rg1 and G-Rb1 varied widely, and there was no significant difference among cultivation ages. The results of the present study might provide useful information for the quality control of raw ginseng and processed white ginseng using marker compound.

Genetic Diversity of Rehmannia glutinosa Genotypes Assessed by Molecular Markers (분자표지자에 의한 지황 유전집단의 유전적 다양성)

  • Random amplified polymorphic DNA (RAPD) markers were used to identify the genetic diversities among and within varieties and landraces of Rehmannia glutinosa. Polymorphic and reproducible bands were produced by 10 primers out of total 20 primers used in the experiment. In RAPD analysis of the 11 genotypes, 64 fragments out of 73 amplified genomic DNA fragments were polymorphic which represented an average 6.4 polymorphic fragments per primer. Number of amplified fragments with random primers ranged from 2 (OPA-1) to 13 (OPA-11) and varied in size from 200 bp to 1,400 bp. Especially, OPA-10, OPA-11 and OPA-19 primers showed specific bands for varieties of Korea Jiwhang and Jiwhang il ho, which could be useful for discriminating from other varieties and landraces of R. glutinosa. Percentage polymorphism ranged from a minimum of 50% (OPA-1) to a maximum of 100% (OPA-11), with an average of 87.7%. Similarity coefficients were higher in the genotypes of Korea Jiwhang and Jiwhang il ho than in other populations. In cluster analysis, genotypes of Korea Jiwhang, Jiwhang il ho, and Japanese accession were separated from those of other varieties and landraces. Average of genetic diversity within the population $(H_S)$ was 0.110, while average of total genetic diversity $(H_T)$ was 0.229. Across all RAPD makers the $G_{ST}$ value was 0.517, indicating that about 52% of the total genetic variation could be explained by RAPDs differences while the remaining 48% might be attributable to differences among samples. Consequently, RAPD analysis was useful method to discriminate different populations such as domestic varieties and other landraces. The results of the present study will be used to understand the population and evolutionary genetics of R. gllutinosa.

The Spatial Patterns of Organic Matter Content and Macrobenthos during Summer in the Muan Bay Intertidal Zone, Korea (하계 무안만 조간대에서의 유기물 함량 및 대형저서동물의 공간 분포특성)

  • The study was performed in order to understand the association between organic matter content in the sediments and the distribution of macrobenthos in the intertidal zone of the Muan bay. The sediment samples obtained from 21 sampling sites in August 2019 were analyzed for sediment composition and organic matter content (Ignition loss; IL and Total organic carbon; TOC). Further the macro benthos was sorted and enumerated. The sediments of inner area of bay has coarser composition where mouth part of bay, the sediments were predominantly silty. The highest organic matter content (both of IL and TOC) was measured at station 10 located near the Mokpo area while the lowest values was measured at station 20 adjacent to the north side of the bay. The 4 most abundant species accounted for more than 10% of all specimens were Assiminea sp., Musculista senhousia, Cerithideopsilla cingulate and Heteromastus filiformis. The maximum number of species and density were observed at station 10. Cluster analysis and non-metric multidimensional scaling (MDS) allow identification of four benthic assemblages based on species abundance. The correlation analysis revealed that there was a significant difference (p<0.05) in the density with TOC. Based on the analysis; it was found that the distribution of macrobenthos varied with the differences in sediment composition and organic matter content.

Variation of leaf morphology among 18 populations of Zelkova serrata Mak. (느티나무 18개 집단의 엽 형질 변이)

  • This study was conducted to examine genetic variation on leaf characteristics of Zelkova serrata populations. Leaf samples were obtained from eighteen populations and fourteen leaf characteristics such as leaf blade length, leaf width, leaf area and etc. were measured. In all leaf characteristics, there were significant differences among populations and among individuals within population. Most variance of leaf characteristics was contributed to among individuals within population except for length from leaf base to maximum width (x3) and the ratio of x3 to leaf blade length (x10). The relatively high variations of leaf characteristics were found at Gurye, Jungeup and Gyungju populations while Youngwol, Cheongsong, Youngchun populations showed the lower variation. There was high correlation among leaf characteristics related to leaf size, but not among the variables of ratio between leaf characteristics. Length from leaf base to maximum width(x3) and the ratio of x3 to x10 showed significant positive correlation with latitude and altitude of populations, which reflect the differences of mean annual temperature among populations. Mean annual rainfall of populations showed negative correlation with leaf blade length, leaf width, length from apex to first serration and leaf area. Four principal components (PC) were deduced from principal component analysis, which explain the 88.5% of total variance of leaf characteristics. Leaf area, length from leaf base to maximum width, serration number and petiole length showed the highest contribution to PC1, PC2, PC3, PC4, respectively. According to cluster analysis, the populations of Z. serrata were divided into two groups, which reflect the difference of mean annual temperature between groups. Within group, however, specific tendency of clustering was not observed among populattions.

Assessment of the Impacts of 'Sea Prince' Oil Spill on the Rocky Intertidal Macrobenthos Community (암반조간대 대형저서동물군집에 대한 씨프린스호 유류 유출사고 영향 평가)

  • This study was aimed to classify the intertidal macrozoobenthic community status after 2 years of Sea Prince oil spill, and oil spill effects along oil spreading track from heavily impacted to unaffected reference site. Field sampling was initiated in late February and continued through November 1998 seasonally, after 2.5 years of oil spill. 7 rocky sampling sites were selected among coastal regions coated and/or affected by the Sea Prince spilled oil. Identified species was 158 species, 65 family, 24 order, 9 class, 5 phylum. Mollusca was the dominant faunal group comprising 100 species (63.3%), and followed by 38 species of Crustacea (24.1%), 12 species of Echinodermata (7.6%), 5 species of Porifera (3.2%), and 3 species of Cnidaria (1.9%). On Dugpo of Sori Island, the fewest species was collected from 28 species to 35 species seasonally among sampling stations. But far away Dugpo toward Gamak Bay, the number of species increased, collecting the maximum on Sohwoenggan Island. At the wreck site of Sori Island, especially the species number of attached animals such as poriferans and anthozoans was very low compared to another site. The density and biomass on the higher tidal zone increased toward the low affected sites, but biomass on middle tidal zone decreased. The invertebrate biomass of study area was dependent on the sessile animals. The major dominant species were small-sized barnacles, Chthmallus challengeri, periwinkles, Littorina brevicula, mussels, Septifer virgatus, and so on. The biomass of C. challengeri and L. brevicula on the higher tidal zone was highest in the wreck site of Sori Island and decreased further and further. However, mussels on the middle tidal zone showed the inverse trends because of the larger individual size of mussel inhabited in Sori Island than those of another sites. As a result of community analysis, the effect of oil spill was not found distinctly. Several ecological indices and cluster analysis did not show the meaningful variation with oil track despite of the conspicuous differences among tidal heights. These indicate that the macrozoobenthic community level of oil spreaded zone recovered in some degree after the Sea Prince oil spill accident, but population or individual levels of dominant sessile animals took more recovery times.

9 Provinces and 5 Secondary Capitals, Myeong-ju(Haseo-ju) - Revolve Around Urban Structure - (구주오소경과 명주(하서주) - 그 도시구조를 중심으로 -)

  • After withdrawal of military troops of Chinese Tang dynasty in the 18th year of King Moon-moo's reign(678), the Silla Kingdom had actually unified the Korean peninsula and had divided the territory into 9 states benchmarking the China's local administrations adjustment system. He had established local administrative units by deploying secondary capitals, counties and prefectures in the nine states. The so-called "9 Provinces and 5 Secondary capitals" are what constitutes the local administrations system. The provinces can be compared to current provinces of the Republic of Korea(hereinafter Korea), and secondary capitals to megalopolises. According to a chapter of the Samkuksaki(三?史記) which had recorded the achievements of king Kyoungdeok in December in his 16th year on the throne(757), the local administrative units had amounted to 5 secondary capitals, 117 counties and 293 prefectures. There are still lots of ambiguous points since there have never been any consultation on locations of provinces and secondary capitals' castles, and on structures of cities because the researches for local cities inside the 9 Provinces and 5 Secondary capitals in the Unified Silla Kingdom has been conducted centering on the historic literatures only. The research for restoring structures of cities seen from an archeological perspective are limited to the studies of Taewoo Park("A study on the local cities in the Unified Kingdom Age" 1987) and that of the author("A study on the restoration of planned cities for the Unified Silla Kingdom in terms of the structures and realities of the castles in the 9 Provinces and 5 Secondary capitals" 2009). The Gangneung city of Gangwon province was originally called Haseoryang(河西良) of the Gogureo Kingdom as an ancient nation of Ye(濊). According to "Samkuksaki", it had evolved from Haseoju(河西州) to a secondary capitals in the 8th year of King Seonduk(639). Afterwards, it had been renamed as Myeongju(溟洲) in the 16th year of King Kyoungduk(757), and then several other names were given to it after Goryo dynasty. Taewoo Park claims that it is being defined as a sanctuary remaining in Myoungjudong because of the vestige of bare castle, and this cannot be ascertained due to the on-going urbanization processes. Also, the Kwandong university authority is suggesting an opinion of regarding Myeongju mountain castle located 3 Kms southwest of the center of Gangwon city as commanding post for the pertinent state. The author has restored the pertinent area into a city composed of villages within a lattice framework like Silla Keumkyoung and many other cities. The structure is depicted next. The downtown of Gangneung is situated on a flat terrain at the west bank of Namdaecheon stream flowing southwest to northeast along the inner area of the city. Though there isn't any hill comparatively higher than others in the vicinity, hills are continuously linked east to west along the northern area of the downtown, and the maximum width of flat terrain is about 1 Km and is not so large. Currently, urbanization is being proceeded into the inner portion of Gangneung city, the lands in all directions from the hub of Gangneung station have been readjusted, and thus previous land-zoning program is almost nullified. However, referring to the topographic chart drawn at the time of Japanese colonial rule, it can be validated that land-zoning program to accord the lattice framework with the length of its one side equaling to 190m leaves its vestige about 0.8Km northwest to southeast and about 1.7Km northeast to southwest of the vicinity of Okcheondong, Imdangdong, Geumhakdong, Myeongjudong, and etcetera which comprize the hub of the downtown. The land-zoning vestige within the lattice framework, compared to other cases related with the '9 states and 5 secondary capitals', is very much likely to be that of the Unified Silla Kingdom. That the length of a side of a lattice framework is 190m as opposed to that of Silla Geumkyoung and other cities with their 140m or 160m long sides is a single survey item in the future. The baseline direction for zoning the lands is tilting approximately 37.5 degrees west of northwest to southeast axis in accordance with the topographic features. It seems that this phenomenon takes place because of the direction of Namdaecheon and the geographic constraints of the hills in the north. Reviewing minimally, a rectangular size of zoned land by 4 Pangs(坊) on the northwest to southeast side multiplied by 7 Pangs(坊) on the northeast to southwest side had been restored within a lattice framework. Otherwise, considering the extent of expansion of the existing zoned lands in the lattice framework and one more Pang(坊) being added to each side, it is likely that the size could have been with 5 Pangs(坊) on the northwest to southeast side multiplied by 8 Pangs(坊) on the northeast to southwest side(950 M on the northwest to southeast side multiplied by 1,520m on the northeast to southwest side). The overall shape is rectangle, but land-zoning programs reminiscent of rebuilt roads(red phoenix road) like Jang-an castle(長安城) of Chinese Tang dynasty or Pyoungseong castle(平城城) in Japan is not to be validated. There are some historic items among the roof tiles and earthen wares excavated at local administrative office sites or Gangneung's town castle in Joseon dynasty inside the area assumed to be containing municipal vestiges even though archeological survey for the vestige of Myeongju has not been made yet, and these items deserve dating back to the Unified Silla Kingdom age. Also, all of the construction sites at local administrative authorities of the Joseon dynasty are showing large degrees of slant in the azimuth. This is a circumstantial evidence indicating the fact that the inherited land-zoning programs to be seen in Gangneung in terms of the lattice framework had ever existed in the past. Also, the author does not decline that Myeongju mountain castle had once been the commanding post when reviewing the roof tiles at the edge of eaves in this stronghold. The ancient municipal castles in the Korean peninsula are composed of castles on the flat terrain as well as hilly areas and the cluster of strongholds like Myounghwal, Namhan, Seohyoung mountain castles built around municipal castle of Geumkyoung based on a lattice framework program. Considering that mountain castles are spread in the vicinity of municipal vestiges in other cities other than the 9 states and 5 secondary capitals, it is estimated that Myeongju was assuming the function of commanding post incorporating cities on the flat terrain and castles on the hills.
