Electrophysiological study on the presence of the electrogenic Na pump of the mouse unfertilized eggs (Mouse 미수정란에서의 electrogenic Na pump 활동여부에 관한 연구)

  • In order to manifest the presence of Na-K pump and its property on the unfertilized egg membranes of mouse, membrane potential was recorded under the physiological condition (at $37^{\circ}C$ and 4mM $Ca^{2+}$). After an induction of superovulation, the fresh eggs with zona pellucida were collected from mouse oviduct. Transient hyperpolarization as pump action was recorded after the switch into the high potassium perfusate (15mM $K^+$) from K-free perfusate, and the difference between membrane potential observed just before the perfusion of high potassium solution and the maximal membrane potenlial during the perfusion of high potassium solution was regard as pump activities. The results observed were as follows, 1. Resting mombrane potential was depolarized under the treatment of $10^{-5}M$ ouabain. 2. Pump activities of the unfertilized mouse eggs were $-3.38{\pm}0.61mV$ ($Mean{\pm}SD$, n=6), recorded as transient hyperpolarization due to the electrogenic property. 3. Pump activities were blocked by both treatment of $10^{-5}M$ ouabain and perfusion of Nafree solution, while increased by high $Na^+$ (300mM) perfusion ($-7.45{\pm}0.75mV$, n =2). 4. Hyperpolarization due to pump activity was not altered by $Mn^{2+}$. 5. Above results confirm the presence of ouabain-sensitive Na-K pump, which affected the membrane potential directly, on the unfertilized egg membranes of mouse.

Optimization of Mutual Information for Multiresolution Image Registration (다해상도 영상정합을 위한 상호정보 최적화)

  • We propose an optimization of mutual information for multiresolution image registration to represent useful information as integrated form obtaining from complementary information of multi modality images. The method applies mutual information as cost function to measure the statistical dependency or information redundancy between the image intensities of corresponding pixels in both images, which is assumed to be maximal if the images are geometrically aligned. As experimental results we validate visual inspection for accuracy, changning initial condition and addictive noise for robustness. Since our method uses the native image rather than prior feature extraction, few user interaction is required to perform the registration. In addition it leads to robust density estimation and convergence as applying non-parametric density estimation and stochastic multiresolution optimization.

  • Alkaline bacterium producing a high pro-tease activity at low temperature was isolated by using enrichment culture from soil samples and identified as Bacil-lus sp. WRD-1 Cell growth was maximal at 10 hours and the optimal initial pH and culture time of culture condition for enzyme production was pH 7 and 10 hours, respectively. Temperature range of high enzyme activity were $10~40^{\circ}C$. The optimal pH and temperature for the enzyme activity were pH9 and $30^{\circ}C$.

Immobilization of α-amylase from Exiguobacterium sp. DAU5 on Chitosan and Chitosan-carbon Bead: Its Properties

  • Glutaraldehyde was used as a cross-linking agent for immobilization of purified ${\alpha}$-amylase from Exiguobacterium sp. DAU5. Befitting concentration of glutaradehyde and cross-linking time is the key to preparation of cross-linking chitosan beads. Based on optimized immobilization condition for ${\alpha}$-amylase, an overall yield of 56% with specific activity of 2,240 U/g on chitosan beads and 58% with specific activity of 2,320 U/g on chitosan-carbon beads was obtained. The optimal temperature and pH of each immobilized enzyme activity were $50^{\circ}C$ and 50 mM glycine-NaOH buffer pH 8.5, respectively. Those retained more than 75 and 90% of its maximal enzyme activity at pH 7.0-9.5 and after incubation at $50^{\circ}C$ for 1 h, respectively. In addition, the immobilization product showed higher organic-solvent tolerance than free enzymes. The mode of hydrolyzing soluble starch revealed that the ${\alpha}$-amylase possessed high hydrolyzing activity. These results indicate that chitosan is good support and has broad application prospects of enzyme immobilization.

Hydrolysis of Inulin by Endo- and Exo-Inulinase (Endo- 및 Exo-Inulinase를 이용한 Inulin 가수분해)

  • Inulin degradation was examined using patially puriiied enzyme mixtures of the Exo-inulinase from a Bacillus spp. and the Endo-inulinase from a Pseudomonas spp.. The highest synergistic xtion of the two cnzymcs was observcd when the Exo- and the Endo-inulinase werc mixed at the ratio of 1 to 13, and the rate of hydrolysis of the above process was enhanced approximately 1.6 times I1ight.1- than that of the reaction catalysed with a single enzyme of the same units. The enzymc mixture showed the maximal activity at pH 6.0 and $55^{\circ}C$, and in the prescncc of 0.5 mM each of $CO^{2+}$ and $Mn^{2+}$. Under the optimal condition described above fructosu was accumulated with the overitll concentration of 84% after 36 hours of the reiiction.

Screenin of Phospholipase D Producing Actinomycetes (방선균으로부터 Phospholipase D 생산균주의 탐색)

  • In order to screen microorganisms producing phospholipase D (PLD) [EC 3.1.4.4], culture broths of about 900 strains of soil bacteria were subjected to examine for the PLD activity. When the hydrolytic activity of PLD (H-activity) in the supernatant was determined, 64 strains produced PLD more than 0.3 unit/ml and all of them were actinomycetes. Among 26 culture broths tested, 6 ones had transphosphatidylation activity (T-activity) of 30~68%. When the strains except one were cultivated on 3 different media at 30$\circ$C for 3 days under aerobic condition, strain # 1090 on medium B (yeast extract 1%, peptone 1%, glucose 1.5%, glycerol 1%, CaCO$_{3}$ 0.4%, and pH 7.2) produced PLD with much higher H- and T-activity, which were 8.3 units/ml and 76.3%, respectively. Subsequently, time course of PLD production of the strain # 1090 during cultivation with aeration of 1 v/v/m and agitation of 400 rpm at 30$\circ$C for 5 days on medium B in jar fermentor was investigated. H-activty of PLD reached almost maximum (about 9 units/ml) after 32 hours and maximal T-activity was found to be about 80%.

Characteristics of Microalgal Growth on Anaerobic Effluent of Animal Waste (축산분뇨 혐기성 처리수에서 미세조류의 성장특성)

  • Characteristics of microalgal growth was investigated using anaerobic effluent from two-phase animal waste digestor as substrate. Batch experiments were carried out to investigate the effect of the initial nitrogen and phosphorus concentrations on growth of Microcystis aeruginosa, Chlorella sp. and Euglena gracilis. In 400 times diluted anaerobic effluent (TN 3 mg/L), single cell growth of the Euglena gracilis population increased twice without delay, although Chlorella sp. and Microcystis aerugenos take over 144 hours. Similar appearance with single cell growth was observed in mixed cultures. However, microalgae population did not increase under condition of 10 times diluted influent (TP 3 mg/L) in both pure and mixed cultures, which was affected by high organic and nitrogen concentration. Logistic growth model successfully fitted to determine biokinetic parameters such as ${\lambda}$: lag time, ${\mu}m$: maximal specific growth rate, A: asymptote of growth.

  • Heteropolysaccharide-7 (PS-7) was produced by Beijerinckia indica HS-2001 under aerobic condition. Production of PS-7 was investigated under various ratios of glucose as carbon source to ammonium nitrate as nitrogen source. Maximal production of PS-7 was 7.13 g/l when concentrations of glucose and ammonium nitrate were 10 g/l and 0.3 g/l, respectively. But its conversion rate from glucose was as low as 7 %. The highest conversion rate of PS-7 was 46% when those of glucose and ammonium nitrate were 1.0 g/l and 0.3 g/l, respectively.

Camptodactyly: An unsolved area of plastic surgery

  • Background Camptodactyly refers to permanent flexion contracture at the proximal interphalangeal joint. Most cases are limited to fifth-finger involvement. Although common, the treatment of camptodactyly is controversial. Many published studies have emphasized conservative treatment, while others have described surgical procedures. The problem with this deformity is that it presents in several forms, which means that there is no single model for effective treatment. The aim of this paper is to present the difficulties encountered with this condition and the management thereof on an individual basis. Methods This is a case series of 14 patients (nine males, five females) who underwent surgical treatment. The results were classified using the method from Mayo Clinic as excellent, good, fair, and poor. Results Fourteen patients with 15 fingers underwent surgery, and the results achieved were as follows: excellent, 0; good, 1; fair, 6; poor, 8. The treatment of camptodactyly still remains controversial, and hence proper planning individualized to each patient is needed to achieve the maximal improvement with realistic goals. Conclusions Although we performed individualised surgery, our careful follow-up was not able to identify any method as superior over another with respect to gain in extension and loss of flexion. We therefore propose that the extensor mechanism should not be disturbed during surgery to treat camptodactyly cases.

Isolation of a Thermophilic Bacillus sp. Producing the Thermostable Cellulase-free Xylanase,and Properties of the Enzyme (내열성 Cellulase-free Xylanase를 생산하는 고온성 Bacillus sp.의 분리 및 효소 특성)

  • A thermophilic bacterium producing the extracellular cellulase-free xylanase was isolated from soil and has been identified as Bacillus sp. The optimal growth temperature was 50$\circ$C and the optimal pH, 7.0. Under the optimal growth condition, maximal xylanase production was 2.2 units/ml in the flask culture. The enzyme production was induced by xylan and xylose, but was repressed by sucrose or trehalose. The partially purified xylanase was most active at 70$\circ$C. It was found that the enzyme was stable at 65$\circ$C for 10 hours with over 75% of the activity. The enzyme was most active at pH 7.0 and retained 90% of its maximum activity between pH 5.0 and pH 9.0 though Bacillus sp. was not grown on alkaline conditions (>pH 8.0). In addition, the activity of xylanase was over 60% at pH 10.0. At the ambient temperature, the enzyme was stable over a pH range of 5.0 to 9.0 for 10 h, indicating that the enzyme is thermostable and alkalotolerant. The activity of xylanase was completely inhibited by metal ions including Hg$^{2+}$ and Fe$^{2+}$, while EDTA, phenylmethylsulfonyl fluoride (PMSF), $\beta$-mercaptoethanol and SDS didn't affect its activity. The enzyme was also identified to exert no activity on carboxymethylcellulose, laminarin, galactomannan, and soluble starch.
