7,8,4'-Trihydroxyisoflavone, a Metabolized Product of Daidzein, Attenuates 6-Hydroxydopamine-Induced Neurotoxicity in SH-SY5Y Cells

  • Daidzein isolated from soybean (Glycine max) has been widely studied for its antioxidant and anti-inflammatory activities. However, the protective effects of 7,8,4'-trihydroxyisoflavone (THIF), a major metabolite of daidzein, on 6-hydroxydopamine (OHDA)-induced neurotoxicity are not well understood. In the current study, 7,8,4'-THIF significantly inhibited neuronal cell death and lactate dehydrogenase (LDH) release induced by 6-OHDA in SH-SY5Y cells, which were used as an in vitro model of Parkinson's disease (PD). Moreover, pretreatment with 7,8,4'-THIF significantly increased the levels of superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH) and decreased malondialdehyde (MDA) activity in 6-OHDA-induced SH-SY5Y cells. In addition, 7,8,4'-THIF significantly recovered 6-OHDA-induced cleaved caspase-3, cleaved caspase-9, cleaved poly-ADP-ribose polymerase (PARP), increased Bax, and decreased Bcl-2 levels. Additionally, 7,8,4'-THIF significantly restored the expression levels of phosphorylated c-Jun N-terminal kinase (JNK), p38 mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinase 1/2 (ERK 1/2), phosphatidylinositol 3-kinases (PI3K)/Akt, and glycogen synthase kinase-3 beta ($GSK-3{\beta}$) in 6-OHDA-induced SH-SY5Y cells. Further, 7,8,4'-THIF significantly increased the reduced tyrosine hydroxylase (TH) level induced by 6-OHDA in SH-SY5Y cells. Collectively, these results suggest that 7,8,4'-THIF protects against 6-OHDA-induced neuronal cell death in cellular PD models. Also, these effects are mediated partly by inhibiting activation of the MAPK and PI3K/Akt/$GSK-3{\beta}$ pathways.

In vivo wear determination of novel CAD/CAM ceramic crowns by using 3D alignment

  • PURPOSE. To determine wear amount of single molar crowns, made from four different restoratives, and opposing natural teeth through computerized fabrication techniques using 3D image alignment. MATERIALS AND METHODS. A total of 24 single crowns (N = 24 patients, age range: 18 - 50) were made from lithium disilicate (IPS E-max CAD), lithium silicate and zirconia based (Vita Suprinity CAD), resin matrix ceramic material (Cerasmart, GC), and dual matrix (Vita Enamic CAD) blocks. After digital impressions (Cerec 3D Bluecam, DentsplySirona), the crowns were designed and manufactured (Cerec 3, DentsplySirona). A dualcuring resin cement was used for cementation (Variolink Esthetic DC, Ivoclar). Then, measurement and recording of crowns and the opposing enamel surfaces with the intraoral scanner were made as well as at the third and sixth month follow-ups. All measurements were superimposed with a software (David-Laserscanner, V3.10.4). Volume loss due to wear was calculated from baseline to follow-up periods with Siemens Unigraphics NX 10 software. Statistical analysis was accomplished by Repeated Measures for ANOVA (SPSS 21) at = .05 significance level. RESULTS. After 6 months, insignificant differences of the glass matrix and resin matrix materials for restoration/enamel wear were observed (P>.05). While there were no significant differences between the glass matrix groups (P>.05), significant differences between the resin matrix group materials (P<.05) were obtained. Although Cerasmart and Enamic were both resin matrix based, they exhibited different wear characteristics. CONCLUSION. Glass matrix materials showed less wear both on their own and opposing enamel surfaces than resin matrix ceramic materials.

Pragmatic Combinations of Acupuncture Points for Lateral Epicondylalgia are Unreliable in the Physiotherapy Setting

  • This study describes the reliability of pragmatic combinations of acupuncture points for lateral epicondylalgia (LE) as prescribed by physiotherapists who were experts in acupuncture. Raters (n = 14; 33-59 years) independently prescribed acupuncture points for 30 simulated human patients with LE who were surveyed via a printed questionnaire. The frequency and cooccurrence of acupuncture points prescribed for patients with lateral epicondylitis were assessed. Absolute agreement and Light's kappa (${\kappa}_{Light}$) with 95% confidence interval (CI) were used to quantify the interrater agreement. Raters prescribed 103 unique acupuncture points in different combinations with a median (min-max) of 5 (0-11) acupuncture points. The most prescribed acupuncture point was LI-11 (297 of 420; 71%), and the most common cooccurring acupuncture points were LI-11 and LI-4 (160 of 420; 38%). The absolute agreement for prescribing the acupuncture points ranged from 70% (point GB-20) to 0% (points LI-10, SP-6, LI-11, GB-34, LI-12, and LI-4). Point LR-3 showed the highest interrater reliability for prescribing the acupuncture points [${\kappa}_{Light}=0.112$, 95% CI = (0.055-0.194)], whereas point LI-4 showed the lowest reliability [${\kappa}_{Light}=-0.003$, 95% CI = (-0.024 to 0.024)]. These findings suggest that pragmatic prescriptions of acupuncture points for LE are unreliable among physiotherapists who are experts in acupuncture. Explicit, high-level evidence-based rules for prescribing and teaching combinations of acupuncture points for LE are warranted.

Quality grading of Hanwoo (Korean native cattle breed) sub-images using convolutional neural network

  • The aim of this study was to develop a marbling classification and prediction model using small parts of sirloin images based on a deep learning algorithm, namely, a convolutional neural network (CNN). Samples were purchased from a commercial slaughterhouse in Korea, images for each grade were acquired, and the total images (n = 500) were assigned according to their grade number: 1++, 1+, 1, and both 2 & 3. The image acquisition system consists of a DSLR camera with a polarization filter to remove diffusive reflectance and two light sources (55 W). To correct the distorted original images, a radial correction algorithm was implemented. Color images of sirloins of Hanwoo (mixed with feeder cattle, steer, and calf) were divided and sub-images with image sizes of 161 × 161 were made to train the marbling prediction model. In this study, the convolutional neural network (CNN) has four convolution layers and yields prediction results in accordance with marbling grades (1++, 1+, 1, and 2&3). Every single layer uses a rectified linear unit (ReLU) function as an activation function and max-pooling is used for extracting the edge between fat and muscle and reducing the variance of the data. Prediction accuracy was measured using an accuracy and kappa coefficient from a confusion matrix. We summed the prediction of sub-images and determined the total average prediction accuracy. Training accuracy was 100% and the test accuracy was 86%, indicating comparably good performance using the CNN. This study provides classification potential for predicting the marbling grade using color images and a convolutional neural network algorithm.

Fabrication of the Alnico Bonded Magnets for Measuring Instruments and its Magnetic Properties (계측기용 알니코 본드자석의 제조 및 자기 특성연구)

  • Alnico magnets can be used as magnetic bearings for the precise electric power measuring instruments such as watt-hour meters because they have high remanence ($B_r$), relatively high maximum energy product ($(BH)_{max}$), and excellent temperature stability. In this study, Alnico composite magnets were fabricated by appropriately mixing alnico alloy powders with epoxy resin and binder. The Alnico powders mixed with epoxy resin and a hardening agent with a mixing ratio of 96:4 were pressed and then cured to be a toroid-type ring magnet with an outer diameter (${\Phi}_{out}$) of 15 mm, an inner diameter (${\Phi}_{in}$) of 6.5 mm and a thickness (t) of 2.5 mm, respectively. The magnetic properties of the Alnico ring magnets were varied with the mixing ratio of Alnico powders that possess different average particle sizes. The Alnico ring magnet prepared by mixing 5 wt% of $50{\mu}m$ (small size) powder, 15~20 wt% of $150{\mu}m$ (medium size) powder, and 75~80 wt% of $300{\mu}m$ (large size) powder showed the best magnetic properties (remanent induction, coercive force, maximum energy product, and surface flux density). In addition, measurements of temperature and moisture characteristics for the Alnico ring magnets showed that the surface flux densities of the N and S poles decreased little and the repulsive distance between the magnets decreased as small as 0.05 mm after 10 days.

Fatty Acid Profile and Thermal Behavior of Fat-Rich Edible Insect Oils Compared to Commonly Consumed Animal and Plant Oils

  • This study compared the physicochemical properties of edible insect oils from silkworm (Bombyx mori) pupa (SP), sago palm weevil (Rhynchophorus ferrugineus) larva (PW), and bamboo caterpillar (Omphisa fuscidentalis; BC) to oils from chicken skin (CK), beef back fat (BF), pork back fat (PF), salmon belly (SB), sea bass belly (BB), coconut (C), and peanut (P). The fatty acid profiles and thermal behaviors (crystallization and melting) of the extracted oils were evaluated. PW and BC oils had more saturated fatty acids (SFAs) than CK, PF, SB, BB, and P oils. SP oil had equivalent SFA content to CK and BB oils. Insect oils exhibited similar monounsaturated fatty acid concentrations in all samples, except C oils. PW and BC oils exhibited a higher content of palmitoleic acid than the other oils. SP oils contained polyunsaturated fatty acids similar to those in SB and BB oils, which were higher than those in PW, BC, CK, BF, and PF oils. SP oil also exhibited the highest concentration of α-linolenic acid (C18:3 n-3). Arachidonic acid (0.01-0.02 g/100 g) in all insect oils was lower level compared to CK, BF, PF, SB, and BB oils. SP oil (0.03 g/100 g) exhibited a slightly higher level of eicosapentaenoic acid compared to PW (0.01 g/100 g) and BC (0.01 g/100 g) oils. The insect oils were liquid at ambient temperature, solid below -15℃, and required less energy (∆Hm-max) for melting than other samples. This study indicated that insects, particularly SP, could serve as an alternative source of fat to meet its growing demand.

Parameters on Physiological Responses of Soybean (Glycine max Merr.) to Salinity (염분에 대한 콩의 생리학적 반응지표 연구)

  • The mechanism imparting salt tolerance to crop plants remains still unsolved, although soybean has been classified as a susceptible plant to NaCl. To determine optimum parameters on physiological responses for improving sensitivity of salinity in breeding program, soybean (Glycine max Merr., cv. "Gwan-gan") plants were grown in a greenhouse, treated 20 days after emergence for 7 days with NaCl at 0, 30, 60, and 90mM, corresponding to electric conductivity of 1.2, 4.4, 7.3, and 10.4 dS/m, respectively, and assessed 30 days after treatment. Chlorophyll contents were significantly decreased by NaCl ($0.4{\sim}1.0\;mg/g$) compared to control (1.2 mg/g). Photosynthesis rate by NaCl treatment at $0{\sim}90\;mM$ at flowering stage was ranged from 5.0 (control) to $9.6\;{\mu}mol/m^2/s$. Oxygen for respiration was consumed from 5.4 to $9.7\;{\mu}mol/m^2/s$ so that the ratio of $O_2$ (evolution:consumption) was increased with the increase of NaCl, indicating that $O_2$ consumption seems to go beyond $O_2$ evolution. Water potential of leaf at vegetative stage II was ranged from -0.6 to -1.8 MPa and the highest level was observed at mid-day. Water potential by salt stress was decreased with range of $-2.1{\sim}-2.7MPa$ compared to control. Transpiration was decreased from 17% to 20% by NaCl stress. Water vapor diffusing resistance of intercellular air space was affected significantly, increasing up to $16{\sim}24%$ compared to control by NaCl treatment. Salt-treated soybean tended to accumulate $Na^+$, specially in root, with reduced absorption of N, P, $K^+$, $Ca^{2+}$, and $Mg^{2+}$ contents. Free proline content of soybean leaf as affected by different NaCl concentrations was increased 4.2 times ($184{\sim}434\;{\mu}g/g$) more than control. NaCl also increased activities of nitrate reductase and peroxidase by $28{\sim}161%$ and $3{\sim}22%$, respectively. The results show that physiological characteristics of soybean plants during assay were useful as the best parameters of salt stress or salt tolerance test to improve sensitivity in screening and breeding program among cultivars or germplasms.

Modification of Vermiculite for Use as a Floating Adsorbent for Copper Removal (수중의 구리 제거를 위한 질석흡착제의 표면개질 연구)

  • The main objective of this study was to evaluate the removal properties of Cu from existing exfoliated vermiculite (EV) coated with a mixed solution of sulfuric acid and glycerol on the $580^{\circ}C$ in heating, which uses coated with glycerol of copper ions can be removed more effectively. Serial batch kinetic tests and batch sorption tests were conducted to determine the removal characteristics for Cu in aqueous solution. The result of batch kinetic test shows that removal rate, $K_{obs}$ (1/hr), of Cu are 0.579, 0.878, 3.459, and 6.578 for MEV weight 1 g (25 g/L), 2 g (50 g/L), 3 g (75 g/L), 4 g (100 g/L), respectively. In this case the initial pH of the solution was 3.26. The removal experiment according to the concentration, $K_{obs}$ (1/hr), of Cu are 1.96, 0.878, 1.25, and 1.04 for the initial concentration of 3 mg/L, 5 mg/L, 8 mg/L, 10 mg/L, and the initial pH of the solution are 3.46, 3.26, 3.10, 2.96, respectively. Influence of initial pHs on Cu removal were tested under 1g of MEV with 5 mg/L of Cu solution. $K_{obs}$ (1/hr) were increased from 0.263 (pH 3) to 0.525 (pH 5). It leads to the conclusion that the removal rates are inversely proportional to the initial Cu concentration and are increased proportional to the initial pHs. Sorption capacity of MEV was determined by batch sorption tests. The maximum sorption capacity ($Q_{max}$) obtained from Langmuir was 0.761 mg/g, Linear and Freundlich partition coefficients were 0.494 L/g and 0.729 L/g (1/n = 0.476). These results show that the MEV could be used as an excellent adsorbent for copper contained in various types of aqueous solutions.

Translucency of ceramic veneers on glazing effect (세라믹 비니어의 글레이징에 따른 반투명도 변화)

  • Purpose: The aim of this study was to compare the translucency of two different laminate ceramic veneers with and without glazing. Materials and methods: Ten millimeter side square-shaped specimens in 0.3 mm and 0.6 mm thick were fabricated for the following materials with and without glazing (n=80): A1 shade IPS e.maxPress (IEM) and Styleveneers (STV). The color coordinates (CIE $L^*a^*b^*$) of the specimens were measured with a colorimeter. The Translucency parameter (TP) was calculated from the color difference of the material on a black versus a white background. For comparisons between materials and between the 'not glazed' and 'glazed' groups, unpaired t-test was used to analyze the data (P=.05). Results: The TP ($Mean{\pm}SD$) of 'not-glazed' and 'glazed' group of IEM specimens at 0.3 mm thickness were $45.99{\pm}3.00$ and $49.53{\pm}2.28$ and the TP at 0.6 mm thickness were $32.82{\pm}2.59$ and $43.02{\pm}0.98$, respectively. Likewise, the TP of 'not-glazed' and 'glazed' group of STV specimens at 0.3 mm thickness were $47.03{\pm}3.65$ and $50.95{\pm}3.05$ and the TP at 0.6 mm thickness group were $34.48{\pm}1.28$ and $43.39{\pm}1.20$, respectively. As the glazing of ceramic veneer differed, the TP of each ceramic veneer showed statistically significant difference. But, the result between the products was not statistically different. Conclusion: Within the limitations of this study, we are concluded that the glazing process changed translucency of laminate ceramic veneers and the TP would not be affected by products.

Dependence of Sub-Cellular Activities of the Blooming and Harmful Dinoflagellate Cochlodinium Polykrikoides on Temperature (수온에 따른 유해성 Cochlodinium polykrikoides 적조생물의 세포생리 변화)

  • Water temperature-dependent fluctuations of biochemical and molecular activities in the harmful dinoflagellate, Cochlodinium polykrikoides were studied. In terms of genomic DNA concentration, a similar value of 0.6 was observed at $12^{\circ}C$ and $15^{\circ}C$. However, DNA significantly increased beyond $18^{\circ}C$ (p<0.05), to a maximum of 1.8 at $24^{\circ}C$. DNA concentration significantly decreased to 0.6. The concentrations of RNA and total protein were likely at their highest values of 1.7 and 0.07 ${\mu}g$ $ml^{-1}$ at $24^{\circ}C$, respectively. RNA and total protein concentrations began to increase at $15^{\circ}C$. Oxygen availability between lower and higher temperatures was significantly different and increased from $18^{\circ}C$ according to light intensity, regardless of wavelengths (p<0.05). At $24^{\circ}C$, the highest value of the maximum electron transport rate ($ETR_{max}$), ranging from 537.9 (Ch 1) to 602.5 ${\mu}mol$ electrons $g^{-1}$ Chl ${\alpha}s^{-1}$ (Ch 4), was also apparent. Nitrate reductase (NR) and ATPase activities were at their highest values of 0.11 ${\mu}mol$ $NO_{2}^{-}$ ${\mu}g^{-1}$ Chl ${\alpha}h^{-1}$ and 0.78 pmol 100 $mg^{-1}$ at $24^{\circ}C$, respectively. In an analysis of CHN, the concentration of C and N also significantly increased (p<0.05). Most of the measurements for the cellular activities at $27^{\circ}C$, however, were less than at $24^{\circ}C$. These results suggest that the sub-cellular activities of C. polykrikoides are sensitive to changes in water temperature. It may be desirable to estimate at $18^{\circ}C$ the initiation of the massive blooming development of C. polykrikoides. In nature, it will be very difficult to maintain the massive blooms beyond $24^{\circ}C$ because of a possibly significant decrease in molecular activity of C. polykrikoides.
