Effects of Sprouting Treatment with Gibberellin to Germination and the Growth of Spring Barley (지벨레린 최아처리가 춘파대맥의 발아생육에 미치는 영향)

  • Germination and field test were made with the spring barley sprouted with gibberellin solution of 5 levels from 5 to 100 ppm. Temperatures of room for germination were classified as ordinary and low. Results obtained will be summarized as follows: 1. Sprouting with Gibberellin promoted the germination by two days in ordinary and three days in low temperature without reducing the germination rate. 2. The higher concentration of gibberellin showed the more promotion of growth in early stage. 3. No retardation of root-growth was found by gibberellin sprouting. 4. No promoting effects to germination by gibberellin sprouting was found in field tests. In high concentration, percentage of germination, number of spike and grain yields reduced remarkably. 5. More remarkable tendency of weak growth and longer duration of recovery were found in the higher concentration of gibberellin, but all plots recovered from weak growth at middle stage of plant growth. 6. In gibberellin concentration of 5 to 10 ppm, retardation of germination, weakness of early growth and reducing of spike number were not remarkable while length of spike and 1000 grain weight were increased. Therefore grain yields showed increasing tendency. 7. Promoting the germination may be expected but hastening of heading and maturing may not be available by gibberellin treatment in sprouting.

Stress-induced Activity of Matrix Metalloproteinase in Tobacco Plants (담배식물체에서 스트레스에 따른 Matrix Metalloproteinase의 활성)

  • Matrix metalloproteinases (MMPs) are zinc-dependent endopeptidases produced by a variety of cell type, and have a fundamental role in the degradation and remodeling of extracellular matrix. In this study, we screened the secretion of MMPs in leaves of different developmental stages and in response to environmental stress using tobacco. Compare with fully maturing leaves and older leaves, the rate of MMPs activity was high in expanding and younger leaves. It is tempting to speculate that MMPs may be involved in tissue modeling, which must occur during leaf expansion. The MMPs activity in tobacco leaves grown in the presence of stressors showed a significantly increase at salinity treatment and pathogen infection. The MMPs activity in salinity and pathogen treatment increased respectively, by 1.2- and 1.5-fold with respect to the control. These results suggest that MMPs may be involved in plant defence against adverse environment and pathogenic infection.

Effect of No-Tillage on Soybean Yield and Weed Emergence in Drained Paddy Field Condition in Jeonnam Province (전남지역 논 조건에서 무경운 재배가 콩의 생육, 수량 및 잡초 발생에 미치는 영향)

  • This study was conducted to compare soybean growth and yield and the degree of weed emergence according to no-tillage and conventional tillage system in two different drained paddy fields, loam of Chilgok series and silty clay loam of Deokpyeong series. In both soil conditions, the maturing time of the soybeans by the no-tillage system was two days earlier than that by the conventional tillage system. In the loam of Chilgok series, the stem length of the soybeans in the no-tillage system was 5.7 cm longer than that in the conventional tillage system. The miss-planted rate and diseased plants of black root rot (Calonectria iliacola) in the no-tillage system were 9.2% and 2.8% lower, respectively than those in the conventional tillage system. Also, the nodulation and seed yield in the no-tillage system were 32% and 13% more, respectively, than those in the conventional tillage system. In the silty clay loam of Deokpyeong series, the stem length of the soybeans in the no-tillage system was 4.6cm shorter than in the conventional tillage system. The diseased plants of black root rot (Calonectria iliacola) in the no-tillage system were 4.2% lower than those in the conventional tillage system. Also, no significant difference in the seed yield between the no-tillage and conventional tillage systems was observed. On the other hand, there was a lower occurrence of weed in the no-tillage system than in the conventional tillage system, and the income increased by 19% owing to yield increase and reduced management costs.

Occurrence and Distribution of Monosporascus Root Rot and Pathogenicity of Monosporascus cannonballus on Cucurbitaceae Plants (박과류 검은점뿌리썩음병의 발생분포 및 분리병원균의 병원성)

  • Root rot of Cucurbitaceae plants, caused by Monosporascus cannonballus, is one of the recently described diseases in Korea. The distribution and pathogenicity of M. cannonballus were examined by field and in vitro experiments. Root rot caused by M. cannonballus occurred on melon, oriental melon, watermelon and cucumber plants. In two years of disease survey, the disease occurred at 10 and 32 fields in 1997 and 1998, respectively, which were located at Kimhae, Chinju, and Namhae in Kyeongnam province, at Kwangyang in Chonnam province, at Kwangju city, and at Yeoju and Inchon in Kyeonggi province. The disease progress in a melon field at Namhae was not observed until the middle stage of plant growth, but rapidly increased at the fruit maturing stage, resulting in more than 50% yield loss. Isolation rate of M. cannonballus was 36.7% from wilted hosts. Fusarium oxysporum and Rhizoctonia solani were also frequently isolated. In vitro test, seedlings wilted after 7∼14 days of inoculation, and perithecia were formed in infected roots 21 days later. Two cucumber cultivars, Baekbong and Eunhwa, were resistant to the disease.

Induced Ovulation by using Human Chorionic Gonadotropin and Gonadotropin-Releasing Hormone Analogue plus Pimozide in Yellow Puffer, Takifugu obscurus (인간의 태반성 성선자극호르몬 또는 성선자극호르몬-방출호르몬 유도체와 Pimozide에 의한 황복의 배란유도)

  • Ovulation of maturing female yellow puffer, Takifugu obscrus, was induced by using single injection of human chorionic gonadotropin (HCG) or gonadotropin releasing hormone-analogue (GnRH-A) $des-Gly^{10}[D-Ala^6]$ GnRH-ethylamide plus pimozide. The response was evaluated using the fertilization and embryo-formation rate after insemination and the gonadotropin (GTH) level in blood plasma using radioimmunoassay. In the fertilization and embryo-formation, maximal effects were recorded by using 1,000 IU/kg HCG or $10\;{mu}g/kg$ GnRH-A plus 5 mg/kr pimozide. Pimozide (1, 5 mg/kg) or GnRH-A treatment alone was not effective in elevation of GTH level, however combinations of these treatments were particularly effective. Injection of dopamine blocked the rapid elevation of plasma GTH levels of blood. These data suggest that yellow puffer secrete GnRH and gonadotropin-releasing-inhibiting factor during the spawning or the other period.

The Effect of Replacing Grass with Urea Treated Fresh Rice Straw in Dairy Cow Diet

  • Nine tons of fresh rice straw from early-maturing varieties was ensiled with 50 g urea $kg^{-1}$ DM straw in plastic bags immediately after threshing. Five months after storage, preserved straw was used to partially replace elephant grass (Pennisetum purpureum, Schumach) for lactating cows. Eight crossbred Holstein lactating cows (75% of Holstein blood) in their second to fourth lactation and in mid-lactation were arranged in a balanced design with two squares consisting of $4\;periods{\times}4\;treatments$ (100% grass ad lib. as a control; 75% grass+urea treated fresh rice straw (UTrFRS) ad lib.; 50% grass+UTrFRS ad lib.; 25% grass+UTrFRS ad libitum) in one square. A concentrate supplement was given at a rate of 400 g per day per kg of milk produced. Samples of fresh straw taken in the field and UTrFRS and elephant grass taken at feeding time were evaluated in a degradation trial with 3 fistulated heifers (undefined blood ratio of crossbred of Sindhi and local yellow cattle). Straw preserved for 5-9 months was in nearly all cases of good quality. Crude protein (CP) content was increased 2.1 fold and 48 h dry matter loss (DML) was 20% higher compared to dry straw. Elephant grass cultivated intensively was low in DM content and 10% higher in 48 h DML compared to UTrFRS. Dry matter intake (DMI) was higher for the mixture of UtrFRS and Elephant grass, and highest when one-third of the roughage was UTrFRS. Higher DMI of mixed roughage diets was probably due to the low DM content of elephant grass in the sole grass roughage diet. Increasing substitution of elephant grass with UTrFRS up to 75% of the roughage component increased milk fat content and had no effect on milk yield and other milk composition parameters. Feeding UTrFRS, partially replacing elephant grass in the diets of lactating cows in the dry season can reduce the cost of roughage.

  • A field experiment was conducted to find out the effects of liming(soil pH) and sources of N on growing characters and yield of burley tobacco. Treatments consisted of liming(nonliming, liming to soil pH5.5 and 6.5) as the main plot and N sources(compound fertilizer of containing 3.9% NH$_4$-N and 6.1% NH$_2$-N, NaNO$_3$,(NH$_2$)$_2$CO and (NH$_4$)$_2$SO$_4$as the sub-plot. The growth of vegetative growing stage of limed plots were delayed(to compare the nonlimed plot) by influence of alkali. When the source on N was NaNO$_3$ the growth of vegetative growing stage was unfavorable and the yellowing of lower leaves of maturing stage was rapid. The yield and value of cured leaf was increased by increasing the rate of Ca(OH)$_2$, but there was no significant differences among the source of N. The yield response to liming was greater when the source of N was (NH$_4$)$_2$SO$_4$ than that of any other plots.

Induced Ovulation in the Mandarin Fish, Siniperca scherzeri by Sex-Maturation Hormones (성성숙 호르몬 처리에 의한 쏘가리의 배란 유도)

  • Ovulation of maturing femal mandarin fish, Siniperca scherzeri was induced using single injection of human chorionic gonadotropin (HCG) or gonadotropin releasing hormone-analogue (GnRH-a), GnRH-a plus prostaglandin F2 (PG$F_2$) or GnRH-a plus pimozide. The response was evaluated by fertilization, embryo-formation and hatching rate after insemination. Those rates were generally higher in GnRH-a group than in HCG group. The higher hatching rat of above 89% was achived using a dosage of 5,000 IU/kg HCG plus 10 ${\mu}$g/kg GnRH-a, 10${\mu}$g/kg GnRH-a plus 500 ng/kg PGF2, and 10 ug/kg GnRH-a plus 1-5 mg/kg pimozide. Ovulation was induced in all female injected with sex-maturation hormones and stimulator, but blocked in female injected with HCG plus GnRH-a plus dopamine combination, and GnRH-a plus PGF2 plus indometacin combination. These results show that the mandarin fish in spawning period secrete a sex-mutruation assosiated hormones and gonadotropin-releasing -inhibiting factor(GRIF).

  • Sixty five soybean varieties were tested to observe varietal response and to get the basic data for resistant variety breeding to soybean cyst nematode (Heterodera glycines I.). They were classified into five groups, from the most resistant to the most susceptible by decreasing rate of seed weight. A few Korean varieties were included in the most resistant group. In general, early maturing varieties were more susceptible. Total plant weight could be used as an important criterion to select a resistant variety to soybean cyst nematode.

Studies on the Method of Reducing of the Photobleaching Leaves and Shortening the Curing Period in Burley Tobacco Stalk Curing (버어리종담배 대말림시 백화엽발생방지 및 건조기간단축에 관한 연구)

  • This study was conducted to find out the stalk curing methods that can reduce photobleaching, and shorten the curing period in burley to bacco stalk curing. The results obtained summarized as follows. In the ventilation window-plot, the temperature within the curing house was high and the curing period was shortened, too. The rate of photobleached leaves of whole plant stalk curing-plot was higher than those of any other plots. In the ventilation window-plot, the labor of operating the curing house was saved and the yield and visual quality of cured leaves were high, too. For reducing the photobleaching of leaves, following procedures are desirable; to prevent the lower leaves from drying up and overripening at maturing stage, cut the stalk after first or second priming, shorten the curing period as possible when the 80% of leaf bodies changed brown.
