Measurement of Antioxidant Activity of Anserine, Taurine, and L-Histidine in vitro and Content of Anserine, Taurine, and L-Histidine in Mature and Juvenile Rainbow Trout (Onchorhynchus mykiss) Muscle

  • The content of anserine, taurine, and L-histidine was measured by HPNC in the muscle of mature(670~690g) and juvenile(80~120g) rainbow trout fatmed in Chungsun, Korea. The concentration of anserine and taurine was higher in mature rainbow trout than in juvenile, but that of L-histidine was lower in mature than in juvenile. When measured with the chemiluminescence(CL) assay, anserine and taurine showed very powerful antioxidative activity above physiological concentration rainbow trout. Taurine still showed antioxidative activity below physiological concentration, while anserine showed prooxidative activity below that. L-Histidine was prooxidative dose-dependently. In TBA method, while taurine showed very week antioxidative effect, anserine appeared very powerful antioxidant and L-histidine prooxidant at physiological concentration. There was no synergism between anserine and taurine and anserine inhibited prooxidative effect of L-histidine.

Initiation of embryogenic callus from mature zygotic embryos in Japanese larch (Larix kaempferi)

  • Embryogenic callus (EC) was created from mature embryos of Larix kaempferi. With the mature embryos, keeping the culture in dark conditions throughout the experiment (38.2%) seemed to give better results than exposing them to 16 h light ($25{\mu}Em^{-2}s^{-1}$) for the first week (21.9%). EC was obtained most frequently from Quoirin and Lepoivre (LP) mediums with 1.0 mg/L 4-amino-3,5,6-trichloropicolinic acid (Picloram), plus 1.0 mg/L benzyladenine (BA) (62.8%) or Litvay's medium (LM) containing 1.0 mg/L p-chlorophenoxyacetic acid (pCPA) plus 1.0 mg/L BA (62.8%) treatment. In both cases, best results were obtained when zygotic embryos were cultured in darkness. As for the effective sucrose concentration on initiation of EC, 29.2 mM sucrose (38.6%) gave the best results.

Quantification of Antidepressant Miquelianin in Mature and Immature Fruits of Korean Rubus Species

  • Antidepressant miquelianin (quercetin 3-O-glucuronide) was isolated from the leaves of Rubus craetaegifolius (Rosaceae) and identified by physical and spectroscopic data. Miquelianin was quantitatively analyzed in the leaves, mature and premature fruits of Korean wild R. craetagifolius, R. pungens var. oldhami, R. coreanus, and R. parvifolius by HPLC. The contents of miquelianin was highest in the mature fruit of R. crataegifolius ($16.29{\pm}0.79mg/g$); however, the content of kaempferol 3-O-glucuronide was $33.88{\pm}7.68mg/g$. These results suggest that the mature fruit of R. crataegifolius would be beneficial for treating depression or stress as a functional food with its sweet taste.

Effects of Rhizobium Inoculant, Compost, and Nitrogen on Nodulation, Growth, and Yield of Pea

  • The effects of Rhizobium inoculant, compost, and nitrogen on nodulation, growth, dry matter production, yield attributes, and yield of pea (Pisum sativum) var, IPSA Motorshuti-3 were assessed by a field experiment. Among the treatments Rhizobium inoculant alone performed best in recording number and dry weight of nodules/plant. The highest green seed yield of 8.38 ton/ha (36.9% increase over control) and mature seed yield of 2.97 ton/ha (73.7% increase over control) were obtained by the application of 90 kg N/ha. The effects of 60 kg N/ha, Rhizobium inoculant alone and Rhizobium inoculant along with 5 ton compost/ha were same as the effect of 90 kg N/ha in recording plant height, root length, dry weight of shoot, and root both at preflowering and pod filling stages, number of mature pods/plant, number of mature seeds/pod, 1000-seed weight, green, and mature seed yields of pea.

  • Malignant transformation of ovarian mature cystic teratomas is rare, and papillary thyroid cancer occurs in 0.1%-0.3% of ovarian teratomas that undergo malignant transformation. We describe a case of successful in vitro fertilization pregnancy and delivery after a fertility-sparing laparoscopic operation in a patient with papillary thyroid carcinoma arising from a mature cystic teratoma.

Reproductive Cycle of BlueStriped Angelfish, Chaetodontoplus septentrionalis in Jeju Coastal Waters

  • Reproductive cycle of the blue-striped angelfish, Chaetodontoplus septentrionalis were histologically investigated. Fish were monthly collected in the coastal waters of Munseom, Seogwipo, Jeju-Island from February to December 2018. The gonadosomatic index (GSI) increased from May and maintained high values in August. The reproductive cycle of female fish can be classified by the characteristics observed during gonadal development as followed: growing stage (November to June), early mature stage (May to June), mature and spawning stage (June to September), and degenerative and recovery stage (September to December). In the male, testicular development period was similar to that of ovarian development period, but mature and spawning period was one month longer from June to October. Fecundity of mature female ranged from 4,601 to 22,840 and was correlated positively with total length and body weight. The histological analysis of gonadal development indicated that the C. septentrionalis was summer-spawning type and is considered a multiple spawner during spawning season.

간척지 갈대의 영양생장과 지상부 모듈의 개체군 동태 (Clonal Growth and Shoot Modules Dynamics of Phragmites longivalvis in a Reclaimed Land)

  • The structure, clonal grwoth, shoot modules dynamics and rhizome respiratory rate of the Phragmites longivalvis circular patch were studied in a 15-year-old reclaimed land on Mado of the western Korea for two growing seasons. Clonal growth by P. longivalvis resulted in subpopulations of shoot modules which may be separated into pioneer, mature and senile growth phase. The radial growth of circular patch was 208 cm per a year. The shoot phytomass, shoot height and spike frequency were the lowest in the senile phase, and were the highest in the mature phase. The maximum shoot phytomass of pioneer, mature and senile phase in August was 347, 536 and 195g/$\m^2$ for the first year, and 609, 748 and 248g/$\m^2$ for the second year, respectively. The birth rates of shoot modules were the highest during the early spring. Of all modules, the percemtage of spring cohort of pioneer, mature and senile growth phase were 52, 82 and 70% for the first year, and 83, 88 and 85% for the second year, respectively. The flowering shoots were only derived from spring cohorts. The death rates of shoot modules of the pioneer and mature phase were the highest in October. The module is an annual semelparous plant though the genet is a perennial iteroparous plant. Their survivorship curve was negatively (Deevey type I). The density of shoot modules of the pioneer, mature and senile growth phase increased as much as 4.0, 1.6 and 2.3 times for one year, respectively. The respiratory rate was the highest in the lowest in the senile growth phase.

Human Embryonic Stem Cell Transplantation in Parkinson′s Disease (PD) Animal Model: II. In Vivo Transplantation in Normal or PD Rat Brain

  • This study was to examine whether the in vitro differentiated neural cells derived from human embryonic stem (hES, MB03) cells can be survived and expressed tyrosin hydroxylase(TH) in grafted normal or PD rat brain. To differentiate in vitro into neural cells, embryoid bodies (EB: for 5 days, without mitogen) were formed from hES cells, neural progenitor cells(neurosphere, for 7-10 days, 20 ng/㎖ of bFGF added N2 medium) were produced from EB, and then finally neurospheres were differentiated into mature neuron cells in N2 medium(without bFGF) for 2 weeks. In normal rat brain, neural progenitor cells or mature neuron cells (1×10/sup 7/ cells/㎖) were grafted to the striatum of normal rats. After 2 weeks, when the survival of grafted hES cells was examined by immunohistochemical analysis, the neural progenitor cell group indicated higher BrdU, NeuN+, MAP2+ and GFAP+ than mature neuron cell group in grafted sites of normal rats. This result demonstrated that the in vivo differentiation of grafted hES cells be increased simultaneously in both of neuronal and glial cell type. Also, neural progenitor cell grafted normal rats expressed more TH pattern than mature neuron cells. Based on this data, as a preliminary test, when the neural progenitor cells were grafted into the striatum of 6-hydroxydopamine lesioned PD rats, we confirmed the cell survival (by double staining of Nissl and NeuN) and TH expression. This result suggested that in vitro differentiated neural progenitor cells derived from hES cells are more usable than mature neuron cells for the neural cell grafting in animal model and those grafted cells were survived and expressed TH in normal or PD rat brain.

Malignant Transformation Rate and P53, and P16 Expression in Teratomatous Skin of Ovarian Mature Cystic Teratoma

  • Objective: To investigate the incidence of malignant transformation and P53 and P16 expression in teratomatous skin of ovarian mature cystic teratoma. Materials and Methods: Data on ovarian teratoma specimens in nearly 10 years were reviewed. P53 and P16 expression were detected by immunohistochemistry in 25 cases of teratomatous skin of ovarian mature cystic teratoma, 20 cases of squamous cell carcinoma and 2 cases of squamous cell carcinoma originated from teratomatous skin. Results: Of 1913 cases of ovarian mature cystic teratoma in nearly 10 years, only two cases of squamous cell carcinoma were found in teratomatous skin, with malignant transformation rate of 0.1045%. P53 expression was detected in 2 cases squamous cell carcinoma originated from teratomatous skin and P16 overexpression in one. There were no expressions of P53 and P16 in 25 cases of teratomatous skin of ovarian mature cystic teratoma. Of 20 cases of squamous cell carcinoma P53 overexpression (positive rate of 55%) was detected in 11 cases, P16 overexpression (positive rate of 35%) in 7 cases. The positive rates of P53 and P16 expression in squamous cell carcinomas were significantly higher than that in the teratomatous skins (p< 0.001, p= 0.002). Conclusions: There was low risk of malignant transformation in teratomatous skin of ovarian mature cystic teratoma which can be explained by lower P53 and P16 expressionin teratomas than that in squamous cell carcinoma.

Efficient Callus Induction and Plant Regeneration from Immature and Mature Embryo Culture of Korean Wheat Genotypes

  • Immature and mature embryos of 18 Korean wheat genotypes were cultured in vitro to develop an efficient method of callus formation and plant regeneration, and to compare the responses of both embryo cultures. Immature and mature embryos were placed on a solid agar medium containing the MS salts and vitamins, 30g/l maltose, 2 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D), and amino acids. The developed calli were maintained on regeneration medium containing MS salts and B5 vitamins, 20 g/l sucrose, and the combination of two plant growth regulators, 6-benzylaminopurine (BAP) and indole-3-acetic acid (IAA). Immature embryos in most genotypes showed high efficiency of callus induction except three genotypes; Eunpamil, Chunggemil, and Namhaemil, and significant differences among the genotypes. Plant regeneration of calli induced from immature embryos showed high efficiency in Geurumil (56.5%), Tapdongmil (50.5%), Gobunmil (45.5%), and Urimil(42.2%). The analysis of variance showed significant differences for regeneration frequency among the genotypes. Mature embryos showed low callus induction frequency compared with that in immature embryos, and significant differences among the genotypes. Plant regeneration of calli induced from mature embryos showed high efficiency in Keumkangmil (33.33%), Tapdongmil(28.13%), and Geurumil (27.78%). The analysis of variance showed significant differences for plant regeneration frequency among the genotypes.
