Effects of Inhalable Microparticles of Seonpyejeongcheon-Tang in an Asthma Mouse Model - Effects of Microparticles of SJT -

  • Objectives: Allergic asthma generally presents with symptoms of wheezing, coughing, breathlessness, and airway inflammation. Seonpyejeongcheon-tang (SJT) consists of 12 herbs. It originated from Jeong-cheon-tang (JT), also known as Ding-chuan-tang, composed of 7 herbs, in She-sheng-zhong-miao-fang. This study aimed to evaluate the effects of local delivery of SJT via inhalable microparticles in an asthma mouse model. Methods: Microparticles containing SJT were produced by spray-drying with leucine as an excipient. SJT microparticles were evaluated with respect to their aerodynamic properties, in vitro cytotoxicity, in vivo toxicity, and therapeutic effects on ovalbumin (OVA)-induced asthma in comparison with orally-administered SJT. Results: SJT microparticles provided desirable aerodynamic properties (fine particle fraction of $48.9%{\pm}6.4%$ and mass median aerodynamic diameter of $3.7{\pm}0.3{\mu}m$). SJT microparticles did not show any cytotoxicity against RAW 264.7 macrophages at concentrations of 0.01 - 3 mg/mL. Inhaled SJT microparticles decreased the levels of IL-4, IL-5, IL-13, IL-17A, eotaxin and OVA-IgE in bronchoalveolar lavage fluid (BALF) in mice with OVA-induced asthma. These effects were verified by histological evaluation of the levels of infiltration of inflammatory cells and collagen, destructions of alveoli and bronchioles, and hyperplasia of goblet cells in lung tissues. The effects of SJT microparticles in the asthma model were equivalent to those of orally-administered SJT extract. Conclusion: This study suggests that SJT is a promising agent for inhalation therapy for patients with asthma.

  • A new cyclone system for the vacuum cleaner to collect dusts has been studied experimentally and numerically to meet the constant suction power, hygienic exhaust and a reduction of maintenance cost. The cyclone system of the vacuum cleaner consists of twin cyclones for improving dust collection efficiency. The first. cyclone catches large dust particles and the second one having two separated flows to decrease pressure drop collects small dust particles. The optimal design factors such as dust collection efficiency, pressure drop, and cut-size are investigated from the experimental results by the Taguchi method. Cyclone cleaner systems designed in this study has a good Performance taking into account the dust collection efficiency of 93% and the cut-size of $1.6{\mu}m$ in mass median diameter at the flow rate of 1 CMM. The cyclone vacuum cleaner showed the potential to be an effective method to collect dusts generated in the household.

  • A visualization test of the fuel-coolant interaction in the Test for Real cOrium Interaction with water (TROI) test facility was carried out. To experimentally simulate the In-Vessel corium Retention (IVR)- External Reactor Vessel Cooling (ERVC) conditions, prototypic corium was released directly into the coolant water without a free fall in a gas phase before making contact with the coolant. Corium (34.39 kg) consisting of uranium oxide and zirconium oxide with a weight ratio of 8:2 was superheated, and 22.54 kg of the 34.39 kg corium was passed through water contained in a transparent interaction vessel. An image of the corium jet behavior in the coolant was taken by a high-speed camera every millisecond. Thermocouple junctions installed in the vertical direction of the coolant were cut sequentially by the falling corium jet. It was clearly observed that the visualization image of the corium jet taken during the fuel-coolant interaction corresponded with the temperature variations in the direction of the falling melt. The corium penetrated through the coolant, and the jet leading edge velocity was 2.0 m/s. Debris smaller than 1 mm was 15% of the total weight of the debris collected after a fuel-coolant interaction test, and the mass median diameter was 2.9 mm.

Effects of Didecyldimethylammonium Chloride on Sprague-Dawley Rats after Two Weeks of Inhalation Exposure

  • Didecyldimethylammonium chloride (DDAC) is used for various purposes, such as a fungicide for coolants, an antiseptic for wood, and disinfectant for cleaning. Despite the increasing likelihood of DDAC inhalation, available data on its toxicity from inhalation are scarce. Therefore, this study was aimed at confirming the toxicity of DDAC after inhalation exposure for 2 wk. Male Sprague-Dawley rats were exposed to approximately $0.15mg/m^3$, $0.6mg/m^3$, and $3.6mg/m^3$ DDAC aerosols in whole-body exposure chambers. After DDAC exposure for 2 wk, effects of DDAC on body weight, blood, bronchoalveolar lavage (BAL), and the lungs were verified. The mass median aerodynamic diameter of DDAC aerosols was $1.86{\mu}m$ and the geometric standard deviation was 2.75. The concentrations of DDAC aerosols for the low, medium, and high groups were $0.15{\pm}0.15mg/m^3$, $0.58{\pm}0.40mg/m^3$, and $3.63{\pm}1.56mg/m^3$, respectively. Body weight gain was significantly influenced by DDAC exposure. In the high group, a body weight decrease of 2.6 g was observed, whereas a 25.8 g increase was observed in the normal control group after the first 3 days. The low and medium groups showed 23.3 g and 20.4 g increases, respectively, after the first 3 days. Decreases in body weight were recovered during the next 4 days. In contrast, no changes were noted in hematological and blood biochemistry parameters after DDAC exposure. Furthermore, only mild effects were observed on bronchoalveolar cell differentiation counts and cell damage parameters in the BAL fluids of the medium and high groups. Although inflammatory cell infiltration and interstitial pneumonia were partially observed, fibrosis was not found in the lungs of the medium and high groups. In conclusion, body weight gain and the lungs were mainly affected by DDAC exposure. The no-observed-adverse-effect level (NOAEL) for DDAC was determined as $0.15mg/m^3$.

Comparison of the Characteristics of 16 Commercial Nebulizer/Compressor Combinations Used in Korea (국내 시판되는 16가지 연무기/압축기의 성능 평가)

  • Purpose : We assessed the dynamic characteristics of 16 nebulizer/compressor combinations currently available in Korea. Methods : The 16 nebulizer/compressor combinations(Pariboy Type 38/Long life, Pariboy Type N/Long life, Pariboy Type N/Salter 8900, Pariboy Type N/LC, Devilbiss pulmoaid-LT/Hudson, Devilbiss pulmoaid/Hudson, Mesmed neb-300/Own, San-up 3040/Hudson, Midas(Basic)/Own, AirJolie 2/Hudson, Thomas 1127/Salter 8900, Noel NE-2000/Salter 8900, Omron CX3/Hudson, Chang Woo CWN-100/Salter 8900, Voyage/Mefar, Chang Woo ASI-Pro/Medel jet pulse) were evaluated in terms of particle size and mass output. In addition, we determined the effects of nebulizer fill volume on mass output. Results : Pariboy Type N/Long life has the highest respirable mass of 0.184 mg/min and Mesmed Neb-300/Own has the lowest 0.019 mg/min. Pariboy Type N/Long life has the highest mass output of 0.68 mg/min and the shortest mass median aerodynamic diameter(MMAD) of $3.76{\mu}m$. All combinations other than Pariboy Type N/Long life produced a MMAD of over $5{\mu}m$. MMAD over a 5 min nebulization ranged 3.76 to $9.83{\mu}m$. There were no significant effects of fill volume on mass output. Conclusion : We concluded that there is a wide variation in performance of nebulizer/compressor combinations. The characteristics of nebulizer/compressor combinations should be considered in selecting products.

Clinical Analysis of 61 Krukenberg Tumors (크루켄버그 종양 61예에 대한 임상적 분석)

  • Purpose: A Krukenberg tumor is an ovarian tumor of a signet-ring cell type. This tumor arises more commonly in young women, and the prognosis is poor. The primary focus of this tumor is often found at gastrointestinal malignancy, especially gastric cancer. We tried to identify the clinical characteristics of this tumor, and in that regard, this report might be helpful. Materials and Methods: We reviewed the 61 patients with Krukenberg tumors, who had been diagnosed at our hospital from 1994 to 2002, and retrospectively analyzed the clinical features. Results: The age distribution ranged from 15 to 59 years, and the mean age was 41 years. The most common symptom was a lower abdominal mass ($46\%$). Fourty-two cases ($77\%$) showed bilateral ovarian involvement, and the size of this tumor was variable, but in 24 cases ($4\%$) the size was $5\∼10$ cm for the largest diameter. Among 54 cases, 40 cases had ascites, and the volume of ascites was variable. The median survival of the 61 patients was 10 months, and Krukenberg tumor developed 19.7 months after the primary operation. The median survival durations of recurrence patterns were 20 months for the Krukenberg tumor alone, and 7 months for the Krukenberg tumor with peritoneal seeding. Conclusion: In young women treated with a gastrectomy, especially one for an advanced tumor, closed observation with abdominal ultrasonography or computed tomography to detect a Krukenberg tumor is recommended. The patient with a Krukenberg tumor alone has a better prognosis than one with a Krukenberg tumor combined peritoneal seeding. We will have to consider more progressive treatment for the patient with a Krukenberg tumor alone.

Characteristics of Size-segregated Mass Concentrations of Indoor Aerosol Particles in University Buildings (대학건물 실내 에어로졸입자의 입경별 질량농도 특성)

  • Objective: Based on the fact that fine particles are more likely to produce negative influences on the health of occupants as well as the quality of indoor air compared to coarse particles, it is critical to determine concentrations of aerosol particles with different sizes. Thus, this study focused on the size distribution and concentrations of aerosol particles in university buildings. Method: Aerosol particles in indoor air were collected from four areas: corridors in buildings(In-CO), lecture rooms(In-RO), laboratories(In-LR), and a cafeteria(In-RE). Samples were also collected from outside for comparison between the concentrations of indoor and outdoor particles. For the collection of the samples, an eight stage non-viable cascade impactor was used. Result: The average concentration of $PM_{10}$ in the samples collected from indoor areas was $34.65-91.08{\mu}g/m^3$,and the average for $PM_{2.5}$ was $22.65-60.40{\mu}g/m^3$. The concentrations of the aerosol particles in the corridors, lecture rooms, and laboratories were relatively higher than the concentrations collected from other areas. Furthermore, in terms of mass median aerodynamic diameter(MMAD), the corridors and lecture rooms had higher numbers due to their characteristics, showing $2.36{\mu}m$ and $2.11{\mu}m$, respectively. Laboratories running an electrolysis experiment showed $1.58{\mu}m$, and the cafeteria with regular maintenance and ventilation had $1.96{\mu}m$. Conclusion: The results showed that the $PM_{10}$ concentrations of all samples did not exceed indoor air quality standards. However, the $PM_{2.5}$ concentration was over the standard and, in particular, the concentration of fine particles collected from the laboratories was relatively higher, which could be an issue for the occupants. Therefore, it is important to improve the quality of the indoor air in university buildings.

Physical, Morphological, and Chemical Analysis of Fly Ash Generated from the Coal Fired Power Plant (석탄 화력발전소에서 발생되는 석탄회 특성과 형성 분석에 관한 연구)

  • Fly ash produced in coal combustion is a fine-grained material consisting mostly of spherical, glassy, and porous particles. A physical, morphological, and chemical characteristic of fly ash has been analyzed. This study may contribute to the data base of domestic fly ash, the improvement of combustion efficiency, ash recycling and ash collection in the electrostatic precipitator. The physical property of fly ash is determined using a particle counter for the measurement of ash size distribution and gravimeter. Morphological characteristic of fly ash is performed using a scanning electron micrograph and an optical microscope. The chemical components of fly ash are determined using an inductively coupled plasma emission spectrometry (ICP). The distribution of fly ash size was ranged from 15 to 25 $\mu$m in mass median diameter. Exposure conditions of flue gas temperature and duration within the combustion zone of the boiler played an important role on the morphological properties of the fly ash such as shape, relative opacity, coloration, cenosphere and plerosphere. The spherical fly ash might be generated at the condition of complete combustion. The size of fly ash was found to be increased the with particle-particle interaction of agglomeration and coagulation. Fly ash consisted of $SiO_2\;Al_2O_3\;and\;Fe_2O_3$ with 85% and carbon with 3~10% of total mass.

Performance Characteristics of PM10 and PM2.5 Samplers with an Advanced Chamber System (챔버 기술 개발을 통한 PM10과 PM2.5 시료채취기의 수행 특성)

  • The purposes of this study are 1) to develop an advanced chamber system within ${\pm}10%$ of air velocity at the particulate matter (PM) collection area, 2) to research theoretical characteristics of PM10 and PM2.5 samplers, 3) to assess the performance characteristics of PM10 and PM2.5 samplers through chamber experiments. The total six one-hour experiments were conducted using the cornstarch with an mass median aerodynamic diameter (MMAD) of $20\;{\mu}m$ and an geometric standard deviation of 2.0 at the two different air velocity conditions of 0.67 m/s and 2.15 m/s in the chamber. The aerosol samplers used in the present study are one APM PM10 and one PM2.5 samplers accordance with the US federal reference methods and specially designed three mini-volume aerosol samplers (two for PM10 and one for PM2.5). The overall results indicate that PM10 and PM2.5 mini-volume samplers need correction factors of 0.25 and 0.39 respectively when APM PM samplers considered as reference samplers and there is significant difference between two mini-volume aerosol samplers when a two-way analysis of variance is tested using the measured PM10 mass concentrations. The PM10 and PM2.5 samplers with the cutpoints and slopes (PM10: $10{\pm}0.5\;{\mu}m$ and $1.5{\pm}0.1$, PM2.5: $2.5{\pm}0.2\;{\mu}m$ and $1.3{\pm}0.03$) theoretically collect the ranges of 86~114% and 64~152% considering the cornstarch characteristics used in this research. Furthermore, the calculated mass concentrations of PM samplers are higher than the ideal mass concentrations when the airborne MMADs for the cornstarch used are smaller than the cutpoints of PM samplers and the PM samplers collected less PM in another case. The chamber experiment also showed that PM10 and PM2.5 samplers had the bigger collection ranges of 37~158% and 55~149% than the theocratical calculated mass concentration ranges and the relatively similar mass concentration ranges were measured at the air velocity of 2.15 m/s comparing with the 0.67 m/s.

Effects of Water Extracts of Persimmon Leaves to Cadmium Toxicity in Rats by Inhalation Exposure (시엽 추출물이 카드뮴에 흡입폭로된 랫드의 독성에 미치는 영향)

  • Experimental animals were divided into 5 groups; normal, cadmium control, and 3 experimental groups. Cadmium control and experimental groups were exposed to 1 mg/㎥ of cadmium aerosol in air by inhalation exposure for 6 hours/day, 5 days/week during 4 weeks. Dosages of 20, 40, and 80mg/kg of extracts of persimmon leaves were intraperitoneally injected to experimental groups respectively and several toxicological parameters and induction of metallothionein were measured from the rats that inhaled cadmium aerosol in air. The results of this study were as follows. Cadmium concentration that cadmium control and experimental groups were inhaled was 0.980±0.061 mg/㎥. Mass median diameter of cadmium aerosol for inhalation exposure was 4.93±0.483㎛. Cadmium content of normal group in lung was 0.088㎍/g and the highest cadmium content in lung, 55.492㎍/g was from 80mg/kg dose group. Cadmium concentration of normal group in blood was 0.348㎍/100㎖ and the highest cadmium concentration in blood, 2.642㎍/100㎖ was from cadmium control. Cadmium concentration of normal group in liver was 0.010㎍/g and the highest cadmium concentration in liver, 31.100㎍/g was from 20mg/kg dose group. Cadmium concentration of normal group in kidney was 0.030㎍/g and the highest cadmium concentration in kidney, 2.526㎍/g was from cadmium control. Cadmium concentration of normal group in intestine was O.064㎍/g and the highest cadmium concentration in intestine, 0.300㎍/g was from 80mg/kg dose group. The highest cadmium concentration in urine by week was 6.080㎍/day from 20mg/kg dose group in the fouth week and the highest cadmium concentration in feces by week was 341.731㎍/day from 20mg/kg dose group in the fouth week. Metallothionein concentration of normal group in lung was 5.769㎍/g and the highest in lung, 30.986㎍/g was from 80mg/kg dose group. Metallothionein concentration of normal group in liver was 38.856㎍/g and the highest in liver, 169.378㎍/g was from 40mg/kg dose group. Metallothionein concentration of normal group in kidney was 22.228㎍/g and the highest in kidney, 47.898㎍/g was from 80mg/kg dose group. Metallothionein concentration of normal group in intestine was 2.170㎍/g and the highest in intestine, 13.642㎍/g was from 80mg dose group.
