국내산 돼지고기의 원산지 검증을 위한 SNP Marker Set 개발 (Development of SNP Markers for Domestic Pork Traceability)

  • 본 연구는 돼지고기 원산지 식별에 활용될 수 있는 최적의 SNP marker set을 개발 및 확립하기 위해 수행되었다. 선발된 51개의 SNP marker들의 효율성, 다형성 및 독립성 검증을 실시하였으며 51개의 SNP marker set은 MassARRAY method에 의해서 Multiplex-PCR panel 4개로 디자인 되었으며, 농장별, 생산조합별, 모돈별, 웅돈별로 효과적으로 고유 유전자형 지문분석이 가능하게 제작되었고, 다른형태의 SNP 유전자형 분석 플랫폼에 적용될 수 있는 적절한 마커 갯수이다. 또한 51개의 SNP marker set을 적용하여 모의 실험 및 친자감별확율을 계산하였을 때 무작위 교배 집단(PI), 반형매 교배집단($PI_{half-sib}$)과 전형매 교배집단($PI_{sibs}$)을 통해 모의 실험을 한 결과 $5.63{\times}10^{-33}$, $4.35{\times}10^{-15}$ 그리고 $1.32{\times}10^{-15}$로 분석되었으며 친자확인률에서도 모두 100%에 가까운 확률값을 나타내었다. 따라서 본 연구에서 개발된 SNP marker set을 이용하여 돈육제품의 원산지를 추적에 이용한다면 개별돼지의 고유한 DNA 지문 정보를 생성할 뿐만 아니라, 이를 통하여 모돈과 웅돈을 식별하여 농장원산지를 확인이 가능 할 수 있을 것으로 사료된다. 따라서 국내산 돼지의 생산에서부터 돈육제품으로의 소비까지 이력추적이 가능한 도구로 제공 될 것이다.

참전복 치패의 최적사육 기술현황 (State of Optimal Rearing Technique on the Abalone (Haliotis discus hannai) Juvenile)

  • In order to standardize the juvenile abalone rearing technique, we selected sample farms by region in East, West and South coasts of Korea and Jeju island. We also have reviewed previous literature and visited farms to survey on the management of abalone juvenile production, spawning, hatching and so forth. Results of investigation are as follows: The light colors of tanks for larvae breeding are good for a frequent examination of larvae behaviour changes during the breeding period. The tank for the abalone juvenile production is a rectangular form in general and its size should amount to 3.5 m in length and 1.2 m width. It also should be built with proper drainage. The best age and size of adult for juvenile production are 3-6 years old individuals, with 9-12 cm separate burial and 125-150 g average weight. To induce spawning, the use of the exposure on air and ultraviolet ray together was the most effective. The density of larvae by plate should be 150-300 individuals and the proper stocking density was est imated and amount to 10-30 individuals. It has been shown that a correlation between water surface size($X_1$) and number of plates ($Y_1$), when producing abalone juveniles, is quite high and it is described by equation $Y_1=138.88X_1-5,736.8\;(R^2=0.9028)$. In addition, it has also been shown that a correlation between production of abalone juveniles ($Y_2$) and number of plates ($X_2$) is high and it is described by equation $Y_2=4.554X_2+12,493\;(R^2=0.8818)$. In Jindo region where a mass production of juveniles abalone has been done, it was shown, that a correlation between rearing water surface size ($X_3$) and production of juveniles abalone ($Y_3$) is very high and this relationship was described by the equation $Y_3=747.03X_2+94,359(R^2=0.9809)$. It has also been shown that a correlation between water surface size ($X_4$) and production of abalone juveniles ($Y_4$) in nationwide is high and the relationship between this variables was described by equation $Y_4=635.85X_4+99,923\;(R^2=0.9020)$.

부채마 (Dioscorea nipponica Makino)의 부정근 배양조건과 Steroidal Saponin의 기내생산 (In vitro Culture of Adventitious Roots from Dioscorea nipponica Makino for the Production of Steroidal Saponins)

  • 액체배양을 통한 부채마의 부정근 배양시 첨가되는 생장 조절제의 종류와 농도에 따른 부정근의 생장과 steroidal saponin 생산과의 관계를 조사하였다. NAA와 BA 농도별 부정근 생장은 NAA를 단독으로 1.0 mg/L를 처리한 곳에서 가장 효과적이었으나 NAA에 BA를 첨가하면 부정근의 생장에 오히려 억제효과를 보였다. 20종의 각각 다른 배양주로부터 dioscin, prosapogenin A 및 prosapogenin C의 함량을 조사한 결과 배양주에 따라 큰 차이를 보였는데, 그 중 dioscin 함량이 건물당 2.5%로 가장 높았던 10번 배양주를 선발하였다. 부정근에 함유된 세 종류의 steroidal saponin 함량은 부정근의 생장이 왕성한 4주까지는 낮으나 부정근의 생장이 거의 정지되는 5주째에 급격히 증가하였다. Dioscin과 prosapogenin C의 함량은 NAA보다 IBA를 처리한곳에서 현저히 높으나 prosapogenin A의 함량은 이들 두성분의 종류와 농도에 영향을 받지 않았다.

극조숙, 다수성 조사료 및 녹비용 호밀 신품종 "이그린" (A New Early-Heading and High Forage Yielding Rye Variety "Egreen")

  • "이그린"은 1995년 조춘 등 10계통을 방임수분시켜 작물과학원 포장에서 타식성 작물의 집단육종법에 의해 선발 육성된 청예용 호밀 품종으로서 2005년~2007년 3개년간 계통명 호밀34호로 지역적응시험을 실시한 결과 그 우수성이 인정되어 2007년 농촌진흥청 농작물 직무육성 신품종 선정위원회에서 새로운 호밀품종으로 결정되었는데 그 주요특성과 수량성을 소개하면 다음과 같다. 1. "이그린"의 출수기는 수원 등 4지역 평균이 4월 22일로서 대비품종 "올호밀" 에 비하여 3일 빠른 극조생 품종이다. 2. "이그린"의 초장은 수확시에 118 cm로서 "올호밀" 보다 6 cm 크며 $m^2$당 수수는 968개로서 "올호밀" 과 비슷하다. 3. "이그린"은 내병성 및 내습성이 "올호밀" 정도로 강한 내재해성 품종으로서 제주도를 포함한 전국의 답리작 및 전작지대에서 추파재배가 가능하다. 4. "이그린"의 청예수량성은 지역적응시험결과 건물수량이 평균 ha당 8.34 MT로서 대비품종 "올호밀" 보다 8%, 주요 도입품종인 "Koolgrazer"에 비하여 10% 증수되는 청예다수성 품종이다.

절화특성이 우수한 백색 심비디움 '웨딩데이' 육성 (Breeding of White Colored Cymbidium spp.'Wedding Day' for Cut Flower)

  • 심비디움 '웨딩데이'는 국립원예특작과학원에서 2021에 육성한 신품종으로 2008년에 C. spp. 'Boksam Holiday'와 C. spp. 'Persicolor'를 인공교배를 통해 개발하였다. 교배 후 기내 파종을 통해 112개의 실생 개체를 배양하여 온실로 순화하였다. 이후 생육 및 개화 특성 평가를 통해 2 계통을 1차 선발하였다. 2선발 계통의 안정성과 균일성을 확인하기 위하여 2017년부터 2021년까지 1, 2차 특성검정을 진행하였다. 그 결과 최종적으로 'C0844-29' 1 계통을 선발하였고, '원교 F1-78'로 계통명을 부여하였다. 이후 육성계통 평가회를 통해 기호성을 인정받았고 '웨딩데이'로 명명하였다. '웨딩데이'(품종보호 등록번호: 제9523호)는 절화수명이 길고 백색의 소화가 11개 이상 달리는 다화성 중형종이다. 꽃대가 직립하고 63.2 cm로 길어 절화로 이용하기에 적합하다. 잎 길이는 65.8 cm로 꽃 높이보다 낮아 꽃을 가리지 않고 감상할 수 있다. 1월 말부터 개화하는 중만생종이며, 기내에서 배양 시 증식함에 있어 문제가 없어 상업적 활용이 가능할 것으로 보인다.

1990년대 이후 패션에 나타난 엘레강스 스타일의 미적 특성 (Aesthetic Characteristics of Elegance Expressed on Fashion Since 1990s)

  • The purpose of this study is to interpret the elegant dressing visualized in modem fashion on the basis of the concept of elegance in dress and its aesthetic characteristics from the holistic viewpoint. Elegance in dress is based upon the idea of aristocratic taste cultivated by good breeding, considering from the documentary study. It is expressed visually through not only the carefully contrived dress but also a sort of aura of dressed body with skillful ease. The aesthetic values of elegance consist of luxury, nobility, refinement, femininity, harmony. To grasp the trends of elegant styles since 1990s, contents analysis of the articles related to elegance in Vogue has been done. As a result, they have been classified into four groups of elegant styles which are Classic, Elaborate Couture, Soft Minimalism, Kitsch Elegance. Classic Elegance and Elaborate Couture Elegance represent traditional ones with conservative viewpoint. Soft Minimalism Elegance is a modem version of elegance. According to postmodernism as a open system, even kitsch has been refined to keep accompany with elegance fur aristocratic taste of high class. As a result of this study, in fashion from the 16th century to the first half of 16th century, elegance has been one of the significant aesthetic categories, resulting from the absolute domination of taste of high society. However in the end of 20th century it seems to start to fade in fashion trends such as mass fashion, youth culture, casual fashion etc. Rather, it can be thought that elegance has been expressed as one of the aesthetic values in dress, by virtue of its value of high class as well as its conservative stability.

A synonymous mutation of uncoupling protein 2 (UCP2) gene is associated with growth performance, carcass characteristics and meat quality in rabbits

  • Background: Uncoupling proteins 2 (UCP2) plays an important role in energy regulation, previous studies suggested that UCP2 is an excellent candidate gene for human obesity and growth-related traits in cattle and chicks. The current study was designed to detect the genetic variation of UCP2 gene, and to explore the association between polymorphism of UCP2 gene and growth, carcass and meat quality traits in rabbits. Results: A synonymous mutation in exon 1 and four variants in the first intron of the UCP2 gene were identified by using PCR-sequencing. The synonymous mutation c.72G>A was subsequently genotyped by MassArray system (Sequenom iPLEXassay) in 248 samples from three meat rabbit breeds (94 Ira rabbits, 83 Champagne rabbits, and 71 Tianfu black rabbits). Association analysis suggested that the individuals with AA and AG genotypes showed greater 70 d body weight (P < 0.05), 84 d body weight (P < 0.01), ADG from 28 to 84 days of age (P < 0.05), eviscerated weight (P < 0.01), semi-eviscerated weight (P < 0.01) and semi-eviscerated slaughter percentage (P < 0.05), respectively. Additionally, the individuals with AA and AG genotype had a lower pH value of longissimus muscle (P < 0.01) and hind leg muscle (P < 0.05) after slaughter 24 h. Conclusions: These findings indicated that UCP2 could be a candidate gene that associated with growth performance, body composition and meat quality in rabbits, and this would contribute to advancements in meat rabbit breeding practice.

Artificial Diet for Mass Rearing the Emma Field Cricket, Teleogryllus emma (Orthoptera: Gryllidae)

  • Nymph of the emma field cricket, Teleogryllus emma, were reared on several types of artificial diets. The development period of nymphs were 55.4 days when only a single food, wheat bran, was provided, and it did not show a significant difference compared to the rearing results of the Danong diet and mixed diet. The supplying period of fish meal as the animal feed, the high emergence rates were obtained at 3rd instar with 90% and 4th instar with 100%. For the added amount test, when more than 40% of the diet was added, it confirmed that the insect weight increased. The characteristics of development according to each added amount of the vegetable food (dry bean-curd residue and corn powder) were investigated to minimize the dangers of the degeneration of diet when rearing with a single feed during the $1st{\sim}3rd$ instar period. First, as the added amount of bean-curd residue increased, nymphal development period became longer and the emergence rate became low. With corn powder as the single diet, all died before becoming adult. However, when corn powder was added up to 30%, no difference existed in the breeding results.

돼지사료 중의 다이옥신 잔류 평가 (Evaluation for Concentration of Residual PCDD/Fs in Pig Feed)

  • Dioxins are chemical substances that are not properly decomposed degradation under natural photochemical, biological and chemical conditions in the environment, and bio-accumulate through the food chain, so pose a risk of causing adverse effects to human health and the environment. Ninety seven percent of human exposure to dioxins comes through meat, fish, dairy consumption and so on. Pork is the largest proportion meat aspect of daily intake in the Korean food. Pigs are three times more than Cattle in terms of breeding population and production amount in the world, and the amount is increasing every year. Therefore, it is necessary to monitor the exposure level of dioxins in pig's body. Also, it is required to evaluate the presence of these chemical substances in the pig's feed grain such as corn, soybeans, and tallow. The purpose of this study was to evaluate PCDD/Fs in pig's feed to search the origin of PCDD/Fs. Feed samples obtained were wheat from East Europe, corn from South America and America, soybean meal from Korea, America, South America and India and tallow from Korea. The preparation of samples was based on the EPA method 1613. Instrumental analysis was based on the use of high resolution gas chromatography coupled to high resolution mass spectrometry (HRGC/HRMS). The concentrations of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and 1,2,3,7,8-pentachlorodibenzo-p-dioxin (PCDD), which are the most toxic potent dioxins, were not observed in any of the samples. However OCDD was detected in most of the samples. The TEQs in the feed samples were ranged from 0 pg TEQ/g to 0.09841 pg TEQ/g. Especially, the high level of TEQs was observed in the samples of wheat from East Europe and soybean meal from Korea.

Genome-wide association study of rice core set related selenium content

  • The purpose of this study was to identify the candidate genes involved in selenium content in brown rice. Rice (Oryza sativa L.) was important crop including diverse functional substance such as carbohydrate, protein, lysine and tocopherol, mineral. Especially, selenium as nutritionally important minerals, it was known to activate the immune system, antioxidant effect and inhibition of carcinogenesis. Also recommended daily requirements of the United States and the United Kingdom were 55 to 90 ug for selenium. Therefore, selenium content in brown rice of core-set were analyzed by using ICP-MS (Inductively Coupled Plasma Mass Spectrometer) and GWAS (Genome Wide Association Study) was conducted to search for candidate genes in this study. The new natural variants identified through haplotyping analysis would be useful to develop new rice varieties with improved storage ability of the valuable mineral through the future molecular breeding.
